      • A Novel Molecular Grading Model: Combination of Ki67 and VEGF in Predicting Tumor Recurrence and Progression in Non-invasive Urothelial Bladder Cancer

        Purpose: To assess efficacy of Ki67 combined with VEGF as a molecular grading model to predict outcomes with non-muscle invasive bladder cancer (NMIBC). Materials: 72 NMIBC patients who underwent transurethral resection (TUR) followed by routine intravesical instillations were retrospectively analyzed in this study. Univariate and multivariate analyses were performed to confirm the prognostic values of the Ki67 labeling index (LI) and VEGF scoring for tumor recurrence and progression. Results: The novel molecular grading model for NMIBC contained three molecular grades including mG1 (Ki67 $LI{\leq}25%$, VEGF $scoring{\leq}8$), mG2 (Ki67 LI>25%, VEGF $scoring{\leq}8$; or Ki67 $LI{\leq}25%$, VEGF scoring > 8), and mG3 (Ki67 LI > 25%, VEGF scoring > 8), which can indicate favorable, intermediate and poor prognosis, respectively. Conclusions: The described novel molecular grading model utilizing Ki67 LI and VEGF scoring is helpful to effectively and accurately predict outcomes and optimize personal therapy.

      • Experimental investigation on the propagation characteristics of pressure oscillation in direct contact condensation with low mass flux steam jet

        <P><B>Abstract</B></P> <P>The propagation characteristics of pressure oscillation in direct contact condensation with low mass flux steam jet have been investigated experimentally. Steam is injected into subcooled water at one atmosphere pressure with steam mass flux and water temperature range of 186–272kg/(m<SUP>2</SUP> s) and 293–343K. The pressure oscillation propagates in the form of wave with stable dominant frequency, however the wave intensity attenuates with the increasing distance from the oscillation source. The root mean square of pressure wave <I>p</I> <SUB>rms</SUB> attenuates rapidly with the increasing dimensionless radial distance from the nozzle exit. At about dimensionless radial distance <I>R</I> =100, the <I>p</I> <SUB>rms</SUB> is attenuated by about 90%. Although the dominant frequency of the pressure oscillation is constant during the propagation, after <I>R</I> =100, there will be not enough energy for the pressure oscillation to resonate with relevant equipment. A correlation equation to calculate the root mean square of pressure oscillation along the radial distance is given. The prediction errors are within ±30% compared with the experimental data.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The pressure oscillation propagates in the form of wave. </LI> <LI> In the propagation process, the frequency remains the same but the intensity attenuates. </LI> <LI> A correlation to calculate the <I>p</I> <SUB>rms</SUB> along the radial distance is given. </LI> <LI> At about R=100, the <I>p</I> <SUB>rms</SUB> is attenuated by about 90% and most of the energy is dissipated. </LI> </UL> </P>

        <P><B>Abstract</B></P> <P>Using X-ray Magnetic Linear Dichroism (XMLD) measurement, we investigate the antiferromagnetic (AFM) NiO spin reorientation transition (SRT) in epitaxial NiO/CoO/MgO(0 0 1) system at room temperature and constructed a complete NiO spin phase diagram. Then utilizing Magneto-Optic Kerr Effect (MOKE) combined with a rotation magnetic field (ROTMOKE), we investigated the effect of NiO SRT on the magnetic anisotropy of a ferromagnetic (FM) Py overlayer in Py/NiO/CoO/MgO(0 0 1) system. We find that the Py coercivity and anisotropy only slightly enhanced for Py on top of out-of-plane NiO spins but greatly enhanced for Py on top of in-plane NiO spins.       • Bathyal records of enhanced silicate erosion and weathering on the exposed Luzon shelf during glacial lowstands and their significance for atmospheric CO<sub>2</sub> sink

        <P><B>Abstract</B></P> <P>We present a new high-resolution multiproxy data set of mass accumulation rates, Sr-Nd isotopes, as well as major and trace elements for the siliciclastic sediment fraction from International Marine Global Change Study Core MD06-3052, located on the continental slope of the western Philippine Sea. We integrate our new data with published grain sizes and sea surface temperatures from the same core, as well as with Equatorial Pacific sea level, and East Asian summer monsoon precipitation, in order to constrain at high-resolution changes in physical erosion and chemical weathering intensities on Luzon, and sediment source-to-sink processes. We assess the potential significance of chemical weathering of arc silicates in regulating global atmospheric CO<SUB>2</SUB> since 156kyr BP. Sr-Nd isotopes show that the siliciclastic sediments were dominantly sourced from volcanic rocks exposed on Luzon (~68–100%), with a lesser contribution from Asian dust (~0–32%). Different indices indicate that stronger physical erosion and chemical weathering occurred during Marine Isotope Stage (MIS) 6 (130–156kyr BP), as well as in the latter stage of MIS 3 and MIS 2 (14–40kyr BP). The large sea-level lowstands and associated significant exposure of continental shelf in the western Philippine Sea during these two cold periods should favor physical erosion and chemical weathering of unconsolidated sediments on the exposed shelf. Furthermore, we notice the relatively good coherence between chemical weathering intensities on Luzon and global atmospheric CO<SUB>2</SUB> concentrations over these cold intervals. We suggest that strengthening of chemical weathering of silicates on Luzon and other tropical arcs (within 20° of the Equator) during the Quaternary glacial sea-level lowstands may significantly contribute to the lowering of atmospheric CO<SUB>2</SUB> concentrations during ice ages. We estimate that a significant fraction, up to ~16% (i.e., ~8ppmv), of all atmospheric CO<SUB>2</SUB> ultimately sequestered by silicate weathering may be processed through an area corresponding to only ~1% of the exorheic drainage area worldwide.</P> <P><B>Highlights</B></P> <P> <UL> <LI> First high-resolution silicate weathering study of the western Philippine Sea. </LI> <LI> Glacial sea-level lowstands favored silicate weathering of sediments on exposed shelf. </LI> <LI> Enhanced weathering on tropical arcs significantly sequestered atmospheric CO2. </LI> </UL> </P>

