        Improvement of the Spectral Reconstruction Process with Pretreatment of Matrix in Convex Optimization

        In this paper, a pretreatment method for a matrix in convex optimization is proposed to optimize the spectral reconstruction process of a disordered dispersion spectrometer. Unlike the reconstruction process of traditional spectrometers using Fourier transforms, the reconstruction process of disordered dispersion spectrometers involves solving a large-scale matrix equation. However, since the matrices in the matrix equation are obtained through measurement, they contain uncertainties due to out of band signals, background noise, rounding errors, temperature variations and so on. It is difficult to solve such a matrix equation by using ordinary nonstationary iterative methods, owing to instability problems. Although the smoothing Tikhonov regularization approach has the ability to approximatively solve the matrix equation and reconstruct most simple spectral shapes, it still suffers the limitations of reconstructing complex and irregular spectral shapes that are commonly used to distinguish different elements of detected targets with mixed substances by characteristic spectral peaks. Therefore, we propose a special pretreatment method for a matrix in convex optimization, which has been proved to be useful for reducing the condition number of matrices in the equation. In comparison with the reconstructed spectra gotten by the previous ordinary iterative method, the spectra obtained by the pretreatment method show obvious accuracy.

        Protocol: The Effect of 12 Weeks of Tai Chi Practice on Anxiety in Healthy but Stressed People Compared to Exercise and Wait-list Comparison Groups: A Randomized Controlled Trial

        Stress is a major problem in today’s fast-paced society and can lead to serious psychoso- matic complications. The ancient Chinese mindebody exercise of Tai Chi may provide an alternative and self-sustaining option to pharmaceutical medication for stressed individ- uals to improve their coping mechanisms. The protocol of this study is designed to eval- uate whether Tai Chi practice is equivalent to standard exercise and whether the Tai Chi group is superior to a wait-list control group in improving stress coping levels. This study is a 6-week, three-arm, parallel, randomized, clinical trial designed to evaluate Tai Chi practice against standard exercise and a Tai Chi group against a nonactive control group over a period of 6 weeks with a 6-week follow-up. A total of 72 healthy adult participants (aged 18e60 years) who are either Tai Chi naı ¨ve or have not practiced Tai Chi in the past 12 months will be randomized into a Tai Chi group (n = 24), an exercise group (n = 24) or a wait-list group (n = 24). The primary outcome measure will be the State Trait Anxiety Inventory with secondary outcome measures being the Perceived Stress Scale 14, heart rate variability, blood pressure, Short Form 36 and a visual analog scale. The protocol is reported using the appropriate Standard Protocol Items: Recommendations for Inter- ventional Trials (SPIRIT) items.

        Development of a Novel Questionnaire for the Traditional Chinese Medicine Pattern Diagnosis of Stress

        Currently, there is no definitive diagnosis or list of signs and symptoms for “stress” in either modern biomedicine or Chinese medicine (CM). While modern theories on stress relate to the neurological interaction of a stressor or stimuli on the autonomic nervous system, it is generally regarded as subjective in nature and as such each individual will likely present varying somatic or cognitive signs and symptoms. A questionnaire was therefore developed, based on textual research, that incorporated both general as well as gender specific signs and symptom responses to determine the most common CM patterns associated with individuals who report as feeling stressed. For the 45 females who completed the questionnaire, the mean percentage of symptoms per CM pattern showed that the pattern with the highest average percentage was heart qi deficiency (61.88%) followed by liver blood deficiency (60.23%) and then heart blood deficiency (60.12%). For males (n Z 16), heart qi deficiency was also the highest scoring CM pattern with a scoring percentage of 54.81%. In males, however, heart blood deficiency was second with 53.29% followed by liver blood deficiency with 51.10%. Of the general non gender-specific symptoms collected (n Z 65 symptoms), the symptom most commonly reported by both men and women was “anxious or racing thoughts”, followed by “constant worrying” and “inability to concentrate”. The CM diagnostic pattern results may prove useful for clinicians as the change in diagnostic understanding will also modify the treatment principle and subsequent treatment with acupuncture or herbal medicine. Future CM research studies should consider including the questionnaire either as a diagnostic aid or as an outcome measure for acupuncture or herbal medicine studies related to stress.

