        정상 한국인에서 연령증가에 따른 혈장 Dehydroepiandrosteronesulfate ( DHEA-S ) 농도 및 DHEA - S / cortisol 비의 변화

        Backgroud: DHEA-S is the most abundant steroid hormone in circulation, and primarily secreted from the adrenal cortex, but its physiological role is little known. One of the characteristic features of DHEA-S is progressive decrement of plasma DHEA-S level with advancing age, in contrast, plasma levels of other adrenal hormones are not chaging or littie decreasing. To grasp the trends of plasma DHEA-S level and DHEA-S/cortisol ratio by age in healthy Korean, we measured the plasma DHEA-S levels and DHEA-S/cortisol ratios in healthy Korean. Methods: Healthy Korean(men: 99, women: 102, age range: 15-97 year old)were studied. Subjects were not taking drugs(such as glucocorticoid or androgenic medication) or cigarettes known to modify the plasma level of DHEA-S and cortisol, and had no evidence of hepatic, renal disease or hyperlipidemia as determined by serum lipid, bilirubin, SGOT, SGPT, BUN, creatinine. Data were analyzed by 10-year age group for men and women: i.e, 10-19, 20-29, 30-39, 40- 49, 50-59, 60-69, 70-79, 80-89 and 90 year or more. Plasma DHEA-S levels were measured by using a commercially available RIA kit with 125I labeled-DHEA-SO4(Coat-A Count DHEA-SO4), and for the measurement of plasma cortisol levels, commercial Gamma Coat TM[125I] Cortisol Radioimmunassay Kit was used. Results:. 1) In both men and women, plasma DHEA-S level showed high interindividual variation within the same age group. 2) There were individual sex differences in plasma levels of DHEA-S, in all age groups, plasma DHEA-S levels were significantly higher values for men than for women. 3) Maximum plasma DHEA-S levels(men; 237+-3.35 ug/dL, women; 108+-17.5 ug/dL) were at third decade in both men and women. 4) Both men and women showed the continuous decline in plasma DHEA-S level with age. These age-related decline was more prominent in men than in women(men; y= -3.152 * +292.6, r2= 0.8459, P$lt;0.05, women; y= -1.417 * +143.3, r2 = 0.7278, P$lt; 0.05). 5) As an index of aging, there was no stastical difference between DHEA-S and DHEA- S/cortisol ratio. Conclusion: In healthy Korean, there were high interindividual variation of plasrna DHEA-S levels. In both men and women plasma DHEA-S level was peak at third decade, and from when it declined progressively with age. These results suggest that although the reliability of single plasma DHEA-S measurement are limited, the decline of DHEA-S with advancing age might be a specific marker of endocrinologic hormonal milieu(aging index). Also, concerning to individual adrenal secreting capacity, we measured DHEA-S/cortisol ratio. But we did not found that plasma DHEA-S/cortisol ratio is superior to the plasma DHEA-S level as an aging index. (J Kor Soc Endocrinol 12:245-254, 1997)

      • T 세포활성항원 CTLA-4의 기능에 관한 연구 : T 세포에서 표적세포 살해능과 CTLA-4 발현과의 연관성 Target Cytotoxicity of T Cell Correlates with CTLA-4 Production

        CTLA-4, a T cell activation antigen and a homologue of CD28, was originally identified as a gene by a series of reverse genetic approaches. While CD28 molecule has been characterized well as a stimulator of T-cell function via enhanced lymphokines production and stablilization of those mRNA, most of the functions of CTLA-4 remain unknown. It has been widely accepted that CTLA-4 functions as an immune suppressor which is down-regualting the function of CD28. We previously showed that 34 KD form of this antigen mainly expressed CD8+ subset, a cytotoxic or suppressor, of activated peripheral blood lymphocyte. Based on our previous finding this study was conducted to further characterize immunological function of CTLA-4 especially in terms of involvement of this molecule in T-cell effector function mediating target cell cytotoxicity. 4 human T cell clones with different target cytotoxicities were employed in this study. NBL46 (CD4+) and NBL77 (CD8+) were cytotoxic and NBL32 (CD4+) and NBL58 (CD 8+) were non-cytotoxic to target LBF cell in target cell chromium release assay. And in Western blot assay 34 kD antigen was detected in NBL46 (CD4+) and NBL77 (CD8+) clones but not in NBL32 (CD4+) and NBL58 (CD8+) clones. It was summarized that expression of the CTLA-4 antigen was associated with cytotoxicity but not with subset phenotypes of T cells. In conclusion CD8+ T subset of PHA-stimulated PBL was major and only CTLA-4 producer and this molecule was induced during mid to late stage of T cell activation. The cytotoxicity of T cell clones to its target cell was directly correlated with its CTLA-4 production and vice versa. And it was highly suggested that primary function of CTLA-4 may involved in T cell effector function which mediates target cell killing.