        Mixtures of Tea and Citrus maxima (pomelo) Alleviate Lipid Deposition in HepG2 Cells Through the AMPK/ACC Signaling Pathway

        Tea and citrus maxima are natural, medicinal homologous plants, typically used for making beverages, which have anticancer, antiobesity, and antioxidation properties. Green tea, yellow tea, and black tea were combined with citrus maxima to obtain green tea and Citrus maxima (GTCM), yellow tea and Citrus maxima (YTCM), and black tea and Citrus maxima (BTCM). The biochemical components of these mixtures were analyzed, and their possible effects and mechanisms on relieving liver lipid deposition were explored. The tea polyphenols, free amino acids, phenolamine ratio, and caffeine were comparable in YTCM and GTCM, being significantly higher than those in BTCM. In addition, the content of esterified catechins, nonesterified catechins, and total catechins in YTCM was significantly higher than those in GTCM and BTCM. All three mixtures of Citrus maxima tea significantly reduced lipid deposition in HepG2 cells, with GTCM and YTCM being slightly more effective than BTCM. Regarding the possible mechanism, Western blot analysis revealed that the three Citrus maxima tea mixtures could activate the AMPK/ACC signaling pathway, upregulate the expression of p-AMPK, p-ACC, and CPT-1 proteins, and downregulate the expression of SREBP1c and fatty acid synthase proteins to inhibit fat synthesis, thereby relieving lipid deposition in liver cells. In conclusion, as a novel and healthy beverage, Citrus maxima tea has the potential to alleviate liver lipid deposition, and further could be responsible for obesity treatment.

        cAMP induction by ouabain promotes endothelin-1 secretion via MAPK/ERK signaling in beating rabbit atria

        Adenosine 3 ,5 -cyclic monophosphate (cAMP) participates in the regulation of numerous cellular functions, including the Na<sup>+</sup>-K<sup>+</sup>-ATPase (sodium pump). Ouabain, used in the treatment of several heart diseases, is known to increase cAMP levels but its effects on the atrium are not understood. The aim of the present study was to examine the effect of ouabain on the regulation of atrial cAMP production and its roles in atrial endothelin-1 (ET-1) secretion in isolated perfused beating rabbit atria. Our results showed that ouabain (3.0 μmol/L) significantly increased atrial dynamics and cAMP levels during recovery period. The ouabainincreased atrial dynamics was blocked by KB-R7943 (3.0 μmol/L), an inhibitor for reverse mode of Na<sup>+</sup>-Ca<sup>2+</sup> exchangers (NCX), but did not by L-type Ca<sup>2+</sup> channel blocker nifedipine (1.0 μmol/L) or protein kinase A (PKA) selective inhibitor H-89 (3.0 μmol/L). Ouabain also enhanced atrial intracellular cAMP production in response to forskolin and theophyline (100.0 μmol/L), an inhibitor of phosphodiesterase, potentiated the ouabain-induced increase in cAMP. Ouabain and 8-Bromo-cAMP (0.5 μmol/L) markedly increased atrial ET-1 secretion, which was blocked by H-89 and by PD98059 (30 μmol/L), an inhibitor of extracellular-signal-regulated kinase (ERK) without changing ouabain-induced atrial dynamics. Our results demonstrated that ouabain increases atrial cAMP levels and promotes atrial ET-1 secretion via the mitogen-activated protein kinase (MAPK)/ERK signaling pathway. These findings may explain the development of cardiac hypertrophy in response to digitalis-like compounds.

        Electrochemical Polymerization of Methoxy Polyethylene Glycol Acrylate on Carbon Fiber for Improved Interfacial Properties

        Methoxy polyethylene glycol acrylate (MPGA) was electrochemical polymerized on carbon fiber (CF) by usingcyclic voltammetry (CV) to enhance the surface properties of CF. Fourier transform infrared spectroscopy (FTIR), scanningelectron microscopy (SEM) and thermogravimetry analysis (TGA) investigations confirmed that MPGA electropolymerizedon CF (MPGA-CF) was obtained. The low frequency capacitance of MPGA-CF (CLF=5.85 mF·cm-2) was higher thanCF(CLF=4.75 mF·cm-2) based on the result of electrochemical impedance spectroscopy(EIS) and the data obtained fitted withan R(Q(R(Q(R(CR))))) equivalent circuit model. Contact angle experiment showed that the contact angle between water andMPGA-CF is significantly lower than that between water and CF. Mechanical experimental results indicated that theinterlaminar shear strength (ILSS) of MPGA-CF/epoxy resin composites (32.24 MPa) has improved by 134.81 % comparewith the CF/epoxy resin composites (13.73 MPa), which significantly enhanced interfacial properties of epoxy resincomposites.

        Isolation of Chemical Constituents from the Aerial Parts of Verbascum thapsus and Their Antiangiogenic and Antiproliferative Activities

        Phytochemical investigation of Verbascum thapsus led to the isolation and identification of one new iridoid compound named verbathasin A, along with ten known compounds. The structure and relative stereochemistry of verbathasin A were elucidated by analysis of spectroscopic data. All the isolates except 10-deoxyeucommiol and ajugol were tested for antiangiogenic and antiproliferative activities, and compounds luteolin and 3-O-fucopyranosylsaikogenin F showed promising antiproliferative activities, with an obvious effect of inducing apoptosis of A549 lung cancer cells.