        A triage strategy in advanced ovarian cancer management based on multiple predictive models for R0 resection: a prospective cohort study

        Objective: To present the surgical outcomes of advanced epithelial ovarian cancer (AEOC) since the implementation of a personalized approach and to validate multiple predictive models for R0 resection. Methods: Personalized strategies included: 1) Non-invasive model: preoperative clinico-radiological assessment according to Suidan criteria with a predictive score for all individuals. Patients with a score 0–2 were recommended for primary debulking surgery (PDS, group A), or otherwise were counseled on the choices of PDS, neoadjuvant chemotherapy (NAC, group B) or staging laparoscopy (S-LPS). 2) Minimally invasive model: S-LPS with a predictive index value (PIV) according to Fagotti. Individuals with a PIV <8 underwent PDS (group C) or otherwise received NAC (group D). Intraoperative assessment (with Eisenkop, peritoneal cancer index [PCI], and Aletti scores) and surgical results were prospectively collected. Results: Between September 2015 and August 2017, 161 pathologically confirmed epithelial ovarian cancer patients were included. A total of 52 (32.3%) patients had a predictive score of 0–2, and 109 (67.7%) patients had a score ≥3. Among these individuals, 41 (25.5%) patients received S-LPS. Finally, 110 (68.3%) patients underwent PDS (A+C), and 51 (31.7%) patients received NAC (B+D). The R0 resection rates in PDS and NAC patients were 56.4% and 60.8%, respectively. The area under the curve (AUC) of Suidan criteria was 0.548 for group (A+C). The AUC of Fagotti score was 0.702 for group C. The AUC of Eisenkop, PCI, and Aletti scores were 0.808, 0.797, and 0.524, respectively. Conclusion: The Suidan criteria were not effective in these AEOC patients. S-LPS was helpful in decision-making for PDS and should be endorsed in the future.

      • Use of autonomous maximal smile to evaluate dental and gingival exposure

        Objective: This study was performed to validate the autonomous maximal smile (AMS) as a new reference for evaluating dental and gingival exposure. Methods: Digital video clips of 100 volunteers showing posed smiles and AMS at different verbal directives were recorded for evaluation a total of three times at 1-week intervals. Lip-teeth relationship width (LTRW) and buccal corridor width (BCW) were measured. LTRW represented the vertical distance between the inferior border of the upper vermilion and the edge of the maxillary central incisors. Intraclass correlation coefficients (ICCs) for reproducibility, and the m-value (minimum number of repeated measurements required for an ICC level over 0.75), were calculated. Results: LTRW and BCW of the AMS were 1.41 and 2.04 mm, respectively, greater than those of the posed smile (p < 0.05), indicating significantly larger dental and gingival exposure in the AMS. The reproducibility of the AMS (0.74 to 0.77) was excellent, and higher than that of the posed smile (0.62 to 0.65), which had fair-to-good reproducibility. Moreover, the m-value of the AMS (0.88 to 1.05) was lower than that of the posed smile (1.59 to 1.85). Conclusions: Compared to the posed smile, the AMS shows significantly larger LTRW and BCW, with significantly higher reproducibility. The AMS might serve as an adjunctive reference, in addition to the posed smile, in orthodontic and other dentomaxillofacial treatments.

        Certain Polymorphisms in SP110 Gene Confer Susceptibility to Tuberculosis: A Comprehensive Review and Updated Meta-Analysis

        Purpose: Numerous studies have assessed the association of SP110 gene variants with tuberculosis (TB), but the results were inconsistent. Through a comprehensive review and meta-analysis, our study aimed to clarify the nature of genetic risks contributedby 11 polymorphisms for the development of TB. Materials and Methods: Through searching PubMed, web of science, China National Knowledge Infrastructure (CNKI) databases,a total of 11 articles including 13 independent studies were selected. The pooled odd ratios (ORs) along with their corresponding95% confidence interval (CI) were estimated for allelic comparisons, additive model (homozygote comparisons; heterozygotecomparisons), dominant model and recessive model. We also assessed the heterogeneity across the studies and publication bias. Results: The results of combined analysis revealed a significantly increased risk of TB for single nucleotide polymorphism (SNP)rs9061 in all five comparisons (allelic comparisons: OR=1.28, 95% CI=1.14–1.44, p<0.0001; homozygote comparisons: OR=2.84, 95%CI=1.84–4.38, p<0.00001; heterozygote comparisons: OR=1.23, 95% CI=1.05–1.43, p=0.009; dominant model: OR=1.32, 95% CI=1.14–1.53, p=0.0003; recessive model: OR=2.26, 95% CI=1.18–4.34, p=0.01). In subgroup analysis, the risk of TB associated with SNP rs9061appeared to be increased. Moreover, increased risk of TB was also found in Asian subgroup of SNP rs11556887, while decreased riskof TB appeared in large sample size subgroup of SNP rs1135791. No significant association was observed between other SNPs andthe risk of TB. Conclusion: Our meta-analysis suggested that the variant of SNP rs9061 might be a risk factor for TB.

