        방사선치료를 시행한 전이성 안구암 1예

        안구의 전이암은 안구의 악성 종양 중에서 가장 흔한 암이다. 많게는 암으로 인해 사망한 환자의 약 12%에서 안구의 전이가 보고되며, 암 환자의 생존 기간이 늘어남에 따라 안구로의 전이 가능성 또한 증가할 것으로 추정된다. 안구 전이는 실명과 같은 심각한 임상적 증상을 야기할 수 있으며, 조기 발견과 치료를 통해 시력을 보존하고 삶의질을 최대화 할 수 있으나, 암 환자에 있어 안구 전이의 중요성은 비교적 낮게 평가되고 있다. 안구 전이에 대한 방사선치료는 국소치료이므로 전신적인 부작용이 없으며, 치료에 대한 반응이 조기에 나타나므로 실명 위기에 있는 안구 전이 환자에게 효과적인 치료법이 될 수 있다. Intraocular metastasis is the most common malignancy of the eye. The frequency of intraocular metastasis in all the patients dying of cancer is about 12% and ocular metastases will be detected more frequently in the future because the patients with malignant tumor are now living longer. Intraocular metastasis can cause a serious clinical problem such as blindness. The early recognition and treatment of intraocular metastasis are very important clinical oncologic issues to maintain vision and to maximize the quality of life. However, significance of intraocular metastasis is still under-evaluated for practicing oncologists. External beam radiotherapy is a safe, effective palliative treatment in terms of preserving both the vision and the eye globe.

        전방 십자 인대 손상 환자에서 인대 재건술 후 정상군과의 보행 양상 비교

        Patients with anterior cruciate ligament deficiency typically do not have quadriceps muscle activity during stance and this gait pattern has been termed $quot;quadriceps avoidance$quot; gait. We performed physical examination and computerized 3-dimensional motion analysis on 7 normal controls and 7 patients who had anterior cruciate ligament reconstruction by having the patients walking in 3 different walking velocity, i.e. a self- selected walking velocity which the patients felts comfortable, a velocity set a metronome with a frequency of 108 per minute, and a fast velocity (the normal controls walk with a frequency of 126 per minute, the patients walk as fast as possible.) All the 7 patients were male and the average age was 32.1 years. The average follow up duration after operation was 21.0 months. The measured average points by the modified Larson scoring scale was 83.14 and the results were 1 excellent, 5 good and 1 fair. In all patients, limping was rarely observed and little limitation was observed in stepping, squatting and running. All the patients complained mild discomfort on the knee joint during exercise. There were also no limitation of range of motion on the knee joint in all patients. On gait analysis using 3-dimensional motion analysis system, external flexion moment of the injured knee joint was decreased compare to normal controls and was more decreased when the patients walked in a faster walking velocity during the midstance and the loading response phases. However net external flexion moment of the knee joints implying the activity of quadriceps muscle was observed. In summary, gait analysis using 3-dimensional motion analysis system may help us to evaluate the present state, objectively and to anticipate the prognosis of the patients who had anterior cruciate ligament reconstruction.

