      • 통풍성 관절염의 임상적 고찰

        통풍은 Purine 대사의 이상으로 발생하는 질환으로, 고뇨산혈증인 사람의 전부가 통풍으로 발현되지 않고 증상의 출현 양상이 다양하여 진단과 치료에 주의가 필요하며, 조기에 적절한 조치를 한다면 충분히 조절이 가능한 질환이다. 본대학 내과학교실에서는 통풍으로 치료한 32명의 환자에서 임상양상, 병력과 검사소견을 분석하여 다음과 같음 결과를 얻었다. 1. 32명 모두 남자이며, 최초 발병 시기는 24세에서 72세로 평균 43.8±11.9세이었으며, 30대에서 40대까지가 19명(59.4%)으로 대부분을 차지하였다. 내원 당시 나이는 27세에서 75세까지로 평균 52.3±10.4세였으며, 내원시 까지 평균 유병기간은 8.5±6.8년으로 나타났다. 2. 동반 질환으로는 고지혈증 12례, 신장질환 10례, 고혈압 12례, 비만 8례, 당뇨 2례 등이었다. 3. 이환된 관절은 단관절 침범이 19례(59.4%), 다관절 침범이 13례 이었으며, 최초 이한된 관절은 족무지 중족골지골 관절로 19례(59.4%)로 가장 많았고, 통풍 결절은 20례(62.5%)에서 관찰되었으며, 유병기간이 10년 이상된 12례중 11례 (91.7%)에서 결절이 관찰되었다. 4. 평균 혈중 요산치는 9.17±1.75 ㎎/dl이었으며, 8.0 ㎎/dl에서 9.9 ㎎/dl 사이가 19명으로 전체의 59.4%를 차지하였다. Objective: The gout is a heterogeneous group of diseases resulting from tissue deposition of monosodium urate or uric acid crystals from extracellular fluids supersaturated with respect to this end product of human purine metabolism. The clinical manifestations are such as hyperuricemia, gouty arthritis, gouty nephropathy, uric acid nephrolithiasis. We analyze of clinical manifestations and associated factors in gout. Method: We have reviewed the medical records, radiologic findings and clinical results of thirty-two patients admitted at our department from April 1996 to July 1997. Result: 1) All patients were male. The mean age at initial attack was 43.8 years old, ranging from 24 to 72 years old. 2) The mean level of serum uric acid was 9.17mg/dl on admission. 3) The first metatarsopharyngeal joint was involved in 19 cases (59.4%). Tophus was observed in 20 cases (62.5%). 4) Hyperuricemia was associated with hypertension, obesity, nephrolithiasis and hyperlipidemia.

      • 인간 재조합 인터루긴-32 면역조절작용에 대한 유세포 분석

        Xenotransplantation of porcine organs has the potential to overcome the severe shortage of human tissues and organ available for human transplantation. however, it remains various hurdles for clinical xenotransplantation. In pig and mouse xenotransplantation, porcine xenograft evoke a strong cellular rejection response in immunocompetent host and grafts are destroyed within a week. This cellular immune response could involved both T cells and NK cells. A number of groups have shown that human NK cells can recognize and damage porcine endothelial cells. In addition, human T cells can respond to porcine endothelial cells through both direct and indirect mechanisms. Cellular rejection of porcine tissues requires T cells, particularly CD4^(+) cells. A new cytokine recombinant human interleukin-32α,β(IL-32α,β) has a role innate and acquired immune system. In order to investigate the role of recombinant mouse IL-18 and recombinant human IL-32α,β in xenograft rejection, we transplanted the PK(15) cells to C57BL/6 mice with or without intraperitoneal injection of recombinant mouse IL-18 or recombinant human IL-32 α,β. It was analyzed the population of NK cell, T cell and B cell in the C57BL/6 mice transplanted with PK(15) cells and recombinant mouse IL-18 or recombinant human IL-32α,β by flow cytometry analysis. As a result, lymph node and thymus of PK15/IL18, PK15/IL32α and PK15/IL32β injected group were increased to T cell activation population than normal injected groups. CD8^(+) T cells were decreased in lymph node of PK15/IL18, PK15/IL32α and PK15/IL32β injected groups. CD4^(+) T cells were increased in lymph node cell of PK15/IL32α and PK15/IL32β injected group and also, B cell population were increased in lymph node cell and spleen of PK15/IL18, PK15/IL32α and PK15/IL32β injected group. Therefore, we suggest that recombinant mouse IL-18 and recombinant human IL-32α,β suppress xenograft rejection in cellular xenotransplantation.

