      • Prognostic Significance of GSTP1, XRCC1 and XRCC3 Polymorphisms in Non-small Cell Lung Cancer Patients

        Aim: Individual differences in chemosensitivity and clinical outcome in non-small cell lung cancer (NSCLC) patients treatment with platinum-based chemotherapy may be due to genetic factors. Our study aimed to investigate the prognostic role of GSTP1, XRCC1 and XRCC3 in NSCLC patients treated with chemotherapy. Methods: A total of 460 cases were consecutively selected from The Affiliated Hospital of Nantong University between Jan. 2003 to Nov. 2006, and all were followed-up until Nov. 2011. Genotyping of GSTP1 Ile105Val, XRCC1 Arg194Trp, XRCC1 Arg399Gln and XRCC3 Thr241Met was conducted by duplex polymerase-chain-reaction with confronting-two-pair primer methods. Results: Patients with GSTP Val/Val exhibited a shorter survival time, and had a 1.89 fold greater risk of death than did those with the IIe/IIe genotype. For XRCC1 Arg194Trp, the variant genotype Trp/Trp was significantly associated with a decreased risk of death from NSCLC when compared with the Arg/Arg. Individuals carrying XRCC1 399Gln/Gln genotype had a longer survival time, with a lowered risk of death from NSCLC. Conclusion: This study indicated that GSTP1 Ile105Val, XRCC1 Arg194Trp and XRCC1Arg399Gln genes have a role in modifying the effect of platinum-based chemotherapy for NSCLC patients in a Chinese population. Our findings provide information for therapeutic decisions for individualized therapy in NSCLC cases.

        Wavelet Multiresolution Complex Network for Analyzing Multivariate Nonlinear Time Series

        <P>Characterizing complicated behavior from time series constitutes a fundamental problem of continuing interest and it has attracted a great deal of attention from a wide variety of fields on account of its significant importance. We in this paper propose a novel wavelet multiresolution complex network (WMCN) for analyzing multivariate nonlinear time series. In particular, we first employ wavelet multiresolution decomposition to obtain the wavelet coefficients series at different resolutions for each time series. We then infer the complex network by regarding each time series as a node and determining the connections in terms of the distance among the feature vectors extracted from wavelet coefficients series. We apply our method to analyze the multivariate nonlinear time series from our oil-water two-phase flow experiment. We construct various wavelet multiresolution complex networks and use the weighted average clustering coefficient and the weighted average shortest path length to characterize the nonlinear dynamical behavior underlying the derived networks. In addition, we calculate the permutation entropy to support the findings from our network analysis. Our results suggest that our method allows characterizing the nonlinear flow behavior underlying the transitions of oil-water flows.</P>

        Collision-avoidance trajectory planning for a virtual kinesthetic feedback system

        This paper proposes the collision-avoidance trajectory design and simulation of a virtual kinesthetic feedback system. The system has two sliders driven in two tracks, respectively, and produces virtual masses when the user holds and moves the system. Trajectory planning considering both the ideal and constrained scenarios in applications is carried out. A collision-avoidance trajectory is designed based on the quintic polynomial, and the influence of the time interval and the switching point is discussed. Simulations were performed to verify the function of the system. The results show that without constraints, the system can produce prescribed virtual masses. The displacements of the slider are affected by the error between the prescribed virtual mass and the real mass of the system. With the constraints of the tracks' lengths, the produced virtual masses are altered from the prescribed. Adjusting the time interval and the switching point could optimize the performance of the system when the geometric parameters of the system are set.

        Isolation and Molecular Characterization of a New CRT Binding Factor Gene from Capsella bursa-pastoris

        A new CRT binding factor (CBF) gene designated Cbcbf25 was cloned from Capsella bursa pastoris, a wild grass, by the rapid amplification of cDNA ends (RACE). The full-length cDNA of Cbcbf25 was 898 bp with a 669 bp open reading frame (ORF) encoding a putative DRE/CRT (LTRE)-binding protein of 223 amino acids. The predicted CbCBF25 protein contained a potential nuclear localization signal (NLS) in its N-terminal region followed by an AP2 DNA-binding motif and a possible acidic activation domain in the C-terminal region. Bioinformatic analysis revealed that Cbcbf25 has a high level of similarity with other CBF genes like cbfl, cbf2, and cbf3 from Arabidopsis thaliana, and Bncbf5, Bncbf7, Bncbfl6, and Bncbfl7 from Brassica napus. A cold acclimation assay showed that Cbcbf25 was expressed immediately after cold triggering, but this expression was transient, suggesting that it concerns cold acclimation. Our study implies that Cbcbf25 is an analogue of other CBF genes and may participate in cold-response, by for example, controlling the expression of cold-regulated genes or increasing the freezing tolerance of plants.

