In this research, experimental and numerical simulations were adopted to investigate the effects of ligament angle on compressive strength and failure mode of rock-like material specimens containing two non-coplanar filled fissures under uniaxial compression. The experimental results show that with the increase of ligament angle, the compressive strength decreases to a nadir at the ligament angle of $60^{\circ}$, before increasing to the maximum at the ligament angle of $120^{\circ}$, while the elastic modulus is not obviously related to the ligament angle. The shear coalescence type easily occurred when ${\alpha}$ < ${\beta}$, although having the same degree difference between the angle of ligament and fissure. Numerical simulations using $PFC^{2D}$ were performed for flawed specimens under uniaxial compression, and the results are in good consistency with the experimental results. By analyzing the crack evolution process and parallel bond force field of rock-like material specimen containing two non-coplanar filled fissures, we can conclude that the coalescence and propagation of crack are mainly derived from parallel bond force, and the crack initiation and propagation also affect the distribution of parallel bond force. Finally, the displacement vectors in ligament region were used to identify the type of coalescence, and the results coincided with that obtained by analyzing parallel bond force field. These experimental and numerical results are expected to improve the understanding of the mechanism of flawed rock engineering structures.
Factors that Influence the Presciption of Antipsychotics for Patients with Schizophrenia in China

Objective: To investigate the patterns of antipsychotic use in China and to analyze the factors that influence antipsychotic prescriptions. Methods: A standardized survey was conducted from May 20 to 24 2002 in five different regions of China with varying economic levels. The patterns of antipsychotic medication use were analyzed in a sample of 4,779 patients with schizophrenia. The survey gathered information on demographic characteristics, clinical profiles, and antipsychotic medications prescribed. Multiple logistic regression was used to analyze factors related to patterns of antipsychotic medication use. Results: A plurality of patients with schizophrenia was treated with clozapine (39%); this was followed by risperidone, sulpride,chlorpromazine, perphenazine, and haloperidol. More than 56.3% of patients were treated with only one atypical antipsychotic. The mean daily dose of chlorpromazine was 365±253 mg (mean±standard deviation), and 6.5% of patients were treated with depot injections of typical antipsychotic medications. A total of 73.7% (n=3,523) of patients with schizophrenia received monotherapy,24.8% (n=1,183) received two antipsychotics, 1.1% (n=52) received three antipsychotics, and one received four different antipsychotics. Patients often simultaneously received other classes of medications including anticholinergic agents, benzodiazepines,β-blockers, antidepressants, and mood stabilizers. Economic status and clinical symptoms were the main factors that contributed to the patterns of antipsychotic prescription. Conclusion: The present study suggests that atypical antipsychotic medications, especially clozapine, are the primary psychiatric treatments of choice in the management of schizophrenia in China. Moreover, the economic status and clinical profile of the patient are the major factors affecting the prescription of antipsychotic medication.

Objective: Clozapine is one of the most commonly used antipsychotic drugs in China. To date, few studies have investigated the patterns the prescription of clozapine nationwide. The present study examined these patterns in China in 2006 and identified the demographic and clinical characteristics associated with the use of clozapine. Methods: Using a standardized protocol and data collection procedure, we surveyed 5,898 patients with schizophrenia in 10provinces with differing levels of economic development. Results: Overall, clozapine had been prescribed for 31.9% (n=1,883) of the patients; however we found considerable variation among the 10 provinces. The frequency of clozapine use was highest in Sichuan (39.3%) and lowest in Beijing (17.3%). The mean daily dose of clozapine was 210.36±128.72 mg/day, and 25.1% of the patients were treated with clozapine in combination with other antipsychotics. Compared with the group not receiving clozapine, clozapine-user had been treated for longer durations and had experienced a greater number of relapses and hospitalizations. Furthermore, those in the clozapine-user had lower family incomes, were less able to seek psychiatric services, and more likely to be male and have a positive family history of schizophrenia. A multiple logistic regression analysis revealed that age, sex, professional help-seeking behaviors, duration of illness, economic status, educational level, and clinical manifestations were associated with the use of clozapine. Conclusion: Clozapine use is common in China. However, use of the antipsychotic varies among provinces, and demographic and clinical factors play important roles in the prescription of clozapine.

