      • Curcumin과 Protoporphyrin류 화합물의 상호작용에 의한 형광특성 변화

        Curcumin, a dietary polyphenol from Curcuma longa L., has been reported to show many beneficial bioactivities. Protoporphyrins are widely distributed in various natural sources. They are also intermediates in the endogenous synthesis of heme and commonly form a complex with metal ions. In the present study, we investigated changes in fluorescence properties of curcumin and protoporphyrin IX (PPIX) through their interactions. Fluorescence intensity of curcumin increased concentration-dependently showing a peak emission at 535 nm with an excitation at 440 nm. Curcumin fluorescence was not changed by PPIX; the fluorescence intensity of PPIX significantly decreased in the presence of curcumin. Curcumin fluorescence decreased time-dependently; the fluorescence from PPIX was rather enhanced according to time, which was less pronounced in the presence of curcumin. Curcumin fluorescence in cells decreased markedly, which is slightly enhanced by co-treatment of PPIX. Curcumin fluorescence also decreased by zinc protoporphyrin; the fluorescence from ZnPP was also quenched by curcumin. The present results suggest a possible interaction of metal porphyrins with curcumin, which may influence on their bioactivities.

        다인종 · 다민족국가인 미국에서 아시아계 미국인은 성공한 이민자 집단으로서의 ‘모델 마이너리티’로 평가받고 있다. 아시아계의 높은 학력수준과 경제적 성취도는 모범적인 이민자집단으로서 비추어지기에 충분했다. 하지만 아시아계 미국인,특히 그 중에서도 재미한인의 높은 학력이 미국사회로의 진출과 성공에 어느 정도의 영향을 미치고 있는지,그리고 그러한 영향력은 백인이나 히스패닉,흑인들과 어떠한 차이를 가지고 있는지에 대한 의문에서 본 연구는 시작되었다. 각종 통계자료와 문헌,그리고 설문조사를 통하여 볼 때,높은 교육수준이 사회 · 경제적 성공과 지위획득에 영향을 줄 것이라는 교육에 대한 높은 기대치는 입시와 취업에 있어서 미국인들과는 다르게 적용되는 기준으로 인하여 더욱 고학력을 지향하던가,아니면 반대로 종족집단으로의 회귀현상으로 나타나게됨을 알 수 있었다. 결국,다문화사회에 대한 반다문화주의의 등장은 종족적 · 인종적 · 계급적인 보편성을 인정하게 되면 사회가 분열할지도 모른다는 걱정과,다른 문화에 대한 우려로 인하여 나타난 것이라 할 수 있다. Asian Americans as ethnic minohties who,despite marginalization, have achieved successin the United States are spoken of as a'model minority' group because the group hasbeen argued to be more successfulcomparatively than other minority groups inthe United States. The new Amehcanimmigration law in 1965 holds preference forskilled workers and those with collegedegrees; as such, many Asian immigrantsare well-educated before coming to Americaand are often in a upper-middle economicbracket. Therefore, Asians especially thosewho are highly educated and skilled startedto settle down in the United States after themid 1960s. For example, Asian Americanshave accounted for a large percentage ofsome of Amehca's most prestigious universities.The median household income of AsianAmehcans is also higher than the totalpopulation's income. The factors of incomeand educational attainment have made theterm 'model minority' pointing out thesuccess of Asian Amehcans such as Chinese,Korean, and Japanese. Compared to Chineseand Japanese, Korean immigrants tend toblend in quickly with the white upper middleclass while descendants of earlier immigrantsfrom China and Japan have a much more different and difficult history.The aim of this study is to examine theinfluence of the Asian Amehcans' academicsuccess on their advance in the United Statesfocusing on Korean Amehcans. Culturalfactors are mainly thought as a one of thereasons why Asian Americans are successfulin the United States. East Asian societiesthemselves, in general, often place moreresources and emphasis on education. Accordingto the statistics and the survey, however,some Asians presume that they are stillfacing racial discrimination even if they aresuccessful in education. Asian Amehcans, asa minority group, tend to depend upon theirethnic business instead of advancing into themainstream of the United States. SinceAsian Amehcans' status in the United Statesis in discord with their educationalattainment, exclusivism germinated alongwith anti-multiculturalism. In other words,anti-multiculturalism is the result ofconcerns that the society might be disunitedor adversed by recognizing the generality inethnic, race, class.

