Here, we report a highly water-soluble colorimetric and fluorescent pH probe that works in the pH range 0.2–3.0. The probe showed a rapid response, high stability, and excellent reversibility to acidity. Moreover, the fluorescence of probe was not influenced by the existence of high concentration of cations. The pKa of probe was calculated to be 2.25 ± 0.06. Furthermore, the pH test papers coated with the probe exhibited a distinct color change in acidic conditions.

We investigated the effects of naringenin and morin on IL-5 and ROS production in PMA+ionomycin-treated EL-4 cells with the corroboration of their antioxidant and anti-inflammatory properties using an asthma-induced mouse model. The EL-4 cell line was used to study the outcomes of naringenin or morin, followed by cell viability studies. Western blot analysis and ELISA test were used to determine Th2 mediated cytokines. In vivo studies were carried out on BALB/c mice to induce allergic asthma using ovalbumin administered intraperitoneally. Intracellular ROS was determined using 2’,7’-dichlorodihydrofluorescein diacetate, followed by serum enzymatic (AST and ALT) estimations and inflammatory cell count in the bronchoalveolar lavage fluid (BALF) and lung tissues. Histopathological studies were conducted to examine lung tissue-stained architecture. Our findings suggested that naringenin and morin significantly suppressed IL-5 and ROS production via various pathways. Interestingly, by reducing NFAT activity, naringenin and morin stimulated HO-1 expression, thereby suppressing IL-5 secretion due to regulating the transcription factor Nrf2 via P13/Akt or ERK/JNK signalling pathways in EL-4 cells, demonstrating the involvement of HO-1 expression in inhibiting asthmatic inflammation. The increased inflammatory cells in the BALF were substantially decreased by both naringenin and morin, followed by inhibition in the elevated Th-2 cytokines levels. The TNF-α protein levels in an allergic asthma mouse model were significantly reduced by suppressing Akt phosphorylation and eosinophil formation. Recent findings confirmed that naringenin and morin possess the potential to control asthma-related immune responses through antioxidant and anti-inflammatory properties, indicating potential therapeutic agents or functional foods
Inhibitory Effects of Syk Transfection on Lung Cancer Cell Invasion

Objective: Spleen tyrosine kinase (Syk) is closely related to tumor invasion and metastasis, and has been shown to have potential inhibitory effects in tumors. In this study, we constructed a eukaryotic expression vector for Syk and analyzed its effects on invasive ability of the A549 non-small cell lung cancer cell line in vitro. Methods: A fragment of Syk was obtained by RT-PCR from human lung cancer cells and cloned into the expression vector pLNCXSyk. After restriction endonuclease digestion, PCR and DNA sequencing confirmation, the recombinant Syk expression plasmid was transfected into A549 human lung cancer cells using lipofectamine protocols. After selection, the cells stably expressed Syk. Detection of Syk expression of the cells by RT-PCR, and invasive ability were examined. Results: The eukaryotic expression plamid pLNCXSyk was constructed and expressed stably in the A549 human lung cancer cells. The RT-PCR results showed that Syk mRNA expression was upregulated significantly (P<0.05). Lower invasion through a basal membrane were apparent after transfection (P<0.05). Conclusions: A eukaryotic expression plasmid to cause Syk expression in lung cancer cells can obviously inhibit their invasive ability in vitro.

