        Direct Interaction between Ras Homolog Enriched in Brain and FK506 Binding Protein 38 in Cashmere Goat Fetal Fibroblast Cells

        Ras homolog enriched in brain (Rheb) and FK506 binding protein 38 (FKBP38) are two important regulatory proteins in the mammalian target of rapamycin (mTOR) pathway. There are contradictory data on the interaction between Rheb and FKBP38 in human cells, but this association has not been examined in cashmere goat cells. To investigate the interaction between Rheb and FKBP38, we overexpressed goat Rheb and FKBP38 in goat fetal fibroblasts, extracted whole proteins, and performed coimmunoprecipitation to detect them by western blot. We found Rheb binds directly to FKBP38. Then, we constructed bait vectors (pGBKT7-Rheb/FKBP38) and prey vectors (pGADT7-Rheb/FKBP38), and examined their interaction by yeast two-hybrid assay. Their direct interaction was observed, regardless of which plasmid served as the prey or bait vector. These results indicate that the 2 proteins interact directly in vivo. Novel evidence is presented on the mTOR signal pathway in Cashmere goat cells.

        Exploring the Influence of Vehicle Mobility on Information Spreading in VANETs

        With the advent of 5G communications, internet of vehicles technology has been widely used in vehicles. Then the dynamic spread of information between vehicles began to come into focus with more research. It is well known that the identification of nodes with great spread influence has always been a hot topic in the field of information spreading. Most of the existing work measures the propagation influence by degree centrality, betweenness centrality and closeness centrality. In this paper, we will identify influential vehicle nodes based on the mobility characteristics of vehicles to explore the information spreading between vehicles in VANETs. Different from the above methods, we mainly explore the influence of the radius of gyration and vehicle kilometers of travel on information spreading. We use a real vehicle trajectory data to simulate the information transmission process between vehicles based on the susceptible-infected-recovered SIR model. The experimental results show that the influence of information spreading does not enhance with increasing radius of gyration and vehicle kilometers of travel. The fact is that both the radius of gyration and the distance travelled have a significant influence on information spreading when they are close to the median. When the value of both is large or small, it has little influence on information spreading. In view of this results, we can use the radius of gyration and vehicle kilometers of travel to better facilitate the transmission of information between vehicles.

        Dysregulated lncRNA and mRNA may promote the progression of ischemic stroke via immune and inflammatory pathways: results from RNA sequencing and bioinformatics analysis

        Background: Long non-coding RNAs (lncRNAs) are widely involved in gene transcription regulation and which act as epigenetic modifiers in many diseases. Objective: To determine whether lncRNAs are involved in ischemic stroke (IS), we analyzed the expression profile of lncRNAs and mRNAs in IS. Methods: RNA sequencing was performed on the blood of three pairs of IS patients and healthy controls. Differential expression analysis was used to identify differentially expressed lncRNAs (DElncRNAs) and mRNAs (DEmRNAs). Based on the co-expression relationships between lncRNA and mRNA, a series of bioinformatics analysis including GO and KEGG enrichment analysis and PPI analysis, were conducted to predict the function of lncRNA. Results: RNA sequencing produced a total of 5 DElncRNAs and 144 DEmRNAs. Influenza A pathway and Herpes simplex infection pathway were the most significant pathways. EP300 and NFKB1 were the most important target proteins, and Human leucocyte antigen (HLA) family were the key genes in IS. Conclusions: Analysis of this study revealed that dysregulated lncRNAs in IS may lead to IS by affecting the immune and inflammation system.