      • 간세포암환자에서 간동맥 화학 색전술 후 발생한 리피오돌에 의한 폐렴 1예

        Hepatocellular carcinoma(HCC)is one of common causes of cancer-related death in Korea where the majority of HCC patients were Hepaitc B virus(HBV)carriers and have cirrhosis. Transarterial chemoembolization(TACE)is commonly applied to the treatment of multinodular HCC in Korea and careful selection of candidate is important for the risk of various side effects. Besides common side effects as fever, nausea, abdominal pain and elevation of liver enzyme, TACE may predispose to hepatic failure, ischemic cholecystitis, pulmonary embolism, cerebral embolism and pneumonitis. In previous studies, some cases of pulmonary and cerebral embolism cases were reported but lipiodol pneumonitis after TACE was rarely reported. A 65-year-old woman with a multinodular HCC associated with HBV infection, was treated with TACE. Seven days after the procedure, nonspecific respiratory symptoms such as dyspnea and dry cough developed. Chest X-ray and chest computed tomography showed diffuse ground glass opacities in whole lung fields, suggestive of lipiodol pneumonitis. After several days of supportive care with steroid administration, radiologic abnormalities and subjective symptoms were much improved, considered that the disease was compatible with lipiodol pneumonitis.

      • Zinc protoporphyrin과 다양한 용매조건이 MTT formazan의 발색 안정성에 미치는 영향

        An assay method using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltertrazolium bromide (MTT) is commonly utilized for analyzing cell viability. In this method, MTT tetrazolium is converted to formazan by mitochondrial reductase of viable cells. The formzan formed is, however, not stable; the stability is affected by various factors. In the present study, effects of zinc protoporphyrin (ZnPP) and different solvents on color response of formazan dye were investigated. MTT formazan was stable under dark; light enhanced the color degradation of formazan. ZnPP induced rapid degradation of formazan under light condition in DMSO solution. Decolorization of formazan was faster in aqueous solvents, including distilled water, HCl as well as Triton X-100 solution. Formazan dye was most unstable in HCl (0.1 N) regardless of light or presence of ZnPP. Color response of formazan dye in NaOH (0.1 N) was enhanced by ~2 folds as compared to one in DMSO. MTT formazan was stable in this condition; the color change was less than 10% even under light during 2 hr. ZnPP, however, induced rapid decolorizaion of formazan dissolved in NaOH under light. H2O2, an oxidizing agent, did not affect color response of MTT formazan. The present results suggest that various factors could affect the color response of MTT formazan, which needs to be considered carefully in MTT assay.         MTA와 4-META/MMA & TBB레진 혼합 재료의 치근단 미세누출에 관한 연구

        이 연구에서는 Mineral Trioxide Aggregate (MTA)와 4-methacryloxyethyl trimellitate anhydride (4-META)/methyl methacrylate (MMA) & tri-n-butylborane (TBB) 레진을 혼합하여 치근단 역충전 재료로 사용하였을 때의 미세누출을 MTA, 포틀랜드 시멘트와 비교하였다. 발치된 사람 치아의 근관형성 및 충전 후 치근단 절제와 역충전 와동을 형성하였다. MTA, Portland cement, MTA와 4-META/MMA & TBB레진을 혼합한 각각의 재료로 치근단을 역충전한 후 methylene blue dye에 72시간 동안 침적하여 염료가 침투한 길이를 10배 현미경 하에서 측정하였다. MTA와 4-META/MMA & TBB 레진 혼합 충전 군에서는 포틀랜드 시멘트나 MTA 충전군보다 통계적으로 유의하게 적은 미세 누출을 보였으며 , 포틀랜드 시멘트와 MTA 충전군의 미세 누출 정도는 통계적으로 차이가 없었다. 이 실험의 결과로 보아 MTA와 4-META/MMA & TBB레진 혼합 재료는 치근단 역충전시 포틀랜드 시멘트나 MTA에 비해 낮은 초기 미세누출을 보여, 치근단 봉쇄 효과가 우수한 것으로 사료된다. We evaluated in vitro microleakage of Mineral Trioxide Aggregate (MTA) powder with 4-methacry-loxyethyl trimellitate anhydride (4-META) / methyl methacrylate (MMA) & tri-n-butylborane (TBB) resin as a retrograde filling material by using methylene blue dye method. Fifty-two single rooted, extracted teeth were instrumented and obturated with gutta percha and AH plus sealer. The apical 3mm of each root was resected and 3mm deep ultrasonic root end preparation was done. External surface of roots was coated with nail varnish. Prepared teeth were randomly divided into five groups: Negative control: completely covered with nail varnish; Positive control: coated with nail varnish except for apical foramen; Group 1 (retrofilled with Portland cement); Group 2 (retrofilled with MTA) ; Group 3 (retrofilled with MTA powder mixed with 4-META/MMA & TBB resin). Immediately after completion of root-end filling, all specimens were submerged in methylene blue dye for 72 hours in 37℃ incubator. The roots were longitudinally sectioned and measured for extent of dye penetration by three different examiners under microscope (×10). The results were statistically analyzed using one way ANOVA and Turkey's HSD test. No leakage was evident in negative control and complete leakage in positive control group. Group 3 showed significantly less leakage than group 1 and 2 (p < 0.01). There was no significant difference between group 1 and 2 (p > 0.01). It was concluded that MTA powder with 4-META/MMA & TBB resin was excellent in reducing initial apical microleakage. 〔J Kor Acad Cons Dent 34(4):371-376, 2009〕

