        Effects of Aqueous Extract of Schizandrae Fructus on Lead-Induced Change of Monoamine Neurotransmitters in Hippocampus

        The effects of aqueous extract of Schizandrae Fructus (AESC) on lead (Pb)-induced changes of monoamine neurotransmitters in the hippocampus (HIP) of adult rats were investigated. Male Sprague-Dawley rats were received intraperitoneal (i.p.) administration of Pb acetate (5 mg/kg/d) for 28 days and sacrificed 7 days after the last administration. Concentrations of norepinephrine (NE), dopamine (DA), serotonin (5-HT), 5-hydroxyindole acetic acid (5-HIAA) in HIP were measured by HPLC. There were significant decreases of NE, DA, 5-HT and 5-HIAA in Pb treated rats (P < 0.05), while pretreatment with AESC (100 mg/kg/d or 300 mg/kg/d, p.o., 2 h before Pb) greatly inhibited the decrease of monoamine transmitters, respectively (P < 0.05). Also, AESC (300 mg/kg/d) significantly increased the reduction of glutathione contents and superoxide dismutase activities in HIP induced by chronic Pb. These results suggest that AESC ameliorates Pb-induced depletion of monoamine neurotransmitters in HIP through its antioxidant activity.

        Signal Transformed Internal Model Control for Non-raster Scanning of Piezo-actuated Nanopositioning Stages

        This paper proposes a new signal transformed internal model control (STIMC) scheme for non-raster scanning patterns of piezo-actuated nanopositioning stages. To smooth the scanning signals superimposed with a ramp or time-varying amplitudes, a signal transformation operator is calculated to convert the references into standard harmonic waveforms. An inverse transformation operator is then added in the control loop to generate the driving signals. For the contained internal model control (IMC) design, an H∞ mixed-sensitivity method is utilized for the first order internal mode synthesis. A second and a third internal modes are included in the IMC for alleviating residual high-frequency errors resulted from the nonlinearity of hysteresis. To verify the proposed STIMC scheme, comparative experiments with conventional IMC are conducted based on a nanopositioning platform. Results prove that the STIMC is effective on non-raster signals’ tracking. A same tracking precision for Lissajous scanning can be obtained by STIMC compared with IMC. An improvement of larger than 50% and 80% of root-mean-square errors can be obtained for cycloid and spiral scanning patterns, respectively.

        In‑situ growth of ZIF‑8/CP with ultra‑high adsorption capacity for removing Malachite green from water

        An environmentally friendly and low-cost chitosan-containing polysaccharide (CP) composite ZIF-8/CP was designed and prepared based on the difficulty of separating the traditional adsorbent from the water phase. ZIF-8/CP was synthesized through in-situ growth approach. The physical, chemical and structure properties of ZIF-8/CP were determined through a series of characterization methods, including SEM, FT-IR and PXRD. The effects of touch time, pH, temperature, and coexisting ions on adsorption were assessed. In addition, kinetics, isotherms of adsorption and thermodynamics were examined. The data of isotherms for adsorption indicated that the adsorption of ZIF-8/CP on MG was similar to the Langmuir model, with a maximum adsorption capacity of 1428.57 mg/g. Moreover, the kinetic parameters were consistent with the pseudo- 2nd-order equation. Thermodynamic studies (ΔG < 0, ΔH > 0) demonstrated a heat-absorbing and spontaneous adsorption process. Our study reveals that ZIF-8/CP has good adsorption properties and environmental properties.

        Research on Modular Permanent Magnet Bias Magnetic Bearing

        In this study, a modular permanent magnet bias magnetic bearing is proposed to solve the problems of complex structure, diffi cult design and cumbersome processing of the existing permanent magnet bias magnetic bearing. Firstly, the structure, magnetic circuit distribution and working principle of modular permanent magnet bias magnetic bearing are introduced. Then, the mathematical formula of suspension force is deduced by using the equivalent magnetic circuit method, and the parameters are designed. The design results are verifi ed by fi nite element simulation, and the magnetic bearing is simulated under diff erent displacement and control current to analyze the current stiff ness characteristics and displacement stiff ness characteristics. Then, the fl oating characteristics are analyzed, and the fl oating force when the rotor is off set by −0.2 and −0.3 mm is simulated and analyzed respectively. Finally, the Model-free Control (MFC) method is used to complete the control experiment of the magnetic bearing, which verifi es the rationality of the control system.

