완두(Pisum sativum L.) 葉肉細胞 原形質體의 分雜 및 融合

완두葉肉細胞의 原形質體分離 및 融合에 있어서 酵素의 處理時間, CaCl₂·2H₂O의 效果, 酸素의 濃度와 葉의 位置에 따른 原形質體의 分離에 미치는 影響과 Mole 濃度에 따른 遠心分離效果, PEG溶液의 處理時間, 酸度에 따른 融合效果 그리고 dimethyl sulfoxide 添加效果를 究明하기 위하여 實施한 實驗의 結果는 다음과 같다. 1. 原形質體의 分離에는 4時間의 酸素處理가 가장 良好했고 CaCl₂·2H₂O의 添加에 따라 原形質體의 分離速度가 늦어졌으며 活性度는 4時間까지 變動이 없었고 CaCl₂·2H₂O에 의해서 滑性度가 長期間 維持되었다. 2. 酸素의 濃度는 1%이상의 處理에서 거의 同一한 原形質體分離率을 나타냈으며 上位 1∼2葉이 가장 좋은 原形質體分離率을 보였으며 遠心分離效果는 0.4M, 0.5M에서 良好하였다. 3. 原形質體融合은 PEG 6000溶液 75分 處理에서 좋은 融合率을 보였다. 그러나 30分 以上의 處理가 活性度에 影響을 미치지 않았으며 pH 5.8인 PEG 6000의 0.2M溶液에서 가장 높은 62.84%의 融合率을 나타내었다. dimethyl sulfoxide에 의한 效果는 融合率이 3∼7% 減少하지만 binucleate의 誘道에 有效한 것으로 推定된다. This experiment was conducted to identify the enzyme treatment time, calcium ion effect, enzyme concentration and leaf position for protoplast isolation. It was also performed to determine the adequate molarity on protoplasts, and to investigate the incubation time, pH, PEG concentration and DMSO effect for protoplast fusion. The results obtained were summarized as follows; The optimal time of incubation in enzyme solution was 4 hours. And the protoplast releasing time was delayed by CaCl₂·2H₂O addition to the enzyme solution compared with no added one. The viability had kept up to above 95% until the 4 hours after digestion. The high viability of the protoplast was preserved more than 16 hours by adding CaCl₂·2H₂O to digestion solution. The enzyme concentration had no effect on protoplast yield in range from 1% to 5% and the first or second leaf from the top of the plant produced the highest protoplast yield among the leaf position tested. The purity of healthy protoplast was better in 0.4M and 0.5M sucrose than in others, and the percentage of protoplast aggregation was more 20% to 50% in PEG 6,000 compared with 4,000 and PEG 1,500. Even though the percentage of protoplast aggregation was less increased by 3% to 7% than without DMSO, its treatment was effective to induce binucleated protoplasts.

This study was conducted to establish a rapid analysis method for determining protein and moisture contents of pea. Ninety and eighty pea (Pisum sativum L.) lines were analyzed to determine protein and moisture contents, respectively using near-infrared reflectance spectroscopy. Simple correlations (~gamma ) of protein content in a ground sample and an intact grain sample by an automatic regression method were 0.978 and 0.910, respectively. Simple correlations by partial least square regression/principal component analysis (PLS/PCA) methods were 0.982 and 0.925, respectively. Standard error of performance (SEP) in protein content was the lowest value, 0.446 in ground sample by PLS/PCA methods. Simple correlation of moisture content was the highest at 0.871 in ground samples. when using a standard regression method. Accuracy for the moisture content was slightly lower than for protein content. It was concluded that the NIRS method would be applicable only for rapid determination of protein content in pea.

“Bora” is a new perilla (Perilla frutescens (L.) Britton) cultivar developed from a cross between Ipdeulkkae1 and YCPL199 at National Yeongnam Agricultural Experiment Station, RDA, in 2001. Purple backside leaf color is one of the most important characteristics for the high quality perilla leaf. “Bora” has purple backside leaf color in all climate condition, while a check variety, Ipdeulkkae1 has too light color during summer season or in high temperature condition. “Bora” matures at October 18th and has 79 cm of the stem length, shorter than that of Ipdeulkkae1. Although leaf yield is 11% less than that of a check variety Ipdeulkkae1, the harvested leaf number of “Bora” is similar with that of Ipdeulkkae1. The cultivar is appropriable in all area of Korea for the leaf harvesting cultivation whereas it is restricted to southern part of Korea South Gyeongsang and South Jeola provinces, for the seed harvesting cultivation.

Designing of the squirrel cage AC Traction Motor has many difficulties which has to be small size in order to be suitable into bogie frame, high efficiency and light weight. It means that induction motor for tractive purpose has to be different magnetic and electric loading ratio from industrial induction motor. This paper is devoted to an examination of how this ratio affects overall design concept and hence the main design points for traction motor. Also studied is the changed coefficients of the magnetic and electric loading ratio squirrel cage induction motor for the traction purpose which has been already identified from the reference book for industrial purpose induction motor.
