        Effect of Run Ze Tonic in Mice with Kidney-Yin Deficiency

        목적: 본 연구는 Run Ze Tonic의 신장-음 결핍 마우스의 갑상선 호르몬, 부신 기능, 성 호르몬 및 주기적 뉴클레오티드에 미치는 영향을 관찰하고자 한다. 방법: ICR 마우스를 정상 대조군, 모델 그룹, Run Ze Tonic 고용량 그룹, 중간 용량 그룹 및 저용량 그룹으로 분류하였으며, 매 그룹은 20마리씩 구성하였다. 신장-음 결핍 마우스 모델은 연속 10 일 동안 1 일 1 회 레보 티록신 나트륨을 위내 직접 투여에 의해 확립되었다. 각 그룹의 마우스의 일반특징 및 체중을 관찰하고 장기지수를 측정하였다. 그리고 ELISA 방법으로 테트라 요오 도티 로닌 (T4), 트리 요오 도티 로닌 (T3), 갑상선 호르몬 (TSH), 코르티코 트로 핀 방출 호르몬 (CRH), 부 신피질 자극 호르몬 (ACTH), 코르티 코스 테론 (CORT), 테스토스테론 (T), 에스트라 디올 (E2), 고리 형 아데노신 모노 포스페이트 (cAMP) , 사 이 클릭 구아노 신 모노 포스페이트 (cGMP)를 측정하였다. 결과: Run Ze Tonic 고용량군은 신장-음결핍 마우스의 일반적인 신체 징후를 개선하고, 체중을 증가시키고(p<0.05), TSH 함량을 증가시킨다(p<0.01). 고용량 군에서 T3 및 T4 수치가 유의하게 감소하 였으며(p<0.01), ACTH 수준도 유의하게 감소하였다(p<0.05). 신장 음 결핍 암컷 마우스는 고용량 군에서 성 호르몬 E2 및 E2/T 비 율이 유의하게 감소(p<0.01)하였다. 신장 음이 결핍 된 수컷 마우스의 경우 고용량 군에서 성 호르몬 T함량이 유의하게 증가되었다 (p<0.05). 그리고 고용량 군에서 고리 형 뉴클레오티드 시스템의 cAMP 수치가 감소하였다(p<0.01). 결론: Run Ze Tonic은 내분비 계에서 호르몬 수치 장애, 부신 피질 기능 및 에너지 대사 균형을 조절하여 음양을 키우고 신장을 강화하는 역할을 할 수 있다고 사료된다. Purpose: To observe the effects of Run Ze Tonic on thyroid hormone, adrenal function, sex hormones and cyclic nucleotide of mice with kidney-yin deficiency. Methods: ICR mice were randomly grouped into normal control group, model group, Run Ze Tonic high dose group, the middle dose group and low dose group, male and female in half. The kidney-yin deficiency mice models were established by intragastric administration of levothyroxine sodium before intragastric administration of corresponding drugs once a day for 10 consecutive days. One day after the last administration, the effects were observed by measuring weight. And tetraiodothyronine (T4), triiodothyronine (T3), thyrotropin (TSH), corticotropin releasing hormone (CRH), adrenocorticotropic hormone (ACTH), corticosterone (CORT), testosterone (T), estradiol (E2), cyclic adenosine monophosphate (cAMP), cyclic guanosine monophosphate (cGMP) were tested by ELISA method. Results: Compared with the model group, the body weight, TSH level increased significantly (p <0.01, p<0.05), and T3, T4, ACTH level decreased significantly in high dose group (p <0.01, p <0.05). The E2 level in female decreased significantly in high dose group (p <0.01), the same as the ratio of E2/T (p <0.01). The T level in male increased significantly in high dose group (p <0.01), the same as the ratio of E2/T (p <0.01). And cAMP level decreased in the high dose group(p <0.05). Conclusion: Run Ze Tonic may play the role of nourishing yin and strengthening kidney by adjusting hormone level disorder, adrenal cortex function and balancing energy metabolism in endocrine system.

