        Functional Characterization of the Genes tauO, tauK, and tauI in the Biosynthesis of Tautomycetin

        Tautomycetin is a specific protein phosphatase I inhibitor. In an effort to elucidate the biosynthetic mechanism of tautomycetin,we inactivated genes of the tautomycetin biosynthetic gene cluster, tauI, tauO, and tauK, which encode for putative P450 oxidase, citryl-CoA lyase, and esterase enzymes,respectively. The mutant STQ0606 (ΔtauO) did not produce any detectable amount of tautomycetin intermediates but could convert dialkylmaleic anhydride to tautomycetin,strongly indicating that TauO was involved in dialkylmaleic anhydride biosynthesis. STQ1211 (ΔtauK) accumulated dialkylmaleic anhydride, whereas the cofermentation of STQ1211(ΔtauK) and STQ0606 (ΔtauO) restored the production of tautomycetin. Together, these results suggest that TauK was responsible for the conjugation of dialkylmaleic anhydride and the polyketide moiety in tautomycetin biosynthesis. The disruption of tauI resulted in the accumulation of 5-des-ketotautomycetin,revealing that TauI was responsible for the oxidation at C5 as the last step. Although the shunt pathways were involved in the biosynthesis of tautomycetin, the main post-polyketide synthase tailoring steps were dehydration,decarboxylation and oxidation, taking place consecutively. This study allowed us to predict the biosynthesis of tautomycetin more accurately and provided novel insights into the mechanism of the biosynthesis of tautomycetin.

        Automatic Detection and Classification of Rib Fractures on Thoracic CT Using Convolutional Neural Network: Accuracy and Feasibility

        Objective: To evaluate the performance of a convolutional neural network (CNN) model that can automatically detect and classify rib fractures, and output structured reports from computed tomography (CT) images. Materials and Methods: This study included 1079 patients (median age, 55 years; men, 718) from three hospitals, between January 2011 and January 2019, who were divided into a monocentric training set (n = 876; median age, 55 years; men, 582), five multicenter/multiparameter validation sets (n = 173; median age, 59 years; men, 118) with different slice thicknesses and image pixels, and a normal control set (n = 30; median age, 53 years; men, 18). Three classifications (fresh, healing, and old fracture) combined with fracture location (corresponding CT layers) were detected automatically and delivered in a structured report. Precision, recall, and F1-score were selected as metrics to measure the optimum CNN model. Detection/diagnosis time, precision, and sensitivity were employed to compare the diagnostic efficiency of the structured report and that of experienced radiologists. Results: A total of 25054 annotations (fresh fracture, 10089; healing fracture, 10922; old fracture, 4043) were labelled for training (18584) and validation (6470). The detection efficiency was higher for fresh fractures and healing fractures than for old fractures (F1-scores, 0.849, 0.856, 0.770, respectively, p = 0.023 for each), and the robustness of the model was good in the five multicenter/multiparameter validation sets (all mean F1-scores > 0.8 except validation set 5 [512 x 512 pixels; F1-score = 0.757]). The precision of the five radiologists improved from 80.3% to 91.1%, and the sensitivity increased from 62.4% to 86.3% with artificial intelligence-assisted diagnosis. On average, the diagnosis time of the radiologists was reduced by 73.9 seconds. Conclusion: Our CNN model for automatic rib fracture detection could assist radiologists in improving diagnostic efficiency, reducing diagnosis time and radiologists’ workload.

        The compressive behavior of CFRP laminates with delamination of different shapes using numerical and experimental approaches

        Composite laminates often produce delamination due to a series of factors during the manufacture and service process. In order to research the effect of containing oblique elliptical and circular initial delamination damage on the compressive strength of composite laminates, numerical and experimental methods are used in this paper. Finite element models (FEMs) and the progressive damage subroutine USDFLD are developed to predict the damage initiation and extension behavior of the intralaminar of the laminate. Interlaminar damage is predicted based on cohesive zone models (CZM). At the same time, four compression tests containing different initial delamination damage are performed. The results show that experimental measurements of compression tests concur with the numerical predictions and validate the FEMs. The strength and stiffness of the specimens gradually decrease with the increase of the initial delamination area. It reveals that, for the compression process, the delamination damage extends from the edge of the initial damage to the surroundings. The bearing capacity of the composite laminates is mainly determined by the material properties of the fibers and matrix, rather than the initial delamination.

        FRP Flexural Strengthened Concrete Member Design Method of Based on Early-Warning Fuses

        The bearing capacity of members strengthened with prestressed carbon fiber reinforced plastic is improved significantly, however, the ductility of the members strengthened are reduced obviously. Moreover, no sign are observed before the failure and the failure process rapidly. The current study proposed a reinforcement method based on early-warning fuses theory to address this issue and took carbon fiber reinforced plastic as an example. First, it's necessary to determine the area of carbon fiber board and the application of prestressed in each part of the member (early-warning fuses part and load-bearing part). This needs to be determined by the basic safety reserve and the additional safety reserve of the carrying capacity, then the calculation formula are derived; second, designing six simulation beams and four test beams. Based on the comparison of the computation of the derived formula with the simulation and test results, the calculated values of the early-warning load agree well with the simulated and experimental values, and the calculated values of the ultimate load are less than the simulated and experimental values. This showed that the theoretical calculation method has high accuracy. In addition, various evaluation index prove that the members strengthened have sufficient reserves.
