        Downstream Neighbor of Son Overexpression is Associated With Breast Cancer Progression and a Poor Prognosis

        Purpose: The incidence rate of breast cancer (BC) has increased annually. Downstream neighbor of son (DONSON) critically affects cell cycle progression and maintains stable genomic properties; however, its relevant effects on BC growth and progression require in-depth investigation. Methods: DONSON upregulation was validated in public databases. DONSON expression in matched BC and adjacent tissues and cell lines (MDA-MB-231, BT-549, and HS-578T) was determined using quantitative reverse transcription polymerase chain reaction. In vitro apoptosis, invasion, migration, and proliferation tests were performed to ascertain the functions of DONSON in BC cell lines. Then, using western blot analysis, the levels of DONSON downstream proteins were determined. Results: Compared to the control, DONSON was expressed at higher levels in BC tissues and cell lines. DONSON knockdown facilitated apoptosis and limited proliferation, migration, invasion, and S/G2 transition of BC cells in vitro. Furthermore, DONSON overexpression promoted BC cell proliferation and inhibited apoptosis in vitro. Moreover, DONSON knockdown reduced cyclin A1 and cyclin-dependent kinase 2 levels. Moreover, DONSON knockdown limited the progression of epithelial-mesenchymal transition. Conclusion: DONSON critically affects BC growth and serves as a possible target and marker for the efficacy of subsequent therapies.

        Fuzzy Approximation-Based Backstepping Control of Permanent Magnet Synchronous Motor

        A permanent magnet synchronous motor(PMSM) control system based on backstepping control can effectively improve the dynamic performance, and the design process is simple and easy to be implemented in engineering. However, factors such as changes in motor parameters due to environmental changes, wear and aging, and external load disturbances can adversely affect the control system, resulting in degraded control performance. To address this problem, this paper proposes a fuzzy approximation-based backstepping control method for PMSM. The method constructs a mathematical model containing the perturbation term of the PMSM body parameters and the load disturbance term. And the universal approximation property of the fuzzy logic system is used to approximate the disturbance terms in the model, based on which a backstepping controller satisfying the stability requirements is designed. Finally, the simulation and experiment are given, which compare with the traditional backstepping method. The results show that the proposed method can effectively suppress the adverse effects of motor parameter changes and load disturbances on the motor control system.

        Improved Pupil-Size Diversity Technology for High-Resolution Imaging with Faint Objects

        Phase aberration is one of the prime factors that degrade the quality of observed images. Pupil-size diversity technology (PSDT) is a newly developed post-processing method for aberration correction and image reconstruction. However, images reconstructed using PSDT suffer from massive grains when imaging astronomical faint objects, which extremely limits its further application. In this paper, we propose an improved PSDT with embedded denoising reprocessing to overcome this drawback. Diversity raw images, generated by modulating the size of the pupil, are firstly processed by blocking-matching and 3D filtering (BM3D), a state-of-art denoising algorithm. Then, the traditional PSDT can be employed to estimate the wavefront and reconstruct a high-resolution image. Both numerical simulations and experiment demonstrations show that our proposed strategy exhibits superior performance for aberrations correction and image restoration of faint objects in view of its efficiency and robustness. Being capable of realizing high-resolution imaging with faint objects, this proposed method may have important application prospects in the fields of astronomical object detection, remote sensing, \textit{etc.}

        Application of galactinol to tomato enhances tolerance to cold and heat stresses

        Galactinol, a galactosyl donor, is the key substrate in raffinose family oligosaccharide (RFO) biosynthesis pathways. Many studies proved that galactinol also regulates some defense-related genes to be transcribed as a sugar signal under biotic and abiotic stresses. There are four galactinol synthase (SlGolS) genes in tomato. In this study, SlGolS1, SlGolS2, and SlGolS4 responded to cold stress, especially SlGolS1 stems treated for 12 h and SlGolS4 stems treated for 24 h. Under heat stress, the expression levels of SlGolS1, SlGolS2, and SlGolS3, especially SlGolS1 and SlGolS2, increased in leaves, roots, and stems. When expressed in E. coli cells, SlGolS2 and SlGolS4 enhanced cold tolerance, whereas SlGolS1 and SlGolS3 improved heat tolerance. These results suggested that SlGolS family members played different roles in tolerance to cold and heat stresses. In addition, the application of galactinol or galactinol + α-galactosidase inhibitor (DGJ) improved the cold and heat tolerances of tomato plants, whereas the single application of DGJ had no effect. Interestingly, the applications of DGJ, galactinol, and galactinol + DGJ also affected the expression levels of SlRS, SlSTS, and SlAGAL under cold and heat stresses. These findings indicated that galactinol was involved in the biosynthesis pathways of RFOs as a galactosyl donor and regulated the expression levels of RFO biosynthesis and breakdown-related genes as a sugar signal under cold and heat stresses.
