        달맞이꽃(Oenothera odorata Jacquin) 뿌리에서 추출한 항균성분의 특성

        An antibacterial component was isolated from the roots of evening primrose (Oenothera odorata Jacquin). The component was purified by repeated preparative TLC of the chloroform-extract with solvent system of toluene(7): ethyl acetate(4) : formic acid(O.8). NMR spectrum indicated that it is a steroid compound containing radical(-C=C-C=0) and aromatic structures. Its antibacterial activity against strains of the genus Streptococcus was relatively high, the MIC value was 50 according to the Hoechst Manual-345, as compared with the other genera such as Staphylococcus and Escherichia.

      • Profiling of the Major Phenolic Compounds and Their Biosynthesis Genes in <i> Sophora flavescens</i> Aiton

        <P>Sophorae Radix (<I>Sophora flavescens </I>Aiton) has long been used in traditional medicine in East Asia due to the various biological activities of its secondary metabolites. Endogenous contents of phenolic compounds (phenolic acid, flavonol, and isoflavone) and the main bioactive compounds of Sophorae Radix were analyzed based on the qualitative HPLC analysis and evaluated in different organs and at different developmental stages. In total, 11 compounds were detected, and the composition of the roots and aerial parts (leaves, stems, and flowers) was significantly different. trans-Cinnamic acid and<I> p</I>-coumaric acid were observed only in the aerial parts. Large amounts of rutin and maackiain were detected in the roots. Four phenolic acid compounds (benzoic acid, caffeic acid, ferulic acid, and chlorogenic acid) and four flavonol compounds (kaempferol, catechin hydrate, epicatechin, and rutin) were higher in aerial parts than in roots. To identify putative genes involved in phenolic compounds biosynthesis, a total of 41 transcripts were investigated. Expression patterns of these selected genes, as well as the multiple isoforms for the genes, varied by organ and developmental stage, implying that they are involved in the biosynthesis of various phenolic compounds both spatially and temporally.</P>

        Morin Protects Acute Liver Damage by Carbon Tetrachloride ($CCl_4$) in Rat

        The purpose of this study was to investigate possible beneficial effects of morin on $CCl_4$-induced acute hepatotoxicity in rats. Rats received a single dose of $CCl_4$ ($150{\mu}L$/100 g 1:1 in corn oil). Morin treatment (20 mg/kg) was given at 48, 24, and 2 h before $CCl_4$ administration. $CCl_4$ challenge elevated serum alanine transaminase (ALT), aspartate transaminase (AST), and alkaline phosphatase (ALP) levels, but these effects were prevented by the pretreatment of rats with morin. To identify the mechanism of protective activity of morin in $CCl_4$-induced hepatotoxicity in rats, we investigated expressions of tumor necrosis factor alpha (TNF-$\alpha$), interleukin-$1{\beta}$ (IL-$1{\beta}$), interleukin-6 (IL-6), and inducible nitric oxide (iNOS). The expressions of TNF-$\alpha$, IL-$1{\beta}$, IL-6, and iNOS were increased by $CCl_4$ treatment and increased expressions of those were decreased by morin. These findings suggest that morin prevents acute liver damage by inhibiting the production of TNF-$\alpha$, IL-$1{\beta}$, IL-6, and iNOS.

      • Comparative proteomic study between tuberous roots of light orange- and purple-fleshed sweetpotato cultivars

