      • Effect of ER signal peptides (KDEL) on expression and function of monoclonal antibody in plant

        Plant genetic engineering led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current animal expression systems. In this study, the expression level of mAbP SO57 between with and without Lys-Asp-Glu-Leu (KDEL) was compared in transgenic plant. PCR and PT-PCR analyse showed stable gene insertion transcription of heavy and light chain genes of mAbP SO57 with or without KDEL in plant, respectively. We validated expression of mAbP SO57 by western blot. Western blot showed the significantly higher expression level of mAbP SO57 with KDEL compared to without KDEL. Flow cytometry (FACS) analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb (mAbM) evidenced similar binding activity to the FcγRI receptor (CD64). High performance liquid chromatography (HPLC) analysed, glycosylation patterns of mAbP SO57 with or without KDEL. The mAbP SO57 with KDEL had glycan profile with both oligomannose type (47.6%) and golgi type (52.4%), while the mAbP SO57 without KDEL had only golgi type (100%) glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest the potential of mAbP SO57 for rabies immunotherapy, regardless of plant specific glycan structures.

        Plant genetic engineering has led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current antibody expression systems. PCR and Reverse Transcription-PCR analyses showed existence of heavy and light chain genes of mAb with or without KEDL and their transcription in plant, respectively. Western blot showed that the expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL. Flow cytometry analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb have similar binding activity to the FcγRI receptor. High-performance liquid chromatography analysis showed that the mAbP SO57 with KDEL had glycan profile with both oligomannose and golgi type, whereas the mAbP SO57 without KDEL had only golgi type glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest that the potential of mAbP SO57 for rabies immunotherapy is regardless of plant specific glycan structures.

        Blue Phosphorescence with High Quantum Efficiency Engaging the Trifluoromethylsulfonyl Group to Iridium Phenylpyridine Complexes

        <P>Incorporation of an electron-withdrawing −SO<SUB>2</SUB>CF<SUB>3</SUB> substituent to cyclometalating C^N-phenylpyridine (ppy) ligand resulted in an expected blue-shifted phosphorescence in the corresponding homoleptic <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB> complex, showing the emission of λ<SUB>em</SUB> = 464 nm at 300 K. One of its heteroleptic derivatives, modified by a pyrazolyl borate LX ligand, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(bor)</B>, exhibited further blue-shifted phosphorescence of λ<SUB>em</SUB> = 460 nm at 300 K. Cyclic voltammograms (CVs) and density-functional theory (DFT) calculations supported the efficacy of the electron-withdrawing capability of the SO<SUB>2</SUB>CF<SUB>3</SUB> substituent lowering HOMO energy and obtained widened bandgaps and resumed blue emissions for all of the iridium complexes studied. The homoleptic complexes of both substituents, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB> and <B>Ir(ppySF)</B><SUB><B>3</B></SUB>, reached the higher quantum yields (<I>Φ</I><SUB>PL</SUB>) of (0.89 and 0.72), respectively. Similarly, emission quantum yields (<I>Φ</I><SUB>PL</SUB>) of the heteroleptic derivatives were reported to be (0.75, 0.83, and 0.87) for <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(acac)</B>, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(bor)</B>, and <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(pic)</B>, respectively. Emission kinetics support the enhanced quantum efficiency when <I>k</I><SUB>r</SUB> and <I>k</I><SUB>nr</SUB> values are compared between <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB> and <B>Ir(ppySF)</B><SUB><B>3</B></SUB>, and both values favorably contribute to attaining a higher quantum efficiency for <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB>. Among solution-processed multilayered devices having an ITO/PEDOT:PSS/TCTA:Ir dopant (10:1, <I>w</I>/<I>w</I>)/TmPyPB/Liq/Al structure, a heteroleptic dopant, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(bor)</B>, exhibited better device performance, reporting an external quantum efficiency (EQE) of 1.14%, current efficiency (CE) of 2.31 cd A<SUP>-1</SUP>, and power efficiency (PE) of 1.21 lm W<SUP>-1</SUP>, together with blue chromaticity of CIE<SUB>x,y</SUB> = (0.16, 0.32).</P><P>Blue phosphorescence of Ir(III) complexes can be tuned with the introducing of strong electron withdrawing substituents and the variation in the ancillary ligands.</P> [FIG OMISSION]</BR>