        MiR-200c-3p aggravates gastric cell carcinoma via KLF6

        Background Gastric cell carcinoma (GCC) is a common and high-incidence malignant gastrointestinal cancer that seriously threatens human life and safety. Evidences suggest that microRNAs (miRNAs) exhibit an essential role in regulating the occurrence and development of GCC, while the efects and possible mechanisms remain to be further explored. Objective This study was designed to explore whether miR-200c-3p exerted its functional role in the growth and metastasis of GCC, and investigate the possible mechanisms. Methods The expression levels of miR-200c-3p in GCC tissues and cell lines were detected by qRT-PCR analysis. The functional role of miR-200c-3p in the viability, proliferation, migration and invasion of GCC cells were evaluated by CCK-8, EdU, wound healing and Transwell assays. In addition, the candidate targets of miR-200c-3p was predicted and confrmed by dual-luciferase reporter assay. Moreover, the relationship between miR-200c-3p and target (Krüppel like factor 6, KLF6) was assessed by qRT-PCR and western blot assays. Besides, the expression levels of KLF6 in GCC cells were determined by qRT-PCR and western blot assays. Furthermore, the role of KLF6 in the viability, proliferation, migration and invasion of GCC cells mediated with miR-200c-3p mimics was evaluated by CCK-8, EdU, wound healing and Transwell assays. Results In the present study, a new tumor promoting function of miR-200c-3p was disclosed in GCC. We found that the expression of miR-200c-3p was obviously increased in clinic GCC tissues and cell lines. In addition, down-regulation of miR-200c-3p suppressed cell viability, proliferation, migration, and invasion in GCC cells. Moreover, KLF6 was verifed as a direct target of miR-200c-3p by binding its 3’-UTR. Additionally, KLF6 was remarkably decreased and was negatively associated with the miR-200c-3p expression in GCC cell lines. Furthermore, over-expression of KLF6 retarded the efects of miR-200c-3p on the growth and metastasis of GCC cell lines. Conclusions MiR-200c-3p potentially played a tumor-promoting role in the occurrence and development of GCC, which may be achieved by targeting KLF6.

        Comparative transcriptome analysis of three chrysanthemums provides insights into flavonoid and terpenoid biosynthesis

        Chrysanthemum indicum var. aromaticum is an important resource plant for breeding excellent aromatic varieties of ornamental and medicinal chrysanthemums. However, the biosynthesis and regulation mechanisms of typically aromatic ingredients in C. indicum var. aromaticum are poorly understood, which restricted the utilization of this plant. Here, the transcriptome of C. indicum var. aromaticum was investigated and comparative analysis was conducted with the other two closely related chrysanthemum taxa C. indicum and C. nankingense. The global transcriptome profiles showed clear difference of gene expression patterns between the three chrysanthemums, of which 52 flavonoid biosynthesis-associated genes, 157 transcription factors, and 24 flavonoid Uridine diphosphate (UDP) -glucuronosyl and -glucosyltransferase genes were significantly differentially expressed. Furthermore, 22 differential expression genes (DEGs) in relation to the terpenoid backbone biosynthesis and 183 DEGs related to terpenoid diversification were identified. Of which, 14 genes involved into TS/CYP gene cluster showed higher expression in CIA than those in CI or CN. These results increased our understanding of flavonoid diversification and aromatic ingredients accumulation in plants, and will also help improve resource conservation and sustainable utilization of the important resource plant C. indicum var. aromaticum.

        Self-validating Pneumatic Actuator Fault Diagnosis Based on Relevance Vector Machine

        In order to solve the fault diagnosis problem of self-validating (SEVA) pneumatic actuator, an actuator fault diagnosis approach based on relevance vector machine (RVM) regression modeling and relevance vector machine (RVM) multi-classifier is proposed. The RVM regression is used to establish the normal models of the SEVA pneumatic actuator. The residuals generated by comparing the output of the models and the actual SEVA actuator are used as the nonlinear features. Then, the structure of the RVM for multi-classification is designed using k-meaning clustering methods, which is used as fault classifier to identify the condition and fault pattern of the SEVA actuator. The proposed approach is verified using fault data generated by DABLib model and actuator data from Lublin Sugar Factory and compared with support vector machine (SVM) fault diagnosis approach. The results indicate that the proposed approach overcomes the drawbacks of SVM and resolves the small sample and nonlinear problem in SEVA pneumatic actuator fault diagnosis.

        Analysis on the Dynamic Characteristics of Cross-Tunnel Structures under Wheel-Rail Impact Forces

        The dynamic behaviour of cross-tunnel structures under the wheel-rail impact forces of heavyhaultrains is complicated. This paper establishes an impact force excitation input model tosimulate the wheel-rail vertical forces of rail welded joints. The calculated results of thecorrection formula of wheel-rail vertical forces for seamless rail joints are in good agreementwith the measured data and meet the accuracy requirements. The present paper describes a1/50 scale model experiment based on the case of the newly-built Jing-Zhang Railwayunderneath the existing Tang-Hu Heavy Haul Railway. Numerical simulation analysis usesFLAC3D to simulate the experiments and investigate the dynamic soil pressure characteristicsunder different factors. The test and numerical results showed that the dynamic responses ofthe upper tunnel monitoring points are stronger than the lower tunnel, but the maximumvertical settlement of the lower tunnel vault is bigger and prone to damage. As the depthincreases, the stress of the surrounding rock interlayer at each monitoring point graduallydecreases. In-depth, the surrounding rock stress decreased approximately non-linearly withincreasing surrounding rock grade and surrounding rock interlayer thickness.

