        An adaptive parameter optimization model and system for sustainable gear dry hobbing in batch production

        Gear hobbing technology is one of the most widely used forming processes of gear teeth. And the development of dry hobbing technologyprovides a solution for realizing productive, economical, and ecological gear production. Since there is no cutting oil for coolingand lubrication in dry hobbing process, the hob tool life, thermal deformation errors of machine tool, and quality of workpiece are sensitiveto the cutting parameters, especially the cutting speed and tip chip thickness. Considering this situation, a dry hobbing parametersoptimization model with the hobbing efficiency as our objective, and the hobbing cost per piece, gear quality, tact time as constraints wasestablished, in which the cutting speed and tip chip thickness were considered as optimal variables and the material of workpiece, coatingof hob, and feed rate were considered comprehensively. An iterative test method is proposed to solve this model. And for the applicationin automated production line, an online adaptive application system was also developed based on SINUMERIK 840D NC system. Theparameters of five different kinds of material gear were optimized by applying this model and system, and the result showed the modeland the system were practical.

        Lmbr1 Expression in Early Embryo Development Stages in White Leghorn and Chinese Silky

        Lmbr1 is regarded as a key gene that controls the digital model formation in early developmental stages of the chicken. However, there are few reports of lmbr1 expression levels and tendencies in 4-toe and 5-toe chicken species. Therefore, the objective of this study was to compare the lmbr1 expression in White Leghorn (4-toe) and Chinese Silky (5-toe). Firstly, total RNA was extracted from 14 different embryonic development stages (HH3 to HH31) in White Leghorn and Chinese Silky. Secondly, dramatic gene expression changes of lmbr1 were monitored by RT-PCR, which indicated a general up-down-up tendency with subtle differences between these two species. Moreover, Q-PCR reactions were performed to quantitate the expression level of lmbr1 in the 14 selected developmental stages. These data demonstrated a first lmbr1 expression peak of 18.68 and 15.32, a lmbr1 expression trough of 6.61 and 1.80, and a second lmbr1 expression peak of 22.33 and 12.48 in White Leghorn and Chinese Silky, respectively. Finally, embryonic in situ hybridization analysis identified that lmbr1 expressed in the ectoderm in HH21, HH23 and HH24 developmental stages in both species.

      • Association of mir-499 and mir-149 Polymorphisms with Cancer Risk in the Chinese Population: Evidence from Published Studies

        Meta-analyses have shown that microRNA polymorphisms have variable effects in different population. Yet, no meta-analysis investigated the association of two common polymorphisms of miRNA, mir-499 rs3746444 polymorphism and mir-149 rs2292832 polymorphism, with cancer risk in the Chinese population. We searched the PubMed, Web of Knowledge, MEDLINE, CNKI databases, as well as Cochrane library, updated on December 31, 2012 for assays regarding cancer risk association with these two common polymorphisms in the present meta-analysis. Odds ratios (ORs) and 95% confidence intervals (95% CIs) were used to explore the strength of associations. The results showed that rs3746444 polymorphism was associated with increased cancer risk (dominant model: GG/AG vs. AA: OR = 1.43, 95% CI: 1.14-1.80; recessive model: GG vs. AG/AA: OR = 1.54, 95% CI: 1.04-2.30; homozygote model: GG vs. AA: OR = 1.69, 95% CI: 1.10-2.60; heterozygote model: AG vs. AA: OR = 1. 35, 95% CI: 1.09-1.67), and rs3746444 was associated with liver cancer in the subgroup of cancer types. For the rs2292832 polymorphism, the results showed no significant risk association in both overall pooled analysis and subgroup of cancer types, smoking status, gender and tea drinking status in the Chinese population. This meta-analysis suggested that the rs3746444 GG genotype is associated with increased cancer risk, especially liver cancer, while the rs2292832 polymorphism showed no association with cancer risk in Chinese.

