        Fluorescent Nanoparticles for Observing Primo Vascular System Along Sciatic Nerve

        The primo vascular system was found in the epineurium along the rat sciatic nerve following subcutaneous injection of fluorescent nanoparticles at the Zusanli acupoint (ST-36). Nanoparticles were injected into the primo-vessel near ST-36 and flowed along the sciatic nerve. Fluorescence revealed a structure in the epineurium that was hardly detectable. Images of the isolated sample stained with 4’,6-diamidino-2-phenylindole were captured using confocal microscopy. These images showed the distinctive nuclei distribution and multi-lumen structure of primo-vessels that differentiate them from lymphatic vessels, blood capillaries and nerves. This study demonstrates a new use for nanoparticles in fluorescence reflectance imaging techniques during in vivo imaging of primo-vessels.

        Study of Novel Threadlike Structures on the Intestinal Fascia of Dogs

        The primo-vascular system was visualized in the mesentery surrounding the small intestine of a dog using Trypan blue. This structure, which was first observed in a rat, formed a network as primo-vessel branches were joined to primo-nodes. Other characteristic features of the primo-vascular system, such as bundles of tubes with fibrous extracellular matrix in a primo-vessel and a broken-line alignment of rodshaped nuclei along the primo-vessel, were observed. Blood vessels, lymph vessels,and primo-vessels were present in the same mesentery, and they could clearly be distinguished by histological differences.

      • Bioremediation of Coastal Saline-Alkali Marshy Soil by the Irrigation with Pulp & Paper Wastewater

        Based on the importance of coastal saline-alkali soil remediation in Yellow River Delta, China, the Cl-、Na+ transferring tendency in soil and the saline-alkali soil eco-remediation effects were researched by measuring soil Cl-、Na+ 、soluble salt、soil respire rate and dry biomass weight of reed, etc. periodically. The results shows: the concentration of irrigated soil Cl-、Na+ and soluble salt were decreased 57.7-70%、45.7-47.1% and 53.2-59.7%, respectively, and soil salt decreasing level were little influenced by the hydraulic load, inundation depth varies in some degree, but it was influenced by dry-wet alternative irrigating mode The concentration of soil total nitrogen、organic mass、microbial respire rate, and the reed biomass above ground were increased averagely to 2.17、1.20、1.46 and 1.34 multiple respectively afterirrigation with wastewater, which have complex remediation effects on the coastal saline-alkali wetland, but there are some differences among the different irrigating crafts.

      • High Expression of MICA in Human Kidney Cancer Tissue and Renal Cell Carcinoma Lines

        The overall incidence and mortality of renal cell carcinoma (RCC), the most common kidney cancer, are steadily increasing for reasons that are not fully explained. Our aim was to explore the expression of membrane MHC class I chain-related gene A (mMICA) in human RCC cell lines and tissue specimens, and to determine expression of soluble MICA (sMICA) in serum of patients with renal cell carcinoma, we used flow cytometry (FCM) and immunohistochemistry as well as an enzyme linked immunosorbent assay (ELISA). The results showed that percentage of mMICA expression was significantly increased in human kidney cancer tissues and RCC cell lines (786-O and Ketr-3) than that in healthy adults and human embryonic kidney 293 (HEK293) cell line individuality (P<0.05). sMICA content in healthy adults was negative, but in renal cancer patients was significantly elevated (P<0.05). Our research showed that high expression of MICA in human kidney cancer, this results show that MICA might serve as potential tumor-associated antigen (TAA) in RCC.

      • Wind tunnel investigation on wind characteristics of flat and mountainous terrain

        Wind tunnel test is often adopted to assess the site-specific wind characteristics for the design of bridges as suggested by current design standards. To investigate the wind characteristics of flat and mountainous terrain, two topographic models are tested in a boundary layer wind tunnel. The wind characteristics, including the vertical and horizontal mean wind speed distributions, the turbulence intensity, and the wind power spectra, are presented. They are investigated intensively in present study with the discussions on the effect of wind direction and the effect of topography. It is indicated that for flat terrain, the wind direction has negligible effect on the wind characteristics, however, the assumption of a homogenous wind field for the mountainous terrain is not applicable. Further, the non-homogeneous wind field can be defined based on a proposed approach if the wind tunnel test or on-site measurement is performed. The calculated turbulence intensities and wind power spectra by using the measured wind speeds are also given. It is shown that for the mountainous terrain, engineers should take into account the variability of the wind characteristics for design considerations

      • Antiproliferative Effects of Celecoxib in Hep-2 Cells through Telomerase Inhibition and Induction of Apoptosis

        Background: To investigate the effect of celecoxib on telomerase activity and apoptosis in a human laryngeal squamous carcinoma cell line (Hep-2 cells). Materials and Methods: The growth inhibition rate of Hep-2 cells in vitro was measured by MTT assay, and apoptosis by TUNEL assay and flow cytometry (FCM). The TRAP-ELISA method was used to determine telomerase activity in Hep-2 cells. The mRNA expression of human telomerase RNA component(hTR), human telomerase reverse transcriptase (hTERT) and human telomerase-associated protein(hTEP1) was determined by RT-PCR assay. Expression of Bax and Bcl-2 proteins was assessed by Western blotting. Results: Celecoxib can inhibit proliferation and induce apoptosis in a dose- and time-dependent manner, repress telomerase activity, decrease hTERT mRNA and Bcl-2 protein expression and increase Bax protein expression, PGE2 had no effect on telomerase. Conclusions: Celecoxib had the antiproliferative and pro-apoptotic effect in Hep-2 cells. Apoptosis was accompanied by a decrease in telomerase activity which was directly correlated with hTERT mRNA and up-regulation of Bax/Bcl-2. Bcl-2 may thus play an important role in telomerase activity as well as apoptosis.

