      • Molecular Cloning, Expression and Genomic Structure of Glyceraldehyde-3-Phosphate Dehydrogenase from the Entomopathogenic fungus, Paecilomyces tenuipes Jocheon-1

        Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces tenuipes Jocheon-1. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. Paecilomyces tenuipes Jocheon-1 cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes Jocheon-1 was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDHis comprised of 1,014bp encoding 338 amino acid residues. The deduced protein sequence of Pt-GAPDH showed higher homology with Beauberia bassiana-GAPDH(93% amino acid identity). Hydropathy analysis revealed that Pt-GAPDH protein is hydrophilic. The major three amino acids in its composition of amino acid residues were alanine(11.54%), valine(9.47%) and glycine(8.88%). The cDNA encoding Pt-GAPDH was expressed as a 37 kDa polypeptide in baculovirus-infected insect Sf9 cells. The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA.

        Fungi belonging to the Paecilomyces spp. have recently been used as food and herbal medicines in Korea and are greatly popular as commercially available powdered supplement or dried fruiting body. Despite this acceptance and its use, little is known of the genes related to its reactive agents. Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces spp. based on the previous identification of ITS1 and ITS2 at the molecular level and collected from Jocheon Miryang, Korea. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes- Jocheon was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDH is comprised of 1,014bp encoding 338 amino acid residues.         <P>Solution‐processable reduced graphene oxide as a hole‐transporting layer for highly efficient and stable organic solar cells is reported on page 4923 by Dong‐Yu Kim, Seok‐In Na, and co‐workers. Introduction of a newly reduced graphene oxide by simple solution processing into solar cells dramatically raises the cell efficiency and cell life‐time. The results will allow full use of chemically reduced graphene and will advance the realization of carbon‐based printable optoelectronic devices. </P>

        This paper was a study on method that transforms a source speaker voice in order to be heard like voice of a specified speaker. It was able to setimate speech signal feature with proposed MFCC method considering property of human auditory. Applying EM algorithm for estimated feature valuse optimal parameter of GMM, we can compose GMM distribution. Composed GMM determined a weighting value of frequency envelope on speech conversion function. With result of experiment applying HNM in order to convert speech, we can verify that speech signal frequency envelope of a source speaker was similarly converted to speech signal frequency envelope fo a specified speaker.

        A 48-year-old male visited the emergency room of the authors’ hospital due to nausea, vomiting, and myalgia for four days. Acute hepatitis A was identified from the serologic marker of the hepatitis A virus. Mild elevation of the serum creatinine and creatinine phosphokinase(CPK) suggested rhabomyolysis, which was confirmed with the serum aldolase, myoglobin, and urine myoglobin. With supportive care, both the liver and renal func- tions were recovered gradually and fully. This case shows that rhabdomyolysis can be one of the mechanisms of renal complication in cases of acute symptomatic hepatitis A.

        현재 고정밀도 저수량 측량에 사용되는 측량장비에는 수평위치결정방법으로 실시간 DGPS측량장비가 많이 활용되고 있고, 수심측량에는 디지털 음향측심기를 많이 사용하고 있다. 본 연구에서는 현재, 수심측량 결과처리 시 문제가 되고 있는 소프트웨어의 가격부담을 줄이고 수심측량과 내업처리에 의한 저수량산출을 통합처리 할 수 있는 국내 자체개발의 저가형 외 · 내업 일체형 저수량산출시스댐을 개발하였다.

