      • Effect of ER signal peptides (KDEL) on expression and function of monoclonal antibody in plant

        Plant genetic engineering led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current animal expression systems. In this study, the expression level of mAbP SO57 between with and without Lys-Asp-Glu-Leu (KDEL) was compared in transgenic plant. PCR and PT-PCR analyse showed stable gene insertion transcription of heavy and light chain genes of mAbP SO57 with or without KDEL in plant, respectively. We validated expression of mAbP SO57 by western blot. Western blot showed the significantly higher expression level of mAbP SO57 with KDEL compared to without KDEL. Flow cytometry (FACS) analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb (mAbM) evidenced similar binding activity to the FcγRI receptor (CD64). High performance liquid chromatography (HPLC) analysed, glycosylation patterns of mAbP SO57 with or without KDEL. The mAbP SO57 with KDEL had glycan profile with both oligomannose type (47.6%) and golgi type (52.4%), while the mAbP SO57 without KDEL had only golgi type (100%) glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest the potential of mAbP SO57 for rabies immunotherapy, regardless of plant specific glycan structures.

        Plant genetic engineering has led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current antibody expression systems. In this study, the expression levels of mAbP SO57 with or without ER-signal peptides (Lys-Asp-Gly-Leu;KDEL) in transgenic tobacco plants were analysed in transgenic plant. PCR and Reverse Transcription-PCR analyses showed existence of heavy and light chain genes of mAb with or without KEDL and their transcription in plant, respectively. Western blot showed that the expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL. Flow cytometry analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb have similar binding activity to the FcγRI receptor. High-performance liquid chromatography analysis showed that the mAbP SO57 with KDEL had glycan profile with both oligomannose and golgi type, whereas the mAbP SO57 without KDEL had only golgi type glycans.         e-Learning can be an extremely flexible delivery system. It is able to present both linear programs and branching programs, but it may be most effective employing other individualized approaches to music instruction. I believe that music enriches our own ways of thinking habits of mind as rich and different from each other botany is different from philosophy, and music is embedded in our daily life often so deeply or subtly that it is part of our subconsciousness. School classes are changing everyday along with developing technology. The transforming power of technology is a force not only in the economy but in the music field as well. This study aims to examine the effects of the e-Learning on music classses. Appreciating is not only a simple listening activity, but also a field of learning. This means appreciation is so important that it should be a theoretical background for music teaching and learning through recognizing and grasping the essence of appreciation teaching Composing is so easy for students to make music using software. It is so enjoying to the students and helps them approach music. As I develop some skills of music appreciation and composing by using lots of web and software(e-Learning), I discovered several benefits, First, students are interested in music through the teaching than without e-Learning. Second, students can learn more easier by himself/herself more information and knowledge through e-Learning. Third, students can concentrate, feel enthusiastic about their learning, and access music information much more quickly. Forth, music and computers are central to the growth of cultural diversity. Finally, intermediate e-Learning - use teachers approached e-Learning. as an integrated system to present course materials effectively. Where as, expert e-Learning - use teachers to seem to value how to search, organize, and synthesize information, and use of technology of communicate each others. Above all, for subject teachers, they need practical training relating to use of e-Learning in their subject areas.

      • 靈樞·寒熱病에 對한 硏究

        The contents of Han Yul Byung(寒熱病) are explained on the basis of the so-called theory "Yin and Yang, the Five Evolutive Phases(음양오행)". The text of the Han Yul Byung(寒熱病) of the Young Chu(靈樞) was written in the Kap Ul Kyung(甲乙經) chapter 5, 7, 8, 10, 11 and 12. Also it was written in the Tae So(太素) volume 10 and 26. Also it was introduced in the Ryu Kyung(類經) volume 20, 21 and 22 by Jang Kae-Pin(張介賓) in the Myung(明) dynasty. Summarizing the contents of the Han Yul Byung in reference to the contents of successive generation note, the contents run as follows: Chapter 1 deals with the symptoms and treatment of diseases which accompany chills and fever(寒熱病). Chapter 2 deals with arthralgia with other joint systems(骨痺) and flaccid paralysis of Iimbs(體惰病). Chapter 3 deals with indications of five points around the Chen You Points(天牖穴). Chapter 4 deals with the treatment of toothache. Chapter 5 deals with the treatment of headache and eye pain. Chapter 6 deals with the acupuncture manipulation according to changes of season. Chapter 7 deals with the serious influence of carbuncle according to the region of body. Chapter 8 deals with the treatment of heat-syndrome. Chapter 9 deals with the side effect of acupuncture. It is too difficult to understand the Han Yul Byung(寒熱病), because it is written in old chinese. Consequence of above reason, the writer of this article has researched to find out the exact meaning of that.

