Students who speak languages other than English comprise a growing population in U.S. schools. As a result, many general education teachers find English language learners (ELLs) in their mainstream classrooms. However, they are often inadequately prepared to work with culturally and linguistically diverse students, resulting in a need for better ELL preparation of mainstream teachers (de Jong & Harper, 2005). Teacher self-efficacy beliefs (TSEB), or teacher perceptions of their confidence and preparedness, can provide important insights relevant to teacher professional development and preparation in teacher education programs. TSEB is defined as a self-assessment of the competence to perform a specific task within a certain context, or a judgement of the ability to perform a desired activity (Tschannen-Moran & Hoy, 2001). Recognizing the importance of investigating TSEB, this dissertation examines elementary preservice teachers and their preparation to work with ELLs through microteaching activities. Two research questions guided this study: 1) What are the self-efficacy beliefs of preservice teachers about teaching ELLs before and after microteaching experiences, and 2) how do four preservice teachers construct their self-efficacy beliefs regarding teaching ELLs through microteaching experiences? The data—consisting of surveys (n=55), individual interviews with preservice teachers (n=4), classroom observations and lesson plans—were analyzed with the use of thematic analysis (Braun & Clarke, 2006). Findings indicated that the self-efficacy beliefs of preservice teachers were enhanced after having participated in ESL microteaching experiences. However, the data also revealed that the preservice teachers still felt some anxiety and emotional concerns about teaching ELLs. Moreover, their self-efficacy beliefs varied by ELL characteristics (situational factors) and setting (contextual factors). Results further revealed that three factors shaped the construction of TSEB, namely ELL-related knowledge from course work, field experiences working with ELLs, personal and professional exposure to linguistic and cultural diversity, including their own language learning experiences. Inquiries about the influence of purposeful field placements and ESL microteaching experiences for teacher candidates in settings with culturally and linguistically diverse students can yield valuable insights. Future studies of teacher preparation coursework for preservice teachers that embed linguistically responsive theories and practices can inform teacher candidates as they strive to enhance their preparation and self-efficacy beliefs for ELLs (Lucas & Villegas, 2013).

This study intends to deriving the equation of non-airconditioned room temperature(hereafter referred to as NART) prediction from the correlation analysis of the influence of environmental design factors on the building. The relation between the NART and environmental design factors was analysed by using correlation coefficients. The equation of NART prediction was derived from the regression analysis by experimentation and simulation. In this study, the main results are summarized as follows: The average NART of experiment and simulation were evaluated to 22.0℃ and 22.6℃, respectively. An error of simulation was the 2.7%. The results from correlation analysis was con-elated with the daily average outdoor temperature, soil temperature, solar intensity, relative humidity, and air velocity in order. Con-elation coefficients of the daily average outdoor temperature, NART, and peak air conditioning load were over the 0.75. According to the sensitivity analysis of environmental design factors and air conditioning load in reference model, the influence of air infiltration and window surface ratio were more than outside insultion.
Ethephon 및 Aminoethoxyvinylglycine 처리가 ‘원황’ 배 모의 수출 환경에서의 품질 및 생리장해 발생에 미치는 영향

We investigate the effect of preharvest spray of aminoethoxyvinylglycine (AVG), an inhibitor of ACC synthase activity, on fruit quality of ‘Wonhwang’ pear (Pyrus pyrifolia Nakai). And the effects were compared with ethephon which releasing ethylene to elucidate treatment efficacy of ethylene biosynthesis inhibitor on Asian pears. Chemicals were sprayed on 30 days before harvest with AVG (75 and 150 mg/L) and ethephon (100 mg/L), respectively. Highest flesh firmness during 21 days of shelf-life at 25℃ after 30 days of cold storage was attained in the fruit treated with 150 mg/L AVG (26.5N) when compared with untreated control (16.7N), while the fruits treated with 100 mg/L ethephon completely lost their marketability within 14 days of shelf-life by mass drop of firmness down to 11.6N. Low incidence of physiological disorders including pithiness and core browning were attained at the fruits treated with 150 mg/L AVG when we compared with untreated control. No occurrence of mealiness found in the fruits treated with AVG but ethephon treated fruits showed 36.4% during 14 days of shelf-life. Consequently, AVG spray at the concentration of 150mg/L on Asian pear ‘Wonhwang’ fruit regarded as useful preharvest management approach for keeping quality during marketing period after simulated exportation.
봉지종류 및 괘대시기가 ‘원황’ 및 ‘화산’ 배 과실의 수확기 품질에 미치는 영향