      • 靑年期 適應狀態와 健康問題 呼訴에 關한 硏究 : 一部 女大生을 中心으로

        Adolescence has been identified as a very stressful stage of development due particularly to the "identity crisis" and difficulty dealing with change that seems to be more constant than stability. To assist adolescents deal with this period, one of the nurses' roles is the identification of life events that contribute to stress. The purposes of this study were: 1. The identification of life events that were the problems of the youth adaptation. 2. The association between the status of youth adaptation and the health problem complaints. 3. The analysis of the characteristics that are affected by youth adaptation. The study sample consisted of 700 Ewha Womans University students who were attended a lecture of the family nursing subject during the first semester, 1985. The tools used for the collection of data were "a youth adaptation rating scale" by Sue Beell, Gayle Schmidt and the Modified Cornell Medical Index. The data was treated by a computer(S.A.S.) used the ratio value(degree of severity), percentage, mean score, analysis of varience The results were as follows: 1. The items of the life events that are the problems of the youth adaptation were "Death of a parent" (4.68±0.80), "parents getting a divorce(Separation)" (4.49±0.84), "getting expelled(suspended)"' (4.34±0.86), "making career decisions(college, majors)" (4.11±0.91). These items were emotional stresses caused by self anxiety and conflict and lead to difficulties of the freedom of selection and realities of life as a social factors. 2. There was a significant association between the physiological & serial basic needs and physical & mental health complaints, that is, the number of the physical & mental health complaints were higher in the students who did not fulfill the physiological & social basic needs. 3. The affected characteristics of the youth adaptation were age, religion." economic status, birth place, father's occupation, family system, relation with parents during the growth. 4. A youth adaptation rating scale used in this study as a tool was cronbach's α=0.91.

      • 형질전환동물세포주에서 벤조피렌의 변이유발성에 대한 천연물질의 항 돌연변이 효과

        인간에게 고퉁을 주고 있는 질병중 많은 부분이 돌연한이로 인한 특정 유전자의 손상과 손상된 윤전자의 발현과 관련되어 있음이 밝혀져 왔다. 특히 암은 대표적인 유전자 돌연변이의 결과로 생기는 빌병으로서, 많은 암 유발 용힌중 화학물질에 의한 돌연변이 유발작용이 발암성과 관련하여 많은 관심의 대상이 되고 잇다. 따라서 본 연구에서는 외부DNA를 자신의 genome내게 삽입시킨 형질전환동물로부터 제조된 세포주를 이용하여 유전자 돌연변이를 검출할 수 있는 최신 유전독성 연구기댄을 확립하고자 하였다. 시험법을 확립하기 위하여 f3c f gene을 target으로 하는 Big Blue· Transgenic Mouse Embryonic Cell을 사용하였으며,시험법의 타당성 및 재현성여분를 확인하기 위하여 음성대조물질로는 1% DMSO, 양성대조물질로는 100rg/mf MNU(rnethrlnitrosourea)를 이웅하여 시험한 결과 mutant frequency(MF)는 각각 0.55±0.39Nle-', 4.26±1.51×10-'으로 나타나 재현성을 확인하였다. 또한 이 시험법의 유용성을 높이기 위하여 B[alP (benzo(alpyrene)를 양성대조물질로 in oifro대사환성화계에서 시험한 결과, 처리농도 20, 40, 60rgjm1에 대하여 Mf값이 9.1츠겪.14×10-", 14.3츠깆.86×10-', 20.St3.78×10-"로서 농도의존성을 보였다. 또한 식품을 비롯한 각종 천연물에는 돌연변이원 물질의 활성을 억제 또는 경감시키는 작용이 많이 보고되고 있어 갈조류의 일종으로서 강력한 항산화효과를 나타내는 것으로 알려진 료」ooi3 rfofonif☞ra(속칭 :곰피)를 사옹하여 꽉립된 시험법으로 항돌연변이 효과를 점색하고자 하였다. 양성대조물질로 사용한 MNU에 대하여 곰릭의 메탄을 추출물을 병용투여하여 항돌연변이 효자를 시험하였다. 100rg/mf MNU와 곰피 25.6, 51.2, 100.3#9/mf을 병용처리한 결과 MF가 각각 67, 79, 74% 감소하여, 형질전환동물세포주께서 공피 추출물치 돌연변이 억제 효과를 관찰하였 다. 그러나 암 예방제 개발의 기초자료로 활용하기 위해서는 돌연변이된 염기 서열을 분석하여 변이의 종류와 특성을 확인하여 그 작용기전을 밝히는 추가연구가 수챙되어야 할 것으로 사료된다. Transgenic animal cell with a A shuttle vector containing a fac ftarget gene is useful as a short-term in oifro mutagenesis assay. The gene is recovered from the cells treated by exposiog mousegenomic DNA to in oifro packaging extracts and plating the rescued phage on agar plates containing S-brorno-4-chloro-3-indolyl fD-galactopyranoside (X-gal). Phage with mutations in the f3c f gene formblue plaques, whereas phage with a nonrnutated fac J form colorless plaques, so mutant is defected easily.In order to estabilish this new transgenic technology, we studied mutagenicity of the direct-ectingalkylating agent N-methyl-N-nitrosourea (MNU ) and indirect mutagen benzora]pyrene in the BigBlue" transgenic mouse emf)ryonic cell line. Treatment of the cells with MNY or benzo[alpyrene causedinduction of mutations over 1% DMSO negative control and we conformed the reproducibility of thetransgenic mutagenicity assay. Also, dose-dependent response has been ohserved in benzo[a]pfrene-treatment cells. Sdrfonia afofonif☞ra, an edible brown algae, haying a strong antioBidizing activity, wasevaluated its antimutagenic effect by performing this assay, the methanol extract of Sffonia sfofonifrrashowed rfuction of mutation frequencies in MNU-treated transgenic mouse cells.

