Morus alba Linne. has long been used as a traditional medicine for the anti-diabetic, anti-hyperlipidemic, anti-oxidant, and anti-inflammatory effects. However, there are no studies on the effect of Morus alba Linne.(MB) related to muscular atrophy caused by oxidative stress. In this study, we conducted DPPH radical scavenging activity in water extracts of candidates medicinal plants from Korea and Mongolia. Also, we investigated the effects of Morus alba Linne. water extract (MBE) on the recovery and prevention of oxidative stress-induced muscular atrophy in mouse skeletal myoblast C2C12 cells. MBE was performed on H2O2-treated myoblasts for anti-atrophic activity; its effects on the growth, toxicity, reactive oxygen species (ROS) production; and apoptosis by antioxidant activity; and on the differentiation of myoblast into myotube were examined. As a results of free radical scavenging activity, MB has shown the highest DPPH radical scavenging activity in Korean herbal medicine, and CA and VA in Mongolian medicinal plants candidates at 0.1 ㎎/㎖. Also, we demonstrated oxidative stress induction that preincubation of C2C12 myoblasts with MBE before stimulated by H2O2 indicated significant inhibition of DNA damaged associated with the inhibition of ROS production. MBE also restored H2O2-induced mitochondrial dysfunction, prevented apopotosis by Bax/Bcl-2 ratio inducing cytosolic release of cytochrome C, and degraded of caspase and PARP in H2O2-stimulated C2C12 myoblastes. In addition, MBE supplementation counteracted H2O2-induced muscle cell atrophy and reversed activation of p-AMPK. Our study demonstrated that MBE can reverse the muscle cell atrophy caused by H2O2 through regulation of the AMPK signaling pathways, followed by inhibition of PARP. AP2α is essential for MUC8 gene expression in human airway epithelial cells

<P>Mucins are high molecular weight proteins that make up the major components of mucus. Hypersecretion of mucus is a feature of several chronic inflammatory airway diseases. MUC8 is an important component of airway mucus, and its gene expression is upregulated in nasal polyp epithelium. Little is known about the molecular mechanisms of MUC8 gene expression. We first observed overexpression of activator protein-2alpha (AP2α) in human nasal polyp epithelium. We hypothesized that AP2α overexpression in nasal polyp epithelium correlates closely with MUC8 gene expression. We demonstrated that phorbol 12-myristate 13-acetate (PMA) treatment of the airway epithelial cell line NCI-H292 increases MUC8 gene and AP2α expression. In this study, we sought to determine which signal pathway is involved in PMA-induced MUC8 gene expression. The results show that the protein kinase C and mitogen-activating protein/ERK kinase (MAPK) pathways modulate MUC8 gene expression. PD98059 or ERK1/2 siRNA and RO-31-8220 or PKC siRNA significantly suppress AP2α as well as MUC8 gene expression in PMA-treated cells. To verify the role of AP2α, we specifically knocked down AP2α expression with siRNA. A significant AP2α knock-down inhibited PMA-induced MUC8 gene expression. While dominant negative AP2α decreased PMA-induced MUC8 gene expression, overexpressing wildtype AP2α increased MUC8 gene expression. Furthermore, using lentiviral vectors for RNA interference in human nasal polyp epithelial cells, we confirmed an essential role for AP2α in MUC8 gene expression. From these results, we concluded that PMA induces MUC8 gene expression through a mechanism involving PKC, ERK1/2, and AP2α activation in human airway epithelial cells. J. Cell. Effects of essential oil from Korean medicinal herb on hair growth and graying

As interest in appearance increases in modern society, hair has become psychologically and socially important meaning and role. Accordingly, many studies have been conducted on therapeutic agents for alopecia and gray hair using natural substances with relatively few side effects. This study investigated the effect of essential oil extracted from Korean medicinal herb with hexane on hair growth and graying. We determined effect of herbal essential oil (OS) on hair growth compared with minoxidil (MNXD) in human hair dermal papilla cells (HDPCs). Also, effect of essential oil on melanogenesis was monitored in the mouse melanoma cells (B16F10) using α-MSH (alpha-melanocyte-stimulating hormone) as a positive control. In addition, cell proliferation was evaluated by MTT assay and cell cycle from flow cytometry in HDPCs. And expressions of hair growth-related gene (IGF-1, Wnt3a, Akt, ERK) were monitored by western blot. We assessed melanogenesis by measuring melanin contents and tyrosinase activity. As the results, cellular protein expression intensities of IGF-1, Wnt3a, Akt, ERK were increased in OS-treated group and MNXD group, compared to DW group, respectively. Also G1 phase and S phase increased in OS-treated group compared to DW group. But melanogenesis didn’t increase significantly in OS-treated group compared to DW group. These results suggest that OS from Korean medicinal herb has promotes hair growth. Therefore, it might be potential hair-promoting material to use for prevention and improvement of alopecia.

<P>ConclusionsThe quality of liver US for hepatocellular carcinoma screening remains suboptimal. Education for physicians who perform liver US examinations is unsatisfactory in Korea. We should make an effort to improve the quality of liver US and teach basic US techniques to physicians.</P>
