        Serum Proteomics Analysis of Feline Mammary Carcinoma Based on Label-free and PRM Techniques

        Background: Feline mammary carcinoma is the third most common cancer that affects female cats. Objectives: The purpose of this study was to screen differential serum proteins in feline and clarify the relationship between them and the occurrence of feline mammary carcinoma. Methods: Chinese pastoral cats were used as experimental animals. Six serum samples from cats with mammary carcinoma (group T) and six serum samples from healthy cats (group C) were selected. Differential protein analysis was performed using a Label-free technique, while parallel reaction monitoring (PRM) was performed to verify the screened differential proteins. Results: A total of 82 differential proteins were detected between group T and group C, of which 55 proteins were down regulated and 27 proteins were up regulated. Apolipoprotein A-I, Apolipoprotein A-II (ApoA-II), Apolipoprotein B (ApoB), Apolipoprotein C-III (ApoC-III), coagulation factor V, coagulation factor X, C1q, albumen (ALB) were all associated with the occurrence of feline mammary carcinoma. Differential proteins were involved in a total of 40 signaling pathways, among which the metabolic pathways associated with feline mammary carcinoma were the complement and coagulation cascade and cholesterol metabolism. According to the Label-free results, ApoB, ApoC-III, ApoA-II, FN1, an uncharacterized protein, and ALB were selected for PRM target verification. The results were consistent with the trend of the label-free. Conclusions: This experimen is the first to confirm ApoA-II and ApoB maybe new feline mammary carcinoma biomarkers and to analyze their mechanisms in the development of such carcinoma in feline.

      • Historical Long-term Exposure to Pentachlorophenol Causing Risk of Cancer - A Community Study

        Background: Pervious studies suggested occupational workers exposure to pentachlorophenol (PCP) might contribute to increased risk of cancer. However, few studies have focused on associations between PCP and cancer risk at the community level. Objective: The present study was to explore the cancer risk for the community population living long-term in a PCP contaminated area. Methods: All the cancer cases diagnosed in 2009-2011 in Tongling City were collected. The cancer patients' residencies were geo-referenced in each district. The historical PCP usage for each district of Tongling was calculated as the PCP pollution index, which was further used to divide into PCP exposure categories. Standardized rate ratios (SRRs) of cancer incidence were applied to detect the cancer risk as exposure grade elevated. Correlation analysis was performed to analyze the relationship between PCP pollution and cancer incidence. Results: A total of 5,288 cancer cases (3,451 male and 1,837 female) were identified. PCP usage was correlated with the incidence of leukemia (r=0.88, P=0.002) for males, and with cancer of the esophagus for males (r=0.83, P=0.008) and females (r=0.71, P=0.020). Compared with the low exposure category, significant SRRs for total cancer sites was obtained for high PCP exposure category (SRR=1.61, 95%CI=1.59-1.62). Most SRR values of the cancer sites were significantly increased as exposure grade elevated and exposure time extended. Conclusion: The present study found that community residents living in the PCP contaminated area had increased risk of cancers. Leukemias, lymphomas and nasopharyngeal and esophageal cancers are most possibly associated with PCP exposure.

        Cold hardiness characteristic of the overwintering pupae of fall webworm Hyphantria cunea (Drury) (Lepidoptera: Arctiidae) in the northeast of China

        The fallwebworm, Hyphantria cunea (Drury) (Lepidoptera: Arctiidae), is an invasive and important pest in China. Understanding the physiological–biochemical characteristics of the overwintering pupae is essential for its cold tolerance and potential distribution in the northeast of China. Results indicated that the supercooling point (SCP) of H. cunea overwintering pupae varied significantly among Jilin city (43°N, 126°E), Jilin Province, Shenyang (41.8°N, 123.4°E), Liaoning Province and Yingkou (40.39°N, 122.13°E), Liaoning Province. The average SCP in Jilin was the lowest (−23.49 ± 0.46 °C) in those of three areas. The water content in overwintering pupae had a significant positive correlation with the SCP (R = 0.848,P = 0.03). Contents of alanine (ALA) (26.00%), lysine (LYS) (30.51%) and arginine (ARG) (22.88%) in the overwintering pupae's hemolymph were higher than others. During the overwintering, the fat content in pupae's bodies decreased gradually. Analysis of variance (ANOVA) indicated that the content of trehalose in the body of H. cunea was remarkably different (P= 0.007) month by month, with the maximum in March (2.88 mg·mL−1) and the minimum (0.40 mg·mL−1) in November. Pearson's correlation test indicated that SCP had a significant negative correlation with the SOD enzyme activity (R = −0.850; P = 0.008). Thermal hysteresis activity (THA) of hemolymph was 0.68°C and 0.7–0.8°C for H. cunea pupae and larva, respectively, using differential scanning calorimetry, which suggests that the H. cunea can improve their cold tolerance through producing antifreeze proteins. These results could help us to predict the distribution of the H. cunea in the northeast of China.