        <P><B>Abstract</B></P> <P>This study compares the differences in proteomes expressed in tuberous roots of a light orange-fleshed sweetpotato (<I>Ipomoea batatas</I> (L.) Lam. cultivar Yulmi) and a purple-fleshed sweetpotato cultivar (Shinjami). More than 370 protein spots were reproducibly detected by two-dimensional gel electrophoresis, in which 35 spots were up-regulated (Yulmi vs. Shinjami) or uniquely expressed (only Yulmi or Shinjami) in either of the two cultivars. Of these 35 protein spots, 23 were expressed in Yulmi and 12 were expressed in Shinjami. These protein spots were analyzed by matrix-assisted laser desorption/ionization-time of flight mass spectrometry and electrospray ionization tandem mass spectrometry. Fifteen proteins in Yulmi and eight proteins in Shinjami were identified from the up-regulated (Yulmi vs. Shinjami) or uniquely expressed (only Yulmi or Shinjami) proteins, respectively. In Yulmi, α-amylase and isomerase precursor–like protein were uniquely expressed or up-regulated and activities of α-amylase, monodehydroascorbate reductase, and dehydroascorbate reductase were higher than in Shinjami. In Shinjami, peroxidase precursor and aldo–keto reductase were uniquely expressed or up-regulated and peroxidase and aldo–keto reductase activities were higher than in Yulmi. PSG-RGH7 uniquely expressed only in Shinjami and the cultivar was evaluated more resistant than Yulmi against the root-knot nematode, <I>Meloidogyne incognita</I> (Kofold and White, 1919) Chitwood 1949 on the basis of shoot and root growth. Egg mass formation was 14.9-fold less in Shinjami than in Yulmi. These results provide important clues that can provide a foundation for sweetpotato proteomics and lead to the characterization of the physiological function of differentially expressed proteins.</P> <P><B>Highlights</B></P> <P>► Proteomics in tuberous roots of two sweetpotato cultivars were compared. ► Over 370 protein spots were reproducibly detected by 2-dimensional electrophoresis. ► Thirty five spots were differentially expressed in either of the two cultivars. ► This study provides new insights to characterize differentially expressed proteins.</P>

        메네트리어병 국내 보고 16예의 임상적 고찰

        Background/Aims: Menetrier's disease is a poorly defined condition that is f unknown origin, characterized by giant folds in the stomach. The histoiogic fentures are foveolar hyperplasia and cystic dilatation of the gland. We presented the characteristic findings of Menetirer's disease in Korea with a review of literatures to understand the Menetirer's disease more precisely, Methods: The sixteen cases of Menetrier's disease was reported in Korea. We analyzed their age, sex, symptoms, signs, laboratory findings and treatments, retrospectively, Results: The average age was 46 years. There were 11 men and 4 women. The most common symptom was epigastric pain (94%). The most common sign were epigastric tenderness (69%) and pretibial pitting edema (63%). Patients were often associated with the hypoalbuminemia (73%). All patients showed hypertrophic folds on either gastrofiberscopy or upper gastrointestinal series, All patients showed foveolar hyperplasia histologically. Three patients were operated to control a massive upper gastrointesinal bleeding. Two patients were operated to control the intractable edema. Two patients were operated to exclude gastrie malignancy, Conclusions: Menetrier's disease showed broad clinical features such as epigastric pain, hypoalbuminemia, massive hematemesis and mimicking gastric malignancy. The giant gastric folds and foveolar hyperplasia were the most commom and important findings in the Menetrier's disease.

        생장조절제에 의한 인삼 ( Panax ginseng C. A. Meyer ) 의 기내 (器內) 화아형성 (花芽形成) 조절

        Ginseng zygotic embryos, seedlings, and excised cotyledonary nodes were cultured on Murashige and Skoog`s(MS) medium, supplemented with 6-benzyladenine(Ba) and gibberellic acid(GA_3) to induce flower buds. As the concentration of nitrogen compounds in MS medium was reduced to half of its strength, the flowering frequency of zygotic embryos increased up to 90%. The optimum concentration of sucrose in the medium for flowering of seedlings was 30-60 g/l. In all cases flower buds were formed on elongated axillary branches from the cotyledonary node, while the apices remained vegetative. When zygotic embryos and excised cotyledonary nodes were cultured on the medium, supplemented with all possible combinations of BA, GA_3, and abscisic acid(ABA) of 5 μM each, flowering was induced on the medium with either BA, BA+GA_3, or BA+GA_3+ABA. However, when seedlings were employed, flowering was induced on the medium with either BA, BA+GA_3, BA+ABA, or BA+GA_3+ABA. In addition, inclusion of 5 μM indole-3-acetic acid(IAA) in the above combinations did not affect flowering. These results suggest that cytokinins, gibberellins, and inhibitors play primary, permissive, and preventive roles, respectively, in the induction of flowering of ginseng.

