      • A Study on the Functionality of Halophytes from Coast area of Jeollanam-do

        This study was undertaken to improve the perceived value of halophytes and contribute to the activity of the local economy by analyzing the antioxidant activity and mineral content of Suaeda japonica, Tetragonia tetragonoides, and Spergularia marina that grow wild on the mud flats and coastal areas. The total phenolic compound content found in each order is as follows: Suaeda japonica (3.674∼5.989 mg TAE/g dry wt.), Tetragonia tetragonoides (2.515∼4.471 mg TAE/g dry wt.), and Spergularia marina (2.450∼4.067 mg TAE/g dry wt.); DPPH radical scavenging and ABTS cation radical scavenging activities were also recorded in the same order. In the case of minerals, Iron(Fe) was relatively high in Suaeda japonica (960.1∼1,346.7 mg/kg), Manganese(Mn) in Tetragonia tetragonoides (197.1∼574.8 mg/kg), and Zinc(Zn) was relatively high in Spergularia marina (63.19∼216.56 mg/kg). It was confirmed that the mineral content of Suaeda japonica, Tetragonia tetragonoides, and Spergularia marina was significantly higher than that of Chinese cabbage, lettuce, leek, and spinach.

      • Targeting Multiple Myeloma Cell Death with Polyphenol EGCG

        (‐)‐Epigallocatechin 3‐gallate (EGCG) is a potent antioxidant polyphenol in green tea that acts as an anticancer agent via both direct and indirect pathways. Although the relationship between EGCG’s anticancer effects and its antioxidant activity is not fully understood, it is known that EGCG stimulates production of reactive oxygen species (ROS), which induce oxidative stress leading to cell death. In IM9 multiple myeloma cells, EGCG acted in a dose‐ and time‐dependent manner to induce apoptotic cell death. Among the antioxidant enzymes expressed in IM9 cells, levels of peroxiredoxin (Prdx) V were selectively and significantly reduced by EGCG. Moreover, the ROS scavenger NAC completely inhibited EGCG‐induced apoptosis and PrdxV reduction, while overexpression of PrdxV, but not a PrdxVC48S mutant, protected IM9 cells from EGCG‐induced apoptosis. EGCG‐induced reductions in cell viability and PrdxV levels were also observed in primary CD138+ multiple myeloma cells from patients. These results suggest that PrdxV is a key target via which EGCG mediates its anticancer effects.

      • Identification of Redox Sensitive Proteins in Prdx II Deficient Red Blood Cells

        Red blood cells (RBCs) have been studied as models for infectious diseases, various symptoms of anemia, hemolysis, and erythrocyte aging. Although do not directly affect RBCs, other diseases may cause RBC physiological alterations that could be advanced for diagnostic aim or to convince better understanding of a certain pathological pattern. In this study, comparative RBC proteomics between healthy and abnormal conditions involve to promote aging related‐biomarker discovery. Peroxiredoxin II (Prdx II; a typical 2‐Cys Prdx) has been originally isolated from erythrocytes, and its structure and peroxidase activity have been adequately studied. Mice absent to Prdx II proteins had heinz bodies in their peripheral blood, and morphologically aged cells were detected in the dense RBC fractions, which contained markedly higher levels of reactive oxygen species (ROS). In addition, a labeling experiment with the thiol‐modifying reagent biotinylated iodoacetamide (BIAM) in Prdx II‒/‒ mice revealed that a variety of RBC proteins were highly oxidized. To identify oxidation‐sensitive proteins in Prdx II‒/‒ mice, we performed RBC comparative proteome analysis by nano‐UPLC‐MSE shotgun proteomics with relative protein quantitative analysis. We found oxidation‐sensitive 18 membrane and 41 cytosol proteins from 32 and 85 peptides containing cysteine oxidation, and analyzed comparative expression pattern in healthy RBCs of Prdx II+/+ mice (W1), healthy RBCs of Prdx II‒/‒ mice (K1), and abnormal RBCs of Prdx II‒/‒ mice (K2). These proteins belonged to cellular functions related with RBC lifespan maintain, such as cytoskeleton, stress‐induced proteins, amino acid/nucleic acid metabolic enzymes, signal transduction, and molecular transporters. Furthermore, protein networks among identified oxidation sensitive proteins were analyzed to associate with aging consequence. Consequently, we expected that RBC proteome may provide clues to understand redox‐imbalanced diseases.