      • Streptozotocin-당뇨병 백서에서 당뇨병 이환기간에 따른 대동맥의 이완능 저하 및 Oxygen-Free Radical에 의한 이완능의 손상정도

        목적: 당뇨병의 이환기간이 혈관의 내피세포 의존성 및 비의존성 이완능의 저하에 미치는 영향과 이환기간에 따라 oxygen-free radical(OFR)에 의한 동맥이완능의 손상정도에 차이가 있는지를 알아보고자 하였다. 방법: Streptozotocin(STZ)으로 당뇨병이 유도된 3 주 및 10주째의 백서군과 연령이 비슷한 정상 대조백서군의 적출 대동맥환을 norepinephrine으로 수축시킨 상태에서 내피세포 의존성 이완제인 acetylcholine(ACh) 및 비의존성 이완제인 nitroglycerine(NTG)에 대한 이완능을 즉정하여 비교 관찬하였다. 또한 xanthine과 xanthine oxidase의 혼합으로 생성된 H₂O₂에 대동맥환을 노출시킨뒤 역시 ACh과 NTG에 대한 이완능의 손상정도를 비교하였다. 결과: 3주및 10주째 당뇨병 백서의 대동맥환은 모두 대조군 백서의 대동맥환에 비해 ACh에 대한 이완능이 유의하게 감소하였으며 10주째 당뇨병 백서군에서의 ACh에대한 이완능의 저하는 3 주째 당뇨병 백서군에 비해 유의하게 심하였다.H₂O₂에 노출시킨 뒤 ACh에대한 이완능은 3주 및 10주 째 당뇨병 백서군에서 모두 대조군에 비해 유의하게 심한 손상을 보였으나 3주와 10주째 당뇨병 백서군 사이의 손상정도의 차이가 통계학적으로 유의하지는 않았다. 한편 3주 및 10주째 당뇨병 백서군 모두 NTH에 대한 대동맥환의 이완능은 대조 백서군과 별다른 차이가 없었으며 또 H₂O₂에 노출시킨 뒤에도 NTG에 대한 이완능의 손상은 관찰되지 않았다. 결론: STZ-당뇨병백서에서 대동맥의 이완능은 당뇨병에 이환된지 3 주째에 이미 ACh에 대한 내피세포 의존성 이완능이 선택적으로 저하되며 당뇨병의 이환기간이 길수록 이완능의 저하가 심하였고, OFR에 의한 동맥 이완능의 손상도 내피세포 의존성 이완능에만 선택적으로 나타나나 이환기간에 따라 OFR에 의한 이완능 손상정도에는 뚜렷한 차이가 없음을 시사하고 있다. Objectives: Diabetes mellitus produces histological evidence of endothelial as well as changes in vascular function in experimental animal. Oxygen free radicals interfere with of destroy endothelium-dependent relaxation in normal and diabetic blood vessels. The present study was investigated whether the duration of diabetic state can affect the severity of impairment in endothelium-dependent and/or independent relaxation and the sensitivity to oxygen free fadical (OFR)-induced damage in diabetic rat vasculature. Methods: The relaxative responses of aortic ring precontracted with norepinephrine in vitro to cumulative concentrations of acetylcholine (ACh) and nitroglycerine (NTG) were conducted in 3- and 10- week streptozotocin (STZ)-diabetic rats and agematched controls. After exposure to H₂O₂generated by mixing xanthine and xanthine oxidase, the degree of impairment in relaxative responses to ACh and NTG were also compared between 3- and 10-week diabetic rats. Results: The relaxative responses of aortic rings produced by cumulative concentrations of ACh were significantly lower in both 3- and 10-week diabetic rats than in the age-matched controls. The impairments of relaxative responses to ACh in 10-week diabetic rats were significantly greater than those in 3-week ones. After H₂O₂ exposure, the impairments of relaxative responses to ACh in 3- and 10-week diabetic aortas was also significantly greater than those in the control in the control ones. However, the severity of H₂O₂-induced damage in 10-week diabetic aortas was not significantly different than that in 3-week ones. In contrast, aortic rings of 3- and 10-week diabetic rats could still be fully relaxed by NTG, even after exposure to H₂O₂. Conclusion: These findings suggest that in SRZ-diabetic rat aortas selective impairment of endothelium-dependent relaxation to Ach can be found early even over a 3-week period after the induction of diabetes and the impairment may be greater in those with the longer duration of the diabetic state and that the impairment of the vascular relaxation by OFR is also selective to endothelium-dependent response but the sensitivity of OFR-induced damage may not be related to the duration of the diabetic state.

