        Modeling and Deep Reinforcement Learning Based Control Parameter Tuning for Voltage Source Converter in a Renewable Energy Generation System

        The fast response and low inertia characteristics of converter-based generation (CBG) lead to a new stability issue that limits renewable energy development. This is due to the fact that, using traditional control theory as basic, there is no unifed and efective way to linearize the electronic device and set the parameters in the existing analysis method. In order to optimize the parameters tuning process of the converter control, a benchmark model is adopted in this paper, and the linearization model is updated by selecting suitable variables and detailed with considering the infuence of resistance of the converter. Based on this model, the stability margin related to the parameters of the system is analyzed. Furthermore, to consider more dynamics and make the converter controller focus on the topologies and scenarios, the dynamic response of a disturbance in the grid is selected as the iteration state to design a controller using Deep Reinforcement Learning (DRL). That is, the cascaded voltage and current control along with droop control are tuned by Deep Deterministic Policy Gradient (DDPG) algorithm in the linearized model. The validation tests are carried out on the nonlinear model in Simulink test and real-time platform. They indicate that the proposed linearization and tuning method provides more accuracy and stability for the power system in various grid conditions.

        Isolation, Culture and Identification of Porcine Skeletal Muscle Satellite Cells

        The objective of this study was to establish the optimum protocol for the isolation and culture of porcine muscle satellite cells. Mononuclear muscle satellite cells are a kind of adult stem cell, which is located between the basal lamina and sarcolemma of muscle fibers and is the primary source of myogenic precursor cells in postnatal muscle. Muscle satellite cells are a useful model to investigate the mechanisms of muscle growth and development. Although the isolation and culture protocols of muscle satellite cells in some species (e.g. mouse) have been established successfully, the culture system for porcine muscle satellite cells is very limited. In this study, we optimized the isolation procedure of porcine muscle satellite cells and elaborated the isolation and culture process in detail. Furthermore, we characterized the porcine muscle satellite cells using the immunofluorecence. Our study provides a reference for the isolation of porcine muscle satellite cells and will be useful for studying the molecular mechanisms in these cells.

      • Genetic Variations in the HIF1A Gene Modulate Response to Adjuvant Chemotherapy after Surgery in Patients with Colorectal Cancer

        Background: Hypoxia-inducible factor $1{\alpha}$ (HIF-$1{\alpha}$) plays an important role in regulating cell survival and angiogenesis, which are critical for tumor growth and metastasis. Genetic variations of HIF1A have been shown to influence the susceptibility to many kinds of human tumors. Increased expression of HIF-$1{\alpha}$ has also been demonstrated to be involved in tumor progression. However, the prognostic value of single nucleotide polymorphisms (SNPs) inthe HIF1A gene remains to be determined in most cancer types, including colorectal cancer (CRC). In this study, we sought to investigate the predictive role of HIF1A SNPs in prognosis of CRC patients and efficacy of chemotherapy. Materials and Methods: We genotyped two functional SNPs in HIF1A gene using the Sequenom iPLEX genotyping system and then assessed their associations with clinicopathological parameters and clinical outcomes of 697 CRC patients receiving radical surgery using Cox logistic regression model and Kaplan Meier curves. Results: Generally, no significant association was found between these 2 SNPs and clinical outcomes of CRC. In stratified analysis of subgroup without adjuvant chemotherapy, patients carrying CT/TT genotypes of rs2057482 exhibited a borderline significant association with better overall survival when compared with those carrying CC genotype [Hazard ratio (HR), 0.47; 95% confidence interval (95% CI): 0.29-0.76; P < 0.01]. Moreover, significant protective effects on CRC outcomes conferred by adjuvant chemotherapy were exclusively observed in patients carrying CC genotype of rs2057482 and in those carrying AC/CC genotype of rs2301113. Conclusions: Genetic variations in HIF1A gene may modulate the efficacy of adjuvant chemotherapy after surgery in CRC patients.

        Copper Phthalocyanine Functionalized Polystyrene Superfine Fibers for the Photodegradation of Methyl Orange

        In this paper, copper phthalocyanine and polystyrene composite superfine fibers were readily prepared by electrospinning technology. After treating these composite fibers with paraformaldehyde in concentrated H2SO4 solution, the copper phthalocyanine molecules were covalently bonded to the polystyrene molecules. Meanwhile, the polystyrene molecules in the fibers were cross-linked to endow these fibers with excellent solvent resistance. The photocatalytic performance of this novel fibrous catalyst was evaluated by photodegradation of methyl orange in the presence of H2O2. The effects of light source, H2O2 dosage, catalyst loading, and temperature on the fiber catalyzed photodegradation of methyl orange were carefully studied. The photodegradation percentage of methyl orange in aqueous solution was up to ~ 97% under optimized reaction conditions. At last, this fibrous catalyst was readily recovered by simple filtration and reused for three times with satisfied photodegradation activities. In all, we have developed a facile way to prepare copper phthalocyanine functionalized polystyrene superfine fibers with excellent photocatalytic performance.

