Calcium intake has been insufficient in all age groups in previous Korean national surveys. This study was conducted to investigate calcium intake in Korean and American populations at different ages. We analyzed two national survey data: the 2007∼2009 Korean National Health and Nutrition Examination Survey (KNHANES) and the 2007∼2008 National Health and Nutrition Examination Survey (NHANES). Statistically, survey procedures were applied using the weight, cluster, and strata variables. The mean calcium intake of Koreans at ages of 1∼2 y, 3∼5 y, 6∼11 y, 12∼18 y, 19∼64 y, and 65+y was lower than American populations at those ages. The sufficient proportions based on Korean Estimated Average Requirement (EAR) of 1∼2 y, 3∼5 y, 6∼11 y, 12∼18 y, 19∼64 y, and 65+y in Koreans were 45.5%, 33.1%, 24.3%, 16.6%, 32.7%, and 19.0%. While the sufficient rates based on American EAR of 1∼3 y, 4∼8 y, 9∼13 y, 14∼18 y, 19∼30 y, 31∼50 y, 51∼70 y, and 71+y in US population were 88.5%, 54.4%, 35.2%, 35.8%, 55.3%, 55.2%, 40.6%, and 24.5%. Overall, the major foods contributing to calcium intakes in Koreans were milk, baechukimchi, and anchovies, whereas, milk products, pasta, or bread were major contributions to calcium in American populations. The calcium supplement intakes in the American population were 5.5 mg (1∼2 y), 15.5 mg (3∼5 y), 13.9 mg (6∼11 y), 35.7 mg (12∼18 y), 150.3 mg (19∼64 y) and 334.4 mg (≥65 y). These results suggest that Korean adolescents and older adults are the most insufficient in dietary calcium intakes among Koreans. In order to accurately estimate calcium intakes in Korean populations, calcium supplements and calcium-fortified foods should be considered.
Comparative proteomics: assessment of biological variability and dataset comparability

<P><B>Background</B></P><P>Comparative proteomics in bacteria are often hampered by the differential nature of dataset quality and/or inherent biological deviations. Although common practice compensates by reproducing and normalizing datasets from a single sample, the degree of certainty is limited in comparison of multiple dataset. To surmount these limitations, we introduce a two-step assessment criterion using: (1) the relative number of total spectra (<I>R</I><SUB><I>TS</I></SUB>) to determine if two LC-MS/MS datasets are comparable and (2) nine glycolytic enzymes as internal standards for a more accurate calculation of relative amount of proteins. <I>Lactococcus lactis</I> HR279 and JHK24 strains expressing high or low levels (respectively) of green fluorescent protein (GFP) were used for the model system. GFP abundance was determined by spectral counting and direct fluorescence measurements. Statistical analysis determined relative GFP quantity obtained from our approach matched values obtained from fluorescence measurements.</P><P><B>Results</B></P><P><I>L. lactis</I> HR279 and JHK24 demonstrates two datasets with an <I>R</I><SUB><I>TS</I></SUB> value less than 1.4 accurately reflects relative differences in GFP levels between high and low expression strains. Without prior consideration of <I>R</I><SUB><I>TS</I></SUB> and the use of internal standards, the relative increase in GFP calculated by spectral counting method was 3.92 ± 1.14 fold, which is not correlated with the value determined by the direct fluorescence measurement (2.86 ± 0.42 fold) with the <I>p</I> = 0.024. In contrast, 2.88 ± 0.92 fold was obtained by our approach showing a statistically insignificant difference (<I>p</I> = 0.95).</P><P><B>Conclusions</B></P><P>Our two-step assessment demonstrates a useful approach to: (1) validate the comparability of two mass spectrometric datasets and (2) accurately calculate the relative amount of proteins between proteomic datasets.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1186/s12859-015-0561-9) contains supplementary material, which is available to authorized users.</P>
Mass Spectrometric Comparison of Cycloamylose Production by 4-Alpha-glucanotransferases

A 4-α-glucanotransferase (DgαGT EC 2.4.1.25, 4αGTase) gene was cloned from Deinococcus geothermalis and expressed in Escherichia coli, then recombinant protein was purified using nickel affinity chromatography. 4αGTase possesses intermolecular and intramolecular glucan transfer activity. 4αGTase have been used to produce cycloamylose(CA) from amylose by intramolecular glucan transfer activity. CA production by DgαGT was compared with those by Thermus scotoductus 4αGTase (TSαGT) and Escherichia coli 4αGTase (MalQ). Optimal CA production conditions were 50℃ and pH 6.0. After the 4αGTase treatment on amylose, residual linear maltooligosaccharides were removed by β-amylase. CA was analyzed using matrix-assisted laser desorption/ionization tandem time-of-flight mass spectroscopy. DgαGT produced CA with similar yield 53% to the other enzymes but needed less time to start CA production. Degree of polymerization (DP) of CA produced by DgαGT ranged broadly from DP 5 to DP 43. Further analyses need to be conducted for concluding prominent characteristics of DgαGT among bacterial αGTs.