      • Pierre Robin syndrome 환아에서 기관내 삽관 경험 : 증례보고

        Pierre Robin syndrome is characterized by micrognathia, glossoptosis, and cleft palate. Neonates with Pierre Robin are at risk of upper airway obstruction and may require surgical fixation of the tongue to the mandible. Such neonates are at high risk of hypoxia and difficult to intubation during induction of anesthesia. We report a case of an 11-day old neonate with Pierre Robin syndrome was intubated in operative room and emergency room. While under general anaesthesia, awake intubation was attempted and facilitated oxygenation. After a 4 day discharge, he revisited to emergency room due to airway obstruction symptom. Repeated attempts at intubation (again with spontaneous breathing) failed. Finally, laryngeal mask airway (LMA) was introduced, and as a result of this ventilation was achieved. The next day, we awake fiberoptic intubation through the laryngeal mask and were easily achieved.

      • 바이오 휘드백 훈련을 통한 이완요법이 생리통 및 스트레스반응경감에 미치는 영향

        The purpose of this study was to investigate the effects of a 4-week relaxation therapy using biofeedback on menstrual pain and stress for 35 sophomore and junior baccalaureate nursing students. A quasi-experimental pretest-posttest control group design was used. The relaxation therapy group included training sessions using respiration therapy augmented with biofeedback techniques for self-relaxation. A significant reduction of pain (P<.05) on the 1st and 2nd day of menstruation was reported on the visual analogue scale by the experimental groups, while that the control groups remained relatively unchanged. Also a significant reduction of pain (P<.05) was reported on the delivery pain scale by the experimental groups on the 2nd day of menstruation, while that of the control groups remained unchanged. However, there were no significant differences of pain on the 3rd and 4th day of menstruation between the two groups. A significant reduction of state of anxiety (P<0.5) was reported by experimental groups, while that of the control groups remained relatively unchanged. There were no significant changes in stress measured by the symptom of stress. The results of the study support the benefits of relaxation therapy using biofeedback on pain relief and stress.

      • 부유대역 용융응고법으로 제조된 YBa_2Cu_3O_x 산화물초전도체의 미세구조와 전기적 특성

        The YBa_2Cu_3O_x-20 wt% Y_2BaCuO_5 composition designed in YBCO system was synthesized by solid state reaction method. The optimum condition of hot zone temperature and growth rate in floating-zone melt were 1080 ℃ and 2.5 mm/hour, respectively. In samples growth as these condition, the low angel boundaries in grain direction and small amount of amorphous phase known as decreasing characteristics of superconducting materials between grain boundaries in microstructure are investigated by polarized light microscopy on the whole. The critical current density(Jc) at 77k of sample using powder fabricated with no CeO_2 and doped 5 wt%CeO_2 were 8,000 A/㎠(Jc : 561A) and 12,000 A/㎠(Jc: 826A) at 0 T by direct current transport method, respectively.