        Effect of weaning age on growth performance, feed efficiency, nutrient digestibility and blood-biochemical parameters in Droughtmaster crossbred beef calves

        Objective: The objective of this study was to determine the effect of weaning age on intake, performance, nutrition metabolism and serum parameters of beef calves. Methods: Sixty Droughtmaster crossbred calves were assigned to 5 groups with 12 calves in each group. The calves in control group remained with the dams till the 22-week age, while the calves weaned at 28 d (4 wk), 42 d (6 wk), 56 d (8 wk), and 70 d (10 wk) of age were sent to group 4 wk, group 6 wk, group 8 wk, and group 10 wk, respectively, and then were fed on milk replacer till the 22-week age. Feed intake and body weight and size were record and blood metabolites were measured. And 24 calves of them (6 in each group) were picked randomly for digestion and metabolism trail. Feed, feces and urine sample were taken and measured. Results: Dry matter intake of calves in group 4 wk was significantly lower than those in the remaining groups from wk 17 to 22 (p<0.05). Feed efficiency of the calves was higher in groups 4 wk and 6 wk than those in groups 8 wk and 10 wk from 11 to 13 wk (p<0.05), and calves had higher feed efficiency in group 4 wk, group 6 wk, and group 8 wk than those in group 10 wk from wk 14 to wk 22. Calves in group 4 wk and 6 wk had lower body weight than group 8 wk and group 10 wk and control group at 10-week age (p<0.05) and 13-week age (p<0.05), and calves in group 6 wk had no significant difference in body weight with control group, group 8 wk and 10 wk (p>0.05) but was higher than that of group 4 wk (p<0.05). Calves in group 6 wk had higher final body weight and total gain than group 4 wk, but no difference of total gain with that of groups 8 wk, 10 wk, and control group. And weaning calves at 6-week age brought higher feed efficiency and average daily gain from wk 14 to wk 22, and higher dry matter and organic matter digestibility at 21 wk. Conclusion: It is concluded that the weaning of calves at 6 weeks of age gave positive results.

        QTLs of Cold Tolerance-Related Traits at the Booting Stage for NIL-RILs in Rice Revealed by SSR

        QTLs for cold tolerance-related traits at the booting stage using balanced population for 1525 recombinant inbred lines of near-isogenic lines (viz. NIL-RILs for BC5F3 and BC5F4 and BC5F5) over 3 years and two locations by backcrossing the strongly cold-tolerant landrace (Kunmingxiaobaigu) and a cold-sensitive cultivar (Towada) was analyzed. In this study, 676 microsatellite markers were employed to identify QTLs conferring cold tolerance at booting stage. Single marker analysis revealed that 12 markers associated with cold tolerance on chromosome 1, 4 and 5. Using a LOD significance threshold of 3.0, compositive interval mapping based on a mixed linear model revealed eight QTLs for 10 cold tolerance-related traits on chromosomes 1, 4, and 5. They were tentatively designated qCTB-1-1, qCTB-4-1, qCTB-4-2, qCTB-4-3, qCTB-4-4, qCTB-4-5, qCTB-4-6, and qCTB-5-1. The marker intervals of them were narrowed to 0.3-6.8 cM. Genetic distances between the peaks of the QTL and nearest markers varied from 0 to 0.04 cM. We were noticed in some traits associated cold tolerance, such as qCTB-1-1 for 5 traits (plant height, panicle exsertion, spike length, blighted grains per spike and spikelet fertility), qCTB-4-1 for 8 traits (plant height, node length under spike, leaf length, leaf width, spike length, full grains per spike, total grains per spike and spikelet fertility), qCTB-4-2 for 3 traits (spike length, full grains per spike and spikelet fertility), qCTB-5-1 for 5 traits (plant height, panicle exsertion, blighted grains per spike, full grains per spike and spikelet fertility). The variance explained by a single QTL ranged from 0.80 to 16.80%. Three QTLs (qCTB-1-1, qCTB-4-1, qCTB-4-2) were detected in two or more trials. Our study sets a foundation for cloning cold-tolerance genes and provides opportunities to understand the mechanism of cold tolerance at the booting stage.
