        Genetic variations in DROSHA and DICER and survival of advanced non-small cell lung cancer: a two-stage study in Chinese population

        MicroRNAs (miRNA) are a class of small, noncoding RNA molecules involved in carcinogenesis. Genetic variations in miRNA processing genes may affect the biogenesis of miRNAs, and consequently affect miRNAs regulation and development and progression of human cancer. Therefore, we hypothesized that polymorphisms in two main miRNA biosynthesis genes (DROSHA and DICER) may modulate the survival of advanced non-small cell lung cancer (NSCLC) patients in China. We selected 36 common tagging SNPs in DROSHA and DICER and evaluated the associations of these SNPs with survival of advanced NSCLC patients by a two-stage study in Chinese Han population (discovery cohort: 303 patients; replication cohort: 340 patients). Thirty-six SNPs were detected in the discovery cohort and 12 promising SNPs were validated in the replication cohort. The results showed that DROSHA rs3805525 was marginally associated with the survival of NSCLC patients in the replication cohort (dominant model: HR 0.69, 95 % CI 0.46–1.03, P = 0.071), which was in the same direction as that in the discovery cohort. When combing all patients into one group, three SNPs (rs3805525, rs17410035 and rs7719497) in DROSHA showed significantly associations with NSCLC survival (additive model: HR 0.82, 95 % CI 0.68–0.99 for rs3805525; HR 0.79, 95 % CI 0.62–1.00 for rs17410035; HR 0.76, 95 % CI 0.62–0.93 for rs7719497). Additionally, the combined analysis of those three SNPs showed a significant locus-dosage effect between number of favorable alleles and death risk of NSCLC (Trend P = 0.002). Genetic variations in DROSHA might be associated with the survival of advanced NSCLC patients in Chinese population.

        Intestine epithelial cell-derived extracellular vesicles alleviate inflammation induced by Clostridioides difficile TcdB through the activity of TGF-β1

        Background Clostridioides diffi cile infection (CDI) has been primarily associated with the toxin B (TcdB), one of the three known protein toxins secreted by C. diffi cile , which can activate the intestinal immune system and lead to pathological damage. Even though the biological functions of intestine epithelial cell-derived extracellular vesicles (I-Evs) have been well documented, the role of I-Evs in the process of CDI is still unknown. Objectives The protective eff ect of I-Evs against C. diffi cile TcdB was investigated both in cultured murine colon carcinoma MC38 cells and a mouse model used in this study. Results Mouse I-Evs with mean diameter ranging from 100 to 200 nm and a density of 1.09–1.17 g/mL were obtained and confi rmed containing the Ev-associated specifi c surface markers CD63 and TSG101 as well as high level of TGF-β1. In MC38 cells, I-Evs were able to decrease the gene expression of IL-6, TNF-α, IL-1β, and IL-22 induced by C. diffi cile TcdB, but to increase both the gene expression and protein levels of TGF-β1. I-Evs treatment via intraperitoneal administration alleviates C. diffi cile TcdB-induced local colon infl ammation in mice and increased their survival rate from 50% up to 80%. Furthermore, I-Evs induced an increase in the proportion of CD4 + Foxp3 + Tregs in vitro and in vivo through a TGF-β1-dependent mechanism by activating the TGF-β1 pathway and prompting phosphorylation of the downstream proteins Smad 2/3. Conclusion For the fi rst time, our study demonstrated that I-Evs originated from intestine epithelial cells can alleviate infl ammation induced by C. diffi cile TcdB both in vitro and in vivo. Therefore, I-Evs might be potentially a novel endogenous candidate for eff ective treatment of CDI.

The surfactants disassembled the probes to turn on significant fluorescence and transform between a pyrene monomer and an excimer, generating a two-dimensional analysis map. The probes were further used to identify different bacterial species.</P>

        LncRNA PVT1 upregulates FBN1 by sponging miR-30b-5p to aggravate pulpitis

        Background Pulpitis is a disease mainly caused by bacterial infection. Long noncoding RNAs (lncRNAs) infl uence the advancement of pulpitis. In this paper, we reconnoitered the characters of lncRNA plasmacytoma variant translocation 1 (PVT1) in pulpitis. Objective PVT1, microRNA-30b-5p (miR-30b-5p) and fi brillin 1 (FBN1) levels were identifi ed by qRT-PCR and western blot. In addition, the cell functions were scrutinized. Additionally, the link between miR-30b-5p and PVT1 or FBN1 was identifi ed by dual-luciferase reporter assay. Result The contents of PVT1 and FBN1 were augmented, but the miR-30b-5p level was declined in pulpitis. Knockdown of PVT1 enhanced cell proliferation and cell vitality, whereas inhibited cell infl ammatory reaction in HDPFs under LPS stimulation. In mechanism, PVT1 acted as a miR-30b-5p sponge to adjust the content of FBN1. Moreover, miR-30b-5p mediated LPS-mediated repression of proliferation and promotion of infl ammatory reaction HDPFs by FBN1. Conclusion PVT1 expedited the improvement of pulpitis via miR-30b-5p/FBN1.

