      • 카드뮴내성군주의 카드뮴 적응기에 나타나는 생리생화학적 변화

        산업폐수에서 분리한 Bacillus sp.를 카드뮴 첨가배지에서 배양하면 2mM 농도까지 내성을 가지며, 세포내 무기인산 중합체의 축적이 증가되는 바, 이들 무기인산의 대사에 관여하는 제반 세포내 생화학적인 변화양상을 조사하고자 하였다. 카드뮴 첨가 배양에 따른 효소 활성도는 poly-Pase 와 ACPase는 상당히 증가되었지만 ALPase 는 그 활성이 카드뮴의 영향으로 감소되었다. 카드뮴 첨가 배양에 따른 세포의 인지질의 변화 양상을 살펴 보면 phosphatidyl glycerol 에 대한 phosphatidyl choline의 상대적 비율이 lyso-phosphatidyl choline의 상대적 비율보다 훨씬 많게 관찰되어 phosphatidyl choline 이 lysophosphatidyl choline으로 분해되는 경로가 카드뮴에 의해 억제되는 것으로 사료된다. 한편, nucleotide pool의 변화 양상을 살펴 보면 카드뮴의 영향으로 adenylate pool을 증가하며 카드뮴 비처리 복합 배지에서 배양한 세포에서는 cytidylate pool이 현저한 증가를 보인 반면 starvation(카드뮴을 첨가하지 않은 당과 인산 결핍 배양)시킨 세포에서는 guanylate pool이 증가됨을 관찰할 수 있었다. Upon cadmium added medium, in the course of culturing Bacillus sp. which was isolated from sewage of Daejon industrial plants and which is cadmium resistant up to the concentration of 2mM, we observed that inorganic polyphosphate was highly accumulated. In order to elucidate regulatory mode of this phenomenon, we tried to check biochemical changes which were correlated with phosphate metabolism ; such as phosphatase activity, phospholipid pattern and nucleotide pool. It found that, depending upon culture with added cadium, enzyme activity was markedly increased in poly-Pase, and in ACPase also, but the effect of cadmium resulted in a decrease in ALPase activity. The variation of cellular content of phospholipids with cadmium addition was showed that relative ratio of phosphatidyl choline for phosphatidyl glycerol was major to that of lysophosphatidyl choline for phosphatidyl glycerol, implicating degradation pathway of phosphatidyl choline into lysophosphatidyl choline was repressed by cadmium. On the order hand, with respect to the change of nucleotide pool, it should be noted that the effect of cadmium induced a increase in cytidylate pool, which considerly increased in Cd-free complex medium, when compared with the starvation of a culture of cells with sugar-phosphate free medium without added cadmium.

      • 적응 뉴럴-퍼지 제어시스템의 설계에 관한 연구

        지능 제어기를 구현하기 위한 한 방법으로 본 논문은 퍼지 논리와 신경망 제어기법을 합성 결합한 새로운 형태의 FNC(Fuzzy-Neural Controller)와 적응 뉴럴-퍼지 제어 방식을 제안한다. 제안되는 뉴럴-퍼지 시스템은 적응성과 학습능력의 두가지 주요한 기능을 갖는다. 먼저 FNC에서의 퍼지 규칙의 전건부와 후건부는 클러스터링 방법과 다층신경망에 의해 구현된다. 그리고 MNN(Model Neural Network)에서의 다층신경망은 수학적 모델링이 어려운 플랜트의 상태를 동정하는데 도입하고 동적인 환경에 적응하도록 제어기에 교사 신호를 제공한다. FNC와 MNN으로 구성된 적응 제어시스템은 학습 알고리듬으로 역전파 학습 알고리듬이 사용된다. 제안된 방식의 효용성은 2 d.o.f 로보트 매니퓰레이터와 cart-pole의 모의 실험으로 입증된다. As an approach to develope the intelligent control scheme, this paper will propose an adaptive neural-fuzzy control scheme. The proposed neural-fuzzy control system, which consists of the Fuzzy-Neural Controller(FNC) and Model Neural Network(MNN), has two important characteristics of adaptation and learning. In the FNC, the antecedence and consequence of the fuzzy rule are constructed by a clustering method and a multi-layer neural network. And in the MNN, a multi-layer neural network is utilized to identify a unknown controlled plant and provides the FNC with the teaching signal. After constructing the adaptive control system using the FNC and the MNN, the error back propagation algorithm has been adopted as a learning technique. The effectiveness of the proposed scheme will be demonstrated by computer simulations of a cart-pole and a two d.o.f. robot manipulator.

