        Association study of protamine 2 (PRM2) gene polymorphism with male infertility in Chinese Han population

        Protamine 2 (PRM2), an essential nuclear protein expressed in sperm, is known to be involved in the spermatogenesis. Although PRM2 defects have been reported to be involved in male infertility, studies for the relationship between male infertility and PRM2 polymorphisms are inconclusive. With the purpose to determine the association of PRM2 variant with male infertility in Chinese Han population, one single nucleotide polymorphism locus G398C in PRM2 which might play a role in semen quality was selected and the variant frequency was analyzed in 144 idiopathic infertile men (case group) and 111 proven-fertile men (control group) in the study. Three genotypes were discovered in the studied population and statistical analysis showed that the frequencies of GG and GC genotypes of PRM2 G398C were significantly different between the fertile and infertile men (P\0.05) and GC genotype was associated with increased risk of male infertility (OR = 1.795, 95 % CI 1.070–3.013, P = 0.026). Further, the C allele distribution was significantly elevated in infertile group (OR = 1.484, 95 % CI 1.001–2.200, P = 0.049). Moreover, we discovered that sperm motility, progressive motility, sperm DNA integrity as well as nuclear maturity rate of GG genotype presented the highest values and were dramatically different with that of CC genotype (P\0.05). Our results gave the first evidence that PRM2 G398C polymorphism was associated with the pathogenesis of male infertility and its genetic variation was in relation to semen quality in Chinese Han population.

        Cytophaga hutchinsonii chu_2177, encoding the O-antigen ligase, is essential for cellulose degradation

        Cytophaga hutchinsonii can efficiently degrade crystalline cellulose, in which the cell surface cellulases secreted by the type IX secretion system (T9SS) play important roles, but the degradation mechanism remains unclear, and the anchor mechanism of cellulases on the outer membrane in C. hutchinsonii has not been studied. Here, chu_2177 was identified by transposon mutagenesis and was proved to be indispensable for cellulose utilization in C. hutchinsonii. Disruption of chu_2177 resulted in O-antigen deficiency and chu_ 177 could confer O-antigen ligase activity upon an Escherichia coli waal mutant, indicating that chu_2177 encoded the Ontigen ligase. Moreover, deletion of chu_2177 caused defects in cellulose utilization, cell motility, biofilm formation, and stress resistance. Further study showed that the endoglucanase activity was markedly decreased in the outer membrane but was increased in the culture fluid without chu_2177. Western blot proved that endoglucanase CHU_1336 was not located on the outer membrane but was released in the culture fluid of the Δ2177 mutant. Further proteomics analysis showed that many cargo proteins of T9SS were missing in the outer membrane of the Δ2177 mutant. Our study revealed that the deletion of chu_2177 affected the localization of many T9SS cargo proteins including cellulases on the outer membrane of C. hutchinsonii.

        Candidate Genes with Ovulation by Differential Display PCR in Small Tail Han Sheep

        To find the candidate genes concerned with ovulation rate of sheep, Differential Display Reverse Transcription Polymerase Chain Reaction was employed to find the differently expressed cDNA controlling ovulation in the Small Tail Han sheep of polyembryony and in Tan sheep of single birth. Twenty-four primer pairs of three anchored primers and eight arbitrary primers were assembled to amplify the specialized bands from these sheep. Positive cross tests were applied to optimize the ascertainable PCR conditions in which different special bands can be identified by silver strain in one PCR tube. After eliminating the false positive PCR products by Northern hybridization, 24 differential display bands were acquired from the ovary in the Small Tail Han sheep. These EST bands were sequenced and 18 different ESTs were found in which five ESTs had several copies and 13 ESTs had only one copy. Comparing these ESTs with homologous sequences by BLAST in the GenBank, there were six ESTs with known open reading frame (ORF) and function, three ESTs with known ORF and no function, and 9 ESTs without homologous sequence. These ESTs partly represent several genes such as NOS2, tensin, TCRA, CDKN1A, ESR1 and ACTB which express especially in Small Tail Han sheep.

      • Effects of probiotics on intestinal health in high-salt diet (HSD) treated mice

        To observe the effect of probiotics on intestinal health in a model of high-salt diet (HSD, 8%) treated mice for 49 days. Cinical (body weight, colon length and colon weight) and historical changes were evaluated. In addition, colon levels of interleukin-1β, IL-6, IL-18, IL-17A, tumor necrosis factor (TNF)-α were detected by enzyme-linked immunosorbent assay. Relative to the HSD model group, the mice in the probiotics treatment group increased in body weight, colon length, and the ratio of colon weight to length was significantly lower (P<0.05). The DAI index of mice in the intervention group was lower than that in the HSD model group (P<0.05). Histological observation also found that the intervention of probiotics is able to reduce the infiltration of inflammatory cells in colon tissue induced by HSD. The results of Masson staining suggest that the intervention of probiotics also reduces the occurrence of intestinal fibrosis in mice with colitis. Our results suggested that probiotics with an activity to improve intestinal health and reduced inflammation in HSD treated mice.