        Effect of Glutinous Rice Slurry on the Unconfined Compressive Strength of Lime-Treated Seasonal Permafrost Subjected to Freeze-Thaw Cycles

        It is important to account for the poor ecological environment of weak seasonal permafrost regions while developing a suitable ground improvement technique. As an organic non-polluting material, glutinous rice slurry improves the unconfined compressive strength of the soil through induction and synergy with lime. The Qinghai-Tibet Plateau has cold weather, large temperature differences between day and night, and a fragile ecological environment. Once damaged, it is extremely difficult to recover. This study investigates the effects of glutinous rice slurry on the unconfined compressive strength (UCS), failure mode, and microstructure of lime-treated seasonally frozen soil subjected to freeze–thaw cycles. Organic glutinous rice slurry with concentrations of 0, 2, 4, 6, and 8% was added to soil specimens that were treated with 10% lime and subjected to 0, 1, 3, 6, and 10 freeze–thaw cycles. In tests, the addition of glutinous rice pulp could increase the UCS of the soil by up to 40%. The first freeze-thaw cycle has the greatest impact on the UCS of the soil sample, with a decrease of 6 – 15%. Glutinous rice slurry can reduce the degradation effects of freeze-thaw cycles by increasing the contact area between soil particles. In a comprehensive evaluation under test conditions, a glutinous rice slurry concentration of 4% led to optimal results. It revealed the change characteristics of the microstructure under the action of freeze-thaw cycles, which has a good corresponding relationship with the macroscopic mechanical properties. The results can provide reference value for the application of glutinous rice slurry in the modification of subgrade soil in seasonal permafrost areas.

      • Manual Acupuncture at PC6 Ameliorates Acute Restraint Stress-Induced Anxiety in Rats by Normalizing Amygdaloid Noradrenergic Response

        <P>Acupuncture improves ethanol withdrawal-induced anxiety in rats in an acupoint-dependent manner. Thus, the present study investigated the effects of acupuncture on acute restraint stress- (ARS-) induced anxiety. Male rats were exposed to ARS for 3 h followed by acupuncture at either PC6 (Neiguan), HT7 (Shenmen), or a nonacupoint (tail) once a day for three consecutive days. Five minutes after the third acupuncture treatment, anxiety-like behavior was evaluated in an elevated plus maze (EPM). Additionally, plasma corticosterone (CORT) levels were measured by radioimmunoassay and the concentrations of norepinephrine (NE) and 3-methoxy-4-hydroxy-phenylglycol (MHPG) in the central nucleus of the amygdala (CeA) were determined using high-performance liquid chromatography. Acupuncture at PC6, but not HT7 or a nonacupoint, attenuated anxiety-like behavior, but this attenuation was abolished by a postacupunctural intra-CeA infusion of NE. Acupuncture at PC6 also reduced the oversecretion of plasma CORT and inhibited increases in amygdaloid NE and MHPG induced by ARS. Further, Western blot analyses and real-time polymerase chain reaction assays revealed that acupuncture at PC6 prevented ARS-induced enhancements in the protein and mRNA expressions of tyrosine hydroxylase in the CeA. These results suggest that acupuncture performed specifically at acupoint PC6 reduces ARS-induced anxiety-like behavior by dampening amygdaloid noradrenergic responses.</P>

        The infertile individual analysis based on whole-exome sequencing in chinese multi-ethnic groups