        Human lactoferrin efficiently targeted into caprine beta-lactoglobulin locus with transcription activator-like effector nucleases

        Objective: To create genetically modified goat as a biopharming source of recombinant human lacotoferrin (hLF) with transcription activator-like effector nucleases. Methods: TALENs and targeting vector were transferred into cultured fibroblasts to insert hLF cDNA in the goat beta-lactoglobulin (BLG) locus with homology-directed repair. The gene targeted efficiency was checked using sequencing and TE7I assay. The bi-allelic gene targeted colonies were isolated and confirmed with polymerase chain reaction, and used as donor cells for somatic cell nuclear transfer (SCNT). Results: The targeted efficiency for BLG gene was approximately 10%. Among 12 Bi-allelic gene targeted colonies, five were used in first round SCNT and 4 recipients (23%) were confirmed pregnant at 30 d. In second round SCNT, 7 (53%), 4 (31%), and 3 (23%) recipients were confirmed to be pregnant by ultrasound on 30 d, 60 d, and 90 d. Conclusion: This finding signifies the combined use of TALENs and SCNT can generate bi-allelic knock-in fibroblasts that can be cloned in a fetus. Therefore, it might lay the foundation for transgenic hLF goat generation and possible use of their mammary gland as a bioreactor for large-scale production of recombinant hLF.

        Chinese herbal medicine for myasthenia gravis: A systematic review and meta-analysis of randomized clinical trials

        Background: Myasthenia Gravis (MG) is a disorder of neuromuscular transmission bringing mild ocular weakness to severe generalized muscle weakness and disability. The conventional treatments have longterm side effects, and Chinese herbal medicines (CHM) have shown possible effect and safety for MG patients, but the existing evidence was not robust enough and the results were out of date. Methods: Searching for randomized controlled trials (RCTs) was conducted in 7 databases and clinical trial registries until July 2021. The Risk of Bias (ROB) 2 tool was used to assess the study quality and GRADE was used to assess the quality of whole evidence. Meta-analyses were conducted and the results were presented as risk ratio (RR) or mean difference (MD) with 95% confidence interval (CI). Results: Nineteen RCTs (1283 participants) testing 13 kinds of CHM with adequate randomization were included and six RCTs investigating Compound Huangqi were included in the meta-analyses. In addition to conventional treatment, nine CHMs reduced symptom scores of MG. Compound Huangqi plus conventional treatment (pyridostigmine bromide or prednisone or both) reduced the symptom scores compared with conventional treatment (MD=-3.56, 95%CI -4.86 to -2.26). Less adverse events happened in the CHM groups (3/247 in the CHM groups, 52/245 in the control groups, RR=0.13, 95%CI 0.06 to 0.30, 9RCTs, a total of 492 participants). The effect on quality of life was inconsistent. Conclusion: Nine CHMs could probably bring benefit for MG symptom improvement. Moderate to low certainty of evidence supported Compound Huangqi added-on conventional treatment probably bring extra benefit of improving MG symptoms. Adding CHMs could be safer than giving only conventional treatment. Study registration: The protocol was registered in PROSPERO (CRD42016032718).

