      • Comparison of controlled ovarian stimulation outcomes for fertility preservation in women with breast cancer according to KI-67, histologic grade, pathologic type, and cancer stage

        Objective: This study aims to analyze that specific prognostic factors such as Ki-67 value, histologic grade, pathology type, and cancer stage can affect controlled ovarian stimulation outcomes(COS) for fertility preservation in women with breast cancer. Methods: This retrospective cohort study included 92 patients with breast cancer aged 21-44 who underwent COS from August 2012 to April 2020, excluding patients with previous gonadotoxic therapy history. All COS cycles were conducted letrozole-combined random start GnRH antagonist protocol. COS outcomes were compared according to prognostic factors;Ki-67 (< 30% vs. ≥ 30%), histologic grade (low vs. high), and pathologic type (intraductal carcinoma(IDC) vs. triple-negative breast cancer(TNBC)), and cancer stage (early (I, II) vs. advanced (III, IV)). Multivariate analysis was also conducted to find any parameter that can impact over 10 mature oocytes acquisition. Results: Among the 92 patients, 42 were Ki-67≥30% compared to 49 of Ki-67<30%. The number of oocytes, and initial mature oocytes were comparable between Ki-67<30% and ≥30 group. The maturation rate was significantly higher in Ki-67≥30% group than Ki-67<30% group (57.4±36.8% vs. 44.8±28.3%, p=0.041). The same results were drawn when divided according to IDC or TNBC. The maturation rate was significantly higher in TNBC than the IDC group (73.0±17.6% vs. 47.0±28.1% p=0.001). However, the histologic grade or stage did not show any difference in COS outcomes between high and low grade or early and advanced stages. With multivariate analysis, age and AMH are parameters associated with the acquisition of over 10 mature oocytes (OR 0.863, 95% CI [0.755-0.987], OR 1.408, 95% CI [1.145-1.732], respectively). Conclusion: Either Ki-67 or pathologic type can be a potential reliable marker for predicting COS outcome, especially maturation rate. Further study with larger sample size with specific conditions is necessary to clarify the correlation between breast cancer prognostic factors and COS outcomes.

      • Possibility of Empowerment in The Surrendered by Chang-rae Lee

        This paper supports the interpretation that the main characters in Chang-rae Lee’s The Surrendered find liberation in being surrendered. As a novel written by a recognized Korean-American writer about The Korean War, The Surrendered received many reviews concentrating on the author’s writing style and artistry. These early critics stated that the novel’s varying temporal and geographical shifts and the fact that the novel revolves around three different characters make the novel unnecessarily complex. In addition, the scholarly works that do analyze the novel concentrate on the trauma, disability, and post-traumatic stress disorder resulting from the violence. Although prior research touched upon the characters and the symbolism within the setting, I would like to do a more in depth analysis. I use the concept of “abjection” to analyze the symbolism of the three main characters: June, Hector, and Sylvia. All three characters have distinct individuals while also representing others who have survived the social situations they faced. Therefore, it is meaningful to analyze three characters and how they use their status as abjects to find a form of liberation and empowerment.

        The Conjugated Double Bond of Coniferyl Aldehyde Is Essential for Heat Shock Factor 1 Mediated Cytotoprotection

        <P>Coniferyl aldehyde (1) is previously reported as a potent inducer of heat shock factor 1 (HSF1). Here, we further examined the active pharmacophore of 1 for activation of HSF1 using the derivatives coniferyl alcohol (2), 4-hydroxy-3-methoxyphenylpropanal (3), and 4-hydroxy-3-methoxyphenylpropanol (4). Both 1 and 2 resulted in increased survival days after a lethal radiation (IR) dose. The decrease in bone marrow (BM) cellularity and Ki67-positive BM cells by IR was also significantly restored by 1 or 2 in mice. These results suggested that the vinyl moiety of 1 and 2 is necessary for inducing HSF1, which may be useful for developing small molecules for cytoprotection of normal cells against damage by cytotoxic drugs and radiation.</P>