        Nephroprotective effect of astaxanthin against trivalent inorganic arsenic-induced renal injury in wistar rats

        Inorganic arsenic (iAs) is a toxic metalloid found ubiquitously in the environment. In humans, exposure to iAs can result in toxicity and cause toxicological manifestations. Arsenic trioxide (As2O3) has been used in the treatment for acute promyelocytic leukemia. The kidney is the critical target organ of trivalent inorganic As (iAs<SUP>Ⅲ</SUP>) toxicity. We examine if oral administration of astaxanthin (AST) has protective effects on nephrotoxicity and oxidative stress induced by As2O3 exposure (via intraperitoneal injection) in rats. Markers of renal function, histopathological changes, Na<SUP>+</SUP>-K<SUP>+</SUP> ATPase, sulfydryl, oxidative stress, and As accumulation in kidneys were evaluated as indicators of As2O3 exposure. AST showed a significant protective effect against As2O3-induced nephrotoxicity. These results suggest that the mechanisms of action, by which AST reduces nephrotoxicity, may include antioxidant protection against oxidative injury and reduction of As accumulation. These findings might be of therapeutic benefit in humans or animals suffering from exposure to iAs<SUP>Ⅲ</SUP> from natural sources or cancer therapy.

        Thermo-mechanical Behaviour of Soft Clay between Operation and Shutdown of Submarine Hot Oil Pipeline

        The interaction between the pipeline and the surrounding soil is a critical factor in thebuckling analysis of the pipeline operating at high temperatures and pressures. The heatingpipeline alters the temperature and induces excess pore pressure in the surrounding soil,which in turn affects the mechanical properties of the soil. Therefore, a comprehensiveunderstanding of the thermo-mechanical response of the soil surrounding the pipeline duringoperation and shutdown is essential. To address this issue, this study developed a temperaturecontrolledpipe model test device that simulates the heating and cooling cycle of the pipe andthe temperature field and pore pressure response of the surrounding kaolin clay. The testresults showed that the soil temperature at the closest location to the pipe (0.1D) reached apeak value of approximately 48.7°C when the pipe was heated from 12.5°C to 55°C. Thepeak temperature decreased with increasing distance, reaching 22.5°C at 0.875D. Heattransfer analysis using ABAQUS software revealed that the influence range of the pipeline onthe soil temperature field during the heating cycle was about 2D. Additionally, a parametricstudy focused on the thermal conductivity (λsoil) of kaolin, and the best-fit value wasdetermined to be 0.6 W/(m·°C). The excess pore pressures were generated in the kaolinsurrounding the pipes during the heating cycle, and negative pore pressures were observedafter cooling cycle. Furthermore, a theoretical solution for calculating the thermally inducedpore pressure was derived, and preliminary verification showed good agreement between thetheoretically computed and experimentally measured results. These findings provide valuableinsights into the thermo-mechanical response of the soil surrounding pipelines duringoperation and shutdown, which can improve the design and safety of oil and gas pipelines.

        Proteasome subunit beta type 1 interacts directly with Rheb and regulates the cell cycle in Cashmere goat fetal fibroblasts

        PSMB1 (proteasome subunit beta type 1) is a core component of the 20S proteasome, and based on its structure, it might have a crucial function in the transcription of certain genes. Rheb (Ras homolog enriched in brain) is a Ras-like small GTPase that acts as an upstream positive regulator of the mechanistic target of rapamycin complex 1 (mTORC1) pathway. We cloned and characterized PSMB1 (KY310590.1) to determine its function in cell cycle progression and proliferation of goat fetal fibroblasts (GFbs). Further, by yeast two-hybrid screen and coimmunoprecipitation, we conformed that PSMB1 interacts directly with Rheb. An siRNA was designed and expressed targeting PSMB1 mRNA in GFbs and inducing cell cycle arrest. Rheb overexpression in GFbs significantly increased the number of S phase cells and growth efficiency compared with control cells. These data indicate that PSMB1 and Rheb have important functions in the cell cycle and proliferation of GFbs, indicating that their interaction governs many processes in GFbs.