Purpose: To investigate the effect of ultra low molecular weight heparin (ULMWH) on glutamate induced apoptosis in rat cortical cells and to explore the possible mechanisms. Materials and Methods: Cell viability was measured using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Apoptosis was first analyzed with Hoechst 33258 and then confirmed by DNA fragmentation. The concentration of free intracellular calcium ([Ca²+]i) was determined with fura-2/AM fluorometry. The expression of Bcl-2 family protein and caspase-3 were evaluated with Western blot. Results: Typical apoptotic morphological change in rat cortical cells treated with 100μmol/L glutamate for 24h was detected by Hoechst 33258 staining, which was then confirmed by the DNA ladder of agarose gel electrophoresis. The apoptotic rate of the glutamate treated cells was up to 33.21%, and 24 h of treatment with glutamate increased [Ca²+]i, down-regulated Bcl-2 expression, up-regulated Bax expression, and increased caspase-3 activation in rat cortical cells. Our research demonstrated that ULMWH pretreatment can prevent the glutamate- induced apoptosis, attenuate the increase of [Ca²+]i not only in medium containing Ca²+ but also in Ca²+-free medium, up-regulate the expression of Bcl-2, down-regulate the expression of Bax, and decrease caspase-3 activation. Conclusion: ULMWH has neuroprotective capacity to antagonize glutamate-induced apoptosis in cortical cells, through decrease of Ca²+ release and modulation of apoptotic processes.

A cDNA encoding a cysteine protease inhibitor (CPI) was isolated from the cDNA library of clamworm Perinereis aibuhitensis Grube. The deduced amino acid sequence analysis showed that the protein had 51%, 48%, and 48% identity with Zgc: 153129 from Danio rerio, cystatin B from Theromyzon tessulatum, and the ChainA, stefin B tetramer from Homo sapiens, respectively. The gene was cloned into the intracellular expression vector pET-15b and expressed in Escherichia coli. The recombinant CPI (PA-CPI) was purified by affinity chromatography on Nicharged resin and ion-exchange chromatography on DEAE-Sepharose FF. The relative molecular mass of PACPI was 16 kDa as deduced by SDS-PAGE. Activity analysis showed that the recombinant protein could inhibit the proteolytic activity of papain. A constitutive and secretive expression vector was also constructed, and the cDNA encoding CPI was subcloned into the vector for extracellular expression. Western blotting analysis results showed that the PA-CPI was secreted into the medium. Bioassay demonstrated that E. coli DH5α harboring pUC18ompAcat-CPI showed a significant difference in mortality to the Asian longhorned beetle Anoplophora glabripennis compared with untransformed E. coli DH5α and control.
Analysis of Pressure Distribution of the Bolted Joints Based on the Finite Element Method

Bolted joints widely exist in the field of the national defense industry. However, its nonlinear stiffness degradation will occur under tangential load which may lead to the reliability reduction. According to Coulomb friction theory, the pressure distribution is the key in the tangential degradation of bolted joints. Therefore, this paper aims to analyze the pressure distribution of bolted joints. Firstly, we established a threaded bolt model and two simplified models. Secondly, the accuracy of the three models describing the contact area is verified, and the influence of preload and material properties on the contact radius is analyzed. Thirdly, we compared the pressure distribution of the three models, and results show that the smooth bolt model is more suitable for pressure analysis. Finally, the accuracy of several different functions to characterize the pressure distribution of bolted joints is analyzed, and results indicated that the Fernlund function is optimal. This paper provides a feasible simplified bolt model for finite element analysis, and the optimal pressure distribution function can be applied in the tangential stiffness degradation modeling.

Under the influence of seasonal freeze-thaw cycles, the concrete lining of channels situated on saline soil foundations is highly vulnerable to an array of deleterious phenomena, such as frost heave, salt expansion, and corrosion. To investigate the synergistic interplay among saline soil substrates, U-shaped channel lining structures, and ambient temperatures during freeze-thaw cycles, a novel Hydraulic-Thermal-Salt-Mechanical (HTSM) model is established in this study. The HTSM model elucidates the migration of moisture and salt solute, ice-water phase transitions, and sodium sulfate crystallization within unsaturated saline soils throughout the freeze-thaw cycles. It considers the variations in volume and pore solution volume before and after the phase transition of sodium sulfate. Temperature and salt frost heaving force measurements obtained from outdoor freeze-thaw cycles are compared with numerical simulation results to validate the efficacy of the HTSM model and investigate the distribution patterns of salt frost heaving force (frost swelling force) along the channel. The results showed that the frost swelling force and depth exhibit a great linear relationship and the maximum stress of the U-shaped channel appears at the bottom of the channel, highlighting the good arching force characteristics of the U-shaped structure which prove that bottom of the channel is also the most prone to fracture. Significantly, the initial water content significantly impacts the frost swelling force—it increases by 4% while the frost swelling force increases by 76%. Comparing the numerical simulation results of temperature variations, moisture changes, and salt-frost heaving forces with the outdoor experimental data, it is evident that the proposed HTSM model effectively simulates the salt-frost heaving patterns observed in U-shaped channel linings.