        초등학교 고학년 학생의 장애아동에 대한 인식 조사

        목적 : 초등학교 고학년 학생의 장애아동에 대한 인식을 알아보는 것이다. 연구방법 : 연구 대상은 김해시에 소재한 통합교육을 실시하지 않는 초등학교의 4, 5, 6학년의 비장애 아동이었다. 연구 기간은 2008년 10월 2일부터 10월 17일까지였으며, 설문지를 사용하였다. 설문지의 하위 영역은 사귐, 활 동, 학업 영역의 세 부분으로 각 10문항으로 이루어져 있으며 SPSS 12.0을 사용하여 분석하였다. 결과 : 장애아동에 대한 인식조사 결과 첫째, 성별에 따른 인식은 남학생보다 여학생의 평균 점수가 높았으나 통계 적으로 유의미한 차이는 없었다. 둘째, 학년에 따른 인식은 4학년의 평균 점수가 가장 높았고 6학년이 가장 낮았으 며 통계적으로 유의미한 차이가 있었다(F=3.546, p<0.05). 셋째, 장애인 접촉 유무에 따른 인식은 접촉 경험이 없 는 학생이 있는 학생보다 평균점수가 높았으나 통계적으로 유의미한 차이는 없었다. 결론 : 비장애학생의 장애아동에 대한 인식은 학년에 따라 차이가 있었다. 본 연구는 작업치료사들에게 장애아동에 대한 또래집단의 인식에 대한 정보를 제공함으로써 학교환경에 대한 이해를 도울 수 있을 것이다. Objective : The purpose of this study was to investigate attitude of grade-schoolers toward children with disabilities. Methods : The subjects of this study were grade-schoolers who have not received an inclusive education in Gim-hae. The study was surveyed from October second to 17th in 2008. The survey was categorized into three parts; comradeship, activity, and study. Each part of the survey consists of 10 questions. Data were analyzed by SPSS (version 12.0). Results : The results were as follows; First, the attitude of girls toward disabled students was non-significant compared to boys. Second, the order of attitude degree in grade of students was 4, 6, and 5. The attitude toward children with disabilities showed statistically significant difference according to grade(F=3.546, p<0.05). Third, the attitude of students who have experience of contact with people with disabilities was non-significant compared to non-experienced students. Conclusion : In conclusion, therefore peer groups in lower class levels showed more positive attitude toward the children with disabilities. The result will be useful information for understanding disabled children and making positive attitude. Also it is expected that occupational therapists help disabled children to participate in school life successfully using these information.

      • Fisetin induces apoptosis in uterine leiomyomas through multiple pathways

        Objective: The aim of this work is to discover a potential natural agent that has pharmacological activities on uterine leiomyomas with fewer adverse effects. Methods: In this study, we investigated the potential of a natural medication, Rhus verniciflua Stokes (RVS), as a new therapeutic agent for leiomyomas. After primary cytotoxicity testing, we analyzed the RVS components that showed pharmacological activity. Leiomyoma cells and myometrium cells were cultured from uterine tissues obtained from patients, under the approval of the institutional review board of Kyung Hee University Hospital (IRB No. KUH 2017-11-064), and were treated with RVS at varying concentrations. Results: RVS was cytotoxic in both leiomyoma and myometrium cells; however, the effects were more prominent in the leiomyoma cells. Among the bioactive components of RVS, fisetin showed significant pharmacological effects on leiomyoma cells. Fisetin showed excellent leiomyoma cell cytotoxicity and induced apoptotic cell death with cell cycle arrest. The apoptotic cell death appeared to involve not one specific pathway but multichannel pathways (the intrinsic, extrinsic, MARK, and p53-mediated pathways), and autophagy. The multichannel apoptosis pathways were activated with a low dose of fisetin (< IC20) and were more vigorously activated at high doses ( > IC50). Conclusion: This is the first demonstration to show the pharmacological activities of fisetin on leiomyoma cells. These findings suggest that fisetin may be used for the prevention and treatment of uterine leiomyomas. Since fisetin can be obtained from plants, it may be a safe and effective alternative treatment for uterine leiomyomas.

        Genomic Analysis of the Extremely Halophilic Archaeon Halobacterium noricense CBA1132 Isolated from Solar Salt That Is an Essential Material for Fermented Foods

        The extremely halophilic archaeon Halobacterium noricense is a member of the genus Halobacterium. Strain CBA1132 (= KCCM 43183, JCM 31150) was isolated from solar salt. The genome of strain CBA1132 assembled with 4 contigs, including three rRNA genes, 44 tRNA genes, and 3,208 open reading frames. Strain CBA1132 had nine putative CRISPRs and the genome contained genes encoding metal resistance determinants: copper-translocating P-type ATPase (CtpA), arsenical pump-driving ATPase (ArsA), arsenate reductase (ArsC), and arsenical resistance operon repressor (ArsR). Strain CBA1132 was related to Halobacterium noricense, with 99.2% 16S rRNA gene sequence similarity. Based on the comparative genomic analysis, strain CBA1132 has distinctly evolved; moreover, essential genes related to nitrogen metabolism were only detected in the genome of strain CBA1132 among the reported genomes in the genus Halobacterium. This genome sequence of Halobacterium noricense CBA1132 may be of use in future molecular biological studies.