      • 초산류프롤리드 투여에 의한 흰쥐의 혈중 testosterone 억제와 뇌 시상하부에서 c-Fos의 발현

        Leuprolide acetate is a potent luteinizing hormone-releasing hormone analogue [leuprorelin, (des-Glyl0-D-Leu6-Pro-NHEt9)-LHRH acetate]. Lorelin depot, which is composed of leuprolide acetate, was designed for one-month release injectable and biodegradable microsphere of multiple high doses Here we compared the efficacy of microsphere lorelin depot with that of Takeda microsphere (Lucrin Depot). Lorelin (leuprolide 375 mg/kg of body weight) was administered sc to male rats, and the serum was obtained from rat tail vein. Enzyme immunoassay (EIA) for testosterone was carried out to assess the effect of lorelin depot. The rats were sacrificed 42 days after the lorelin β injection. and c-Fos changes of brain were observed by indirect immunofluorescence method. Results showed that the testosterone secretion was inhibited and the expression of c-Fos was also inhibited in the brain.

        Various cytokines including IL-1β, IL-2, IL-4, IL-10, IL-12p40, IFN-γ, and TNF-α have been recently reported to play important roles in both host and donor cell activations during xenotransplantation. Interleukin (IL)-18, a proinflammatory cytokine, can induce IL-4 and IL-13 production in T cells, NK cells, B cells, mast cells, and basophils. IL-18 has the capacity to stimulate innate immunity and both Th1-and Th2- mediated responses, but little is known to involve in the xenograft rejection. In order to investigate the role of IL-18 in xenograft rejection, we transplanted the pig kidney (PK15) cells to C57BL/6 mouse with or without intraperitoneal injection of mouse recombinatant IL-18. It was analyzed the population of T cell, B cell and NK cell in the mice transplanted with PK 15 cells and recombinant IL-18 by flow cytometry. We found that splenic CD3+ T cells were increased in mice injected PK15 cells with recombinant IL-18. These results suggest that recombinant IL-18 is critical in the xenograft rejection by increasing T cell population of the recipients.

        Leuprolide acetate is a potent luteinizing hormone-releasing hormone analogue [leuprorelin, (des-Glyl0-D-Leu6-Pro-NHEt9)-LHRH acetate]. Lorelin depot, which was composed of leuprolide acetate, was designed for one-month release injectable and biodegradable microsphere of multiple high doses. Here we examined the effect of microsphere lorelin depot, in comparison with Takeda microsphere (Lucrin Depot). Lorelin (leuprolide 3.75 ㎎/㎏ of body weight) was administered subcutaneous to male rats. The rats were sacrificed after 42 days and morphological changes of brain were observed by light microscopy. Leuprolide acetate has been shown to have an inhibitory effect on the growth of several hormone-dependent human tumors, So, leuprolide actate was examined for the cell cytotoxicity of the human endometrial cancer cell line, SNU-685, which was characterized in primary tumors of Korean patient and other cancer cell lines, MCF-7, T-24 and FB-l3P Cells were cultured with indicated concentrations of leuprolide acetate for 48 hr. Antiproliferative effect on SNU-685 cell which were determined by colorimetric method, MTT and MTS/PMS, was shown 20%, but those effects on MCF-7, T24, FB-23P cells were little shown.

