Background: Korean Red Ginseng (KRG) is known to have antianxiety properties. This study was conducted to investigate the anxiolytic effects of KRG extract (KRGE) during ethanol withdrawal (EW) and the involvement of the mesoamygdaloid dopamine (DA) system in it. Methods: Rats were treated with 3 g/kg/d of ethanol for 28 d, and subjected to 3 d of withdrawal. During EW, KRGE (20 mg/kg/d or 60 mg/kg/d, p.o.) was given to rats once/d for 3 d. Thirty min after the final dose of KRGE, anxiety-like behavior was evaluated in an elevated plus maze (EPM), and plasma corticosterone (CORT) levels were determined by a radioimmunoassay (RIA). In addition, concentrations of DA and 3,4-dihydroxyphenylacetic acid (DOPAC) in the central nucleus of the amygdala (CeA) were also measured by high performance liquid chromatography (HPLC). Results: The EPM test and RIA revealed KRGE inhibited anxiety-like behavior and the over secretion of plasma CORT during EW. Furthermore, the behavioral effect was blocked by a selective DA D2 receptor (D2R) antagonist (eticlopride) but not by a selective DA D1 receptor (D1R) antagonist (SCH23390). HPLC analyses showed KRGE reversed EW-induced decreases of DA and DOPAC in a dose-dependent way. Additionally, Western blotting and real-time polymerase chain reaction (PCR) assays showed that KRGE prevented the EW-induced reductions in tyrosine hydroxylase (TH) protein expression in the CeA and TH mRNA expression in the ventral tegmental area (VTA). Conclusion: These results suggest that KRGE has anxiolytic effects during EW by improving the mesoamygdaloid DA system.

To further enhance the role of connected vehicles with the Human-Machine Interface (HMI) in helping drivers in foggy weather, it is meaningful to optimize the HMI based on the driver’s needs. In order to explore the method of HMI optimization, this paper builds a driving simulation experimental test platform of freeway connected vehicle system, designing two experimental scenarios according to the technical conditions (without HMI or with HMI). After that, the paper uses Markov chain to explore the drivers’ fixation transition to identify the driver’s needs in different sections at different conditions. Besides, combined with the visual trajectory results, the paper provides suggestions for optimizing HMI. The results show the review rate of drivers in each section for the straight upper front area is very high. The highest value occurs in the heavy fog zone, suggesting that HMI can provide more road information. HMI should offer a prompt to the driver before entering the warning zone, and remind the driver of changes in the speed limit before entering the fog area. The prompt module of HMI should shorten the horizontal length. In conclusion, this paper aims to propose a general diagnosis method by diagnosing the self-designed HMI.

The Potentials in Solar Cells for MEH-PPV Derivatives: Molecular Design and Performance Prediction

In this contribution, the polymer poly(2-methoxy-5(2?-ethyl-hexyloxy)-1,4-phenylene vinylene) (MEH-PPV) is researched at the aspect of electronic and structural properties to locate the relationships between structures and the experimental properties by employing DFT and TD-DFT method. The studied properties refer to the geometry features, absorption spectra, frontier molecular orbital properties, hole transport properties, and exciton dissociation ability at polymer/PC61BM interface. In addition, four MEH-PPV derivatives, MEH-PPV substituted with F atoms, are designed and studied through comparing with the properties of MEH-PPV. The results indicate that MEH-PPV has low HOMO levels and large hole transport property, which result in its preferable performance. Besides, the designed polymers are promising candidates as PSC donors due to better exciton dissociation ability and larger theoretical VOC owing to lower highest occupied molecule levels, in special, MEH-PPV-Fc has better hole transport properties than MEH-PPV due to smaller m*.

<P>Rutin is an important indicator for evaluating the quality of buckwheat. In this study, flavonoid biosynthesis was compared between two common cultivars (an original and a high-rutin line) of buckwheat, <I>Fagopyrum esculentum</I> Moench. Transcriptional levels of the main flavonoid biosynthetic genes were analyzed by real-time PCR, and main flavonoid metabolites were detected by high-performance liquid chromatography (HPLC); levels of gene expression varied among organs of the two cultivars. Significantly higher transcription levels of most flavonoid biosynthetic genes, except <I>FeFLS1</I>, were detected in stems of the high-rutin line than in stems of the original line. <I>FeCHI</I> and <I>FeFLS2</I> genes also showed higher expression levels in seeds of the high-rutin cultivar. In contrast, <I>FePAL</I>, <I>FeC4H</I>, <I>Fe4CL1</I>, <I>FeCHS</I>, <I>FeF3H</I>, <I>FeF3</I>′<I>H</I>, <I>FeFLS2</I>, and <I>FeDFR</I> were highly detected in the roots of the original line. The HPLC results indicated 1.73-, 1.62-, and 1.77-fold higher accumulation of rutin (the primary flavonoid compound) in leaves, stems, and mature seeds of the high-rutin cultivar (24.86, 1.46, and 1.36 μg/mg, respectively) compared with the original cultivar (14.40, 0.90, and 0.77 μg/mg, respectively). A total of 46 metabolites were identified from seeds by gas chromatography–time-of-flight mass spectrometry. The metabolite profiles were subjected to principal component analysis (PCA). PCA could clearly differentiate the original and high-rutin cultivars. Immune Effect of Newcastle Disease Virus DNA Vaccine with C3d as a Molecular Adjuvant

Newcastle disease is a serious infectious disease in the poultry industry. The commercial vaccines can only offer limited protection and some of them are expensive and need adjuvants. At present, DNA vaccines are widely used. However, the immune responses induced by DNA vaccines are too slow and low. Here, we constructed the transfer vectors with a different number of C3d as molecular adjuvants (n = 1, 2, 4, or 6), and the vectors were cloned into the optimal eukaryotic expression plasmid (pVAXI-optiF) that expressed the F gene of Newcastle disease virus (NDV), and named pVAXI-F(o)-C3d1, pVAXI -F(o)-C3d2, pVAXI-F(o)-C3d4, and pVAXI-F(o)-C3d6, respectively. Cell transfection test indicated that pVAXI-F(o)-C3d6 showed the highest expression. In vivo immunization showed that the chickens immunized with pVAXI-F(o)-C3d6 intramuscularly induced better immune responses than the chickens immunized with the other plasmids. The protective efficacy of pVAXI-F(o)-C3d6 was 80% after challenge with the highly virulent NDV strain F48E9. The results in this study showed that C3d6 could be used as a molecular adjuvant to quickly induce an effective immune response to control NDV.