<P>The metabolic changes that occur in <I>Dunaliella tertiolecta</I> upon exposure to low temperatures and nitrate deficiency were analyzed by exploring the fatty acid composition and lipid profile of two strains that were acclimated to different temperatures. The results indicate that the levels of linolenic acid (C18:3) and diacylglyceryl-<I>N</I>,<I>N</I>,<I>N</I>-trimethylhomoserine (DGTS) were significantly higher in the low-temperature (15 °C) strain (SCCAP K-0591) than in a strain grown at 21 °C (UTEX LB999). In addition, DGTS accumulated in LB999 under nitrate-deficient conditions, while the levels of most lipids, including DGTS, remained almost consistent in K-0591. The higher levels of DGTS in K-0591 suggest that DGTS could play a role in adaptation to low temperatures and nitrate deficiency in this organism. Highbush blueberries (Vaccinium corymbosum L.) are cultivated worldwide for their fruit with unique taste and potential health benefits. Blueray, Bluecrop, and Spartan are prominent among the various blueberry cultivars. We performed gas chromatography-mass spectrometry (GC-MS)-based metabolic profiling to differentiate the fruits of these three cultivars, and built an optimal partial least squares-discriminant analysis (PLS-DA) model to separate them. Amino acids, fatty acids, organic acids, phenolic compounds, and sugars were identified in the fruits. The optimized PLS-DA model for different cultivars of the fruits was obtained by selecting variables based on a variable importance in the projection (VIP) cut-off value of 1.0. Caffeic acid, aspartic acid, acetic acid, threonolactone, inositol, xylose, glucoside, linolenic acid, mannose, altrose, glycine alanine, and valine were found to be relevant and contributing compounds for differentiating cultivars. In addition, a hierarchical cluster analyses dendrogram pattern was correlated with the PLS-DA. This study suggested that GC-MS-based metabolic profiling coupled with multivariate statistical analysis could be used to differentiate the fruits of three major highbush blueberry cultivars.
Inflexible eye fixation pattern in schizophrenia affecting decision-making on daily life

<P><B>Abstract</B></P> <P>Patients with schizophrenia have difficulties in real life due to impairment in ability to make decisions. The purpose of this study was to elucidate the relationship between impaired decision-making processes with real life stimuli and abnormal eye gaze patterns in patients with schizophrenia. Each of 23 patients with schizophrenia and 23 healthy controls performed an apparel purchase decision task including the influencing factors such as preference, fit, and price, during which the eye gaze was traced. Fixation time and fixation time ratio on areas of interest, which were set for participant faces and clothing, were compared between the two groups. Compared with controls, patients made purchase decisions at a higher rate and showed significantly shorter fixation time on clothing in the preference, fit, and price phases and on faces in the purchase phase. Fixation time ratio of face over clothing did not change over purchase decisions in patients, whereas controls showed significantly higher fixation time ratio in not-to-buy decisions than in to-buy decisions. These results suggest that aberrant decision-making behaviors in patients with schizophrenia are closely related to inflexible visual information gathering patterns because they apportion the same amount of attention to objects regardless of purchase intention.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Patients with schizophrenia made purchase decisions at a higher rate. </LI> <LI> Patients showed shorter fixation on clothing or faces during purchase decisions. </LI> <LI> In patients, fixation time ratio did not change over purchase decisions. </LI> <LI> In controls, fixation time ratio was higher in not-to-buy than to-buy decisions. </LI> <LI> Aberrant decision-making behaviors are closely related to inflexible gaze patterns. </LI> </UL> </P>

<P>In this study, Chlorella vulgaris (C. vulgaris) was treated with ethephon at low (50 mu M) and high (200 mu M) concentrations in medium and harvested at 0, 7, and 14 days, respectively. The presence of ethephon led to significant metabolic changes in C. vulgaris, with significantly higher levels of alpha-tocopherol, gamma-aminobutyric acid (GABA), asparagine, and proline, but lower levels of glycine, citrate, and galactose relative to control. Ethephon induced increases in saturated fatty acids but decreases in unsaturated fatty acids. The levels of highly saturated sulfoquinovosyldiacylglycerol species and palmitic acid bound phospholipids were increased on day 7 of ethephon treatment. Among the metabolites, the productivities of alpha-tocopherol (0.70 mu g/L/day) and GABA (1.90 mu g/L/day) were highest for 50 and 200 mu M ethephon on day 7, respectively. We propose that ethephon treatment involves various metabolic processes in C. vulgaris and can be an efficient way to enrich the contents of alpha-tocopherol and GABA.</P>
