TORC2 Regulates Hepatic Insulin Signaling via a Mammalian Phosphatidic Acid Phosphatase, LIPIN1

<P><B>Summary</B></P><P>TORC2 is a major transcriptional coactivator for hepatic glucose production. Insulin impedes gluconeogenesis by inhibiting TORC2 via SIK2-dependent phosphorylation at Ser171. Interruption of this process greatly perturbs hepatic glucose metabolism, thus promoting hyperglycemia in rodents. Here, we show that hyperactivation of TORC2 would exacerbate insulin resistance by enhancing expression of LIPIN1, a mammalian phosphatidic acid phosphatase for diacylglycerol (DAG) synthesis. Diet-induced or genetic obesity increases <I>LIPIN1</I> expression in mouse liver, and TORC2 is responsible for its transcriptional activation. While overexpression of LIPIN1 disturbs hepatic insulin signaling, knockdown of LIPIN1 ameliorates hyperglycemia and insulin resistance by reducing DAG and PKCϵ activity in <I>db/db</I> mice. Finally, TORC2-mediated insulin resistance is partially rescued by concomitant knockdown of LIPIN1, confirming the critical role of LIPIN1 in the perturbation of hepatic insulin signaling. These data propose that dysregulation of TORC2 would further exaggerate insulin resistance and promote type 2 diabetes in a LIPIN1-dependent manner.</P>
Identification of differentially expressed genes by DNA methylation in cervical cancer

The objective of this study is to find novel cervical cancer-related genes which are regulated by DNA methylation. We performed the integrated analysis of genome-wide DNA methylation and RNA expression profiles using normal and tumor regions of cervical cancer patient`s tissues. For the validation of epigenetic regulation of selected genes, we treated the cervical cancer cell line, HeLa with demethylating reagent (decitabine: 5-Aza-2`-deoxycytidine) and measured the methylation and expression status by methylation-specific PCR (MS-PCR) and realtime PCR. We identified 19 novel cervical cancer related genes showing differential RNA expression by DNA methylation. Many of selected genes have not been reported in the cervical cancer gene database (CCDB), but showed differential expression in cancer versus normal in the microarray database (GENT). Among candidate genes, the epigenetic regulation of three genes, CAMK2N1, ALDH1A3 and PPP1R3C, was validated in cervical cancer cell line The results indicate that expression of CAMK2N1, ALDH1A3 and PPP1R3C genes are suppressed in cervical cancers by DNA methylation.
Clinical characteristics of brain metastasis form ovarian cancer: A study of 51 cases

The aim of this report is to describe the characteristics, pathologic features, treatments and survival of patients with brain metastasis from ovarian cancer at our single institution. Between January, 1989 and April, 2011, a total of 4625 women with ovarian cancer were evaluated at Asan Medical Center. A total of 71 patients were identified using the ICD-10 code for malignant neoplasm of ovary, cross-matched with the medical records for brain metastasis. Patients with other synchronous primary tumor were excluded. The clinical and pathologic characteristics, treatment and outcome of patients with brain metastases from epithelial ovarian carcinoma were analyzed using SPSS 12.0.using SPSS 12.0. The incidence of brain metastasis from ovarian cancer in our population was 1.1%(51/4625). The median survival from the diagnosis of brain metastases was 7 months (range: 0-72). Prior cancer relapse before the diagnosis of brain metastases, and multimodal treatment were related to the duration of survival. Although the prognosis for patients with brain metastases from epithelial ovarian carcinoma is generally poor, palliative therapy may offer symptomatic relief and improvement in the quality of life on ovarian cancer patients with brain metstasis.

Endocervicosis refers to the presence of benign mucinous glands of endocervical type in ectopic site. It is rare and usually detected incidentally by histologic examination. The urinary bladder is the most common site, but peritoneum, small bowel, abdominal scar, outer wall of the cervix, vagina, and lymph nodes have been documented. Because this is the first documented report of endocervicosis arising in the rectus muscle in korea, we report this with a brief review of the concerned literatures.

