      • Mathematical Simulation of Spray Forming Process

        In this paper, an integrated approach for modeling the entire spray forming process is presented and the program for the simulation is written by hand. The whole spray forming process can be divided into four calculate process and the basis for the analysis is classical k-ε turbulence model which used to simulate the flow field of gas formed in the chamber. The other three mathematical models are atomization model,deposition model and solidification model. In the atomization model coupled the flow field of gas and formation、velocity and location of droplet. By means of integrate above mathematical model the process of spraying forming was simulated in detail.

        Identification of novel rheumatoid arthritis-associated MiRNA-204-5p from plasma exosomes

        Rheumatoid arthritis (RA) is an autoimmune disease characterized by infiltration of immune cells in the synovium. However, the crosstalk of immune cells and synovial fibroblasts is still largely unknown. Here, global miRNA screening in plasma exosomes was carried out with a custom microarray (RA patients vs. healthy controls = 9:9). A total of 14 exosomal miRNAs were abnormally expressed in the RA patients. Then, downregulated expression of exosomal miR-204-5p was confirmed in both the replication (RA patients vs. healthy controls = 30:30) and validation groups (RA patients vs. healthy controls = 56:60). Similar to the findings obtained in humans, a decreased abundance of exosomal miR-204-5p was observed in mice with collagen-induced arthritis (CIA). Furthermore, Spearman correlation analysis indicated that plasma exosomal miR-204-5p expression was inversely correlated with disease parameters of RA patients, such as rheumatoid factor, erythrocyte sedimentation rate, and C-reactive protein. In vitro, our data showed that human T lymphocytes released exosomes containing large amounts of miR-204-5p, which can be transferred into synovial fibroblasts, inhibiting cell proliferation. Overexpression of miR-204-5p in synovial fibroblasts suppressed synovial fibroblast activation by targeting genes related to cell proliferation and invasion. In vivo assays found that administration of lentiviruses expressing miR-204-5p markedly alleviated the disease progression of the mice with CIA. Collectively, this study identified a novel RA-associated plasma exosomal miRNA-204-5p that mediates the communication between immune cells and synovial fibroblasts and can be used as a potential biomarker for RA diagnosis and treatment.

        Effects of Phosphate Dispersants on the Liquid Limit, Sediment Volume and Apparent Viscosity of Clayey Soil/Calcium-Bentonite Slurry Wall Backfills

        Soil-bentonite slurry-trench wall, usually consisting of sandy soil and Na-bentonite, is used extensively as in-situ engineered barriers for contaminant containment. Clayey soil/Ca-bentonite may be considered as an alternative backfill when Na-bentonite is scarce at some sites. Adding trace amount of phosphate dispersant to clayey soil/Ca-bentonite backfills may be advantageous to maintain the deflocculated structure of bentonite, which is benefit to minimizing increases in hydraulic conductivity of the backfills when attacked by contaminants. However, studies on the application of phosphate dispersants to the clayey soil/Ca-bentonite backfills are very limited. A series of laboratory tests are conducted for the measurements of liquid limit, sediment volume and apparent viscosity of the phosphate dispersant-amended backfills. The phosphate dispersants tested are sodium hexametaphosphate, sodium tripolyphosphate and sodium pyrophosphate with contents ranging from 0 to 2%. The results demonstrate that the values of liquid limit, sediment volume and apparent viscosity of the backfills decrease sharply when the dispersant content is relatively low (≤ 0.1 to 0.5%), while they change slightly at relatively high dispersant content (> 0.1 to 0.5%). The type and content of the dispersant as well as Ca-bentonite content have significant effects on the liquid limit, sediment volume and apparent viscosity of the backfill. The sodium hexametaphosphate is shown to have higher dispersibility compared with the others in terms of greater reduction in apparent viscosity, and its optimum content is suggested in a range of 0.1% to 0.5%. A power function is proposed which well quantifies the relationship between the measured apparent viscosity and liquid limit.

