Acidic endo-polygalacturonases are the major part of pectinase preparations and extensively applied in the clarification of fruits juice, vegetables extracts, and wines. However, most of the reported fungal endo-polygalacturonases are active and stable under narrow pH range and low temperatures. In this study, an acidic endo-polygalacturonase (EPG4) was purified and characterized from a mutant strain of Penicillium oxalicum. The N-terminal amino acid sequence of EPG4 (ATTCTFSGSNGAASASKSQT) was different from those of reported endopolygalacturonases. EPG4 displayed optimal pH and temperature at 5.0 and 60-70°C towards polygalacturonic acid (PGA), respectively, and was notably stable at pH 2.2-7.0. When tested against pectins, EPG4 showed enzyme activity over a broad acidic pH range (>15.0% activity at pH 2.2-6.0 towards citrus pectin; and >26.6% activity at pH 2.2-7.0 towards apple pectin). The Km and Vmax values were determined as 1.27 mg/ml and 5,504.6 U/mg, respectively. The enzyme hydrolyzed PGA in endo-manner, releasing oligo-galacturonates from PGA, as determined by TLC. Addition of EPG4 (3.6 U/ml) significantly reduced the viscosity (by 42.4%) and increased the light transmittance (by 29.5%) of the papaya pulp, and increased the recovery (by 24.4%) of the papaya extraction. All of these properties make the enzyme a potential application in the beverage industry.
Time-dependent Nonlinear Theory and Numerical Simulation of Folded Waveguide Traveling Wave Tubes

In this paper, a time-dependent nonlinear theory including the generalized time-dependent radiofrequency (RF) field equations is presented to simulate the beam and wave interaction (BWI) of folded waveguide (FWG) traveling wave tubes (TWTs). The analytical RF fields in FWG TWTs are replaced by digitized RF field profiles obtained from electromagnetic simulations. A W-band FWG TWT is studied by using a self-consistent one-dimensional code based on the theory. The numerical results show good predictions when compared with the experimental tests.

An endo-polygalacturonase (endo-PGase) exhibiting excellent performance during acidic fruit juice production would be highly attractive to the fruit juice industry. However, candidate endo-PGases for this purpose have rarely been reported. In this study, we expressed a gene from Penicillium oxalicum in Pichia pastoris. The recombinant enzyme PoxaEnPG28C had an optimal enzyme activity at pH 4.5 and 45℃ and was stable at pH 3.0-6.5 and < 45℃. The enzyme had a specific activity of 4,377.65 ± 55.37 U/mg towards polygalacturonic acid, and the K<sub>m</sub> and V<sub>max</sub> values of PoxaEnPG28C were calculated as 1.64 g/l and 6127.45 U/mg, respectively. PoxaEnPG28C increased the light transmittance of orange, lemon, strawberry and hawthorn juice by 13.9 ± 0.3%, 29.4 ± 3.8%, 95.7 ± 10.2% and 79.8 ± 1.7%, respectively; it reduced the viscosity of the same juices by 25.7 ± 1.6%, 52.0 ± 4.5%, 48.2 ± 0.7% and 80.5 ± 2.3%, respectively, and it increased the yield of the juices by 24.5 ± 0.7%, 12.7 ± 2.2%, 48.5 ± 4.2% and 104.5 ± 6.4%, respectively. Thus, PoxaEnPG28C could be considered an excellent candidate enzyme for acidic fruit juice production. Remarkably, fruit juice production using hawthorn as an material was reported for the first time.
Friction Characteristics of a Cylinder Based on a Bridge-Type Pneumatic Energy-saving Circuit

A bridge-type energy-saving circuit is a new type of pneumatic system that uses four on-off valves tocontrol the inlet and exhaust of two cylinder chambers. It saves energy through the open-and-close sequence of thefour control valves. Cylinder friction is the key factor in accuracy and stability of the bridge-type pneumatic energysavingcircuit. This paper focuses on research of the circuit’s friction characteristics. Based on friction theory andthe classic Stribeck model, a composite dynamic friction model of a cylinder in a circuit system is established, anda cylinder friction test platform is constructed. The Nelder-Mead algorithm is used to identify static parametersof the model through the relationship between friction and velocity while the piston is moving. Friction modelverification with error analysis is achieved by comparison with the traditional friction model. Experiments withthe energy-saving circuit under certain conditions are carried out to illustrate the effectiveness of the compositedynamic friction model. Finally, compared with existing friction model, the validity of the model is proved to beapplicable to different working conditions.

