        The periplasmic chaperone protein Psg_2795 contributes to the virulence of Pseudomonas savastanoi pv. glycinea: the causal agent of bacterial blight of soybean

        Pseudomonas savastanoi pv. glycinea (Psg, also named P. syringae pv. glycinea and P. amygdali pv. glycinea) is the causative agent of bacterial blight in soybean. The identification of virulence factors is essential for understanding the pathogenesis of Psg. In this study, a mini-Tn5 transposon mutant library of Psg strain PsgNC12 was screened on soybean, and one low-virulent mini-Tn5 mutant, designated as 4573, was identified. Sequence analysis of the 4573-mutant revealed that the mini-Tn5 transposon was inserted in the Psg_2795 gene. Psg_2795 encodes a FimC-domain protein that is highly conserved in Pseudomonas. Further analysis revealed that the mutation and knockout of Psg_2795 results in a reduced virulence phenotype on soybean, decreased motility, weakened bacterial attachment to a glass surface and delayed the population growth within soybean leaves. The phenotype of the 4573-mutant could be complemented nearly to wild-type levels using an intact Psg_2795 gene. Collectively, our results demonstrate that Psg_2795 plays an important role in the virulence, motility, attachment and the population growth of PsgNC12 in soybean. This finding provides a new insight into the function of periplasmic chaperone proteins in a type I pilus and provides reference information for identifying Psg_2795 homologues in P. savastanoi and other bacteria.

        A Fuzzy Identity-Based Signcryption Scheme from Lattices

        Fuzzy identity-based cryptography introduces the threshold structure into identity-based cryptography, changes the receiver of a ciphertext from exact one to dynamic many, makes a cryptographic scheme more efficient and flexible. In this paper, we propose the first fuzzy identity-based signcryption scheme in lattice-based cryptography. Firstly, we give a fuzzy identity-based signcryption scheme that is indistinguishable against chosen plaintext attack under selective identity model. Then we apply Fujisaki-Okamoto method to obtain a fuzzy identity-based signcryption scheme that is indistinguishable against adaptive chosen ciphertext attack under selective identity model. Thirdly, we prove our scheme is existentially unforgeable against chosen message attack under selective identity model. As far as we know, our scheme is the first fuzzy identity-based signcryption scheme that is secure even in the quantum environment.

        PREPARATION OF FOLATE-CONJUGATED BOVINE SERUM ALBUMIN NANOPARTICLES ADSORBING EPIRUBICIN HYDROCHLORIDE

        This work investigated the preparation process of folate-conjugated bovine serum albumin nanoparticles (FA–BSANPs) adsorbing epirubicin hydrochloride (EPI) nanoparticles (FA–EPI–BSANPs), a specific-targeting drug delivery system in cancer chemotherapy. The BSANPs were prepared by desolvation as a drug carrier system and conjugated with folate to produce FA–EPI–BSANPs that specifically target tumors by cross-linking. EPI, an anticancer drug, was adsorbed by this drug carrier system. The influences of six experimental parameters, namely, the adsorption time, FA–BSANPs solution-adsorbed EPI concentration, stirring speed, FA–BSANPs solution pH, the ratio of glutaraldehyde and BSA, and mass ratio of FA–BSANPs to EPI, on the drug loading efficiency (DLR) and drug entrapment efficiency (DER) of FA–EPI–BSANPs were investigated via the single factor method. The results indicated that the optimum operation conditions were obtained with 145.4 nm±0.5 nm MPS, 23.41% DLR and 98.93% DER. The N-hydroxysuccinimide-folate content associated with BSANPs was up to 0.9757% (wt). The DLR and DER of EPI increased with increasing adsorption time, FA–BSANPs solution concentration, and pH value, peaking at 1750 rpm with increasing stirring speed, but decreasing thereafter. The FA–EPI–BSANPs obtained were characterized by laser light scattering, scanning electron microscopy, Fourier transform infrared spectroscopy, differential scanning calorimetry, X-ray diffraction and thermogravimetric analysis. Drug release in vitro was investigated, as well. The characterization results showed that EPI in FA–EPI–BSANPs existed in an amorphous, instead of crystalline state. Most of the EPI was enclosed by FA–BSANPs, and a small amount was adsorbed onto the surface of the FA–BSANPs. The FA–EPI–BSANPs particles are nearly ellipsoidal and significantly affect sustained release. The inhibitory rate of FA–EPI–BSANP was mensurated by MTT method. The inhibitory rate of FA–EPI–BSANPs for SMMC 7721 cell developed with raise of concentration and was higher than other samples. The IC50 values of FA–EPI–BSANPs and EPI were 11.5 μg/mL and 18.8 μg/mL, respectively. The target ability of FA–EPI–BSANP for SMMC 7721 cell was mensurated by fluorescence (FITC) modified albumin techniques. The uptake rate of FA–EPI–BSANPs was higher than samples without folate conjugated, and increased with increased concentration.