        Fabrication of carbon nanofiber electrodes using poly(acrylonitrile‑co‑vinylimidazole) and their energy storage performance

        For electrodes in electrochemical double-layer capacitors, carbon nanofibers (CNFs) were prepared by thermal treatment of precursor polymer nanofibers, fabricated by electrospinning. Poly(acrylonitrile-co-vinylimidazole) (PAV) was employed as a precursor polymer of carbon nanofibers due to the effective cyclization of PAV polymer chains during thermal treatment compared to a typical precursor, polyacrylonitrile (PAN). PAV solutions with different comonomer compositions were prepared and electrospun to produce precursor nanofibers. Surface images obtained from scanning electron microscopy showed that their nanofibrous structure was well preserved after carbonization. It was also confirmed that electrospun PAV nanofibers were successfully converted to carbon nanofibers after the carbonization step by Raman spectroscopy. Carbon nanofiber electrodes derived from PAV showed higher specific capacitances and energy/power densities than those from PAN, which was tested by coin-type cells. It was also shown that PAV with an acrylonitrile/vinylimidazole composition of 83:17 is most promising for the carbon nanofiber precursor exhibiting a specific capacitance of 114 F/g. Their energy and power density are 70.1 Wh/kg at 1 A/g and 9.5 W/kg at 6 A/g, respectively. In addition, pouch cells were assembled to load the higher amount of electrode materials in the cells, and a box-like cyclic voltammetry was obtained with high capacitances.

        기단 역궤적분석에 의한 경주시 대기오염물질의 농도 변화

        Gyeongju, which was the central city of the ancient civilization at Silla Kingdom, has various kinds of stone cultural properties. It is significantly important to preserve historical sources of Korea. However, recent air quality data measured in Gyeongju did not show good air quality level. In order to investigate variation of the concentration of the air pollutants with meteorological condition, an air quality monitoring and an aerosol sampling were conducted during the intensive monitoring period in Gyeongju. Impacts of the meteorological factors on the air pollutants were also analyzed based on the air mass pathway categories using HYSPLIT model and the local wind patterns using MM5 model. The prevailing air mass pathways were classified into four categories as following; category Ⅰ affected by easterly marine aerosols, category Ⅱ affected by northwesterly continental aerosols, category Ⅲ affected by southwesterly continental aerosols, and category Ⅳ affected by northerly continental aerosols. The concentrations of the air quality standards were relatively lower during the fall intensive monitoring period. At that time, the easterly marine air mass pattern was dominated. The seasonal average mass concentration of PM<SUB>10,Opt</SUB>, which optically measured at the monitoring site, was the highest value of 77.6±28.3 ㎍ m?³ during the spring intensive monitoring period but the lowest value of 20.1±5.3 ㎍ m?³ during the fall intensive monitoring period. The concentrations of SO₂ and CO were relatively higher when the air mass came from the northwestern continent or the northern continent. The concentrations of SO₄²? and NO₃? - increased under the northwesterly continental condition. It was estimated that the acidic aerosols were dominated in the atmosphere of Gyeongju when the air mass came from the continental regions.

      • 가족생활교육 프로그램의 개발에 관한 연구 동향 분석

        Korean society with low fertility rate and high divorce rate crisis has been tried very hard to find out alternatives to solve these social problems. These efforts resulted in increasing the development of family life education programs. One of the characteristics for family life education is multidisciplinary so that family life education programs have been development in various different academic fields. Therefor, this is the time to establish database for family life education programs which were developed so far. The purpose of this study is to examine research trend in the field of family life education programs in Korea. For this purpose, 228 family life education programs developed during the last 10 years were analyzed. Frequencies, percentages were used for this analysis. The results of this study were as followed: First, family life education programs were very actively developed in Home Management field compared to other academic fields such as Social Work, Education, Psychology, and so on. Second, in terms of family life education program subjects, a family dynamics subject was mainly used. Third, with program participative, most family life education programs were focused on participants in early adulthood and families with childbearing. Finally, with respect to the fact that whether or not effective study was published, 71.5% of family life education programs had effective study results.