        Preventative effects of Lactobacillus plantarum YS-3 on oxazolone-induced BALB/c colitis in mice

        In this study, the preventative effects of Lactobacillus plantarum YS-3 (LP-YS3) on colitis were studied using an in vitro animal experiment. Oxazolone was administered to BALB/c mice to induce colitis, and the preventive effects of LP-YS3 were determined using serum- and tissue-related indexes. The mice treated with LP-YS3 showed a significant decrease (p<0.05) in disease activity index for colitis by inhibiting colon shortening and increasing colon weight/length ratio. The application of LP-YS3 resulted in a significant reduction in myeloperoxidase, nitric oxide, and malondialdehyde activities and a decrease in glutathione activity (p<0.05) in mouse colon tissues. LP-YS3 also significantly increased serum interleukin-2 and reduced interleukin-10 cytokines levels in mice (p<0.05). Reverse transcription-polymerase chain reaction and western blot assays showed that LP-YS3 application resulted in a significant increase in neuronal nitric oxide synthase, endothelial nitric oxide synthase, c-Kit, and stem cell factor expressions and a decrease in inducible nitric oxide synthase, interleukin-8, and C-X-C motif chemokine receptor 2 expressions in mouse colonic tissues (p<0.05). These findings indicate that LP-YS3 imparts preventative effects on oxazolone-induced colitis in mice.

        Protective effect Malus pumila Mill leaf polyphenols in reserpineinduced gastric ulcer in mice

        The protective effect of polyphenols from Malus pumila leaf (MALP) on reserpine (10 mg/kg)- induced gastric ulcer in Kunming (KM) mice was investigated. Gastric juice secretion, pH of gastric juice, serum neuropeptides, including motilin (MOT), substance P (SP), vasoactive intestinal peptide (VIP), and somatostatin (SS) levels, as well as serum levels of inflammatory cytokines (TNF-a, IL-6, IL-12, and IFN-c) were measured. Gastric occludin and p38MAPK phosphorylation was analyzed by RT-PCR or Western blotting. MALP reduced reserpineinduced gastric juice secretion and increased the pH of gastric juice. MALP increased the serum levels of VIP and SS and reduced MOT, SP, and inflammatory cytokines in serum. It also modulated the gastric expression of occludin and p38MAPK phosphorylation in mice. These results suggest that MALP showed a protective effect against reserpine-induced ulcer in mice by reducing gastric juice secretion, modulating serum neuropeptide levels, attenuating serum inflammatory cytokines, and regulating gastric levels of occludin and p38MAPK phosphorylation.

        White Tea (Camellia sinenesis (L.)) Ethanol Extracts Attenuate Reserpine-induced Gastric Ulcers in Mice

        The gastroprotective effect of white tea extract ethanol (WTEE) on reserpine induced gastric ulcers in KM mice was investigated. Gastric juice secretion, the pH of gastric juice, serum neuropeptides, including motilin (MOT), substance P (SP), vasoactive intestinal peptide (VIP), and somatostatin (SS) levels, and serum levels of the inflammatory cytokines TNF-α, IL-6, IL-12, and IFN-γ were measured. WTEE dose-dependently reduced reserpine-induced gastric juice secretion and increased the pH of gastric juice. WTEE increased serum levels of VIP and SS and reduced levels of MOT, SP, and inflammatory cytokines in the serum. WTEE modulated gastric expression of occludin and p38 phosphorylation in ulcer-bearing mice. WTEE has a gastroprotective effect against reserpine-induced gastric ulcers in KM mice via reduction of gastric juice secretion, modulation of serum neuropeptide levels, attenuation of serum inflammatory cytokines, and regulation of gastric levels of occludin and p38 phosphorylation.

        A putative plastidial adenine nucleotide transporter, BRITTLE1-3, plays an essential role in regulating chloroplast development in rice (Oryza sativa L.)

        Differentiation from proplastids into chloroplasts isa light- and energy-dependent process. How this process isregulated is still poorly understood at the molecular level. We herein report a new putative plastidial adenine nucleotidetransporter, BRITTLE1-3 (referred to as OsBT1-3), encoded bythe rice (Oryza sativa) White Stripe Leaf 2 (WSL2) gene. Loss of OsBT1-3 function results in defective chloroplastbiogenesis, severely reduced photosynthetic efficiency, andfinally a white stripe leaf phenotype in the first four leaves. The expression levels of genes related to chlorophyllbiosynthesis and photosynthesis are drastically reduced,accompanied with over accumulation of reactive oxygenspecies (ROS) in the wsl2 mutant. OsBT1-3 is targeted tothe chloroplasts and it expresses in almost all tissues inplants, especially in young leaves. OsBT1-3 consists of 419amino acids and exhibits features of all mitochondrialcarrier proteins, including a typical transmembrane-spanningdomain and a highly conserved sequence motif designatedas the ‘mitochondrial energy transfer signatures’. Phylogeneticanalysis shows that OsBT1-3 is a putative plastidialadenine nucleotide transporter and is most closely relatedto ZmBT1-2. Together, these observations suggest that thenew putative adenine nucleotide transporter, OsBT1-3,plays an essential role in regulating chloroplast biogenesisand maintenance of ROS homeostasis during rice seedlingde-etiolation.
