Adzuki Bean(팥)의 Callus형성과 분화에 관한 연구

This experiment was carried out for the analysis of the ability of callus formation and differentiation in the eight Korean local varieties of adzuki beans (Phaseolus angularis). KLA 102 has the highest ability on the formation of callus (all tissues have 90 % over). Morphological heterogeneity (color and shape) of callus was well reflected to the cell constitution and cell meristematic activities. Meristemoid tissues and rooting were derived from only creamish-white color and soft region of callus.
單色光의 前處理 照射가 양상치 種子의 發芽特性 誘導에 미치는 影響

This experiments were intended to obtain a detailed information on the induction of different germination behavior of two cultivars(MSU 15, MSU 16)under pretreatments of monochromatic light irradiation during seed formation period. Pretreatment of red light irradiation induced high level of seed phytochrome(Pfr) which considered responsible for dark germination in both cultivars. However, pretreatment of blue light or far- red light irradiation modified germination behaviors of dark germinable seed to light requiring seed in both cultivars. Above results suggested that the pretreatment of monochromatic light irradiation given to the immature seed adhered or detached to a mother plant brought about some modification of its native seed phytochrome systems of both cultivars through the photoequilibrium of phytochrome ( Pr ⇔ ????? Pfr).
포도와 미국자리공 세포현탁배양계 안토시아닌과 베타시아닌에 미치는 광의 영향

본 실험은 포도와 미국자리공의 세포현탁배양계에서 세포생장과 athocyaninn 및 betacyanin 색소생성에 미치는 광의 영향을 조사하였다. 그 결과, 포도의 세포현탁배양계에서의 세포증식 패턴은 광의 효과가 미약하였으나, 미국자리공의 경우는 암배양에 비하여 광조사구에서 뚜렷한 세포증식 패턴을 나타내었다. 포도세포는 12일간의 배양기간중 한번의 생장 피크를 보여주는데 비하여 미국자리공의 배양세포는 두 번의 증식 피크를 보여 광이 세포의 생장 및 분열 주기를 촉진시킬 뿐만 아니라 색소의 축적도 촉진시키는 것으로 나타났으며, 미국자리공 배양세포는 배양 후 4일째에 액포화에 따른 색소의 축적이 확인되었다. 한편, 색소생성에 미치는 광의 영향은 포도 현탁배양계에서는 배양후 6일째부터 생성되기 시작하여 배양 후 10일 전후에 최고치를 보이는데 비하여, 미국자리공의 현탁배양계에서는 세포증식 패턴에 따라 배양 후 4일째와 8일째에 두 번의 피크를 보였다. 그러나 이들 두 가지 모두 암배양하에서는 거의 색소 생합성이 이루어지지 않고 광상태하에서만 색소의 축적을 볼 수 있었다. 한편, 포도 세포배양계에서는 배양 후 4일째에 처리한 광은 약간의 효과가 있었으나, 배양 후 7일째 12시간 이상의 광에 의하여 색소생성이 크게 촉진되는 것을 알 수 있었다. 미국자리공의 경우에는 배양 후 7일 째보다는 오히려 배양 후 4일째에 광이 더욱 효과적이었다. 이상의 사실은 이들 두 가지 색소의 생합성과정에는 광이 절대적으로 필수적이지만 광을 필요로 하는 시기 및 요구하는 광량 등이 서로 다르다는 것을 시사해 준다. The effects of light on the production of anthocyanin and betacyanin in cell suspension cultures of Vitis vinifera and Phytolacca americana were investigated. The cell growth of V.vinifera was little affected by exposure to light, but that of P.americana was markedly increased by light than in the dark In suspension cultures of V vinifera maximum accumulation of anthocyanin was observed during the stationary phase in continuous light By contrast, in suspension cultures of R americana, accumulation of betacyanin occured in parallel with cell division which showed two peaks after 4 days and 8 days of culture in continuous light whereas in continuous dark accumulation of anthocyanin and betacyanin did not occured However treatment of light interrupting for l, 12, and 24 h after 4 days in cell suspension. cultures of remarkably showed a slight anthocyanin accumulation, but after 8 days of culture remarkably accumulated by light interrupting for more than 12 h. In cultures of P. americana, the light treatment was more effective at 4th day than at 7th day after culture, but betacyanin accumulation was decreased again in the dark after light treatment These result indicate that the difference of light responses exist between the V.vinifera and the betacyanin of P. americana though cell suspension culture systems.

In the present paper, we intented to analyze and to compare the different seed dormancy of two legume plants, Albizzia and Cercis. We found two different types of seed dormancy: one type of dormancy in Albizzia seed was caused by the water impermeability, and other type of seed dormancy was found in Cercis and it was induced by inhibitors. We confirmed that the extract substances from Cercis seed seemed to have strong regulational functions on the inhibition of seed germination and primary root growth of lettuce seed.

결구상치의 종자 형성기에 있어서의 일장(日長)과 온도조건(溫度條件)이 종자(種子)의 형태형성(形態形成)에 미치는 영향에 관하여 조사하였다. 종자(種子)의 크기, 무게 및 종피의 두께는 종자형성기의 온도조건에 의하여 크게 영향을 받았으나, 친식물(親植物)의 생체중(生體重) 및 건물량(乾物量)등은 일장(日長)의 영향을 크게 받았다. 또한 본실험(本實驗)을 통하여 결구상치종자의 형태형성(形態形成)은 종자형성기(種子形成期)의 기상조건과 밀접한 관계가 있는 것으로 생각되었다. 환경조건(環境條件)에 대응(對應)하여 얼어나는 종자형태상(種子形態上)의 다양성은 종(種) 또는 품종(品種)의 유전적 요인에 의하여 어느정도 한계(限界)가 정(定)해져 있는것으로 생각 되었으나 이에 관한 자세한 정보(情報)는 추후(追後)의 연구(硏究)에 기대하는 바가 크다. 또한 본보(本報)에 있어서는 종자(種子)의 형태형성(形態形成)에 있어서 일어나는 다형현상(多形現象)을 육안(肉眼) 및 현미경을 통하여 관찰하였으며 여기에 관한 직접적인 몇가지 정보(情報)를 제공(提供)하는 바이다. Interactions between the adaptive polymorphism and climatic conditions were also discussed. Environmental conditions affected the morphology of lettuce seed and this variation of seed morphology resulted from interaction between genetic potentialities and environmental conditions.
상치 種子의 胚乳 細胞間 原形質 連絡絲 및 外皮角質層왁스에 對한 電子顯微鏡的 觀察

This electron microscopical study was carried out to observe the plasmodesmata of endosperm cells and epicuticular waxes of pericarp ribs on lettuce seed. The morphological modification of pericarp and pericarp ribs was detected by SEM observation. The seed which formed under low temperature, has well developed, suberized cell walls and epicuticular waxes while the seed fromed under high temperature condition has very poorly developed epicuticular waxes. Epicuticular waxes was considered to take over the function of seed protection and keeping the good quality during storage period. The plasmodesmata of endosperm cells in lettuce seed was observed at 4hrs after imbibition for the first time. This phenomenon indicates that the lettuce seed starts its physiological activities in the very early imbibition stages.