        갈색세포종의 임상 표현을 보인 부신 피질 선종 1예

        하나의 부신 세포에서 피질과 수질의 특징이 함께 공존하는 피질-수질세포에 관한 예들이 1960년대 이후로 보고되어 왔다. 저자들은 갈색세포종의 임상적 특징을 보이지만, 수술 후 갈색세포종의 임상증상과 생화학적 특징이 정상으로 회복되었으며 광학현미경상 부신 피질 선종의 특징을 보이지만, 면역조직화학 검사와 전자현미경 검사에서 피질-수질세포를 보인 1예를 경험하였기에 문헌고찰과 함께 국내 처음으로 보고하는 바이다. The coexpression of cortical and medullary features in a single adrenal cortical cell has been recognized, leading to terms such as cortico-medullary cells. Here, we reported a case of adrenocortical adenoma consisting of cortico-medullary cells that clinically mimicked pheochromocytoma. A 52-year-old woman was admitted to our hospital complaining of an 8-month history of paroxysmal palpitation with refractory hypertension. A 24-hour urine study revealed increased norepinephrine and metanephrine levels. Computed tomography of the abdomen revealed a 1.0×0.9-cm mass in the left adrenal gland. The patient subsequently underwent unilateral laparoscopic adrenalectomy for a presumptive pheochromocytoma. Light microscopic findings of the left adrenal mass indicated an adrenocortical adenoma, but electron microscopy identified lipid vacuoles and smooth endoplasmic reticulum, along with dense core neurosecretory granules, so-called cortico-medullary cells. This is the first report of the detection of cortico-medullary cells in adrenocortical adenoma presenting as pheochromocytoma in Korea. (Korean J Med 75:479-483, 2008)

      • 남자대학생의 흡연태도와 행동에 관한 연구

        There have been so many researches about problems of cigarettes and relation of smoking and heath since 1950's and generally known that smoking is one of the worst factor of increasing disease and mortality. No matter what such many proofs about various problems of smoking have been researched, smoking rate of current male collage students has not been decreased. Besides, most of the researches about cigarette use and its addiction have been focused on the adolescent or adult women smokers or adult male, not male collage student. Therefore, the subject of this study is about smoking attitude and smoking behavior of male collage students by general features. Smoking attitude was analyzed by modifying 'Guidelines for the content of tobacco smoking surveys of the general population'(WHO, 1982). Two hundred of male collage students participated to the distributed questionnaire for one week in May. The smoking attitude was categorized into duration of smoking and number of cigarette per day(CPD), and nicotine dependency, investigated by Fagerstrom Tolerance Questionnaire(FTQ) (Fagerstorm & Schneider, 1989). All statistics were performed by procedure of SAS according to number, percentage, mean, standard deviation, and ANOVA. The results are as follows: 1. Smoking attitude level was mean 25.73±5.15 marks, ranging from 13 to 39 marks of the total 40 marks. 2. Duration of cigarettes, number of cigarette per day, and nicotine dependency were mean 5.14±2.84 years, 12.88±6.85 pieces, and 4.78±1.69, respectively. 3. Smoking attitude showed significance according to general features such as marital state and allowance, but the significance according to marital state is not valuable because the numerical difference is so big between married one and single that they cannot be compared with each other. 4. Duration of smoking was significant according to general features such as age and allowance. 5. Smoking amount showed significance the factors of age only. 6. ll1ere was no significance in nicotine dependency according to general features. As a result, smoking attitude of male collage student in this study shows moderate level. Smoking attitude and duration is significant according to allowance, and smoking amount is significant according to age. Nicotine dependency according to general features is not significant but comparing with the preceding study about adult male, male collage students in this study are respectively more nicotine-dependent than adult male smokers. In conclusion, the right allowance usage strategies and effective heath education focused on the young are needed. In addition, helpful programs and information spreading strategies are necessary for establishing higher level of smoking attitude. At last, systemic regulations in daily living for ""anti-nico-dependent"" and perspective reforming of male university student are needed for reducing nicotine dependency.

      • Interacelluar reprogramming of expression, glycosylation, and function of plant-derived anti-virus therapeutic monoclonal antibody

        Plant genetic engineering has led to production of plant-derived mAb (mAbP), which can provides a safe and economically effective alternative to the conventional antibody expression methods. In this study, the expression levels of mAbP SO57 with or without ER-retention peptide extensions signal (Lys-Asp-Glu-Leu; KDEL) in transgenic tobacco plants were analysed in transgenic plant. The expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL regardless of transcription level. mAbP SO57 with KDEL localized surround to the nucleus suggesting that the mAbP with KDEL is localized in ER. The mAbP without KDEL and mAbH had mainly Golgi type glycans, whereas the ER-localized mAbP with KDEL had glycan profile with both oligomannose type (47.6%) and Golgi type (52.4%). The Fc domains of both purified mAbP (with and without KDEL) and human-derived mAb (mAbH) had similar bingding activity to the Fcγ RI receptor (CD64). Both mAbP (with and without KDEL) had a shorter half-life than mAbH. However, both mAbP with and without KDEL was as effective at neutralizing activity of the rabies virus CVS-11 as the mAbH. These results suggest that ER localization of recombinant mAbP by KDEL reprograms glycosylation and enhances production of the functional antivirus therapeutic antibody in the plant.