        Peroxiredoxin V, an atypical thioredoxin peroxidase, is widely expressed in mammalian tissues. In addition, Prdx V is localized in mitochondria, peroxisome, cytosol, and nucleus. Prdx V has been reported to protect a wide range of cellular environments as antioxidant enzyme, and its dysfunctions may be implicated in several diseases, such as cancer, inflammation, and neurodegenerative disease. Identification and relative quantification of proteins affected by Prdx V may help identify novel signaling mechanisms that are important for oxidative stress response. However, the role of Prdx V in the modulation of hypoxia‐related cellular response is not studied yet. In order to examine the function of endogenous Prdx V in hypoxic condition in vivo, we generated a transgenic mouse model with Prdx V siRNA expression controlled by U6 promoter. Of many tissues, the knockdown of Prdx V expression was displayed in kidney, lung, and liver, but not spleen and skin. We conducted on the basis of nano‐UPLC‐MSE proteomic study to identify the Prdx V‐affected protein networks in hypoxic kidneys. In this study, we identified protein networks associated with oxidative stress, fatty acid metabolism, and mitochondrial dysfunction. Our results indicated that Prdx V affected to regulation of kidney homeostasis under hypoxia stress.

      • 産婦人科 患者에서 分離된 Ureaplasma urealyticum의 生物學的 性狀

        Isolation rates of Ureaplasma urealyticum from vaginal swab of 216 patients, 105 prostitutes and 100 college females was investigated. Also, minimal inhibitory concentration of isolated strains about tetracycline and minocycline was studied. The results were summarized as follows; 1. Out of 70 vaginitis patients, isolation rates of Ureaplasma urealyticum and Mycoplasma hominis were 34.3% and 15.7%, respectively. 2. Out of 54 cervicitis patients, isolation rates of Ureplasma urealyticum and Mycoplasma hominis were 37.0% and 11.0%, respectively. 3. Out of 92 other gynecological patients, isolation rates of Ureaplasma urealyticum and mycoplasma hominis were 44.6% and 16.3%, respectively. 4. Out of 105 prostitues, isolation rates of Ureaplasma urealyticum and Mycoplasma hominis were 54.3% and 75.2%, respectively. 5. Out of 100 college students, isolation rates of Ureaplasma urealyticum and Mycoplasma hominis were 23.0% and 7.0% respectively. 6. In the susceptibility patterns of isolated Ureaplasma urealyticum, resistant strains about tetracycline in patients, prostitues and college students were 75.2%, 73.6% and 69.5%, respectively 7. In the susceptibility patterns of isolated Ureaplasma urealyticum, resistant strains about minocycline in patients, prostitues and college students were 91.7%, 87.7% and 73.9%, respectively.

