Chitin deacetylase (CDA) is an insect chitin degradation enzyme that catalyzes the deacetylation of chitin to form chitosan. In this study, combination of rapid-amplification of cDNA ends (RACE) technology with Cnaphalocrocis medinalis transcriptome database analysis revealed the presence of at least five C. medinalis CDAs (CmCDAs), which were CmCDA1, CmCDA2, CmCDA4, CmCDA5, and CmCDA6. The cDNA sequences of CmCDA1, CmCDA2, and CmCDA4 hadwhole open reading frame (ORF) for further analysis. Phylogenetic analysis indicated that insect CDAs could be categorized into five groups. CmCDAs' structural domain analysis revealed that all three CDAs contained the chitin deacetylase-like catalytic domain. CmCDA1 and CmCDA2 belong to Group I because they both contain the chitin-binding peritrophin-A domain (ChBD), low-density lipoprotein receptor class A domain (LDLa), and chitin deacetylase-like catalytic domain. CmCDA4 only contains ChBD and chitin deacetylase-like catalytic domain thus belongs to Group III. Tissue and developmental stage expression analysis showed that the expression levels of CmCDA1, CmCDA2, and CmCDA4 are significantly higher in the head than other tissues and also significantly higher in adults than in larvae. CmCDA5 had significantly higher expression in the integument than other tissues, suggesting potential roles in the process of degradation of chitin. In contrast, CmCDA5 showed relatively high expression in larvae. In conclusion, this study analyzed the cDNA sequences of three CDA genes and determined their expression and molecular characteristics, which provided a new sequence resource and improved the development of bio-pesticides and the biological pest control and contributed to management of this important agricultural pest.
A case study on safety integrity level analysis for shale gas station
Yuan-Jian Yang,Chao Huang,Qiu-Yang Zhong,Hai-Jun Chen,Hong-Zhong Huang 대한기계학회 2021 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.35 No.12
Safety integrity level (SIL), as defined in IEC 61511, is a widely used safety performance measure for safety instrumented functions. Shale gas station is an important part of shale gas gathering and transportation system. To ensure the safety of its instrument system is of great significance to the safe and stable operation of the whole gathering and transportation system. This paper is motivated to conduct a case study on safety integrity analysis for shale gas station by using the layer of protection analysis method (LOPA). Firstly, the equipment under control (EUC) is identified. It is followed by the analysis of reaction reasons on safety instrumented function (SIF) from safety instrumented system (SIS). Finally, according to the identification of independent protection layer (IPL) and probability of failure on demand (PFD), the SIL analysis for every SIF is carried out. The result shows that the SIS of this shale gas station has reached the required of SIL and there is no need for further increase in SIS. Utilizing this method can accurately determine the reliability of the SIS in the station, and it can also be the reference of safe operation for this shale gas station.

We have characterized a new male-sterile mutant in Arabidopsis that exhibits conditional sterility but has restored fertility when drought-stressed. This mutant, multiple impairments in male reproduction 1 (mimr1),shows pleiotropic defects in both vegetative and reproductive development. Examination with dissecting and scanning electron microscopes revealed that its pollen grains are not effectively released from the anther locule after dehiscence, and anther differentiation is defective. Growth of the style and stamen filaments are also abnormal. Histological analysis demonstrated that these phenomena are due not only to a noticeably reduced extension of the stamen but also greater elongation of the pistil. Genetic analysis indicated that mimr1 is a single locus recessive nuclear mutant. The mutation can be mapped to a locus strongly linked to a 1200-kb region on Chromosome 3. Meta-analysis of expression patterning presented several candidate genes in that region. No mutants with similar phenotypes have previously been reported, suggesting that mimr1 is a novel male-sterile locus. Characterization of MIMR1 will provide further insights into the molecular basis for the development of plant reproductive organs.