        차아염소산나트륨 처리와 멸균법이 근관 치료용 파일의 부식에 미치는 영향에 관한 연구

        근관 치료시 많이 사용되고 있는 K-flex S-S 파일 (Maillefer, USA)과 Profile Ni-Ti 파일 (Maillefer, USA), K-3 Ni-Ti파일 (SybronEndo, USA)의 부식 정도를 보기 위하여 총 360개의 20번 file을 1) 멸균소독 방법 (고압가열멸균소독, E-O gas 소독) ,2) 근관세척 액 (5.25% 차아염소산나트륨, 생리식염수) ,3) 멸균소독 횟수 (1, 5, 10회 ) 에 따라 36개 실험군으로 나누었다. 각각의 파일을 각 군별로 처리한 뒤, 세 명의 검사자가 광학 현미경 하에서 25배의 배율로 검사하여 0; 부식 없음, 1; 경도의 부식, 2; 중등도의 부식, 3; 심한 부식으로 점수화 하였다. 심한 부식을 보이는 파일의 표면은 SEM으로 관찰하였고 Kruskal-Wallis test (p < 0.05)를 이용하여 통계 분석 처리하여 차아염소산나트륨 접촉 여부와 멸균소독 방법의 차이에 의한 부식 정도를 평가하였다. 그 결과 고압가열멸균소독을 10회 시행한 파일은 통계적으로 유의한 차이를 보여 부식 정도가 심하였고, 1회나 5회 고압가열멸균소독을 시행한 파일과 E-O 가스로 소독한 파일들은 부식의 정도가 경미하여 통계적으로 유의하지 않았다. 차아염소산나트륨 접촉 여부와 제조사 혹은 파일 재료에 따른 유의성 있는 차이는 없었다 A variety files made of stainless steel (S-S) or nickel-titanium (Ni-Ti) are used during endodontic treatment. The purpose of tt)is study was to evaluate the corrosion susceptibility of S-S and Ni-Ti endodontic files. Three brands of files were used for this study: K-flex S-S files (Maillefer, USA), Profile Ni-Ti files (Maillefer, USA), K-3 Ni-Ti files (SybronEndo. USA). 120 files of each brands (21mm, ISO size #20) were divided into 12 groups according to 1) sterilization methods using Autoclave or Ethylene Oxide (E-O) gas, 2) Irrigation solutions using 5.25% NaOCl or Saline, 3) the number of sterilization (1, 5, 10 times), After above procedures, each of the files was inspected by three examiners with a light microscope and camera at X25. Each file was judged and ranked according to the following criteria: 0;, no corrosion, 1; mild corrosion, 2; moderate corrosion, and 3; severe corrosion. The files of high score were examined under the Scanning Electron Microscope. Data were statistically analyzed with the Kruskal-Wallis test (p < 0.05). Most of the ten time-autoclaved files had showed mild to moderate corrosion. But, one or five time-autoclaved files did not show corrosive surface. NaOCl treatment and E-O gas sterilization did not influence on corrosion. There was a significant difference in corrosion susceptibility between sterilization methods and the number of autoclaving. However, there was no significant difference between brands and file materials.

        Pathogenomics of Streptococcus ilei sp. nov., a newly identified pathogen ubiquitous in human microbiome

        Viridans group streptococci are a serious health concern because most of these bacteria cause life-threatening infections, especially in immunocompromised and hospitalized individuals. We focused on two alpha-hemolytic Streptococcus strains (I-G2 and I-P16) newly isolated from an ileostomy effluent of a colorectal cancer patient. We examined their pathogenic potential by investigating their prevalence in human and assessing their pathogenicity in a mouse model. We also predicted their virulence factors and pathogenic features by using comparative genomic analysis and in vitro tests. Using polyphasic and systematic approaches, we identified the isolates as belonging to a novel Streptococcus species and designated it as Streptococcus ilei. Metagenomic survey based on taxonomic assignment of datasets from the Human Microbiome Project revealed that S. ilei is present in most human population and at various body sites but is especially abundant in the oral cavity. Intraperitoneal injection of S. ilei was lethal to otherwise healthy C57BL/6J mice. Pathogenomics and in vitro assays revealed that S. ilei possesses a unique set of virulence factors. In agreement with the in vivo and in vitro data, which indicated that S. ilei strain I-G2 is more pathogenic than strain I-P16, only the former displayed the streptococcal group A antigen. We here newly identified S. ilei sp. nov., and described its prevalence in human, virulence factors, and pathogenicity. This will help to prevent S. ilei strain misidentification in the future, and improve the understanding and management of streptococcal infections.