        Synthesis and BiologicalEvaluation of Pyrimidine-Based Dual Inhibitors of Human EpidermalGrowth Factor Receptor 1 (HER-1) and HER-2 Tyrosine Kinases

        <P>A novel series of N-4-(3-chlorophenyl)-5-(oxazol-2-yl)pyrimidine-4,6-diamines were synthesized and evaluated as dual inhibitors of HER-1/HER-2 tyrosine kinases. In contrast to the currently approved HER-2-targeted agent (lapatinib, 1), our irreversible HER-1/HER-2 inhibitors have the potential to overcome the clinically relevant and mutation-induced drug resistance. The selected compound (19a) showed excellent inhibitory activity toward HER-1/HER-2 tyrosine lcinases with selectivity over 20 other kinases and inhibited the proliferation of both cancer cell types: lapatinib-sensitive cell lines (SK-Br3, MDA-MB-175, and N87) and lapatinib-resistant cell lines (MDA-MB-453, H1781, and H1975). The excellent pharmacokinetic profiles of 19a in mice and rats led us to further investigation of a novel therapeutic agent for HER-2-targeting treatment of solid tumors, especially HER-2-positive breast/gastric cancer and HER-2-mutated lung cancer.</P>

        Growth, quality, and yield characteristics of transgenic potato (Solanum tuberosum L.) overexpressing StMyb1R-1 under water deficit

        This study was conducted to evaluate agronomic characteristics such as growth, quality, and yields of StMyb1R-1 transgenic potato and also to obtain the basic data for establishing assessment guidelines of transgenic potato. Three transgenic lines (Myb 1, Myb 2, and Myb 8) were cultivated under conventional irrigation, drought condition, and severe drought condition and were analyzed by comparing with wild type, non-transgenic cv. Superior. Myb 2 showed a different flower color from wild type and Myb 1 had much bigger secondary leaflets than wild type. Myb 1 and Myb 2 showed higher $P_2O_5$ content in both top and root zone and longer shaped tubers than wild type. In yield factors, transgenic lines had more tubers than wild type, however their yield decreases were severe because of the poor enlargement of tuber under water deficit condition. This tendency was noticeable in Myb 1 and Myb 2. In TR ratio, chlorophyll content, dry matter rate, and relative water content, there were no big differences between transgenic lines and wild type. Meanwhile, in phenotype, growth, quality, and yield factors, substantial equivalent was confirmed between Myb 8 and wild type. Then, Myb 8 showed the highest marketable tuber yield under conventional irrigation, while showed lower level than wild type under water deficit. Judged by this result, the enhancing droughttolerance by StMyb1R-1 gene might actually not mean the enhancement of photosynthesis or starch accumulation in tuber and, furthermore, not the yield improvement. More detailed research will be required to accurately understand the relationship between StMyb1R-1 and yield factors.