        Differences in the biological properties of mesenchymal stromal cells from traumatic temporomandibular joint fibrous and bony ankylosis: a comparative study

        The aim of this study was to compare the functional characteristics of mesenchymal stromal cells (MSCs) from a sheep model of traumatic temporomandibular joint (TMJ) fibrous and bony ankylosis. A sheep model of bilateral TMJ trauma-induced fibrous ankylosis on one side and bony ankylosis on the contralateral side was used. MSCs from fibrous ankylosed callus (FAMSCs) or bony ankylosed callus (BA-MSCs) at weeks 1, 2, 4, and 8 after surgery were isolated and cultured. MSCs derived from the bone marrow of the mandibular condyle (BM-MSCs) were used as controls. The MSCs from the different sources were characterized morphologically, phenotypically, and functionally. Adherence and trilineage differentiation potential were presented in the ovine MSCs. These cell populations highly positively expressed MSC-associated specific markers, namely CD29, CD44, and CD166, but lacked CD31 and CD45 expressions. The BA-MSCs had higher clonogenic and proliferative potentials than the FA-MSCs. The BA-MSCs also showed higher osteogenic and chondrogenic potentials, but lower adipogenic capacity than the FA-MSCs. In addition, the BA-MSCs demonstrated higher chondrogenic, but lower osteogenic capacity than the BM-MSCs. Our study suggests that inhibition of the osteogenic and chondrogenic differentiations of MSCs might be a promising strategy for preventing bony ankylosis in the future.

      • Development of a Cloud Model for Improving Detection of Driver Visual Distraction

        Distracted driving is a serious threat to traffic safety as evidenced by traffic accident records. Detection of driver distraction is not straightforward due to the complex nature of driver behaviors. In this paper, we develop a cloud model to detect driver visual distraction effectively and efficiently. The cloud model is a new mathematical representation of linguistic concepts, which implements uncertainty transformation between qualitative concepts and their quantitative values based on mathematical statistics and fuzzy mathematics. The model has three numerical characteristics: Expected Value_ Ex, Entropy_ En and Hyper-entropy_ He. The Ex represents the most suitable point to represent the domain of a concept which has been quantified. The En represents the uncertainty measurement of qualitative concept, reflecting the range of domain space: the larger the entropy, the larger the granularity, the concept is more macro). The He represents the uncertainty measurement of entropy, representing the relationship between randomness and fuzziness. We conducted a driving simulator experiment to obtain data for distracted driving based on manipulation of a navigation system during driving. We chose the standard deviation of steering wheel angles and lateral lane position as the distraction indicators. We drew the receiver operating characteristic curves of the two variables to confirm their effectiveness for detecting driver distraction, and also to determine threshold values for the cloud model. We trained the cloud model using the simulator experiment results. We then tested the cloud model using the new simulator experiment results. The results show that the cloud model can detect driver visual distraction effectively and efficiently because the model extracts three numerical characteristics from one variable, and therefore enables us to use fewer variables for reliable detection while avoiding possible conflicts among many variables.

        Upregulation of PITX2 Promotes Letrozole Resistance Via Transcriptional Activation of IFITM1 Signaling in Breast Cancer Cells

        Purpose Although the interferon  (IFN) signaling and the paired-like homeodomain transcription factor 2 (PITX2) have both been implicated in the progression of breast cancer (BCa), it remains obscure whether these two pathways act in a coordinated manner. We therefore aimed to elucidate the expression and function of PITX2 during the pathogenesis of endocrine resistance in BCa. Materials and Methods PITX2 expression was assessed in BCa tissues using quantitative reverse transcription polymerase chain reaction (RT-qPCR) and immunohistochemistry and in experimentally induced letrozole-resistant BCa cells using RT-qPCR and immunoblotting. Effects of PITX2 deregulation on BCa progression was determined by assessing MTT, apoptosis and xenograft model. Finally, using multiple assays, the transcriptional regulation of interferon-inducible transmembrane protein 1 (IFITM1) by PITX2 was studied at both molecular and functional levels. Results PITX2 expression was induced in letrozole-resistant BCa tissues and cells, and PITX2 induction by IFN signaling powerfully protected BCa cells against letrozole insult and potentiated letrozole-resistance. Mechanistically, PITX2 enhanced IFN-induced AKT activation by transactivating the transcription of IFITM1, thus rendering BCa cells unresponsive to letrozoleelicited cell death. Additionally, ablation of IFITM1 expression using siRNA substantially abolished IFN-elicited AKT phosphorylation, even in the presence of PITX2 overexpression, thus sensitizing BCa cells to letrozole treatment. Conclusion These results demonstrate that constitutive upregulation of PITX2/IFITM1 cascade is an intrinsic adaptive mechanism during the pathogenesis of letrozole-resistance, and modulation of PITX2/IFITM1 level using different genetic and pharmacological means would thus have a novel therapeutic potential against letrozole resistance in BCa.
