        Molecular Cloning and Expression of a Cu/Zn-Containing Superoxide Dismutase from Thellungiella halophila

        Superoxide dismutases (SODs) constitute the first line of cellular defense against oxidative stress in plants. SODs generally occur in three different forms with Cu/Zn, Fe, or Mn as prosthetic metals. We cloned the full-length cDNA of the Thellungiella halophila Cu/Zn-SOD gene ThCSD using degenerate RT-PCR and rapid amplification of cDNA ends (RACE). Sequence analysis indicated that the ThCSD gene (GenBank accession number EF405867) had an open reading frame of 456 bp. The deduced 152-amino acid polypeptide had a predicted molecular weight of 15.1 kDa, an estimated pI of 5.4, and a putative Cu/Zn-binding site. Recombinant ThCSD protein was expressed in Escherichia coli and assayed for SOD enzymatic activity in a native polyacrylamide gel. The SOD activity of ThCSD was inactivated by potassium cyanide and hydrogen peroxide but not by sodium azide, confirming that ThCSD is a Cu/Zn-SOD. Northern blotting demonstrated that ThCSD is expressed in roots, stems, and leaves. ThCSD mRNA levels increased by about 30-fold when plants were treated with sodium chloride (NaCl), abscisic acid (ABA), and indole-acetic acid (IAA) and by about 50-fold when treated with UVB light. These results indicate that ThCSD is involved in physiological pathways activated by a variety of environmental conditions.

      • Golgi Phosphoprotein 2 Is a Novel Regulator of IL-12 Production and Macrophage Polarization

        <P>Golgi phosphoprotein 2 (GOLPH2), a widely expressed Golgi type II transmembrane protein, has been implicated in several important physiological and pathological processes, including virus infections, cancer cell proliferation, and metastasis. However, its biological functions and mechanisms, particularly in the immune system, remain highly obscure. In this study, we report the biochemical identification of GOLPH2 from B cell lymphoma culture supernatant and show that the secreted protein could inhibit IL-12 production by dendritic cells (DCs) and IL-12–induced IFN-γ production by activated T cells. Further molecular analysis revealed that GOLPH2’s IL-12–inhibiting activity was mediated through a proximal <I>IL12p35</I> promoter element involving a previously identified transcriptional repressor named GC-binding protein that is induced during phagocytosis of apoptotic cells by macrophages. We subsequently generated global <I>golph2</I> knockout mice, which exhibited little developmental abnormality but were more susceptible to LPS-induced endotoxic shock than were wild-type mice with elevated serum IL-12 levels. Furthermore, we found that GOLPH2 played a regulatory role in macrophage polarization toward the M2 type. A comprehensive analysis of gene expression profiles of activated wild-type and GOLPH2-deficient DCs by RNA sequencing uncovered mechanistic insights into the way GOLPH2 potentially modulates DC function during inflammatory insults. Our functional study of GOLPH2 helps advance the scientific understanding of the biological and pathogenic roles of this novel and intriguing molecule with great potential as a diagnostic and prognostic marker as well as a therapeutic target in many acute and chronic inflammatory disorders.</P>

      • The model analysis of the evolutionary route of rural financial cooperation in China

        Although central government has implemented a series of reforms, development and promotion in the rural financial field since rural reform, under a completely isolated state, it could only cause "performance noncooperation" between the rural finance support system to the rural support policy. This paper has set up an angle balanced model, and the analysis on the model indicated that the entire process growing gradually from nothing of occurrence, development and maturation of the financial coooperation went with the harmonious advantage which the government offering harmoniy service obtained from the financial service offered by its own. Therefore, the reasonable system arrangement could reduce the transaction cost which was brought by the resource supply restraint and the development of the finnancial market, reduce the difference between the individual income and the social efficiency, promote the individual to engage in the coordinative activities ans finally cause the growth of the financial gross.