        Gene expression of MC3T3-E1 osteoblastic cells on titanium and zirconia surface

        PURPOSE This study was performed to define attachment and growth behavior of osteoblast-like cells and evaluate the gene expression on zirconia compared to titanium. MATERIALS AND METHODS MC3T3-E1 cells were cultured on (1) titanium and (2) zirconia discs. The tetrazolium-based colorimetric assay (MTT test) was used for examining the attachment of cells. Cellular morphology was examined by scanning electron microscopy (SEM) and alkaline phosphatase (ALP) activity was measured to evaluate the cell differentiation rate. Mann-Whitney test was used to assess the significance level of the differences between the experimental groups. cDNA microarray was used for comparing the 20215 gene expressions on titanium and zirconia. RESULTS From the MTT assay, there was no significant difference between titanium and zirconia (P>.05). From the SEM image, after 4 hours of culture, cells on both discs were triangular or elongated in shape with formation of filopodia. After 24 hours of culture, cells on both discs were more flattened and well spread compared to 4 hours of culture. From the ALP activity assay, the optical density of E1 cells on titanium was slightly higher than that of E1 cells on zirconia but there was no significant difference (P>.05). Most of the genes related to cell adhesion showed similar expression level between titanium and zirconia. CONCLUSION Zirconia showed comparable biological responses of osteoblast-like cells to titanium for a short time during cell culture period. Most of the genes related to cell adhesion and signal showed similar expression level between titanium and zirconia.

        인삼 ( Panax ginseng C. A. Meyer ) 의 성숙한 배 (胚)로부터 체세포 배발생을 (胚發生) 통한 재분화 및 유식물체의 개화

        Mature zygotic embryos dissected from ginseng(Panax ginseng C.A. Meyer) seeds were cultured on Murashige and Skoog`s (MS) medium containing various concentrations of 2,4-dichlorophenoxyacetic acid(2,4-D) and kinetin. Somatic embryos were induced directly from cotyledonary tissue or from intervening callus. The induction frequency of somatic embryos was up to 55%. Upon transfer to half-strength MS medium supplemented with 1 ㎎/l 6-benzyladenine(BA) and 1 ㎎/l GA_3, most somatic embryos dveloped into plantlets. Over 50% of the plantlets flowered after 4 weeks of culture and then a few bore immature fruits in vitro. Therefore, it is suggested that the juvenility of the ginseng tissue which give rise to somatic embryos does not interfere with in vitro flowering of their regenerated plantlets.

        한식당에서 인지된 한국문화가 한식당 및 한국문화 이미지와 한식당 만족에 미치는 영향에 관한 연구

        The aim of the study is to explore how foreigners recognize Korean culture through Korean food and how it influences the perception of Korean restaurants, culture and satisfaction. As a result of analyzing the data, three factors in the Korean culture of restaurants were obtained: style, flavor and affection. With multiple regression analysis, all three hypotheses were supported. Style was the most important contributor to the image of Korean restaurants and culture, followed by affection and flavor. The image of Korean culture has signifiant influence on satisfaction. Major recommendations of this study can be outlined as follows: First, among style, flavor and affection, style was a major determinant of the image of Korean culture; hence, in order to enhance the image of Korean restaurants and culture, style needs to be emphasized. Second, as the image of Korean culture has significant influence on satisfaction, the images of Korean culture should be well displayed and maintained to boost this satisfaction.