        MIR210HG Aggravates Sepsis-Induced Inflammatory Response of Proximal Tubular Epithelial Cell via the NF-κB Signaling Pathway

        Purpose: Acute kidney injury (AKI) is a serious complication of sepsis and is characterized by inflammatory response. MicroRNA-210 host gene (MIR210HG) is upregulated in human proximal tubular epithelial cells under treatment of inflammatory cytokines. This study aimed to explore the role of MIR210HG in sepsis-induced AKI. Materials and Methods: Cell viability was detected by a cell counting kit 8 assay. The levels of proinflammatory cytokines weredetected by enzyme-linked immunosorbent assay kits. The protein levels of p65, IκBα, and p-IκBα were examined by western blotanalysis. The nuclear translocation of nuclear factor kappa B (NF-κB) was detected by immunofluorescence assay. The histologicalchanges of kidneys were analyzed by hematoxylin and eosin staining assay. Results: Lipopolysaccharide (LPS) treatment significantly inhibited cell viability and increased productions of proinflammatorycytokines in proximal tubular epithelial cells (HKC-8). Additionally, MIR210HG levels in HKC-8 cells were increased by LPS treatment. MIR210HG silencing inhibited the LPS-induced cell inflammatory response. MIR210HG activated the NF-κB signalingpathway by promoting the phosphorylation of IκBα and nuclear translocation of p65. Rescue assays revealed that the MIR210HGinducedincrease of cytokines levels and decline of cell viability were rescued by QNZ treatment. Knockdown of MIR210HG decreasedblood urea nitrogen, serum creatinine, and proinflammatory cytokine levels in AKI rats. Moreover, the knockdown ofMIR210HG protected against AKI-induced histological changes of kidneys in rats. Conclusion: MIR210HG promotes sepsis-induced inflammatory response of HKC-8 cells by activating the NF-κB signaling pathway. This novel discovery may be helpful for the improvement of sepsis-induced AKI.

        Ratiometric and discriminative visualization of autophagic processes with a novel dual-responded lysosome-specific fluorescent probe

        Background Autophagy is a critical self-eating pathway involved in numerous physiological and pathological processes. Lysosomal degradation of dysfunctional organelles and invading microorganisms is central to the autophagy mechanism and essential for combating disease-related conditions. Therefore, monitoring fluctuations in the lysosomal microenvironment is vital for tracking the dynamic process of autophagy. Although much effort has been put into designing probes for measuring lysosomal viscosity or pH separately, there is a need to validate the concurrent imaging of the two elements to enhance the understanding of the dynamic progression of autophagy. Methods Probe HFI was synthesized in three steps and was developed to visualize changes in viscosity and pH within lysosomes for real-time autophagy tracking. Then, the spectrometric determination was carried out. Next, the probe was applied to image autophagy in cells under nutrient-deprivation or external stress. Additionally, the performance of HFI to monitor autophagy was employed to evaluate acetaminophen-induced liver injury. Results We constructed a ratiometric dual-responsive probe, HFI, with a large Stokes shift over 200 nm, dualwavelength emission, and small background interference. The ratiometric fluorescent signal (R = I610/I460) of HFI had an excellent correlation with both viscosity and pH. More importantly, high viscosity and low pH had a synergistic promotion effect on the emission intensity of HFI, which enabled it to specially lit lysosomes without disturbing the inherent microenvironment. We then successfully used HFI to monitor intracellular autophagy induced by starvation or drugs in real-time. Interestingly, HFI also enabled us to visualize the occurrence of autophagy in the liver tissue of a DILI model, as well as the reversible effect of hepatoprotective drugs on this event. Conclusions In this study, we developed the first ratiometric dual-responsive fluorescent probe, HFI, for real-time revealing autophagic details. It could image lysosomes with minimal perturbation to their inherent pH, allowing us to track changes in lysosomal viscosity and pH in living cells. Ultimately, HFI has great potential to serve as a useful indicator for autophagic changes in viscosity and pH in complex biological samples and can also be used to assess drug safety.