        Sweet Dream Liquid Chinese Medicine Ameliorates Learning and Memory Deficit in a Rat Model of Paradoxical Sleep Deprivation through the ERK/CREB Signaling Pathway

        Sweet dream oral liquid (SDOL), a traditional Chinese herbal compound contains 17 traditional Chinese medicines. It has various pharmacological effects, such as improving brain dysfunction and increasing sleeping quality. This study investigated the neuroprotective effect and the underlying mechanisms of SDOL-impaired hippocampus learning and memory-induced paradoxical sleep deprivation (PSD) in rats. Sixty Male Wistar rats were randomly divided into six groups. Before PSD, SDOL treatment group rats were intragastrically administered SDOL for 25 days at dose of 2.1, 4.2, and 8.4mL/kg body weight per day. Normal control group, large platform control group, and PSD groups were treated with normal saline instead of SDOL. After 25 days treatment, PSD and SDOL groups were deprived of paradoxical sleep for 72 h. Then two behavioral studies were conducted to test the spatial learning and memory ability using the open field test and Morris water maze test. Expression of the c-fos, c-jun, cyclic AMP response element binding protein (CREB), extracellular signal-regulated protein kinase (ERK), mitogen-activated protein kinases (MAPK)/ERK kinase (MEK), and p-CREB, p-ERK, and p-MEK in the hippocampus were also assayed by western blot. In this study, PSD decreased the levels of p-CREB, p-ERK, p-MEK, c-fos, and c-jun. However, SDOL treatment increased expressions of these proteins. Our results showed that SDOL improved 72-h PSD-induced cognitive impairment. These affects may be mediated by increasing the contents of c-fos, c-jun, and p-CREB/ERK signaling.

        Astragaloside IV reversed the autophagy and oxidative stress induced by the intestinal microbiota of AIS in mice

        Acute ischaemic stroke (AIS) seriously affects patient quality of life. We explored the role of the intestinal microbiota on oxidative stress and autophagy in stroke, and Astragaloside IV (AS-IV) reversed the changes induced by intestinal microbiota. We determined the characteristics of the intestinal microbiota of AIS and transient ischaemic attack (TIA) patients by 16S sequencing and found that the structure and diversity of the intestinal microbiota in patients with AIS and TIA were significantly different from those in healthy subjects. Specifically, the abundance of genus Bifidobacterium, Megamonas, Blautia, Holdemanella, and Clostridium, content of homocysteine and triglyceride was increased significantly, thus it may be as a potential mechanism of AIS and TIA. Furthermore, germ-free mice were infused intracolonically with fecal supernatants of TIA and AIS with/without feed AS-IV for 12 weeks, and we found that the feces of AIS up-regulated the autophagy markers Beclin-1, light chain 3 (LC3)-II and autophagy-related gene (Atg)12, and the expression of reactive oxygen species (ROS) and NADPH oxidase 2/4 (NOX2/4), malondialdehyde (MDA), however, the expression of total antioxidant capacity (T-AOC) and activity of superoxide dismutase (SOD) and glutathione (GSH) was down-regulated in brain tissue, the content of homocysteine and free fatty acids (FFA) in serum of the mice. Meanwhile, AS-IV could reverse the above phenomenon, however, it does not affect the motor function of mice. AS-IV reversed these changes and it may be a potential drug for AIS therapeutics.