        Vibration response and parameter influence of TBM cutterhead system under extreme conditions

        A cutterhead endures complex random loads when the TBM tunnels, which leads to structure excessive vibration, due to the extreme working conditions. So anti-vibration design is key in the cutterhead system design. In this study we established a virtual prototype of TBM mainframe system, comprehensively considering the spatial multi-point random excitations, split cutterhead structure and multidirectional support stiffness of the main bearing, etc., based on multi-body system dynamics and virtual prototype technology. For verifying the virtual prototype model, a TBM cutterhead vibration field detection system was constructed, to test the cutterhead accelerations and compare the simulation and measured results. The results show that the acceleration change rule by the two approaches is similar, and the maximum relative error is 55 %, keeping on the same order of magnitude, which illustrates the established model has certain reliability. Accordingly, an orthogonal test scheme for cutterhead radial vibration analysis was designed, to analyze the influence of different parameters on cutterhead vibration, based on the virtual prototype model. As indicated in the analysis, the parameter of cutter force had the greatest influence on cutterhead vibration, and then the pinion support stiffness and cutterhead rotating speed. Last, the cutterhead vibration optimal schemes in different directions were confirmed. The research results and methods can provide reference for TBM system parameters matching.

        A study of synergetic carrier emulsion liquid membrane for the extraction of amoxicillin from aqueous phase using response surface methodology

        Amoxicillin (AMX) is the most commonly used antibiotic in life. AMX was extracted from aqueoussolution by synergetic carrier emulsion liquid membrane (ELM) technology for thefirst time. Themembrane phase of ELM was mainly composed of kerosene and butyl acetate. The carrier has consisted ofAliquat-336 and TOA, the surfactant was Span-80, and the membrane enhancer was liquid paraffin. AndNa2CO3 and NaCl were used as the internal aqueous phase for extraction. Firstly, the basic structure andmechanism of emulsion were characterized and analyzed. And then, the center composite design (CCD)of response surface methodology (RSM) was used to optimize the operating conditions. The optimalvalues were: Aliquat-336 concentration (1.1% v/v), trioctylamine (TOA) concentration (8.6% v/v), andextraction time (15 min), AMX concentration (75 mg/L). A polynomial model wasfitted to predict theextraction yield of AMX. Under the optimal operating conditions, the optimal extraction prediction valuewas 98.58%, the extraction actual value was 98.2%, and the recovery rate of AMX was 75.3%.

        A variational nodal formulation for multi-dimensional unstructured neutron diffusion problems

        A variational nodal method (VNM) with unstructured-mesh is presented for solving steady-state and dynamic neutron diffusion equations. Orthogonal polynomials are employed for spatial discretization, and the stiffness confinement method (SCM) is implemented for temporal discretization. Coordinate transformation relations are derived to map unstructured triangular nodes to a standard node. Methods for constructing triangular prism space trial functions and identifying unique nodes are elaborated. Additionally, the partitioned matrix (PM) and generalized partitioned matrix (GPM) methods are proposed to accelerate the within-group and power iterations. Neutron diffusion problems with different fuel assembly geometries validate the method. With less than 5 pcm eigenvalue (k<sub>eff</sub>) error and 1% relative power error, the accuracy is comparable to reference methods. In addition, a test case based on the kilowatt heat pipe reactor, KRUSTY, is created, simulated, and evaluated to illustrate the method's precision and geometrical flexibility. The Dodds problem with a step transient perturbation proves that the SCM allows for sufficiently accurate power predictions even with a large time-step of approximately 0.1 s. In addition, combining the PM and GPM results in a speedup ratio of 2-3.

        Proteomic Analysis on Exosomes Derived from Patients’ Sera Infected with Echinococcus granulosus

        Cystic echinococcosis (CE), a zoonotic disease caused by Echinococcus granulosus at the larval stage, predominantly develops in the liver and lungs of intermediate hosts and eventually results in organ malfunction or even death. The interaction between E. granulosus and human body is incompletely understood. Exosomes are nanosized particles ubiquitously present in human body fluids. Exosomes carry biomolecules that facilitate communication between cells. To the best of our knowledge, the role of exosomes in patients with CE is not reported. Here, we isolated exosomes from the sera of patients with CE (CE-exo) and healthy donors and subjected them to liquid chromatography-tandem mass spectrometry analysis. Proteomic analysis identified 49 proteins specifically expressed in CE-exo, including 4 proteins of parasitic origin. The most valuable parasitic proteins included tubulin alpha-1C chain and histone H4. And 8 proteins were differentially regulated in CE-exo (fold change>1.5), as analyzed with bioinformatic methods such as annotation and functional enrichment analyses. These findings may improve our understanding about the interaction between E. granulosus and human body, and may contribute to the diagnosis and prevention of CE.