        Synergistically enhanced osteoconductivity and anti-inflammation of PLGA/β-TCP/Mg(OH)<sub>2</sub> composite for orthopedic applications

        <P><B>Abstract</B></P> <P>Synthetic biodegradable polymers including poly(lactide-<I>co</I>-glycolide) (PLGA) have been widely used as alternatives to metallic implantable materials in the orthopedic field due to their superior biocompatibility and biodegradability. However, weak mechanical properties of the biodegradable polymers and inflammatory reaction caused by the acidic degradation products have limited their biomedical applications. In this study, we have developed a PLGA composite containing beta-tricalcium phosphate (β-TCP) and magnesium hydroxide (Mg(OH)<SUB>2</SUB>) as additives to improve mechanical, osteoconductivity, and anti-inflammation property of the biopolymer composite simultaneously. The β-TCP has an osteoconductive effect and the Mg(OH)<SUB>2</SUB> has a pH neutralizing effect. The PLGA/inorganic composites were uniformly blended <I>via</I> a twin extrusion process. The mechanical property of the PLGA/β-TCP/Mg(OH)<SUB>2</SUB> composite was improved compared to the pure PLGA. In particular, the addition of Mg(OH)<SUB>2</SUB> suppressed the inflammatory reaction of normal human osteoblast (NHOst) cells and also inhibited the differentiation of pre-osteoclastic cells into osteoclasts. Moreover, synergistically upregulated late osteogenic differentiation of NHOst cells was observed on the PLGA/β-TCP/Mg(OH)<SUB>2</SUB> composite. Taken all together, we believe that the use of β-TCP and Mg(OH)<SUB>2</SUB> as additives with synthetic biodegradable polymers has great potential by the synergistic effect in orthopedic applications.</P> <P><B>Highlights</B></P> <P> <UL> <LI> PLGA composites containing β-TCP and Mg(OH)<SUB>2</SUB> were successfully fabricated by a twin screw extrusion technique. </LI> <LI> The PLGA/inorganic composites exhibited improved mechanical properties and superior pH neutralization effect. </LI> <LI> PLGA composite showed significantly improved osteogenesis as well as reduced inflammation and osteoclastogenesis. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Vulnerability, Affect, and Bio-power in James Hannaham’s Delicious Foods

        Delicious Foods by James Hannaham tells the story of a mother and son, Darlene and Eddie Hardison, who become involved in a debt slavery scam that preys upon drug-addicts, immigrants, and African-Americans. More importantly, it sheds light on racist institutions and the failure of representative democracy that lead them to a life of contemporary slavery. In the novel, many of the tragedies that happen arise as a result of the characters’ love for each other. Love in the novel is represented in the grief that Darlene feels for her husband Nat and the need that Eddie has for Darlene to be a proper mother. Their emotional attachment to each other’s bodies and the control of their affective responses by racist institutions cause the tragedy of this novel. This paper will employ the concepts of vulnerability, affect, and bio-power as defined by Judith Butler in Precarious Life to examine how love operates in the context of “the social vulnerability” of bodies. While the novel does not disprove Butler’s argument, I argue that Delicious Foods illustrates what happens when a person’s vulnerability and existence under the law is unacknowledged.

      • Identification of Human Complement Factor B as a Novel Biomarker Candidate for Pancreatic Ductal Adenocarcinoma

        <P>Pancreatic cancer (PC; pancreatic ductal adenocarcinoma) is characterized by significant morbidity and mortality worldwide. Although carbohydrate antigen (CA) 19-9 has been known as a PC biomarker, it is not commonly used for general screening because of its low sensitivity and specificity. Therefore, there is an urgent need to develop a new biomarker for PC diagnosis in the earlier stage of cancer. To search for a novel serologic PC biomarker, we carried out an integrated proteomic analysis for a total of 185 pooled or individual plasma from healthy donors and patients with five disease groups including chronic pancreatitis (CP), PC, and other cancers (e.g., hepatocellular carcinoma, cholangiocarcinoma, and gastric cancer) and identified complement factor b (CFB) as a candidate serologic biomarker for PC diagnosis. Immunoblot analysis of CFB revealed more than two times higher expression in plasma samples from PC patients compared with plasma from individuals without PC. Immunoprecipitation coupled to mass spectrometry analysis confirmed both molecular identity and higher expression of CFB in PC samples. CFB showed distinctly higher specificity than CA 19-9 for PC against other types of digestive cancers and in discriminating PC patients from non-PC patients (<I>p</I> < 0.0001). In receiver operator characteristic curve analysis, CFB showed an area under curve of 0.958 (95% CI: 0.956 to 0.959) compared with 0.833 (95% CI: 0.829 to 0.837) for CA 19-9. Furthermore, the Y-index of CFB was much higher than that of CA 19-9 (71.0 vs 50.4), suggesting that CFB outperforms CA 19-9 in discriminating PC from CP and other gastrointestinal cancers. This was further supported by immunoprecipitation and qRT-PCR assays showing higher expression of CFB in PC cell lines than in normal cell lines. A combination of CFB and CA 19-9 showed markedly improved sensitivity (90.1 vs 73.1%) over that of CFB alone in the diagnosis of PC against non-PC, with similar specificity (97.2 vs 97.9%).