        Aloin protects against chronic alcoholic liver injury via attenuating lipid accumulation, oxidative stress and inflammation in mice

        The present study was designed to investigatethe protective effect of aloin against alcoholic liver disease ina chronic alcohol feeding mouse model. Mice were givenalcohol twice a day by intragastric administration for11 weeks (4.0, 4.7, 5.5 g/kg bw/day for the first 3 weeksrespectively, 6.3 g/kg bw/day for the following 8 weeks). Aloin (10, 30 mg/kg bw) or vehicle was given by gavage tomice after each alcohol administration. Alcohol elevated theserum transaminases alanine aminotransferase, aspartateaminotransferase, total cholesterol and triglyceride levelswhich were significantly attenuated by the co-administrationof aloin (p\0.05). Histopathological observations wereconsistent with these indices. Co-administration of aloinsignificantly suppressed the alcohol-dependent induction ofsterol regulatory element-binding protein-1c expression(p\0.01) and remarkably up-regulated themRNA levels ofAMP-activated protein kinase-a2 (p\0.001). Furthermore,aloin supplementation significantly inhibited the alcoholdependentelevation of malondialdehyde and cytochromeP4502E1 expression (p\0.05), and significantly elevatedsuperoxide dismutase activity (p\0.01). The up-regulationof serum lipopolysaccharide (LPS), hepatic nitric oxide,tumor necrosis factor a, toll-like receptor-4, and myeloiddifferentiation primary response gene 88 were also markedlysuppressed by the co-administration of aloin (p\0.05) inalcohol-treated mice. These results suggest that aloin mayrepresent a novel, protective strategy against chronic alcoholicliver injury by attenuating lipid accumulation, oxidativestress and LPS-induced inflammatory response.

        A hierarchical identity-based broadcast encryption (HIBBE)scheme is an identity-based broadcast encryption (IBBE)scheme in a hierarchical environment. In order to obtain secure HIBBEschemes in the quantum era, we propose an H-IBBE schemebased on the learning with errors problemassumption.Our schemeachieves indistinguishability from random under adaptive chosenplaintextand chosen-identity attacks in the random oracle model.

        Antiviral effects of Bovine antimicrobial peptide against TGEV in vivo and in vitro

        Background: In suckling piglets, transmissible gastroenteritis virus (TGEV) causes lethal diarrhea accompanied by high infection and mortality rates, leading to considerable economic losses. This study explored methods of preventing or inhibiting their production. Bovine antimicrobial peptide-13 (APB-13) has antibacterial, antiviral, and immune functions. Objectives: This study analyzed the efficacy of APB-13 against TGEV through in vivo and in vitro experiments. Methods: The effects of APB-13 toxicity and virus inhibition rate on swine testicular (ST) cells were detected using 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT). The impact of APB-13 on virus replication was examined through the 50% tissue culture infective dose (TCID50). The mRNA and protein levels were investigated by real-time quantitative polymerase chain reaction and western blot (WB). Tissue sections were used to detect intestinal morphological development. Results: The safe and effective concentration range of APB-13 on ST cells ranged from 0 to 62.5 μg/mL, and the highest viral inhibitory rate of APB-13 was 74.1%. The log10TCID50 of 62.5 μg/mL APB-13 was 3.63 lower than that of the virus control. The mRNA and protein expression at 62.5 μg/mL APB-13 was significantly lower than that of the virus control at 24 hpi. Piglets in the APB-13 group showed significantly lower viral shedding than that in the virus control group, and the pathological tissue sections of the jejunum morphology revealed significant differences between the groups. Conclusions: APB-13 exhibited good antiviral effects on TGEV in vivo and in vitro.