Ischemia-reperfusion injury (IRI) is a major complication in liver transplantation (LT) and it is closely related to the recovery of grafts' function. Researches has verified that both innate and adaptive immune system are involved in the development of IRI and Kupffer cell (KC), the resident macrophages in the liver, play a pivotal role both in triggering and sustaining the sterile inflammation. Damage-associated molecular patterns (DAMPs), released by the initial dead cell because of the ischemia insult, firstly activate the KC through pattern recognition receptors (PRRs) such as toll-like receptors. Activated KCs is the dominant players in the IRI as it can secret various pro-inflammatory cytokines to exacerbate the injury and recruit other types of immune cells from the circulation. On the other hand, KCs can also serve in a contrary way to ameliorate IRI by upregulating the anti-inflammatory factors. Moreover, new standpoint has been put forward that KCs and macrophages from the circulation may function in different way to influence the inflammation. Managements towards KCs are expected to be the effective way to improve the IRI.

In the case study of this paper, sensitivity experiments are carried out using the mesoscale non-hydrostaticWeather Research and Forecasting (WRF) model to investigate the impact of tropical cyclone (TC) Soudelor (2003) on the East Asian subtropical upper-level jet (EASJ) before TC Soudelor transformed into an extratropical cyclone. The physical mechanism for changes in the EASJ intensity and position caused by TC Soudelor is explored. Results indicate that TC Soudelor would warm the air in the middle and upper troposphere over the Japan Sea and the adjacent areas through stimulating northward propagating teleconnection pattern as well as releasing large amounts of latent heat, which led to increase (decrease) the meridional air temperature gradient to the south (north) below the EASJ axis. As a result, the geopotential height abnormally increased in the upper troposphere, resulting in an anomalous anticyclonic circulation belt along the EASJ axis. Correspondingly, the westerly winds to the north (south) of the EASJ axis intensified (weakened) and the EASJ axis shifted northward by one degree. The case study also suggests that before the extratropical cyclone transition of TC Soudelor, the TC activities had exerted significant impacts on the EASJ through thermodynamic processes.
Improved photocatalytic performance of Pd-doped ZnO

ZnO and Pd-doped ZnO photocatalysts with different molar ratio of Pd/Zn (1/100, 2/100, 3/100 and 4/100) were prepared by a solegel method. The photocatalysts prepared were characterized by BET surface area, X-ray diffraction (XRD), UV/vis diffuse reflectance (DRS) and surface photovoltage spectroscopy (SPS), respectively. The results show that doping Pd into ZnO decreases the BET surface area. The XRD spectra of the Pd-doped ZnO catalysts calcined at 773 K show only the characteristic peaks of wurtzite-type. Doping Pd into ZnO increases the absorbance of ZnO in visible region and enhances the photoinduced charge separation rate. The photocatalytic activity of Pd-doped ZnO photocatalysts for decolorization of methyl orange (MO) solution was evaluated, of all the photocatalysts prepared, the Pddoped ZnO with 3/100 possesses the best photocatalytic activity. The results of further experiments show that increased adsorption ability of light and high separation rate of photoinduced charge carriers all play an important role in promotion of photocatalytic activity of Pd-doped ZnO nanostructure.

Purpose Platelet-related indices, including mean platelet volume (MPV) and plateletocrit (PCT), have been reported as new prognostic factors of overall survival (OS) in many cancers, but not yet in biliary tract cancer (BTC). We intended to assess these indices in predicting OS in BTC patients with the aim to build a new prognostic model for patients with BTC after surgical resection. Materials and Methods Survival analysis and time receiver operating characteristic analysis were applied to screen the platelet indices. Univariate and multivariate Cox analyses were used to identify independent prognostic factors and develop a new prognostic model. Harrell’s C-statistics, calibration curves, and decisive curve analysis were used to assess the model. Results MPV and platelet distribution width (PDW)/PCT showed the best prognostic accuracy among the platelet indices. In multivariable analysis, factors predictive of poor OS were presence of nodal involvement, Non-radical surgery, poor tumor differentiation, carbohydrate antigen 19-9 > 100 U/mL, MPV > 8.1 fl, and PDW/PCT > 190. The new model was found to be superior to the TNM staging system and our new staging system showed higher discriminative power.Conclusion MPV and PDW/PCT have high prognostic value in BTC patients, and the novel staging system based on these two indices showed good discrimination and accuracy compared with the American Joint Committee on Cancer 7th TNM staging system.