        Microstructure control of Fe catalyst films for the growth of multiwalled carbon nanotube arrays

        Fe catalyst films were deposited on silicon substrates by using a metal vapor vacuum arc (MEVVA) ion deposition system for the growth of aligned multiwalled carbon nanotubes. The effects of deposition angle and film thickness on the morphologies of catalyst films before and after thermal treatment were investigated. The results show that the uniformities of both 5-nm and 10-nm films are enhanced as the deposition angle decreases. With increase from 5 nm to 10 nm, the surface uniformities of pristine films prepared at higher deposition angles (60˚ and 90˚) are increased, while that of the films produced at 30˚ deposition angle is slightly decreased. The uniformity of Fe catalyst particle sizes after thermal treatment on the whole, clearly decreases as the thickness is increased from 5 nm to 10 nm in the case of the same deposition angle. 5-nm films deposited at 30˚ deposition angle show the most uniform features before and after thermal treatment and can be used for the synthesis of high-quality carbon nanotube arrays.         Effect of the microstructure of Ag catalysts in the fabricating process of silicon nanowires

        In this paper, well-aligned silicon nanowire (SiNW) arrays have been directly synthesized on single crystal silicon wafer at room temperature via a simple chemical etching method, and the effect of the microstructure of Ag catalysts on the fabricating process of the silicon nanowires has been systematically investigated. The experimental results show that different microstructures of Ag catalysts have been observed with different concentration of AgNO3/HF solution for different deposition times. The diameters of Ag particles and the mean interstitial sizes among Ag particles are relatively small for a short deposition time, and some large particles are formed when the deposition time increases. A much longer deposition time caused the substrate to be covered with a large amount of Ag dendrite. It can be inferred that 4 min is a good deposition time, because of the proportioned structure of particles and interstitial size, according to our experimental results. Nanostructures of Ag catalyst also vary with a change of concentration of AgNO3/HF solution. Ag dendrite may be formed at high concentration and affect the fabrication of SiNWs.         Long-term Stable Field Electron Transfer from Carbon Nanotube Arrays at High Emission Current Densities

        The stability behaviors of multiwalled carbon nanotube arrays during field electron emission are studied. The results indicate that the stability, even at a high emission current density, has been greatly improved by an aging process, and a degradation of about 0.66% in the emission current density at 21.86 mA/cm_2 during a 10-hour stability test has been obtained. A detailed analysis of the deterioration of the field electron emission characteristics is given, and the generation of Joule heat during field emission is found to be able to burn off the extruded carbon nanotubes, which will directly reduce the number of emission sites. On the other hand, the Joule heating effect may induce an annealing of the defects existing in the carbon nanotubes and may influence the distribution of electron energy states, both having a bad influence on the field emission characteristics. An aging process, especially aging at high emission current densities, can greatly reduce the influence of Jouleheating induced current degradation. Hence, an aging process at high emission current densities provides an effective way to realize long-term stable field electron emission from carbon nanotube arrays.

        Morphology-dependent Optical Properties of One-dimensional Nanostructure-arrayed Silicon

        The optical properties of one-dimensional nanostructure-arrayed silicon (1DNSASi), which wasfabricated by the metal assisted electroless chemical etching method under different conditions,were characterized in the wavelength range of 220 - 1000 nm. Whether the optical absorption ofthe 1DNSASi was enhanced relative to that of the polished Si was determined from the detailedmorphology of the 1D nanostructures. For the yellow 1DNSASi prepared at a high etchant concentrationand high temperatures, its optical absorption was relatively nice in the ultraviolet lightregion, while a gradual attenuation was shown in the visible and the near-infrared regions, and theoptical absorption was lower than that of the polished Si at wavelengths above 800 nm. When theeffects of zeroth-order reflectance and zero transmission were combined, the optical absorption ofthe black 1DNSASi prepared at a low etchant concentration and room temperature was very high(> 99%) in the wavelength range of 220 - 1030 nm and displayed a slight decrease at wavelengthsabove 1030 nm. Our results demonstrate that the optical absorption of the black 1DNSASi couldbe further improved by increasing the etching depth and exhibited its measurable maximum valuewhen the etching depth was large enough. These results indicate that the 1DNSASi may be apromising candidate for high-efficiency photovoltaic devices, high-sensitivity sensors and detectors.