        扶正抗癌湯의 抗腫瘍效果에 關한 實驗的 硏究

        腫瘍으로 인한 死亡率은 多樣한 名種 治療에도 不拘하고 循環器疾患 다음으로 2위를 차지하고 있다. 이에 著者는 韓醫學과 西醫學의 結合治療를 通해서 抗腫瘍藥物의 效果를 增進시키고 副作用을 줄이는 藥物의 開發을 爲하여 健脾 益氣 祛痰 補腎 杭癌效能을 갖고 잇는 扶正杭癌湯抽出液을 使用하여 Colony 形成抑制實驗, Sulforhoda-mine B(SRB) assay를 觀察하였고, 扶正抗癌湯抽出液과 抗癌化學療法劑인 Mitomycin C(MMC)를 Ehrlich carcinoma의 solid tumor와 ascites tumor에 各各 單獨 및 倂用 投與하여 抗腫瘍效果 및 Iysosomal enzymes의 活性에 대한 效果를 觀察하였다. 먼저 扶正抗癌湯抽出液을 Caki-1 cell, Hep 3B 및 A549에 투여한 후 增殖抑制作用을 colony 形成抑制實驗과 SRB assay를 통하여 觀察한 결과 濃度依存的으로 腫瘍細胞의 成長을 抑制하였다. Ascites form of Ehrlich carcinoma에 대한 抗腫瘍效果 實驗에서 扶正抗癌湯抽出液과 MMC를 병용투여한 결과, 扶正抗癌湯抽出液에 의하여 MMC의 抗腫瘍效果가 增加하였다. Solid form of Ehrlich carcinoma에 대한 抗腫瘍效果 實驗에서 扶正抗癌湯抽出液과 MMC를 병용 투여하였을 경우, MMC를 단독 투여하였을 경우보다 腫瘍의 크기가 현저하게 減少하는 결과를 보였다. Ehrlich ascites carcinoma cell에 扶正抗癌湯抽出液과 MMC를 병용 투여하였을 경우, MMC를 단독 투여하였을 경우보다 lysosomal enzymes의 活性이 强하게 나타났다. 本 實驗을 통하여 扶正抗癌湯抽出液은 MMC의 抗腫瘍效果를 증가시키는 效果를 보였다. 또한 扶正抗癌湯抽出液은 마우스에서 solid form of Ehrlich ascites carcinoma의 크기를 감소시키는 效果를 나타냈으며, 생존기간 實驗에서는 扶正抗癌湯抽出液과 MMC를 병용투여시 ascites tumor에 MMC를 단독으로 투여하였을 때와 비교하여 유의하게 생존기간을 증가시키는 결과를 보였다. 이상의 결과를 종합하여 볼 때 扶正抗癌湯抽出液은 用量에 依存的으로 直接的인 抗腫瘍效果가 있으며, 抗癌療法劑인 MMC의 效果를 亢進시키는 것으로 보아 間接的인 抗腫瘍效果도 있을 것으로 思科되나 正確한 效果를 究明하기 위해서는 보다 正確한 抗腫瘍機轉에 대한 硏究와 藥物學的 硏究 및 臨床的 硏究가 追加되어야 할 것이다. In order to investigate the effects of Bujeonghangamtang Extract(扶正抗癌湯抽出液) on antitumor effects after human cell lines(A549, Hep3B, Caki-1, Ehrlich) transplantation into the peritoneal cavity or right groin in mice induced by RPMI 1640 and DMEM etc, the extracts of its herbal medicines were orally administered for 10 or 12 days. Experimental studies were performed for measurement of antitumor effect of Mitomycin C(MMC) and lysosomal enzyme's activities using colony forming efficiency, SRB assay which were regarded as a valuable method for the measurement of antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows: 1. The change of colony-forming efficiency and SRB assay of Caki-1 cells, Hep3B and A549 Cells after exposure to the extract of Bujeonghangamtang extract depressed the growth of tumor cells by concentration of Bujeonghangamtang. 2. Antitumor activity of the ethanol extract of Bujeonghangamtang extract and MMC on ascites form of Ehrlich carcinoma in mice is slightly improved. Especially the mean of survival times in the group of 200㎎/㎏ and MMC 0.1㎎/㎏ is improved over 30.9%. 3. When Bujeonghangamtang extract and MMC are administered together, the weight of tumor is more decreased than MMC alone. 4. The lysosomal enzyme's activities of the Bujeonghangamtang extract and MMC are more significantly improved than MMC alone. According to the above result, it could be suggested that Bujeonghangamtang extract has indirect antitumor effect by the increase of MMC uptake.