Aim: To take a deeper insight into the relationship between the root of the inferior mesenteric artery (IMA) and the autonomic nerve plexuses around it by cadaveric anatomy and explore anatomical evidence of autonomic nerve preservation in high ligation of the IMA in laparoscopic surgery for colorectal cancer. Methods: Anatomical dissection was performed on 11 formalin-fixed cadavers and 12 fresh cadavers. Anatomical evidence-based autonomic nerve preservation in high ligation of the IMA was performed in 22 laparoscopic curative resections of colorectal cancer. Results: As the upward continuation of the presacral nerves, the bilateral trunks of SHP had close but different relationships with the root of the IMA. The right trunk of SHP ran relatively far away from the root of IMA. When the apical lymph nodes were dissected close to the root of the IMA along the fascia space in front of the anterior renal fascia, the right trunk of SHP could be kept in suit under the anterior renal fascia. The left descending branches to SHP constituted a natural and constant anatomical landmark of the relationship between the root of IMA and the left autonomic nerves. Proximal to this, the left autonomic nerves surrounded the root of the IMA. Distally, the left trunk of the SHP departed from the root of IMA under the anterior renal fascia. When high ligation of the IMA was performed distal to it, the left trunk of SHP could be preserved. The distance between the left descending branches to SHP and the origin of IMA varied widely from 1.3 cm to 2.3 cm. Conclusions: The divergences of the bilateral autonomic nerve preservation around the root of the IMA may contribute to provide anatomical evidence for more precise evaluation of the optimal position of high ligation of the IMA in the future.
An Observer and Post Filter Based Scheme for Fault Estimation of Nonlinear Systems

The problem of actuator fault estimation for a kind of nonlinear discrete time varying (NDTV) systems is investigated. Norm bounded disturbance is considered and the nonlinear function is assumed to fulfill a Lipschitzlike condition. Fault estimation is accomplished by a novel nonlinear state observer and a dynamic post filter. First, the nonlinear state observer is designed with the help of the small gain theorem and the H∞ filtering approach. Then for the error dynamic of the observer, the dynamic post filter is constructed to estimate the fault. In this scheme, no approximation of the nonlinear function is taken, and the problem of infeasibility resulting from multiple synthesis conditions is considerably improved. Simulation studies are carried out with a nonlinear unmanned aerial vehicle (UAV) model. The turbulent condition is considered and the feed back control loop is employed. Simulation results show that the proposed method can accomplish the estimation work, while the residual evaluation based approach fails to detect the fault.

1-Deoxynojirimycin (DNJ) as a kind of alkaloid has been confirmed that could modulate glycometabolism andhas toxicity for the eri-silkworm in our previous research. On the contrary, what is the potentially defensivemechanism when the DNJ enters the eri-silkworm. Based on comparative transcriptome sequencing, we foundthat ATP-binding cassette (ABC) transporter genes could be induced significantly by DNJ. In this study, a total of16 putative ABC transporter genes were identified, which can be classified into seven subfamilies, namely oneABCA, four ABCBs, three ABCCs, two ABCDs, one ABCE, three ABCFs, and two ABCGs. Phylogenetic analysisrevealed that ScABCs had strong conservation with Bombyx mori. Reverse transcription quantitative PCR (RTqPCR)suggested that 6 ABC transporters had a strong positive correlation between RT-qPCR and transcriptomedata. Additionally, S. c. ricini ABC transporter C-subfamily 4 (ScABCC4), S. c. ricini ABC transporter G-subfamily4 (ScABCG4), S. c. ricini ABC transporter A-subfamily 3 (ScABCA3) and S. c. ricini ABC transporter C-subfamily10 (ScABCC10) showed different expression pattern in two feed dose (1% and 2% DNJ) and three time points(6h, 12 h, 48 h). This study provides the first study on identification, characterization and expression patterns ofABC transporter genes in S. c. ricini response to DNJ, and lays a foundation for further understanding of theirphysiological roles response to the alkaloid.

Epidermolysis bullosa (EB) is a very rare inherited disease featured with skin blistering resulting from minor trauma. Sometimes the esophageal mucosa could also be involved, which leads to esophageal strictures. Here we report two cases of EB-related esophageal strictures who were successfully treated with esophageal balloon dilations. The two cases with EB had severe dysphagia. Clinical examination showed signs of malnutrition, skin blisters and loss of toenails due to EB. They underwent careful fluoroscopic balloon dilation with 10- and 16-mm-sized balloon catheters, respectively. They could ingest soft and some solid foods after the procedure and maintained during the 20 months and 16 months follow-up periods.

Photodynamic therapy (PDT) is a promising cancer treatment modality that uses dye-sensitized photooxidation of biologic matter in target tissue. This study explored effects of the photosensitizer BCPD-17 during PDT for osteosarcoma. LM-8 osteosarcoma cells were treated with BCPD-17 and cell viability after laser irradiation was assessed in vitro with the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay. The effects of BCPD-17 during PDT recurrence were then examined on tumor-bearing mice in vivo. BCPD-17 had dosedependent cytotoxic effects on LM-8 osteosarcoma cells after laser irradiation which also had energy-dependent effects on the cells. The rate of local recurrence was reduced when marginal resection of mice tumors was followed by BCPD-17-mediated PDT. Our results indicated BCPD-17-mediated PDT in combination with marginal resection of tumors is a potentially new effective treatment for osteosarcoma.