        Indole-3-acetic acid ameliorates dextran sulfate sodium-induced colitis via the ERK signaling pathway

        Microbiota-derived catabolism of nutrients is closely related to ulcerative colitis (UC). The level of indole-3-acetic acid(IAA), a microbiota-dependent metabolite of tryptophan, was decreased signifi cantly in the feces of UC patients. Thus supplementationwith IAA could be a potential therapeutic method for ameliorating colitis. In this work, the protective eff ect ofsupplementation with IAA on dextran sulfate sodium (DSS)-induced colitis was evaluated, and the underlying mechanismwas elucidated. The results indicated that the administration of IAA signifi cantly relieved DSS-induced weight loss, reducedthe disease activity index (DAI), restored colon length, alleviated intestinal injury, and improved the intestinal tight junctionbarrier. Furthermore, IAA inhibited intestinal infl ammation by reducing the expression of proinfl ammatory cytokines andpromoting the production of IL-10 and TGF-β1. In addition, the ERK signaling pathway is an important mediator of variousphysiological processes including infl ammatory responses and is closely associated with the expression of IL-10. Notably,IAA treatment induced the activation of extracellular signal-regulated kinase (ERK), which is involved in the progressionof colitis, while the ERK inhibitor U0126 attenuated the benefi cial eff ects of IAA. In summary, IAA could attenuate theclinical symptoms of colitis, and the ERK signaling pathway was involved in the underlying mechanism. Supplementationwith IAA could be a potential option for preventing or ameliorating UC.

      • Special Transmission Gear Invalidation Analysis Coupled with Finite Element Method Based on Meshless Local Petrov-Galerkin Method

        Aiming at high nonlinear problem of special transmission gear invalidation analysis, provided a method which based on Meshless local Petrov-Galerkin method coupled with finite element method and method to solve fracture problem of special transmission gear. Simulation calculation has been done to non-involute beveloid gear developed by project team. Calculation result verified efficiency of the simulation method. The method has important meaning to novel gear development and research.

        모바일 환경에서 적용 가능한 SIP기반 인터넷전화(VoIP) 보안 통신 프로토콜 성능 평가

        국내 인터넷전화(VoIP) 서비스는 저렴한 요금, 다양한 부가서비스 제공 등의 이점으로 인해 공공기관, 기업 및 일반 가정에서 도입이 지속적으로 증가하고 있다. 또한 스마트폰 이용 증가로 모바일 환경에서 인터넷 이용 역시 크게 확산될 전망이다. 그러나 VoIP 서비스 확산에 따라 취약성을 이용한 침해사고 우려 또한 증가하고 있다. 특히 도청의 경우에는 개인 사용자 프라이버시 침해는 물론이고 기업의 중요 정보가 유출될 수 있어 VoIP 보안 통신 기술의 적용은 필수적이다. 공공기관 인터넷전화에는 2010년부터 보안통신 기술이 적용되고 있으나, 일반 사용자들 대상으로는 아직 적용되지 못하고 있다. 더욱이 모바일 환경에서 인터넷전화는 스마트폰의 제한된 성능으로 인해, 기존 보안 프로토콜을 적용 가능한지 검증이 필수적이다. 본 논문에서는 기존에 유선단말에 적용 가능한 보안 프로토콜들을 모바일 환경에서 적용가능한지 성능 평가를 통해 비교하였다. 성능평가 결과 표준에서 제시하고 있는 보안 프로토콜들을 모바일 환경에서 적용하였을 때, 사용자가 불편함을 느낄 정도의 문제점은 없는 것으로 나타나 모바일 환경에서 기존 보안 프로토콜을 그대로 사용할 수 있음을 확인하였다. The adoption of VoIP is continuously increasing in public institutions, private enterprises and households due to cheaper cost and various supplementary services. Also, it is expected to spread widely the use of VoIP in mobile environment through the increasing use of smartphone. With the growing concern over the incidents of VoIP service while the VoIP service has become increasingly. Especially eavesdropping, it is possible to invade user privacy and drain the secret of company. So, it is important to adopt the protocols for VoIP secure communication. VoIP security protocols are already adopted in public institutions, but it is not adopted in private enterprises and households. In addition, it is necessary to verify whether the VoIP security protocol could be adopted or not in mobile VoIP due to its limited computing power. This paper compared the VoIP security protocol under fixed network and mobile network through performance evaluation. Finally, we found that it is possible to adopt the VoIP security protocols in mobile network.
