An Interactive Perspective Scene Completion Framework Guided by Complanate Mesh

This paper presents an efficient interactive framework for perspective scene completion and editing tasks, which are available largely in the real world but rarely studied in the field of image completion. Considering that it is quite hard to extract perspective information from a single image, this work starts from a friendly and portable interactive platform to obtain the basic perspective data. Then, in order to make this interface less sensitive, easier and more flexible, a perspective-rectification based correction mechanism is proposed to iteratively update the locations of the initial points selected by users. At last, a complanate mesh is generated by the geometry calculations from these corrected initial positions. This mesh must approximate the perspective direction and the structure topology as much as possible so that the filling process can be conducted under the constraint of the perspective effects of the original image. Our experiments show the results with good qualities and performances, and also demonstrate the validity of our approaches by various perspective scenes and images.

Mammalian target of rapamycin complex 1 (mTORC1) is a central regulator of cell growth and metabolism and is sufficient to induce specific metabolic processes, including de novo lipid biosynthesis. Elongation of very-long-chain fatty acids 1 (ELOVL1) is a ubiquitously expressed gene and the product of which was thought to be associated with elongation of carbon (C) chain in fatty acids. In the present study, we examined the effects of rapamycin, a specific inhibitor of mTORC1, on ELOVL1 expression and docosahexaenoic acid (DHA, C22:6 n-3) synthesis in bovine mammary epithelial cells (BMECs). We found that rapamycin decreased the relative abundance of ELOVL1 mRNA, ELOVL1 expression and the level of DHA in a time-dependent manner. These data indicate that ELOVL1 expression and DHA synthesis are regulated by mTORC1 in BMECs.

As a synthetic glucocorticoid, dexamethasone is widely used to treat potential premature delivery and related diseases. Our previous studies have shown that prenatal dexamethasone exposure (PDE) can cause bone dysplasia and susceptibility to osteoporosis in female rat offspring. However, whether the effect of PDE on bone development can be extended to the third generation (F3 generation) and its multigenerational mechanism of inheritance have not been reported. In this study, we found that PDE delayed fetal bone development and reduced adult bone mass in female rat offspring of the F1 generation, and this effect of low bone mass caused by PDE even continued to the F2 and F3 generations. Furthermore, we found that PDE increases the expression of miR-98-3p but decreases JAG1/Notch1 signaling in the bone tissue of female fetal rats. Moreover, the expression changes of miR-98-3p/JAG1/Notch1 caused by PDE continued from the F1 to F3 adult offspring. Furthermore, the expression levels of miR-98-3p in oocytes of the F1 and F2 generations were increased. We also confirmed that dexamethasone upregulates the expression of miR-98-3p in vitro and shows targeted inhibition of JAG1/Notch1 signaling, leading to poor osteogenic differentiation of bone marrow mesenchymal stem cells. In conclusion, maternal dexamethasone exposure caused low bone mass in female rat offspring with a multigenerational inheritance effect, the mechanism of which is related to the inhibition of JAG1/Notch1 signaling caused by the continuous upregulation of miR-98-3p expression in bone tissues transmitted by F2 and F3 oocytes.

PSMB1 (proteasome subunit beta type 1) is a core component of the 20S proteasome, and based on its structure, it might have a crucial function in the transcription of certain genes. Rheb (Ras homolog enriched in brain) is a Ras-like small GTPase that acts as an upstream positive regulator of the mechanistic target of rapamycin complex 1 (mTORC1) pathway. We cloned and characterized PSMB1 (KY310590.1) to determine its function in cell cycle progression and proliferation of goat fetal fibroblasts (GFbs). Further, by yeast two-hybrid screen and coimmunoprecipitation, we conformed that PSMB1 interacts directly with Rheb. An siRNA was designed and expressed targeting PSMB1 mRNA in GFbs and inducing cell cycle arrest. Rheb overexpression in GFbs significantly increased the number of S phase cells and growth efficiency compared with control cells. These data indicate that PSMB1 and Rheb have important functions in the cell cycle and proliferation of GFbs, indicating that their interaction governs many processes in GFbs.

We present a study of summer precipitation changes overthe Yangtze River Valley (YRV) and North China (NC) simulatedfrom 20 models of the CMIP3 (phase 3 of the Coupled Model IntercomparisonProject). It is found that the LASG-FGOALS-g1.0 (fgoals)model has the highest ability in simulating both the interannualvariability of individual regions and the seesaw pattern of the tworegions observed during the past few decades. Analyses of atmosphericcirculations indicate that the variability in precipitation isclosely associated with the 850 hPa meridional winds over the tworegions. Wetness in the YRV and dryness in NC are corresponding tostrong meridional wind gradient and weak meridional wind overthese two regions, respectively. The ability of a coupled generalcirculation model (CGCM) to simulate precipitation changes in theYRV and NC depends on how well the model reproduces bothobserved associations of precipitation with overlying meridionalwinds and observed meridional wind features in summer. Analysis offuture precipitation changes over the two regions projected by thefgoals model under the IPCC scenarios B1 and A1B suggests asignificant increase of 7-15% for NC after 2040s due to thestrengthened meridional winds, and a slight increase over the YRVdue to less significant intensification of the Mei-yu front.