        A High-Throughput Method Based on Microculture Technology for Screening of High-Yield Strains of Tylosin-Producing Streptomyces fradiae

        Tylosin is a potent veterinary macrolide antibiotic produced by the fermentation of Streptomyces fradiae; however, it is necessary to modify S. fradiae strains to improve tylosin production. In this study, we established a high-throughput, 24-well plate screening method for identifying S. fradiae strains that produce increased yields of tylosin. Additionally, we constructed mutant libraries of S. fradiae via ultraviolet (UV) irradiation and/or sodium nitrite mutagenesis. A primary screening of the libraries in 24-well plates and UV spectrophotometry identified S. fradiae mutants producing increased yields of tylosin. Mutants with tylosin yield 10% higher than the wild-type strain were inoculated into shake flasks, and the tylosin concentrations produced were determined by highperformance liquid chromatography (HPLC). Joint (UV irradiation and sodium nitrite) mutagenesis resulted in higher yields of mutants with enhanced tylosin production. Finally, 10 mutants showing higher tylosin yield were re-screened in shake flasks. The yield of tylosin A by strains UN-C183 (6767.64 ± 82.43 μg/ml) and UN-C137 (6889.72 ± 70.25 μg/ml) was significantly higher than that of the wild-type strain (6617.99 ± 22.67 μg/ml). These mutant strains will form the basis for further strain breeding in tylosin production.

        Selection of inspection path optimization scheme based on analytic hierarchy process and inspection experimental study

        This paper mainly studies the optimization of the patrol route of a fire inspection robot, based on analytic hierarchy process (AHP) and fire source detection. First, the A* algorithm is improved based on two aspects of the heuristic function and of the obstacle boundary setting, while the suboptimal path is obtained in MATLAB. Next, the path planned according to the improved A* algorithm is smoothed and optimized, by means of gradient descent method, Bezier curve and B-spline curve, while the index parameters are optimized by means of MATLAB simulation. In view of the simulation results, the trajectory optimization performance index evaluation system, established by five decision criteria, including running time, path length, ride comfort, no-collision effect and quadratic optimization space, is put forward. The three kinds of optimization methods are analyzed qualitatively and quantitatively, and the results show that, in the total hierarchical ranking, the B-spline curve trajectory optimization scheme has the largest weight and is more important than the other two schemes. Finally, the superiority of B-spline curve is verified experimentally.

        Bias Adjustment of Satellite Precipitation Estimation Using Ground-Based Observation: Mei-Yu Front Case Studies in Taiwan

        The Global Satellite Mapping of Precipitation (GSMaP) was used to estimate the accumulated rainfall in May from the Mei-Yu front in Taiwan. Rainfall estimation from GSMaP during 2002–2017 were evaluated using more than 400 local gauge observations, collected from the Taiwan CentralWeather Bureau (CWB). Studies have demonstrated that the GSMaP rainfall estimation estimates can be biased, depending on the target region, elevation, and season. In this experiment, we have evaluated GSMaP over three elevation ranges. The GSMaP systemic errors for each elevation range were identified and corrected using regression analysis. The results indicated that GSMaP estimation can be improved significantly through adjustment over three elevation ranges (elevation less than 50 m, elevation of 50–100 m, and elevation higher than 100 m). For these three elevation ranges, the correlation coefficient between the GSMaP estimations and CWB rainfall data was 0.76, 0.78, and 0.59, respectively. This indicated that the GSMaP estimation was more accurate for low-elevation regions than high-elevation regions. After the proposed approaches were employed to correct the errors, the bias errors were respectively improved by 5.64(13.7%), 7.33(38.4%) and 10.52(31.2%)mmfor low-, mid- and high-elevation regions. This study demonstrated that the local correction approaches can be used to improve GSMaP estimation of Mei-Yu rainfall in Taiwan.

      • ZnO Nanowire Arrays on 3D Hierachical Graphene Foam: Biomarker Detection of Parkinson’s Disease

        <P>We report that vertically aligned ZnO nanowire arrays (ZnO NWAs) were fabricated on 3D graphene foam (GF) and used to selectively detect uric acid (UA), dopamine (DA), and ascorbic acid (AA) by a differential pulse voltammetry method. The optimized ZnO NWA/GF electrode provided a high surface area and high selectivity with a detection limit of 1 nM for UA and DA. The high selectivity in the oxidation potential was explained by the gap difference between the lowest unoccupied and highest occupied molecular orbitals of a biomolecule for a set of given electrodes. This method was further used to detect UA levels in the serum of patients with Parkinson’s disease (PD). The UA level was 25% lower in PD patients than in healthy individuals.         A detection method of Escherichia coli O157:H7 based on immunomagnetic separation and aptamers-gold nanoparticle probe quenching Rhodamine B’s fluorescence

