      • Research on the Influence of Chinese New Media on Cross-cultural Communication

        With the development of economic globalization, cross-cultural communication has become the best way to highlight the soft power of national culture. For China, under the background of the vigorous development of cultural industry and the growing support of the international community for the Belt and Road initiative, the pace of globalization in many fields is accelerating, which can be seen from the aspects of society, economy and culture. Under the current situation of spreading Chinese culture throughout the world, the new media created by “Internet plus initiative” has become an ideal carrier of cross-cultural communication and thus plays a vital role. Based on these, this paper combines communication, journalism, psychology and other related knowledge, while adopting literature reading, data analysis, case analysis and other research methods to carry out research. Starting with the general situation of the development of new media and cross-cultural communication, this paper discusses the influence of new media on cross-cultural communication and points out the challenges which are faced in the application of new media. Moreover, the paper discusses the effective ways to develop cross-cultural communication under the help of new media, for the purpose of making contributions to the cause of cross-cultural communication in China by making full use of new media.

        LncRNA PVT1 influences breast cancer cells glycolysis through sponging miR-145-5p

        Background Long-non-coding RNA PVT1 (lncRNA PVT1) can be used as an oncogenic regulatory non-coding RNA (ncRNA) for many cancers. However, its function and mechanism in breast cancer (BRCA) are still not clearly elucidated. Objective We attempt to explain the mechanism of PVT1’s role in breast cancer from different perspectives. Methods We analyzed the expression of PVT1 and its correlation with the breast cancer related clinical data in the The Cancer Genome Atlas (TCGA) database. We used PVT1 overexpression and knockdown lentivirus to infect breast cancer MDA-MB-231 cell line for cell function verification, in vitro using CCK-8 to measure proliferation, flow cytometry to measure apoptosis, transwell test to measure invasion and migration ability, detecting cell extracellular acidification rate (ECAR) to assess glycolysis metabolism and explore the biological functions of PVT1 in breast cancer cells. Transcriptome sequencing was used to analyze the changes of related genes in cells after overexpression of PVT1. In vivo we used a xenograft model to study the effect of PVT1 on breast cancer. Results PVT1 was up-regulated in breast cancer tissues and was positively correlated with the clinical stage of breast cancer patients. Overexpression of PVT1 in vitro promoted cell proliferation, migration and invasion, and promoted tumor growth in vivo. Knockdown of PVT1 led to the opposite biological consequence. Further bioinformatics analysis showed that PVT1 changes the glycolysis metabolism of tumors through regulation of glycolysis-related genes. In addition, the expression of miR-145-5p is negatively correlated with PVT1. We consider the possibility of PVT1 promoting cell proliferation and metastasis by regulating the aerobic glucose metabolism in breast cancer cells through sponging the miR-145-5p. Conclusion Our results reveal a potential pathway for competing endogenous RNA to regulate breast cancer glucose metabolism. PVT1 regulates glycolysis related genes expression by competitively binding to endogenous miR-145-5p in breast cancer cells to change the metabolic phenotype. This may Provide new ideas for precise molecular therapy targets for breast cancer.

        Enhanced photocatalytic performance of TiO2 nanowires by substituting noble metal particles with reduced graphene oxide

        Noble metal particles have been embedded in semiconductors to improve photocatalysis efficiently, but the high cost made this approach difficult to apply widely in industry. Herein titanium dioxide/reduced graphene oxide (TiO2/rGO) nanowires in a core-shell structure were prepared. The physicochemical properties and photocatalytic performance of the specimen were characterized in comparison with TiO2 and TiO2/Pt nanowires. The rGO layer and Pt nanoparticles increased chemical states of the components, reduced bandgap energy of the nanowires, enhanced visible light absorption, improved conductance and capacitance significantly. The methylene blue as catalyzed by TiO2/Pt and TiO2/rGO nanowires was degraded to 7.9% and 8.4% in an hour, but retained 25.7% by the TiO2 nanowires. The properties and function of TiO2/rGO nanowires were close to those of TiO2/Pt nanowires, while the rGO price was much lower than that of Pt, which was of great significance for the photocatalytic application of TiO2 heterojunction materials in industry.

      • Optomizing Transfection Efficiency of Cervical Cancer Cells Transfected by Cationic Liposomes Lipofectamine<sup>TM</sup>2000