        Current state of research about acupuncture for the treatment of COVID-19: A scoping review

        Background Since the outbreak of coronavirus disease (COVID-19), acupuncture has been widely used in the treatment of COVID-19. The research community has responded rapidly and has already published many research articles about this topic. Methods We searched PubMed, Embase, Cochrane Library as well as CNKI, Wanfang and VIP from January 1, 2020 to July 31, 2021. The dates of publication, language of publication, methodological characteristics and the key findings were analyzed separately. The data are presented as bar graphs, structured tables and figures. Results In this scoping review, 16 research articles were included: 7 case reports, 6 observational studies, 1 review, 1 RCT and 1 nonrandomized clinical trial. The majority of the articles (81.3%) were published by Chinese scholars, 12.5% articles were by scholars in the United States, and 6.3% articles were by scholars in Iran. The included studies reported that acupuncture could alleviate the symptoms of COVID-19 patients, shorten their hospitalization days, and is effective for the elderly. There were no side effects reported. The most frequent acupoints used were LI4, PC6, ST36 and KI3. They reported many obstacles in implementing acupuncture therapy for treating COVID-19 patients. Conclusion Acupuncture has a good effect for the treatment of COVID-19, but high-quality evidence support is still lacking. Coupled with the difficulties that acupuncturists experienced during the process of treatment, the promotion of acupuncture treatment for COVID-19 faces many obstacles.         Mycobacterium tuberculosis PE_PGRS45 (Rv2615c) Promotes Recombinant Mycobacteria Intracellular Survival via Regulation of Innate Immunity, and Inhibition of Cell Apoptosis

        Tuberculosis (TB), a bacterial infectious disease caused by Mycobacterium tuberculosis (M. tuberculosis), is a significant global public health problem. Mycobacterium tuberculosis expresses a unique family of PE_PGRS proteins that have been implicated in pathogenesis. Despite numerous studies, the functions of most PE_PGRS proteins in the pathogenesis of mycobacterium infections remain unclear. PE_PGRS45 (Rv2615c) is only found in pathogenic mycobacteria. In this study, we successfully constructed a recombinant Mycobacterium smegmatis (M. smegmatis) strain which heterologously expresses the PE_PGRS45 protein. We found that overexpression of this cell wall-associated protein enhanced bacterial viability under stress in vitro and cell survival in macrophages. MS_PE_PGRS45 decreased the secretion of pro-inflammatory cytokines such as IL-1β, IL-6, IL-12p40, and TNF-α. We also found that MS_PE_PGRS45 increased the expression of the anti-inflammatory cytokine IL-10 and altered macrophage-mediated immune responses. Furthermore, PE_PGRS45 enhanced the survival rate of M. smegmatis in macrophages by inhibiting cell apoptosis. Collectively, our findings show that PE_PGRS45 is a virulent factor actively involved in the interaction with the host macrophage.

        民间美术是民族文化的一个重要组成部分,具有悠久的历史和丰富的资源。本研究以金秀瑶族自治县A 幼儿园为例着重于探讨瑶族民间美术资源在幼儿园环境创设应用中存在的问题及优化策略。本文首先阐述了金秀瑶族民间美术资源在幼儿园环境设计中的应用现状,并指出了其中存在的问题,进而提出了一系列有效的优化策略。 Folk art is an important component of national culture, with a long history and abundant resources. This study takes Kindergarten A in Jinxiu Yao Autonomous County as an example to focus on exploring the problems and optimization strategies of Yao folk art resources in the application of kindergarten environment creation. This article first elaborates on the current application status of Jinxiu Yao folk art resources in kindergarten environmental design, points out the existing problems, and then proposes a series of effective optimization strategies.