        This research aimed to detect Escherichia coli O157:H7 in milk based on immunomagnetic probe separation technology and quenching effect of gold nanoparticles to Rhodamine B. Streptavidin-modified magnetic beads (MBs) were combined with biotin-modified antibodies to capture E. coli O157:H7 specifically. Gold nanoparticle (AuNPs) was incubated with sulfhydryl-modified aptamers (SH-Aptamers) to obtain the Aptamers-AuNPs probe. After magnetic beads captured target bacteria and formed a sandwich structure with the gold nanoprobe, Rhodamine B was added into complex to obtain fluorescent signal changes. Our results demonstrated that the established method could detect E. coli O157:H7 in the range of 101–107 CFU/mL, and the limit of detection (LOD) was 0.35 CFU/mL in TBST buffer (pH = 7.4). In milk simulation samples, the LOD of this method was 1.03 CFU/mL. Our research provides a promising approach on the detection of E. coli O157:H7.

        Highly aligned indium zinc oxide nanowire-based artificial synapses with low-energy consumption

        Emulating neural activities at individual synapse level has recently attracted tremendous attention. Here,we demonstrate design and fabrication of thefirst digitally aligned nanowire (NW)-based three-terminalsynaptic transistor. The highly-aligned and individually-position-controlled long continuous indium zincoxide (IZO) NW arrays were directly printed on a large area with low cost using a specialelectrohydrodynamic nanowire printing (e-NWP) process. The NWs printed by e-NWP with a diameterof 250 nm emulated the structure of nervefibers. The device emulates plasticity of biological synapsesand showed remarkable advantages in energy consumption compared with previous reports. Uniqueband-edge modulation along the NW axial direction underlies makes excellent electrical properties ofthe device. This approach paves the way to easy fabrication of printed-NW-based artificial neuralnetworks.

      • Benzochloroporphyrin Derivative Induced Cytotoxicity and Inhibition of Tumor Recurrence During Photodynamic Therapy for Osteosarcoma

        Photodynamic therapy (PDT) is a promising cancer treatment modality that uses dye-sensitized photooxidation of biologic matter in target tissue. This study explored effects of the photosensitizer BCPD-17 during PDT for osteosarcoma. LM-8 osteosarcoma cells were treated with BCPD-17 and cell viability after laser irradiation was assessed in vitro with the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay. The effects of BCPD-17 during PDT recurrence were then examined on tumor-bearing mice in vivo. BCPD-17 had dosedependent cytotoxic effects on LM-8 osteosarcoma cells after laser irradiation which also had energy-dependent effects on the cells. The rate of local recurrence was reduced when marginal resection of mice tumors was followed by BCPD-17-mediated PDT. Our results indicated BCPD-17-mediated PDT in combination with marginal resection of tumors is a potentially new effective treatment for osteosarcoma.

        Study on preparation of inorganic binder stabilized material with large dosage of phosphogypsum

        With the rapid development of the phosphorus chemical industry, a large amount of phosphogypsum (PG) is discharged. The impurities in PG pollute the surrounding soil, the groundwater and the air in the long-term storage. In this paper, PG as the main raw material is used to study the stable material of inorganic binder in the condition of large dosage of PG. With PG and graded gravel as stabilized materials, and cement and fly ash as binder, materials are mixed to determine the influence of PG ratio and the amount of activator on the mixture. The results show that the mechanical strength of the mixture increased first and then decreased with the increase in the proportion of PG replacing graded gravel in the mixture. The activator significantly improves the properties of the material system and makes a significant contribution to the curing of PG. The best ratio of pretreated PG: cement: fly ash: aggregate is 50:6:5:39. The amount of activator added by the external mixing method is 0.3%, which can make the unconfined compressive strength reach 4.1 MPa after 7d. Finally, water-soluble phosphorus leaching experiment is conducted for all the samples. The results show that phosphorus leaching is so little that no secondary pollution of the environment is caused.