      • 대인ㆍ대내인간 통합적 의사소통 프로그램 개발

        연구는 대인, 대내인간 통합적 의사소통 훈련 프로그램을 개발하기 위하여 실시되었다. 연구목적을 달성하기 위해 먼저 선행연구와 임상경험을 토대로 프로그램을 구성하였다. 구성된 프로그램의 효과는 동신대학교 남·여 대학생 30명을 실험집단과 통제집단으로 구분한 후, 총 24시간(6X4)의 프로그램을 실시하여 사전-사후간의 차이를 비교, 검증하였다. 측정도구는 문제해결에 대한 자기평가검사와 자아존중감검사를 사용하였는데, 결과를 요약하면 다음과 같다. 첫째, 대인간·대내인간 통합적 의사소통 훈련 프로그램이 지각된 문제해결능력에 미치는 효과를 검증한 결과, 유의수준 .001에서 통계적으로 유의미한 차이가 있었다. 둘째, 대내인간 통합적 의사소통 훈련 프로그램이 자아존중감에 미치는 효과를 검증한 결과, 유의수준 .001에서 통계적으로 유의미한 차이가 있었다. 따라서 대인간·대내인간 통합적 의사소통 훈련 프로그램은 문제해결능력과 자아존중감 향상에 효과가 있다고 할 수 있다. This study has a goal to develop of the Integrated(Inter·Intra) Communication Training Program. The Integrated(Inter·Intra) Communication Training Program based on preceding research and clinical experience was developed. The subject of this study were 30 Dongsin university students. They divided into experimental group and control group, and then experienced 24 hours program. to examine the effects of Measurement instruments were Problem Solving Inventory and Self-Esteem Inventory. The results are as follow: 1) The results of The Integrated(Inter·Intra) Communication Training Program at a problem solving ability is revealed on significant difference(p<.001). 2) The results of The Integrated(Inter·Intra) Communication Training Program at increasing of self-esteem is revealed on significant difference(p<.001). For that reason The Integrated(Inter·Intra) Communication Training Program is effective to problem solving ability and increasing of self-esteem.

        Congenital Insensitivity to Pain with Anhidrosis 환아의 치험례

        본 병원에 동통을 느끼지 못하는 것으로 인한 혀나 손가락 깨물기 등이 자가 손상,무한증,반복적 발열등의 임상증상을 보이며 유전성이 없이 CIPA로 내원한 환아에서 보호자에게 세심한 보호와 우발사고 방지에 대한 교육이 무엇보다도 중요하며, 치면세마,불소도포,소와열구전색등의 예방적인 처치와 주기적으로 내원시켜 치과적인 문제를 조기에 해결하는 것이 필요하다고 사료된다. Congenital Insensivity to Pain with Anhidrosis(CIPA) is rare autosomal recessive disorder which is known to be hereditary sensory and autonomic neuropathies(HSAN) typeⅣ. It is predilection for male and observed in early infant. Its clinical features include congenital analgesia, which leads to self-mutilation; inability to sweat, which leads to defective thermoregulation; and mild to moderate mental retardation. Its dental features include self-mutilation by nail peeling, which leads to gingival ulcer, tongue ulcer due to tongue biting, and enamel hypoplasia. Partial anodontia is often observed as well. Seventeen-months-old boy with CIPA was reported. This is the case in which the dental characteristics are described and the dental treatment of patient is discussed.

      • 알루미늄을 첨가한 브레이크 마찰재 개발

        먼저 소형 배합기를 사용하여 특정 방식을 가지고 혼합한 후 일정 성형 조건하에서 브레이크 마찰재 시편을 제작한다. 제조된 브레이크 마찰재 시편을 가지고, 정량적인 마모시험기를 통하여 마모계수를 측정하고 그 마모면을 SEM과 광학현미경을 이용하여 관찰하여 마모 기구를 조사하고, 마찰재의 열전도도 측정, 온도에 따른 TGA, DSC 측정, 분석 등 다양한 열적 특성을 분석하여 열 안정성을 평가하였다. By utilizing small mixer we can make brake frictional materials's specimen with special formulas in the condition of constant pressure and forming processes. The frictional coefficients were measured quantitatively with utilizing tribometer (TE77 reciprocating configuration), the worn surfaces were observed by a SEM (scanning electron microscopy), and the wear mechanism will be investigated. The thermal conductivity of the frictional materials was examined, and, TGA (thermal gravitation analysis), and DSC (differencial scanning calorimeter analysis) were used to study thermal properties and to evaluate the thermal stability of the frictional materials). On the bases of results from these experiments, the quantitative characteristics of frictional materials can be obtained.