Background: Smoking behavior is influenced by multiple genes, including the bitter taste gene TAS2R38. It has been reported that the correlation between TAS2R38 and smoking behavior has ethnicity-based differences. However, the TAS2R38 status in Chinese smokers is still unclear. Objective: This study aims to investigate the possible relationship between genetic variations in TAS2R38 (A49P, V262A and I296V) and smoking behaviors in the Han Chinese population. Methods: The haplotype analyses were performed and smoking behavior questionnaire was completed by 1271 individuals. Genetic association analyses for smoking behavior were analyzed using chi-square test. Further, for investigating the molecular mechanism of TAS2R38 variants effect on smoking behavior, we conducted TAS2R38-PAV and TAS2R38-AVI expression plasmids and tested the cellular calcium assay by cigarette smoke compounds stimulus in HEK293. Results: Significant associations of genetic variants within TAS2R38 were identified with smoking behavior. We found a higher PAV/PAV frequency than AVI/AVI in moderate and high nicotine dependence (FTND ≥ 4; X2 = 4.611, 1 df, p = 0.032) and strong cigarette smoke flavor intensity preference (X2 = 4.5383, 1 df, p = 0.033) in participants. Furthermore, in the in vitro cellular calcium assay, total particle matter (TPM), N-formylnornicotine and cotinine, existing in cigarette smoke, activated TAS2R38-PAV but not TAS2R38-AVI-transfected cells. Conclusion: Our data highlights that genetic variations in TAS2R38 are related to smoking behavior, especially nicotine dependence and cigarette smoke flavor intensity preference. Our findings may encourage further consideration of the taste process to identify individuals susceptible to nicotine dependence, particularly Han Chinese smokers.

This paper investigates the influences of change recipients’ supportive behaviors toward the national reform in the Chinese football sector. Qualitative data were collected through semi-structured interviews (n = 29), which were conducted with change recipients from national and local football associations and commercial football clubs. Drawing on an integrated conceptual framework, the findings suggest that the change-supportive behaviors demonstrated by the change recipients were influenced and incentivised by managerial factors (i.e., management competency, communication channels, participation in decision-making, leaders’ commitment to change, and principal support); and contextual factors (i.e., an amenable football environment and the perceived political pressure to change). Three manifestations of change-supportive behaviors were identified: a) showing understanding of the change but pessimistic about the outcome; b) supporting the change and being willing to take risks; and c) supporting the change and actively seeking alternative solutions.

Curcumol is a sesquiterpene originally isolated from curcuma rhizomes, a component of herbal remedies commonly used in oriental medicine. Its beneficial pharmacological activities have attract significant interest recently. In this study, anti-cancer activity of curcumol was examined with both in vitro and in vivo models. It was found that curcumol exhibited time- and concentration-dependent anti-proliferative effects in SPC-A-1 human lung adenocarcinoma cells with cell cycle arrest in the G0/G1 phase while apoptosis-induction was also confirmed with flow cytometry and morphological analyses. Interestingly, curcumol did not display growth inhibition in MRC-5 human embryonic lung fibroblasts, suggesting the anti-proliferative effects of curcumol were specific to cancer cells. Anti-neoplastic effects of curcumol were also confirmed in tumor bearing mice. Curcumol (60 mg/ kg daily) significantly reduced tumor size without causing notable toxicity. In conclusion, curcumol appears a favorable anti-cancer candidate for further development.

Experimental research on flow characteristics of open natural circulation system was performed, tofigure out the mechanism of the open natural circulation behaviors. The influence factors, such as theheating power, the inlet subcooled and the level of cooling tank on the flow characteristics of the systemwere examined. It was shown that within the scope of the experimental conditions, there are five flowtypes: single-phase stable flow, flash and geyser coexisting unstable flow, flash stable flow, flash unstableflow, and flash and boiling coexisting unstable flow. The geyser flow in flash and geyser coexisting unstable flow is different from classic geysers flow. The flow oscillation period and amplitude of the formerare more regular, is a newly discovered flow pattern. By drawing the flow instability boundary diagramand sorting out the flow types, it is found that the two-phase unstable flow is mainly characterized byboiling and flash, which determine the behavior of open natural circulation respectively or jointly. Moreover, compared with full liquid level system, non-full liquid level system is more prone to boilingphenomenon, and the range of heat flux density and undercooling degree corresponding to unstableflow is larger

This paper compares and analyzes the determinants of foreign direct investment and trade in China with OECD-member countries, Hong Kong, Singapore and Taiwan spanning the period from 2003 through 2011. Analysis used included the basic and extension gravity model. Referring to advanced research data, various control variables were utilized. Two main determinants of FDI and trade in China were found and these were GDP and geographical distance. GDP and the geographical distance of the countries under study from China have a significant influence on foreign direct investment and trade. This paper analyzed two main factors of economic development in China. That is FDI and trade. In this perspective, we provided not only a prediction of China's future economic development but also significant meaning for policy making and strategizing in developing countries.