        Fabrication of hierarchical structures on concrete surfaces with superhydrophobicity using replicated micro-nano dendritic structures

        Concrete with superhydrophobicity can protect it from being eroded by water and reduce accumulationof dust. Herein, a green and efficient method was proposed for fabricating hierarchical structures on concretesurfaces with superhydrophobicity by directly replicating micro-nano dendritic structures fromsuperhydrophobic copper mesh to fresh concrete. Microstructures, chemical compositions and wettabilityof the as-fabricated concrete were characterized. Superhydrophobicity of the as-fabricated concretewas studied. Effect of water jet on the as-fabricated concrete was investigated. Wetting durability ofthe as-fabricated concrete were studied. Self-cleaning property was confirmed. It was found that owingto the successful replication of micro-nano dendritic structures and low surface energy stearic acid, theas-fabricated concrete displayed good superhydrophobicity, wetting durability and self-cleaningproperty.

        The Influence of Cultural Intelligence and Institutional Distance of Chinese and Korean Contractors on the Performance of International Construction Projects

        Cultural intelligence can effectively solve cross-cultural problems, which encourages contractors to adapt to the new international market environment. However, in international projects, the effectiveness of cultural intelligence can be complicated or moderated by the host country's institutional environment. This study explores the impact of cultural intelligence on the performance of international construction projects and the moderating effect of institutional distance, then compares the differences between Chinese contractors and Korean contractors. In this paper, eleven hypotheses were proposed, and an empirical analysis was conducted to test the hypotheses and measure the impact of cultural intelligence on the performance of international construction projects and the moderating effect of institutional distance. The results from 144 valid questionnaires indicated that process capability and path capability in cultural intelligence have a significant positive impact on the performance of international construction projects, while formal institutional distance can positively moderate the relationship between position capability and international construction project performance. Moreover, Chinese and Korean contractors display significant differences in the moderating role of institutional distance. These findings provide theoretical support and strategic suggestions for not only contractors in China and Korea but for all international contractors seeking to improve project performance through cultural intelligence and institutional distance.

        Multidrug Resistance Mechanisms of Carbapenem Resistant Klebsiella pneumoniae Strains Isolated in Chongqing, China

        Background: Carbapenem-resistant Klebsiella pneumoniae (CRKP) is considered a serious global threat. However, little is known regarding the multidrug resistance (MDR) mechanisms of CRKP. This study investigated the phenotypes and MDR mechanisms of CRKP and identified their clonal characteristics. Methods: PCR and sequencing were utilized to identify antibiotic resistance determinants. Integron gene cassette arrays were determined by restriction fragment length polymorphism (RFLP) analysis. Multi-locus sequence typing (MLST) and pulsed-field gel electrophoresis (PFGE) were used for epidemiological analysis. Plasmids were typed by using a PCR-based replicon typing and analyzed by conjugation and transformation assays. Results: Seventy-eight strains were identified as resistant to at least one carbapenem; these CRKP strains had a high prevalence rate (38.5%, 30/78) of carbapenemase producers. Additionally, most isolates harbored MDR genes, including Extended spectrum β-lactamases (ESBLs), AmpC, and quinolone and aminoglycoside resistance genes. Loss of porin genes was observed, and Class 1 integron was detected in 66.7% of the investigated isolates. PFGE and MLST results excluded the occurrence of clonal dissemination among these isolates. Conclusions: A high prevalence of NDM-1 genes encoding carbapenem resistance determinants was demonstrated among the K. pneumoniae isolates. Importantly, this is the first report of blaNDM-1 carriage in a K. pneumoniae ST1383 clone in China and of a MDR CRKP isolate co-harboring blaNDM-1, blaKPC-2, blaCTX-M, blaSHV, acc(6’)-Ib, rmtB, qnrB, and acc(6’)-Ib-cr.

        Stayed Buckle Cable Recycling for Seismic Upgrading of Super-long-span Concrete Filled Steel Tube Arch Bridges

        A seismic upgrading approach for super-large-span upper-deck concrete-filled steel tube arch bridges using the stayed buckle cables (SBCs, which are temporary facilities for construction of long-span arch bridges) was proposed. The arrangement of SBCs most beneficial to the seismic resistance of arch bridges was determined. The results show that the reasonable arrangement of SBCs can achieve significant seismic performance improvement, reduce the stress demand on the diagonal of the arch rib, and improve the capacity-demand ratios of the weak sections of the arch rib. In upgrading model, the max displacement response of the structure is decreased by 41.3%, the max tensile stress on the diagonal is decreased by 30.1%, the max compression stress demand on the diagonal decreased by 18.0%, the CDRs of sections of the arch ribs are greatly improved.

        A Game Theory Based Interaction Strategy between Residential Users and an Electric Company

        With the development of smart grid technology, it has become a hotspot to increase benefits of both residential users and electric power companies through demand response technology and interactive technology. In this paper, the game theory is introduced to the interaction between residential users and an electric company, making a mutually beneficial situation for the two. This paper solves the problem of electricity pricing and load shifting in the interactive behavior by building the game-theoretic process, proposing the interaction strategy and doing the optimization. In the simulation results, the residential users decrease their cost by 11% mainly through shifting the thermal loads and the electric company improves its benefits by 5.6% though electricity pricing. Simulation analysis verifies the validity of the proposed method and shows great revenue for the economy of both sides.