        pH 변화에 따른 클로라민 생성과 분해 특성

        본 연구의 시료는 현재 서울특별시와 수도권 도시들에서 상수원수로 이용하고 있는 한강수를 대상으로 수행되었다. 배·급수계통의 2차 소독을 위한 클로라민소독의 특성에 관하여 고찰하였으며 그 결과는 다음과 같다. 1) 25 ℃, Cl_2 / NH_3-N비 1:1 ~ 14:1에서 pH6, 7, 8의 불연속점은 각각 Cl_2 / NH_3-N비 11:1, 9:1, 10:1이었다. pH6의 경우, Cl_2 / NH_3-N비가 9:1일 때 총 잔류염소량이 최대인 반면, pH7, 8의 경우 Cl_2 / NH_3-N비가 각각 6:1, 5:1일 때 최대가 되었다. 2) pH가 6, 7, 8로 증가함에 따라 모노클로라민이 최대로 생성되는 Cl_2 / NH_3-N비는 7:1에서 5:1로 이동하였으며, pH6 ~ 8 범위에서 pH가 증가함에 따라 모노클로라민의 생성량이 많았다. 3) 클로라민의 분해는 Cl_2 / NH_3-N비가 증가하고 pH가 낮을수록 증가하였다. 클로라민의 잔류성은 유리염소보다 월등히 좋았다. pH6에서 모노클로라민과 디클로라민이 생성된 이후, 접촉시간이 경과함에 따라 디클로라민의 분포비가 증가하였다. This study was conducted to investigate the characteristics of chloramination as a secondary disinfection in a drinking water distribution system. At the range from pH 6 to pH 8, monochloramine was predominant with a trace of dichloramine, and the free chlorine was detected after breakpoint. At 25℃, the breakpoints of pH 6, 7 and 8 appeared when the weight ratios of chlorine to ammonia nitrogen were 11:1, 9:1 and 10:1 respectively, and the peak points on the breakpoint curves at pH 6, 7 and 8 were in the Cl_2 / NH_3-N ratio of 9:1, 6:1 and 5:1 respectively. As pH increased from 6 to 8, maximum point of monochloramine on the breakpoint curve was moved from 7:1 to 5:1 in the weight ratio of chlorine to ammonia nitrogen. The maximum concentration of monochloramine was formed at the pH values of 7~8 and in the Cl_2 / NH_3-N ratio below 5:1. As the Cl_2 / NH_3-N ratio increased and the pH lowered, chloramines decay proceeded at an increased rate, and residual chloramines lasted longer than the residual free chlorine. The monochloramine and the dichloramine were formed at pH 6, and then the dichloramine continued increasing with contact lime.

      • 동결 보존에 의한 돼지 정자 세포질 칼슘 신호의 변화

        Although mammalian sperms are cryopreserved for in vitro fertilization a process of cryopreservation decreases the fertility. Acrosome reaction requires depolarization-induced ca^(2+) influx and ca^(2+) releases from the ca^(2+) stores. To examine whether the cellular ca^(2+) mobilization is altered by a sperm cryopreservation we compared cytosolic ca^(2+) signals between fresh and cryopreserved pig sperms using confocal ca^(2+) imaging. The magnitudes of depolarization-induced ca^(2+) increases were significantly smaller in cryopreserved sperms. Exposures to 10 mM caffeine or 5 μM thapsigargin elicited less ca^(2+) increases in the cryopreserved sperms compared to fresh sperms. In addition, progesterone-triggered ca^(2+) rises, that are thought to enhance acrosome reaction, were completely abolished in the cryopreserved sperms. These results suggest that storage and(/or) release of ca^(2+) from the intracellular ca^(2+) stores in pig sperms are significantly impaired by the process of cryopreservation.

        액체-고체 순환유동층의 상승관에서 액상의 점도가 고체체류량 및 열전달계수에 미치는 영향

        직경이 0.102 m이고 높이가 3.5 m인 액체-고체 순환유동층에서 액체의 점도가 고체입자의 체류량과 열전달계수에 미치는 영향을 검토하였다. 밀도가 2,500 kg/㎥인 유리구슬(d_(p)=1.0, 1.7, 2.1 또는 3.0 mm)과 CMC 용액(μ_(L)=0.96-38 mPas)을 각각 고체상과 액체상으로 사용하였다. 액체-고체 순환유동층에서 고체입자의 체류량은 액체의 유속이나 점도가 증가함에 따라 감소하였고 유동입자의 크기나 입자의 순환속도가 증가함에 따라 증가하였다. 액체-고체 순환유동층에서 열전달계수는 액체의 점도가 증가함에 따라 감소하였고 유동입자의 크기나 순환속도가 증가함에 따라 증가하였으나, 액체의 유속이 증가함에 따라서는 큰 변화가 없었다. 액체-고체 순환유동층에서 고체입자의 체류량과 열전달계수는 다음과 같이 실험변수와 무차원군에 의해 상관식으로 얻을 수 있었다. Glass beads (d_(p)=1.0, 1.7, 2.1 or 3.0 ㎜) whose density is 2,500 ㎏/㎥ and aqueous solutions of carboxymethy cellulose (CMC)(μ_(L)=0.96-38 mPas) have been used as the solid and liquid phase, respectively. It has been found that the solid holdup decreases with increasing liquid velocity or viscosity, but it increases with increasing solid particle size or solid circulation rate. The heat transfer coefficient decreases with increasing liquid viscosity but it increases with increasing particle size or solid circulation rate, however, it does not change considerablely with increasing liquid velocity. The solid holdup and heat transfer coefficient have been well correlated in terms of operating variables or dimensionless groups as fellow: ◁수식원문참조▷