        Background Infertility is a common and rapidly growing health issue around the world. The genetic analysis based on the infertile population is crucial for intervention and treatment. Objective To find candidate gene locus led to azoospermia in Chinese multi-ethnic groups and provide theoretical guidance for the diagnosis of genetic diseases to progressively aggravated infertility patients and sterile offspring with ART. Methods The study based on whole-exome sequencing (WES) was presented for genetic characteristic analysis of multi-ethnics and identification of variants related to infertility in Xinjiang area of China. Results The frequency of pathogenic variants showed significant ethnic differences among four main ethnics in Xinjiang. The population structure analysis confirmed that the Hui was close to the Han population, the Kazak was close to the Uygur population, and there are three ancestry components in the four ethnics. In addition, ten candidate variants potentially regulated azoospermia were detected, and KNTC1 (rs7968222: G > T) was chosen to validate the association. Through the analysis in the valid group, the frequency of rs7968222 (G > T) has a significant difference in the azoospermia population (11.76%, 8/68) and normospermia population (4.63%, 35/756) (P < 0.001). Interestingly, the proportion of people with abnormal follicle-stimulating hormone (FSH) level in the group carrying rs7968222 (G > T) was significantly higher than non-carriers (P < 0.05). Therefore, rs7968222 may regulate spermatogenesis through affecting hormone level. Conclusion Our study establishes the genetics analysis of Northwest China and finds a candidate gene locus KNTC1 (rs7968222: G > T), which is one of the genetic susceptibility factors for male azoospermia.

        KLF9 promotes autophagy and apoptosis in T-cell acute lymphoblastic leukemia cells by inhibiting AKT/mTOR signaling pathway

        Background T -cell acute lymphoblastic leukemia (T-ALL) is considered a malignant tumor with a high mortality rate. To combat this disease, exploring the mechanism of T-ALL progression is urgently needed. Krüppel-like factors (KLFs) are known as the transcription factors and mediate series of biological processes. KLF9 is a member of the KLF family which could serve as a tumor suppressor gene in most solid tumors. GEO Database analysis showed that KLF9 expression in normal T cells was higher than T-ALL cell lines and patients. However, the possible role of KLF9 in T-ALL progression is still unclear. Objective To uncover the possible eff ects of Krüppel-like transcription factor 9 (KLF9) on the progression of T-Acute lymphoblastic leukemia (T-ALL). Results The expression of KLF9 was low in human T-ALL cells. KLF9 suppressed the viability of T-ALL cells. In addition, KLF9 stimulated the apoptosis as well as autophagy of T-ALL cells. Mechanically, KLF9 suppressed AKT/mTOR pathway in T-ALL cells. Conclusion KLF9 suppressed viability and promoted autophagy as well as apoptosis in T-ALL cells by inhibiting AKT/ mTOR pathway.

        Korean Red Ginseng attenuates anxiety-like behavior during ethanol withdrawal in rats

        Background: Korean Red Ginseng (KRG) is known to have antianxiety properties. This study was conducted to investigate the anxiolytic effects of KRG extract (KRGE) during ethanol withdrawal (EW) and the involvement of the mesoamygdaloid dopamine (DA) system in it. Methods: Rats were treated with 3 g/kg/d of ethanol for 28 d, and subjected to 3 d of withdrawal. During EW, KRGE (20 mg/kg/d or 60 mg/kg/d, p.o.) was given to rats once/d for 3 d. Thirty min after the final dose of KRGE, anxiety-like behavior was evaluated in an elevated plus maze (EPM), and plasma corticosterone (CORT) levels were determined by a radioimmunoassay (RIA). In addition, concentrations of DA and 3,4-dihydroxyphenylacetic acid (DOPAC) in the central nucleus of the amygdala (CeA) were also measured by high performance liquid chromatography (HPLC). Results: The EPM test and RIA revealed KRGE inhibited anxiety-like behavior and the over secretion of plasma CORT during EW. Furthermore, the behavioral effect was blocked by a selective DA D2 receptor (D2R) antagonist (eticlopride) but not by a selective DA D1 receptor (D1R) antagonist (SCH23390). HPLC analyses showed KRGE reversed EW-induced decreases of DA and DOPAC in a dose-dependent way. Additionally, Western blotting and real-time polymerase chain reaction (PCR) assays showed that KRGE prevented the EW-induced reductions in tyrosine hydroxylase (TH) protein expression in the CeA and TH mRNA expression in the ventral tegmental area (VTA). Conclusion: These results suggest that KRGE has anxiolytic effects during EW by improving the mesoamygdaloid DA system.
