        Inhibitory Effect of Clavicepitaceae on Serotonin Release out of Human Platelets and Human Platelet Aggregation

        We have investigated the effects of hypha-water extracts (HWE), fruit body-water extracts (FWE) and cordycepin from Cordyceps militaris on serotonin release out of human platelets and human platelet aggregation. HWE and FWE inhibited the release of [^(3)H]-serotonin from human platelet stimulated by thrombin (2 U/ml) or collagen (20 ㎍/ml) in a dose-dependent manner. Furthermore, cordycepin, a major component of Cordyceps militaris, inhibited the human platelet aggregation induced by collagen (10 ㎍/ml) in a dose-dependent manner. These results suggest that cordycepin containing in HWE and FWE may inhibit the serotonin release by suppressing the collagen-induced human platelet aggregation. Accordingly, our data demonstrate that HWE and FWE containing much cordycepin might have antithrombotic and antimigrainous functions.

        Multi‑tissue lipotoxicity caused by high‑fat diet feeding is attenuated by the supplementation of Korean red ginseng in mice

        Background Excessive intake of fat, one of the causes of obesity, is associated with low-grade infammation in various susceptible organs and eventually causes tissue toxicity. This study examines the multifaceted suppressive efects of Korean red ginseng extract (KRG) on high-fat diet (HFD)-induced lipotoxicity and infammatory responses in the aorta, liver, and brain. Methods Male C57BL/6 mice were fed HFD with or without KRG for 12 weeks. The improvement efect in KRG on lipotoxicity and infammatory potential was determined in the blood and the aorta, liver, and brain tissues. Results KRG signifcantly inhibited 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity by >20% in vitro. KRG supplementation suppressed HFD-associated body weight gain, lipid profle changes, and excessive fat deposition in the liver and increased leptin, insulin, and ALT levels in the blood. Infammatory markers in the aorta, liver, and brain were also signifcantly reduced by KRG treatment. In microvascular endothelial cells, the 15% cyclic stretch-mediated upregulation of ICAM-1 and vascular cell adhesion protein-1 (VCAM-1) expression was signifcantly attenuated in the presence of KRG. Conclusion KRG supplementation attenuates HFD-mediated body weight gain, lipid profle changes, and multi-tissue infammatory responses.

        BTCA 처리 면직물의 질소계양이온화제 첨가에 의한 염색성 향상에 관한 연구

        The purpose of this study is to produce cotton fabric having improved dyeability and colorfastness as well as retaining the desirable durable press property. When cotton fabric was treated with additives, nitrogen content was increased as the additive concentration was increased. DP ratings and wrinkle recovery properties of BTCA treated cotton fabrics were greatly increased. But DP rating was lowered slightly by adding additives and wrinkle recovery property was decreased with the increase of additive concentration. Tensile strength was decreased by BTCA treatment but increased as additive concentration was increased. Various BTCA/additive treatments produced great affinity for reactive and acid dyes when dyebaths were adjusted at pH 3. It was showed that dyeability was increased as additive concentration was increased. The most effective additive for improving the dyeing properties of the crosslinked cotton was TEA·HCl In case of reactive dye, there was almost no improvement on the dyeability of finished cotton by adding salt. Color fastness to washing and light of finished and dyed cotton were low except for the washfastness of reactive dye. Dyeing the BTCA/additive treated cotton fabrics with reactive dye had no effect on the physical properties.

        We have used molecular biology technique to label the side chains and backbone of HBV preS1(1-56) with stable isotope (??) during E. coli over-expression and purified the protein to near homogeneity. The expression level of ?? labelled preS1(1-56) seems to be ∼15% of the total protein. to characterize the secondary structure of the labelled protein, circular dichroism was used. In different buffer conditions including pH 3.3 and pH 7.4 the protein seems to be devoid of any stable secondary structure probably suggesting that the N-terminal portion (preS1(1-56)) of HBV preS1 might be unstructured before binding to its target cells.

