        Folate-decorated Polysaccharide-doxorubicin Polymer: Synthesis, Characterization, and Activity in HeLa Cells

        In order to develop the application of the natural polysaccharide lentinan (LNT) and decrease the side effects of doxorubicin, we successfully synthesized a novel folate-modified maleilated lentinan-doxorubicin (FA-M.LNT-DOX) polymer and used it for tumor-targeted drug delivery. The release efficiency and cytotoxicity of the prodrugs were evaluated in vitro. Although DOX release from FA-M.LNT-DOX was quite slow in a neutral buffer, it was particularly fast in an acidic solution with a pH of 5.0. Compared with DOX, FA-M.LNT-DOX showed higher cytotoxicity in HeLa cells and significantly lower cytotoxicity in normal cells. These results suggested that FA-M.LNT-DOX could be considered as a potential drug delivery candidate for folate receptor-positive cancer therapy.

        A Novel Menthol-Cementing Sampling Technique for Cohesionless Coarse-Grained Fillers

        It is difficult to test the engineering properties of undisturbed cohesionless coarse-grained fillers in the laboratory, especially those with low water contents because most sampling methods disturb the natural structure of the filler. This study proposes a novel sampling method that uses menthol as a temporary cement to obtain samples that are more resistant to disturbance. Liquid menthol infiltrates the interparticle spaces and cements the particles together as it cools, forming a high-strength structure. The samples can then be cut with a cutting ring (for grain sizes ≤ 0.5 mm) or a cutting machine (for grain sizes > 0.5 mm) to prepare the final samples, from which the menthol is removed by heating. To demonstrate the feasibility of the method, a number of laboratory experiments were performed, including sampling tests, confined compressive strength tests, vibration tests, drop tests, and CT scans. The optimal particle temperature for introducing the liquid menthol is 70°C, while 90°C is optimal for removing the cemented menthol. For both cost and anti-disturbance reasons, 20°C is suggested as a suitable storage temperature for menthol-cemented samples. The interspace structure undergoes less change with increases in sample depth and with larger grain sizes. We recommend that the lower two-third of the depth of a menthol-cemented block is used as the final sample for laboratory testing.

        Fabrication of hierarchical structures on concrete surfaces with superhydrophobicity using replicated micro-nano dendritic structures

        Concrete with superhydrophobicity can protect it from being eroded by water and reduce accumulationof dust. Herein, a green and efficient method was proposed for fabricating hierarchical structures on concretesurfaces with superhydrophobicity by directly replicating micro-nano dendritic structures fromsuperhydrophobic copper mesh to fresh concrete. Microstructures, chemical compositions and wettabilityof the as-fabricated concrete were characterized. Superhydrophobicity of the as-fabricated concretewas studied. Effect of water jet on the as-fabricated concrete was investigated. Wetting durability ofthe as-fabricated concrete were studied. Self-cleaning property was confirmed. It was found that owingto the successful replication of micro-nano dendritic structures and low surface energy stearic acid, theas-fabricated concrete displayed good superhydrophobicity, wetting durability and self-cleaningproperty.

        Cloning of porcine chemerin, ChemR23 and GPR1 and their involvement in regulation of Lipogenesis

        Chemerin is a novel adipokine which is abundant in adipose tissue to promote adipocyte differentiation and with significant relativity to BMI and insulin sensitivity. We report here the molecular characterization of porcine chemerin and its receptors ChemR23 and GPR1, as well as their transcriptional regulation during lipogenesis. Chemerin was mainly expressed in liver, intestine, kidney and adipose tissue, consistent with the expression pattern of GPR1, but not ChemR23, which was predominantly present in spleen and temperately in adipose tissue. We further investigated the lipogenesis-related transcriptional activation of PPARγ and KLF15 on chemerin and its receptors. The data showed that KLF15, but not PPARγ, can up-regulate the mRNA level of chemerin, ChemR23 and GPR1, which was consistent with the results of luciferase assay that confirmed the effect of KLF15 on ChemR23 promoter. Taken together, our data provide basic molecular information for the further investigation on the function of chemerin in lipogenesis. [BMB reports 2010; 43(7): 491-498]