      • 社會體育施設 實態調査 硏究 : 行政地域을 中心으로 Focus on Administrative Districts

        In modern societies, there are trends for people to engage in more positive and creative physical activities in their leisure time, which Leads to rather participant sports than spectator sports. This study was undertaken to explore the actual state of " Sports for all" facilities in Korea. The implications of the study are as follows: 1. The total numbers of "Sport for all" facilities in Korea is 20,080 ; there are 328 public facilities, 14,696 school facilities, 1,386 job-site facilities and 3,596 civil vacilitles, which indicates that Korea has got smaller number of those facilitles than the other developed countries. 2. The actual state of the facilities by each citles and counties shows the fact that Seoul has got 2,869 facilities, Incheon 434, facilities and Jeja 290 facilities. 3. The square measure for each Korean People is 0.795㎡. 4. The facilities have been set up in servral particalar events and districts. In conclusion, it is requested that all the facilities should be opened to the publics in order to anltivate the healthy mental and physical condition of peoples and to host various types of competitions, thereby promoting the sports, sport-for-all sports in particular, by means of civil-directed activities.

      • Z-Map기법을 이용한 머시닝센터용 CAD/CAM기술에 관한 연구

        본 연구에서는 확장형 Z-Map모델을 토대로 3차원 제품의 형상설계에서 머시닝센터에 의한 CNC가공까지를 일괄적으로 처리할 수 있는 CAD/CAM시스템을 IBM PC와 Borland C++4.5를 이용하여 Windows환경에서 개발하였다. 3차원 제품의 형상은 기본 입체의 생성기능, 자유곡면형 입체의 생성기능, 평행이동, 회전스위프기능 등과 같은 입체정의기능, 그리고 스플리트기능, 불리안기능 등과 같은 입체조직기능을 통해서 쉽게 설계 및 구형할 수 있다. 또한 제품과 소재의 형상정보, 밀링공구의 제원, 그리고 절삭조건을 토대로 황삭가공과 정삭가공에 대한 공구경로의 자동생성, 머시닝센터용 CNC프로그햄의 자동생성, 시뮬레이션, 표면조도 확인 등과 같은 여러 가지 기능들을 갖추고 있다. 특히 과대절삭이 방지된 공구경로는 밀링공구의 제원과 절삭조건을 토대로 제품형상을 옵셋시켜서 생성한 CL Z-Map모델을 이용해서 도출하였다. In this paper, the extended Z-Map model based CAD/CAM system which is able to process systematically from the three dimensional shape design of products to the CNC cutting of products by the machining center, is realized on the Windows environment by using IBM PC and Borland C++4.5. The three dimensional shape of products can be easily designed and constructed by means of the solid definition functions such as the creation operation of primitives and solids with free-formed surfaces besides the translational and rotational sweep operations, and the solid manipulation functions such as the split operation and the Boolean operation. And the system has the various capabilities related to the automatic generation of tool path for the rough and finish cutting processes, the automatic elimination of overcut, the automatic generation of CNC program for a machining center, the cutting simulation, and the surface roughness evaluation of machined product. Especially the overcut-free tool path is obtained by using the CL Z-Map models which are composed of the offset surfaces of the geometric shape of product.

        This study is to analyze the flood damages such as an inundation by overflow levee during rainfall duration. HEC-HMS and HEC-RAS are used to analyze rainfall-runoff and inundated area, respectively. Yeongwoon area is a study catchment for runoff effect analysis. As the result, it was understood that inundated area was magnified gradually according to the increase of rainfall recurrence interval in years. Inaddition GIS technique is an effective tool for calculating the exact flood damage area.