We tested fruits quality and skin coloration of two pear cultivar ‘Wonhwang’ and ‘Whasan’ produced by bagging with different kinds of paper bags in the Cheonan area, Chungnam Province, Korea. The fruits were bagged at 35, 45, 55 or 65 days after full bloom (DAFB) with 3 kinds of double layer paper bag which consisted of grey color, yellow color and newspaper for outer layer. The great extent of light interruption was observed in grey colored outer paperbag among three kinds of paperbag which showed only 0.46% of lowest light transmittance when compared with 43.7% and 40.0% of yellow paper and newspaper, respectively. The development of skin redness (a*) increased with the delay of bagging time in two pear cultivars. But the fruits treated at 65 DAFB showed uneven coloration and excessive development of redness which represented low fruit external appearance value. Uniform flesh firmness was attained at the fruits bagged with grey outer color paperbag regardless of bagging time, although the fruit quality indices including soluble solids and titratable acidity did not change significantly by using different kind of fruit bags and bagging time in two pear cultivar. Based on our results, it was explained that the grey-colored fruit bag had positive effect on the development of skin coloration without any detrimental effect on fruit quality factors in ‘Wonhwang’ and ‘Whasan’ pears.

<P><I>Aster glehni </I>(AG) has been used in cooking and as a medicine to treat various diseases for over hundreds of years in Korea. To speculate the protective effects of AG on skin barrier, we estimated the protein levels of biomarkers related to skin barrier protection in human keratinocytes, HaCaT cells treated with sodium dodecyl sulfate (SDS), or 2,4-dinitrochlorobenzene (DNCB). The protein levels for keratin, involucrin, defensin, tumor necrosis factor alpha (TNF<I>α</I>), peroxisome proliferator-activated receptor delta (PPAR<I>δ</I>), 5′ adenosine monophosphate-activated protein kinase (AMPK), serine palmitoyltransferase long chain base subunit 2 (SPTLC2), and transient receptor potential cation channel subfamily V member 4 (TRPV4) were evaluated using western blotting or immunocytochemistry in HaCaT cells. AG extract increased the protein levels of PPAR<I>δ</I>, phosphorylated AMPK, SPTLC2, keratin, involucrin, and defensin compared to the SDS or DNCB control group. However, TNF<I>α</I> expression increased by SDS or DNCB was decreased with AG extract. The order of action of each regulatory biomarker in AG pathway was identified TRPV4→PPAR<I>δ</I>→AMPK from antagonist and siRNA treatment studies. AG can ameliorate the injury of keratinocytes caused by SDS or DNCB through the sequential regulation of TRPV4→PPAR<I>δ</I>→AMPK pathway.</P>
Construction of Expression Vector for Functional Analysis of Target Protein in Streptomyces sp.

Streptomycetes are gram-positive filamentous bacteria that are well-known for producing a vast array of bioactive compounds, including more than 70 % of commercially important antibiotics. For the research about Streptomyces sp., the protoplast and electroporation transformation method have been the general techniques for the construction of transformants. However, these techniques have low efficiency and are time-consuming. Another option is intergenic conjugation, which is used for DNA transfer using methylation-deficient E. coli as a DNA donor to avoid the methylated- DNA-dependent restriction systems of actinomycetes. This conjugation method has been widely improved and applied to many other actinomycetes. In this research, an effective transformation procedure for the construction of expression vector by using gateway system was established to avoid limit of restriction enzyme site for cloning of target gene based on transconjugation by Escherichia coli ET12567/pUZ8002 with a pSET152 integration vector.