        금강수역내에서 분리된 몇 종의 Ampicillin 내성균의 β-Lactamase 발현양상

        In order to characterize the bacterial β-lactamase types occurred in Kum river area, this research was designed to analyze with identification upon ampicillin (Amp) resistant bacteria, and was investigated into relationships between β-lactamase expression (inducibility) and growth phase under ampicillin shift and with restriction patterns of R-plasmids. As the identification result upon 4 strains of Amp resistant bacteria, KL 601 is identified as Pasteurella maltocida, KL 602 as Enterobacter agglomerans, KL 609 as Pseudomonas maltophilia and KL 611 as Acinetobactor calcoaceticus, respectively. In order to investigate Amp effect upon growth, when the Amp resistant bacteria were cultivated in nutrient liquid LB medium, in LB medium added with Amp, or in LB medium in the manner of Amp shift, the changes in growth rate and lag phase could not be observed. However, upon the prototrophic strains, KL 602 and KL 609, examining Amp effect upon these two strains in minimal GM63 liquid media, even though there was no change of maximum growth yield, the latent lag phase become very lengthened. In nutrient medium, β-lactamase activities of these 4 strains were metabolically regulated in accordance with growth phase, on the other hand, in minimal medium were differently regulated in accordance with growth phase and with strains. It could be postulated that KL 609 strain has the M.W.24,000, β-lactamase type which E. coli comprise, however, that KL 601, KL 602 and KL 611 have the M.W.35,000, β-lactamase type which E. cloacae or C. freundii comprise. Through the whole growth phase, since Amp did not affect to phosholipid metabolism, there is no change in phospholipid compisition upon Amp effect. Main phospholipid are the phosphatidyl glycerol and cardiolipin in all 4 Amp resistant strains. So, it was very significant that there were no changes in phospholipid composition and in growth rate upon nutrient medium under Amp stress. In order to recognize whether Amp resistant strains comprise the plasmid or not, preparation of plasmids and restriction digestion with Bam H1 were carried out, KL 601 strain has about 30 Kb of plasmid, KL 609 and KL 611 strain have about 25 Kb of plasmid and KL 602 does not have the plasmid, however. Hence, it will be predicted strongly that β-lactamase gene in KL 602 might be encoded chromosomally. Moreover, considering the results upon the changes of β-lactamase acticity, upon β-lactamase inducibility and upon changes of periplasmic protein profile under Amp stress, its prediction is very recognizable.

      • Two-dimensional electrophoresis of Bacillus sphaericus toxin : Relationship between activated and non-activated toxin

        곤충류에 병원성을 갖는(Entomopathogenic) Bacillus sphaericus(BS) 균주들은 43kD 크기의 단백질을 생성하여 활성화 과정을 거친 후 곤충에 치사성을 갖는 40kD 크기의 독성물질로 전환된다. 서로 다른 4종류의 곤충병원성 BS 균주들로부터 분리된 이들 단백질을 2차원 전기영동을 통한 분석 결과, 이들 단백질들은 등전점의 항상성을 나타내었으며, 활성 독성물질과 비활성 독성물질간에 분자량의 차이를 보여 주었다. 이러한 결과는 이들 물질의 활성화, 기작이 균주간에 일정함을 시사한다. Entomopathogenic Bacillus sphaericus(BS) strains produce a 43kD entomocidal protein, converted through activation to a 40kD toxin. Two-dimensional electrophoresis of these proteins from 4 entomopathogenic BS strains reveals constance in pI and molecular weight differences between activated and non-activated toxin. This suggests a common activation mechanism.

        Hopfield like neural network is presented which performs optimization subject to bound constraints on each of the optimization variables. It is shown to be globally convergent, and conditions on the quadratic problem and the network parameter are established under which exponential asymptotic stability is achieved. We can choose a suitable network parameter and the system of differential equations governing the network activation is preconditioned in order to reduce its sensitivity to noise and round-off error. Condition number is used to check a instability of system parameter which is concern to network local state.

        Trichoderma 속의 염색체에 관한 연구

        The life cycle of 17 species of Trichoderma was elucidated to seize the proper stage for observing the nuclear behavior and chromosome count. The most convenient stage for the purpose in their life cycle was the stage just before producing the asexual spore. Of the 17 species in the genus Trichoderma the haploid chromosome numbers were counted 5, 6, 7 and 10. Six chromosomes were most frequently observed. It is believed that the basic chromosome number is placed between 4 and 10, and that the number might be 6, referring to the related papers. It appears necessary to reclassify the single genus of Trichoderma into at least two or three genera.