Aujeszky’s disease (AD), also called pseudorabies, is an infectious viral disease, caused by an alpha herpes virus and has domestic and wild pigs, as well as a wide range of domestic and wild animals, as the natural host. AD affects many countries and regions in the world, causing important economic losses, mainly due to international trade restrictions. In this study, to determine the characteristics of the Aujeszky’s disease virus (ADV), NYJ strain, which was isolated from the serum of an infected pig in 1987, we investigated the nucleotide sequence and expression of the glycoproteins gB, gC, and gD using the bBpGOZA system. We found that the glycoproteins gB, gC, and gD of NYJ consisted of 2751 bp, 1443 bp, and 1203 bp, respectively. Comparison of the NYJ with the other strains revealed nucleotide sequence identity ranging from 91.tito 99.0%. To better understand the genetic relationships between other strains, phylogenetic analyses were performed. The NYJ strain was formed a distinct branch with high bootstrap support. The expression of glycoprotein gD in insect cells was characterized by SDS-PAGE and Western blotting with an anti-ADV polyclonal antibody. Glycoprotein gD of approximately 45 kDa was detected. The results of this study have implications for both the taxonomy of ADV and vaccine development.

Classical swine fever virus (CSFV) envelope glycoprotein E2 is the main target for inducing neutralizing antibodies and protective immunity in swine. Here, we report a novel strategy forthe large-scale production of a CSFV E2 subunit vaccine that demonstrates a high immunogenic capability in the larvae of a baculovirus-infected silkworm, Bombyx mori. We constructed a recombinant B. mori nucleopolyhedrovirus (BmNPV) that expressed recombinant polyhedra together with the N-terminal 179 amino acids of CSFV E2 (CSFV E2ΔC). BmNPV-E2ΔC-infected silkworm larvae expressed an approximately 44-kDa fusion protein that was detected using both anti-polyhedrin and anti-CSFV E2 antibodies. Electron and confocal microscopy both demonstrated that the recombinant polyhedra were morphologically normal and contained CSFV E2ΔC. The CSFV E2ΔC antigen produced in BmNPV-E2ΔC-infected silkworm larvae reached 0.68 mg per ml of hemolymph and 0.53 mg per larva at 6 days post-infection. Mice that were immunized with the granule form of recombinant polyhedra or the soluble form of the fusion protein elicited CSFV E2 antibodies, which indicated that the recombinant polyhedra carrying CSFV E2ΔC were immunogenic. The virus neutralization test showed that the serum from mice that were treated with recombinant polyhedra or the soluble form of the fusion protein contained significant levels of virus neutralization activity. These results demonstrate that the present strategy can be used for the large-scale production of CSFV E2 antigen and that the recombinant polyhedra containing CSFV E2ΔC as a granule antigen can be used as a potential subunit vaccine against CSFV.

The performance tests for an 87 PS 4-cylinder 4-cycle gasoline engine were carried out in order to measure the heat balances in various operating conditions. The experimental apparatus is equiped with 130kW eddy current dynamometer. In the heat balance diagram of the engine, mechanical losses could be quite accurately estimated by substracting shaft power from indicated power of the engine since the indicated power was obtained from indicator diagram by a piezo-electric pressure sensor located at the cylinder head and the shaft power was measured by the eddy current dynamometer. The typical results obtained in this study can be summarized as follows. First, The best engine performance shows around 2000 rpm of the engine speed in most running conditions. Second, The heat transfer rate to cooling water and to exhaust gas as well as output and frictional works change considerably with the engine speed and boost pressure. However, the extra losses(those are, heat transfers to oil, air, ete.) do not change much with respect to them. Third, The rate of mechanical loss increases as engine speed increases, but, at the same engine speed, the rate of mechanical loss does not change much as boost pressure varies.