        Inhibitory Effect of Cordycepin on Human Platelet Aggregation

        Cordycepin separated from Cordyceps militaris is a major physiologic active component in Cordyceps militaris. The platelet aggregation is stimulated by Ca^(2+), which is either mobilized from intracellular endoplasmic reticulum or transported from extracellular space. cGMP antagonizes the actions of Ca^(2+). Based on these facts, we have investigated the effects of cordycepin on the mobilization of Ca^(2+) and the production of cGMP on collagen (10 ㎍/ml)-induced human platelet aggregation. Cordycepin potently stimulated the human platelet aggregation induced by collagen (10 ㎍/ml) in a dose-dependent manner. Cordycepin (500 μM) inhibited also the collagen-induced human platelet aggregation in the presence both 1 mM and 2 mM of CaCl_(2). These are in accord with the results that cordycepin inhibited the Ca^(2+)-influx on collagen-induced human platelet aggregation. These results suggest that cordycepin decrease the intracellular Ca^(2+) concentration to inhibit collagen-induced human platelet aggregation. Besides, cordycepin increased the level of cGMP on collagen-induced human platelet aggregation. This result is related with the decrease of intracellular Ca^(2+) concentration, because cGMP inhibits the mobilization of Ca^(2+). In addition, cordycepin inhibited the human platelet aggregation induced by LY-83583, inhibitor of guanylate cyclase. This result suggested that cordycepin inhibit the platelet aggregation by stimulating the activity of guanylate cyclase. In conclusion, we demonstrated that cordycepin might have the antiplatelet function by inhibiting Ca^(2+)-mobilization via the stimulation of the production of cGMP.

        유산소 리듬 운동프로그램이 노인의 혈압, 심박수 및 생활만족도에 미치는 효과

        The purpose of this study was to test the effects of aerobic rhythmical exercise program on blood pressure, pulse and the life satisfaction of the elderly. This quai-experimental study was designed as a non-equivalent control group pretest-posttest study. The subjects were 36 subjects(experimental group: 19 subjects, control group: 17 subjects), aged between 65 and 73 years who have normal cognition, sensory function and cerebellum function. Aerobic rhythmical exercise program was developed on the basis of aerobic exercise, Korean tradititional dance and music by the author. The blood pressure, pulse rate and life satisfaction were measured prior to and following the experimental treatment. Data were analyzed with x^2-test, t-test, mean, standard deviation, percentage of change and ANCOVA test using SAS program. The results were as follows. 1. There was no significant difference of systolic blood pressure between experimental and control group following the aerobic rhythmical exercise program(F=1.39, P=0.247). 2. The diastolic blood pressure of the experimental group was significantly lower than that of control group following the aerobic rhythmical exercise program(F=2.96, p<0.1). 3. There was no significant difference of resting heart rate between experimental and control group following the aerobic rhythmical exercise program(F=0.07, P=0.793). 4. The life satisfaction of the experimental group was significantly higher than that of control group following the aerobic rhythmical exercise program(F=9.94, p<0.01). On the basis of the above findings, aerobic rhythmical exercise program is effective in reducing diastolic blood pressure and promoting life satisfaction of the elderly. Thus, this program can be recommended as an effective nursing intervention for the elderly.

        Based on the data interpretation on an artificial marsh built in Chun-cheon Lake, the study analyzed flow characteristics and found that flow was unstable due to sediment of natural river but the effect of artificial marsh was similar with that of river improvement works. Flow velocity in the section of artificial marsh was found to be 1m/sec. Therefore flow velocity was stable, which could contribute to improving water quality. A flow velocity as well as stream vector was improved.

        홍국 풀을 이용한 김치 제조 및 품질 특성

        Kimchi was prepared with the addition of 2.5% and 5.0% Monascus purpureus koji(MPK) paste (20%) and were fermented at 20C for 18 days. The quality and sensory characteristics of the kimchi were evaluated by analyzing the pH, acidity, number of viable cells, the concentration of reducing sugar, and sensory properties during fermentation. The pH and titratable acidity of the kimchi prepared with MPK(MPK kimchi) were higher and lower, respectively, than those of the control kimchi. The MPK kimchi showed high 'L' and 'b' values during storage, but the 'a' values were low. The contents of the reducing sugar of the MPK kimchi tended to increase during fermentation, particularly after six days. The number of total microbial cells, lactic acid bacteria and yeast in the MPK kimchi were lower than those of the control kimchi until 3 days of fermentation. However, the number of these bacteria in the MPK kimchi and the control kimchi after six days of fermentation was similar. The sensory score of the kimchi with 2.5% and 5.0% added MPK paste were significantly higher than the control groups in terms of the sweetness and overall acceptability.

        To evaluate tensile behavior of groove-welded joints with alien substance, poor welding conditions, inserting an alien substance, are intentionally introduced in groove-welded joints which are widely used for joint connections in steel construction. Based on the results from the tensile tests, modulus of elasticity, tensile strength, failure mechanism, and the percent of elongation for the joints are investigated in this study. Tensile test results proved that modulus of elasticity was not affected by inserting an alien substance. Form the tensile test results on these joints with an alien substance, however, tensile strength, failure mechanism and the percent of elongation are strongly affected by the size and shape of alien substance introduced in the welded joints.