        Background: Currently, cationic liposome has become the commonly used vehicles for gene transfection. Furthermore, one of the most significant steps in microRNAs expression studies is transferring microRNAs into cell cultures successfully. In this study we aim to approach the feasibility of transfection of cervical cancer cell lines mediated by liposome and to obtain the optimized transfection condition for cervical cancer cell lines. Materials and Methods: $Lipofectamine^{TM}2000$ as the carrier, miR-101 mimic was transfected into Hela cells and Siha cells. Using green fluorescent protein as reporter gene, to set different groups according to cell seeding density, the amount of miRNA, miRNA and the proportion of Liposomes, Whether to add serum into medium to study their impact on the liposomal transfection efficiency. Finally, MTT assay was used to analyze the relative minimal cell toxicity of liposome reagents. Results: The seeding density of Hela cell line and Siha are $1.5{\times}10^4$ (per well of 24 well plates), miRNA amount is 1ul of both, the ratio of miRNA and liposome is 1:0.5 of Hela cell line; 1:0.7 of Siha cell line respectively, after 24 hours we can get the highest transfection efficiency. Compared with serum medium, only Siha cells cultured with serum-free medium obtained higher transfection efficiency before transfection (P<0.01). MTT assay showed that according to the above conditions which has the lowest cytotoxicity. Conclusions: The method of Liposome to transfected is a suitable way and it can be an efficient reagent for miRNA delivery for Hela cells and Siha cells in vitro. It may serve as a reference for the further research or application.

        Development of Characteristics Measurement System for the Micro-Wall of Flexible Joints

        Characteristics of the micro-wall of flexible joints seriously impact the performance of gyro. Hence, measurements of these characteristics become very important. The present study carries out a system to measure the characteristics, including the stiffness and thickness, of the micro-wall of flexible joints. The force sensor deemed as a core component is mainly expounded in the paper. It is developed based on the principle of resistance strain gauge and good performance is shown on measuring the stiffness and the thickness. The measurement accuracy is calculated though repeated experiments and measurement theory and results are elaborated. The measurement results help to reveal the rule of structural design so as to enhance the performance of gyro. Finally, some improvements of this measurement system are proposed for further applications.

        MicroRNA-373 Inhibits Cell Proliferation and Invasion via Targeting BRF2 in Human Non-small Cell Lung Cancer A549 Cell Line

        Purpose The purpose of this study was to investigate the biological role and mechanism of miR-373 targeting of TFIIB-related factor 2 (BRF2) in the regulation of non-small cell lung cancer (NSCLC) cells. Materials and Methods miRNA microarray chip analysis of four paired NSCLC and adjacent non-tumor tissues was performed. Quantitative real-time polymerase chain reaction (qRT-PCR) and western blotting were used to detect the expression levels of miR-373 and BRF2 in NSCLC tissues and cell lines. The dual-luciferase reporter method was performed to determine if BRF2 is a target of miR-373. MTT, wound-healing, Transwell, and flow cytometric assays were conducted to examine the proliferation, migration, invasion, and cell cycle progression of NSCLC A549 cells, respectively; western blotting was used to detect the expression of epithelial-mesenchymal transition (EMT)–related proteins. Results The miRNA microarray chip analysis demonstrated that miR-373 was down-regulated in NSCLC tissues, and this result was confirmed by qRT-PCR. Additionally, miR-373 was confirmed to target BRF2. Moreover, miR-373 expression was inversely correlated with BRF2 expression in NSCLC tissues and cell lines; both miR-373 down-regulation and BRF2 up-regulation were strongly associated with the clinicopathological features and prognosis of NSCLC patients. In vitro, overexpression of miR-373 markedly inhibited cell proliferation, migration, and invasion; up-regulated the expression of E-cadherin; and down-regulated the expression of N-cadherin and Snail in A549 cell. Knockdown BRF2 by siRNA resulted in effects similar to those caused by overexpression of miR-373. Conclusion MiR-373 is decreased in NSCLC, and overexpression of miR-373 can suppress cell EMT, and inhibit the proliferation, migration, and invasion of NSCLC A549 cells by targeting BRF2.

        Continuous coating of Y2O3 buffer layers on Ni and NiW tapes by electron beam evaporation for YBCO coated conductors

        In research on YBa2Cu3O7 coated conductors, the development of buffer layers for epitaxial growth of YBa2Cu3O7 is very important. In our work, epitaxial buffer layers of Y2O3 have been deposited on biaxially textured Ni and NiW substrates using a continuous electron beam evaporation technique. The surface morphology and texture of Y2O3 buffer layers were characterized using SEM and XRD. The results show a sharp (2 0 0) orientation distribution and a smooth surface. Textured, crack-free, continuous Y2O3 buffer layers were obtained on moving tape using electron beam evaporation.         Properties of Fe-based Cr3C2 Coatings Produced with a High-Velocity Arc-Spraying Process

        Fe-based Cr3C2 coatings were sprayed with a high-velocity spraying process. The main phases of the coatings were α-Fe, (Fe, Cr)23C6, (Fe, Cr)7C3, FeCr2O4, Cr3C2 and a small amount of Fe3Si. The abrasive wear performances of the coatings were tested by using a rubber wheel abrasive tester. The wear mechanisms of the coatings were mainly plastic cutting and brittle fracture of some ceramic phases. At 650℃ the erosion rate of the coatings had a peak value at 40℃ impingement angles. The coatings exhibited excellent erosion resistance at high temperatures and were suitable for use in circulating fluid-bed boilers.