        A New Furostanol Saponin from Asparagus cochinchinensis

        A new furostanol saponin, (25S)-26-O-β-D-glucopyranosyl-5β-furost-20(22)-en-3β, 15β,26-triol-3-O-[α-L-rhamnopyranosyl-(1-4)]-β-D-glucopyranoside, namely, aspacochioside D (1) were isolated from Asparagus cochinchinensis (Lour.) Merr, along with three known saponins, aspacochioside C (2), (25S)-5β-spirostan-3β-yl-O-[O-α-L-rhamnopyranosyl-(1-4)]-β-D-glucopyranoside (3), and pseudoprotoneodioscin (4). The structure of 1 was elucidated on the basis of chemical reactions and spectral analysis (IR, GC, ESI-MS, ^1H-NMR, ^13C-NMR, DEPT, HMBC, HMQC and NOESY). The antiproliferative effects of 1-4 were evaluated in a cytotoxicity assay against the human tumor cell line, A549. Compound 2 (Aspacochioside C) exhibited moderate cytotoxicity against A-549, with an IC_50 value of 3.87 μg/mL.

        iTRAQ-based proteomics analysis of diethylstilbestrol induced dysfunction of testicular gubernaculum development in mouse

        Background Diethylstilbestrol (DES) has been shown to disrupt the morphology and proliferation of gubernaculum testis cells. Objective This study aimed to elucidate the mechanism underlying DES induced damage to gubernaculum testis based on proteome analysis. Results Neonatal mice were exposed to DES or control vehicle and the gonads were collected for isobaric tags for relative and absolute quantitation based proteomics analysis. We found that at the early and late stages of gubernaculum development, treatment with different concentrations of DES upregulated the expression of proteins which were generally involved in several function categories including single-organism process, cellular process, binding, catalytic activity, cell part and organelle. KEGG pathway analysis showed that cardiac muscle contraction, oxidative phosphorylation, calcium signaling and cGMP-PKG signaling pathway were the most dysregulated pathways associated with DES in the late stage of gubernaculum development, while steroid biosynthesis was the only enriched pathway in the early stage of gubernaculum development. Conclusion Proteomics profi ling and functional analysis reveal new clues on how DES interferes with cellular processes and disrupts the development of gubernaculum. These findings have potential applications in the treatment of genitourinary diseases such as cryptorchidism.

        Oxidation behavior of C/C composites with B4C-SiCZrC-ZrB2 coating prepared by infiltration and pyrolysis

        Polycarbosilane, B-Si and B-Si-Zr modification coal tar pitch were used as impregnate agent for infiltration and pyrolysis formation of carbon/carbon (abbreviated as C/C) composites with B4C-SiC-ZrC-ZrB2 coating. The density of the composites was examined to be 1.0 g·cm-3, 1.4 g·cm-3 and 1.8 g·cm-3, respectively. The phase compositions, surface morphologies and element distributions of the coating were analyzed by X-ray diffraction (XRD) scanning electron microscopy (SEM) and energy dispervive spectroscopy (EDS), respectively. The result showed that the two composites with a density of 1.4 g·cm-3 and 1.8 g·cm-3 displayed compact microstructures. The oxidation behavior of the composites was studied by oxidation in air at temperatures from 800 to 1,550 ℃ for 1 h. The composites with a density of 1.8 g·cm-3 exhibited a lower weight loss of 1.8 % after oxidation. B4C-SiC-ZrC-ZrB2 coating was found to provide the best protection by the precipitated B2O3, SiO2 and ZrO2 on the surface of the composites during the oxidation process, which were characterized by self-healing and antioxidation. The C/C composites with B4C-SiC-ZrC-ZrB2 coating performed well at high temperatures with the formation of complex oxides glass film that prevented oxygen from further spreading into the matrix.

        Upregulation of PITX2 Promotes Letrozole Resistance Via Transcriptional Activation of IFITM1 Signaling in Breast Cancer Cells

        Purpose Although the interferon  (IFN) signaling and the paired-like homeodomain transcription factor 2 (PITX2) have both been implicated in the progression of breast cancer (BCa), it remains obscure whether these two pathways act in a coordinated manner. We therefore aimed to elucidate the expression and function of PITX2 during the pathogenesis of endocrine resistance in BCa. Materials and Methods PITX2 expression was assessed in BCa tissues using quantitative reverse transcription polymerase chain reaction (RT-qPCR) and immunohistochemistry and in experimentally induced letrozole-resistant BCa cells using RT-qPCR and immunoblotting. Effects of PITX2 deregulation on BCa progression was determined by assessing MTT, apoptosis and xenograft model. Finally, using multiple assays, the transcriptional regulation of interferon-inducible transmembrane protein 1 (IFITM1) by PITX2 was studied at both molecular and functional levels. Results PITX2 expression was induced in letrozole-resistant BCa tissues and cells, and PITX2 induction by IFN signaling powerfully protected BCa cells against letrozole insult and potentiated letrozole-resistance. Mechanistically, PITX2 enhanced IFN-induced AKT activation by transactivating the transcription of IFITM1, thus rendering BCa cells unresponsive to letrozoleelicited cell death. Additionally, ablation of IFITM1 expression using siRNA substantially abolished IFN-elicited AKT phosphorylation, even in the presence of PITX2 overexpression, thus sensitizing BCa cells to letrozole treatment. Conclusion These results demonstrate that constitutive upregulation of PITX2/IFITM1 cascade is an intrinsic adaptive mechanism during the pathogenesis of letrozole-resistance, and modulation of PITX2/IFITM1 level using different genetic and pharmacological means would thus have a novel therapeutic potential against letrozole resistance in BCa.
