      • Zn₄SnSe_(6):Co^(2+) 단결정의 물성에 관한 연구

        In this paper, author describe the undoped and Co^(2+)-doped Zn₄SnSe_(6) single crystals were grown by the chemical transport reaction(CTR) method. For the crystal growth, the temperature gradient of the CTR furnace was kep a t 700℃ for the source aone and at 820℃ for the growth zone for 7-days. I t was found from the analysis of x-ray diffraction that undoped and Co^(2+)-doped Zn₄SnSe_(6) compounds have a monoclinic structure. The optical absorption spectra obtained near the fundamental absorption edge showed that these compounds have a direct energy gaps.

        LB layers of L-α-DLPC deposited by LB method were deposited onto n-type silicon wafer as Z-type film. Films made up of 8, 16 layers of lipid with long alkyl chain and the thickness of monolayer and multilayers was determined by ellipsometry. It was deposited Ag and Al onto LB layers and silicon wafer for electrode and exhibit high capacitance with low frequency. The MLS capacitor shows very high value of capacitance. The result indicated the lower leakage current and very high saturation value of capacitance was reached within 700-800 ㎊ when the thickness was 275Å.

        Exercise index(EI) was measured on 933 pneumoconiosis patients (865 patients with small rounded opacities and 68 patients with large opacities) and 176 suspected pneumoconiosis patients from Korean anthracitic mines, in order to evaluate the ventilatory impairment of pulmonary function according to the chest radiographic findings of pneumoconiosis. Exercise index was obtained from the maximum breathing capacity (MBC) and the exercise ventilation volume(EV) which were measured by spirometer and two step-test. Classification of pneumoconiosis was based on the international classification of ILO U/C. The results were as follows: 1. The mean value of E1 was significantly increased in pneumoconiosis patients with large opacities(46.0%) than in patients with small rounded opacities(26.5%) and with suspected pneumoconiosis(25.0%). The number of jpatients in substandard EI was 97.1% in patients with large opacities but 49.4% in patients with small rounded opacities and 40.3% in patients with suspected pneumoconiosis. 2. The mean values of EI and the number of patients in substandard EI were markedly increased in pneumoconiosis patients with small rounded opacities and suspected over 50 years of age. However, the number of patients in substandard EI showed no association with age in pneumoconiosis patients with large opacitis. 3. In pneumoconiosis patients with small rounded opacities, the mean values of EI and the number of patients in substandard EI showed significant association with the profusion (category) but no significant association with the type of small rounded opacities. 4. In pneumoconiosis patients with large opacities, the mean values of EI and the number of patients in substandard EI showed no significant difference between patients complicated with and without pulmonary tuberculosis. 5. No significant association of EI and the number of patients in substandard EI with the size of large opacities was observed in pneumoconiosis patients with large opacities, however, the mean values of EI showed increasing tendenccy in category B and C compared to category A.

      • Helicobacter pylori와 대장균의 Shuttle Vector 개발

        In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.

        화학 이론의 전통 과학적 연구

        According to the system of the oriental traditional science, everything has to be harmony and interact with one another. The fundamental notions of the traditional natural philosophy are "do(doctrine)", "taegeuk(the great absolute)", "yin-yang," "five primary substance", "four events", and "eight signs for divination". Taegeuk, especially, explains the formation and the death of all things in the universe through circulation of yin-yang, and shows the balance of nature and universe. Yin-yang, relative ideas produces four events, and four events also is divided into the eight signs for divination. 'four events', 'eight signs for divination' like 'yin-yang' indicates the cycle of nature. The essential notions of the traditional science are not different from those of modern chemistry. Taegeuk which is the origin of all things in the universe means the situation of harmony and balance through ceaseless circulation and connotes dynamic equilibrium on neutralization that chemical reaction get to. A number of opponent concepts such as acid-base, oxidation-reduction, atomic nucleus-electron are given an explanation by ying-yang(the positive and negative) that reaches Taegeuk through chemical bonding or reaction over simple antagonistic relationship. 'Yin-yang', 'four-events', 'eight signs for divination' do not imply a substance or a phenomenon in itself but the properties of them. That is why it is possible to see into various chemical concepts like a relative strength of acid-base, groups of periodic table with traditional scientific philosophy. We have regretfully neglected studies in traditional science. It is time to perform the concrete work connecting theory of traditional science with that of modern science for advisable appreciating of traditional science. Moreover, It is necessary to do research and development of educational programs or teaching-learning materials using the traditional science properly. 동양 과학 철학이나 자연 철학은 태극,음양오행,사상팔괘를 이용해 우주 만물의 운동과 자연의 순환을 설명하는데 있어 그 학문적 우수성을 부인할 수 없으나 현대의 화학식이나 수식이 현상을 미시적 관점에서 구체적으로 설명할 수 있는 것과는 달리 전통 과학이론은 거시적 관점에서 그 전체적 흐름만을 나타낸다는 한계를 가지고 있다. 그럼에도 불구하고 태극,음양오행 그리고 사상팔괘론을 중심으로 한 전통 과학 이론은 현대인들에게 과학에 대한 새로운 시각을 제공함으로써 무분별하고 비윤리적인 과학 기술 활용으로 인한 전 지구적인 문제들을 해결하는데 있어 핵심적인 역할을 수행함은 물론 오늘날의 한국 과학이 한 걸음더 전진하는데 이바지 할 수 있을 것이다. 그러므로 서구 중심의 기존의 과학문명에만 머무를 것이 아니라 동양의 전통 과학 철학을 연구하여 민족의 과학 체계를 수립하고 과학의 토착화를 이루어야 한다. 이를 성공적으로 수행하기 위해서는 전통 과학과 현대 과학 사이에 다리를 놓는 구체적인 작업이 본격적으로 진행되어야 한다. 기본적인 화학 개념들을 전통 과학적 관점에서 재해석 하는 것은 물론이고 더 나아가 전통 과학의 이론을 이용하여 화학 반응 과정을 구체적으로 설명하려는 시도가 필요할 것이다.

      • Helicobacter pylori 편모 유전자의 클로닝 및 염기서열 분석

        A λgt11 expression libary of H. pylori DNA in E. coli Y1090 was screened with flagellin-specific rabbit antiserum for molecular cloning of the flagellin gene of H. pylori. A positive clone, λHPF4, was obtained and the recombinant antigen expressed from λHPF4 was a fusion protein with the molecular weight of 168kd. Sequence analysis of antigen-encoding DNA showed that an open reading frame composed of 1,536 nucleotides encodes a polypepride with a oredicted molecular size of 54kd. This open reading frame did not show the homology with flaA gene encoding 56kd protein of H. pylori and was confirmed as a unique sequence through homoligy searching. Therefore, the cloned antigen is supposed to be the carboxy-terminal region of the other flagellin protein of H. pylori, flaB, with the molecular weight of 58kd.