        Protectin DX increases alveolar fluid clearance in rats with lipopolysaccharideinduced acute lung injury

        Acute respiratory distress syndrome is a life-threatening critical syndrome resulting largely from the accumulation of and the inability to clear pulmonary edema. Protectin DX, an endogenously produced lipid mediator, is believed to exert anti-inflammatory and pro-resolution effects. Protectin DX (5 μg/kg) was injected i.v. 8 h after LPS (14 mg/kg) administration, and alveolar fluid clearance was measured in live rats (n = 8). In primary rat ATII epithelial cells, protectin DX (3.605 × 10−3 mg/l) was added to the culture medium with LPS for 6 h. Protectin DX improved alveolar fluid clearance (9.65 ± 1.60 vs. 15.85 ± 1.49, p < 0.0001) and decreased pulmonary edema and lung injury in LPSinduced lung injury in rats. Protectin DX markedly regulated alveolar fluid clearance by upregulating sodium channel and Na, K-ATPase protein expression levels in vivo and in vitro. Protectin DX also increased the activity of Na, K-ATPase and upregulated P-Akt via inhibiting Nedd4–2 in vivo. In addition, protectin DX enhanced the subcellular distribution of sodium channels and Na, K-ATPase, which were specifically localized to the apical and basal membranes of primary rat ATII cells. Furthermore, BOC-2, Rp-cAMP, and LY294002 blocked the increased alveolar fluid clearance in response to protectin DX. Protectin DX stimulates alveolar fluid clearance through a mechanism partly dependent on alveolar epithelial sodium channel and Na, K-ATPase activation via the ALX/PI3K/Nedd4–2 signaling pathway.

        The Synthesis of Eu<sup>3+</sup> Doped with TiO<sub>2</sub> Nano-Powder and Application as a Pesticide Sensor

        Using tetrabutyl titanate as precursor, $Eu^{3+}$ doped $TiO_2$ nano-powder was prepared by sol-gel method, the nature of luminescence of nano-powder was studied. The interaction of chlorpyrifos with $Eu^{3+}$ doped $TiO_2$ was studied by absorption and fluorescence spectroscopy. The results indicated the fluorescence intensity of $Eu^{3+}$ doped $TiO_2$ was quenched by chlorpyrifos and the quenching rate constant ($k_q$) was $1.24{\times}10^{11}\;L/mol{\cdot}s$ according to the Stern-Volmer equation. The dynamics of photoinduced electron transfer from chlorpyrifos to conduction band of $TiO_2$ nanoparticle was observed and the mechanism of electron transfer had been confirmed by the calculation of free energy change (${\Delta}G_{et}$) by applying Rehm-Weller equation as well as energy level diagram. A new rapid method for detection of chlorpyrifos was established according to the fluorescence intensity of $Eu^{3+}$ doped $TiO_2$ was proportional to chlorpyrifos concentration. The range of detection was $5.0{\times}10^{-10}-2.5{\times}10^{-7}mol/L$ and the election limit ($3{\sigma}$) was $3.2{\times}10^{-11}$ mol/L.

        LSKL, a Peptide Antagonist of Thrombospondin-1, Attenuates Renal Interstitial Fibrosis in Rats with Unilateral Ureteral Obstruction

        The effects of LSKL, the peptide antagonist of thrombospondin-1 (TSP-1), on renal interstitial fibrosis in rats subjected to unilateral ureteral obstruction (UUO) were investigated. Rats were divided randomly into three groups (n = 20 each): UUO group, sham-operation group and UUO plus LSKL treatment group. Collagen deposition was studied using histopathology and reverse transcription polymerase chain reaction analysis (RT-PCR). TSP-1, transforming growth factor beta 1 (TGF-β1), phosphorylated Smad2 (pSsmad2) and α-smooth muscle actin (α-SMA) in the kidney were measured using immunocytochemistry, western blotting analysis, RT-PCR and enzyme-linked immunosorbent assay. Biochemical analyses in the serum and urine were made. Histopathology showed severe tubular dilatation and atrophy, interstitial inflammation and collagen accumulation after surgery and LSKL significantly inhibited interstitial fibrosis including tubular injury as well as collagen deposition. The protein and mRNA levels of TSP-1 increased notably at different time point and significantly decreased in the presence of LSKL. The expression of TGF-β1 and pSmad2 were upregulated in the obstructed kidney and substantially suppressed by LSKL treatment. Myofibroblast accumulation could be alleviated after administration of LSKL. Biochemical parameters did not show differences among the three groups. As TSP-1 is the major activator of TGF-β1, we demonstrate that LSKL can attenuate renal interstitial fibrosis in vivo by preventing TSP-1-mediated TGF-β1 activation.

      • Deregulated Expression of Cry1 and Cry2 in Human Gliomas

        Growing evidence shows that deregulation of the circadian clock plays an important role in the development of malignant tumors, including gliomas. However, the molecular mechanisms of gene chnages controlling circadian rhythm in glioma cells have not been explored. Using real time polymerase chain reaction and immunohistochemistry techniques, we examined the expression of two important clock genes, cry1 and cry2, in 69 gliomas. In this study, out of 69 gliomas, 38 were cry1-positive, and 51 were cry2-positive. The expression levels of cry1 and cry2 in glioma cells were significantly different from the surrounding non-glioma cells (P<0.01). The difference in the expression rate of cry1 and cry 2 in high-grade (grade III and IV) and low-grade (grade 1 and II) gliomas was non-significant (P>0.05) but there was a difference in the intensity of immunoactivity for cry 2 between high-grade gliomas and low-grade gliomas (r=-0.384, P=0.021). In this study, we found that the expression of cry1 and cry2 in glioma cells was much lower than in the surrounding non-glioma cells. Therefore, we suggest that disturbances in cry1 and cry2 expression may result in the disruption of the control of normal circadian rhythm, thus benefiting the survival of glioma cells. Differential expression of circadian clock genes in glioma and non-glioma cells may provide a molecular basis for the chemotherapy of gliomas.

      • Quantitative Presentation of the Systematic Relationships Between S. sudanense and S. bicolor

        Fifty-four biological characters were used for quantitative classification of sorghum and sudangrass and their relationship. Discriminant analysis of all 240 individuals of 48 accessions suggested that 100% of originally grouped cases were correctly classified. Analysis of variance results showed that the differences among 48 accessions in characters were significant at the 0.01 level. Cluster analysis showed that the 48 accessions were obviously divided into two clusters by a Euclidean distance of 17.46 as the classification standards, i.e. the genus maize and genus sorghum. The 48 accessions were divided into four clusters by a Euclidean distance of 9.86, i.e. Sorghum-Sudangrass, Sorghum related species (S. propinquum and S. halepense), Zea mexicana and Z. mays. All accessions were divided into five clusters by a Euclidean distance of 8.91, namely group Ⅰ (Sudangrass-Chinese sorghum), group Ⅱ (Non-Chinese sorghum), group Ⅲ (Sorghum related species), group Ⅳ (Zea mexicana), and group Ⅴ (Z mays).

        The genome of the cucumber, Cucumis sativus L.

        Cucumber is an economically important crop as well as a model system for sex determination studies and plant vascular biology. Here we report the draft genome sequence of Cucumis sativus var. sativus L., assembled using a novel combination of traditional Sanger and next-generation Illumina GA sequencing technologies to obtain 72.2-fold genome coverage. The absence of recent whole-genome duplication, along with the presence of few tandem duplications, explains the small number of genes in the cucumber. Our study establishes that five of the cucumber's seven chromosomes arose from fusions of ten ancestral chromosomes after divergence from Cucumis melo. The sequenced cucumber genome affords insight into traits such as its sex expression, disease resistance, biosynthesis of cucurbitacin and 'fresh green' odor. We also identify 686 gene clusters related to phloem function. The cucumber genome provides a valuable resource for developing elite cultivars and for studying the evolution and function of the plant vascular system.