      • 작약의 RAPD 분석을 위한 PCR 최적조건 구명

        To optimize the PCR condition is one of the most important steps for RAPD analysis. The purpose of this study was to optimize PCR condition in Paeonia. 3×3×3 factorial experiment for template DNA concentration, MgCl2 concentration, and amount of taq polymerase was conducted. Another factorial experiment for reaction temperature(denature, annealing, and extension) was also conducted. The most appropriate template DNA concentration was 60ng. Clear bands were observed from 2.5mM and 4.5mM of MgCl2 if template DNA concentration and amount of taq polymerase were proper. Amount of taq polymerase for the optimal PCR condition was 0.5unit and 1unit. In the consideration of results from template DNA concentration, MgCl2 concentration, and amount of taq polymerase, 4 conditions (60ng of template DNA, 2.5mM MgCl2 and 0.5unit taq polymerase; 60ng of template DNA, 4.5mM MgCl2 and 0.5unit taq polymerase; 60ng of template DNA, 4.5mM MgCl2 and lunit taq polymerase; and 40ng of template DNA, 4.5mM MgCl2 and 1unit taq polymerase) were best combinations for the optimal PCR condition. Reaction temperature for the optimal PCR condition was 92℃, 36℃, 72℃.

      • 다이아몬드상 탄소 박막이 코팅된 실리콘 전계 방출 소자의 특성

        We have fabricated the field emitter arrays (FEAs) coated with diamond-like carbon (DLC) films. The turn-on field of DLC-coated FEAs was rapidly decreased and the emission current was remarkably increased compared with the non-coated silicon FEAs. In the nitrogen doped FEAs, the turn-on field decreased and the emission current increased with nitrogen doping concentration. From the Fowler-Nordheim plot, the work functions of the DLC-coated FEAs are much lower than those of the non-coated silicon FEAs. As nitrogen doping concentrations are increased, the work function of FEAs is decreased and the field emission properties are improved in nitrogen doped DLC-coated FEAs. This phenomenon is due to the fact that the Fermi energy level moves to the conduction band by increasing nitrogen doping concentration.

        적외선 열화상을 이용한 비파괴시험 활용 및 결함 진단

        본 고는 적외선 열화상의 개념과 적외선일화상 측정원리 및 카메라 셋팅을 기술하고 적외선일화상(IRT)에 의한 비파괴검사(NDT)의 활용 및 비파괴검사의 진단에 대하여 살펴보았다. 적외선 일화상은 주어진 표면을 따라 온도에 관련된 일패턴의 평가를 통한 정기적인 비접촉, 비파괴 시험의 수행으로 초기에 장비 고장의 예방이 가능하다. 진단 활용으로서, 열적으로 가열된 내부결함이 있는 블록에 대하여 적외선열화상을 이용한 열화상 패턴을 비파괴 기법으로 평가하고, 결함과 열화상패턴간의 특성을 분석하여 열화상 평가 기법에 대하여 논하였다. In this paper, the concept of infrared thermography(IRT), the principle of measurement of IRT and how to set up the 111 camera for the nondestructive testing are described in detail. Also, its utilization and non-destructive testing(NDT) diagnosis are reviewed. By performing the periodic non-touched NDT through the estimation of thermal patterns related with the temperature for the surface targeted, IRT can be applied to the early prevention of the device failure. For the diagnosis utilization, thermal imaging patterns obtained from IRT for heated blocks with internal defects were estimated through the non-destructive method and discussed the way of IRT estimation from the analysis of characteristics between material defects and thermal imaging patterns.

        헬리코박터 파이로리 균 진단용 ^13C-요소 캅셀의 개발

        The purpose of this study was to develop a new ^13C-urea- containing capsule for diagnosis of H. pylori. The urea-containing capsules were prepared with various diluents such as polyethylene glycol (PEG), microcrystalline cellulose, sodium lauryl sulfate and citric acid. The dissolution test, ^13C-urea breath test and stability test were then performed on the capsules. Microcrystalline cellulose and sodium lauryl sulfate retarded the initial dissolution rates of urea. However, PEG increased the initial dissolution rates of urea. Furthermore, two formulae composed of PEG, [^13C-urea/PEG (38/1.9 mg/cap)] and [^13C-urea/PEG/citric acid (38/1.9/1.9 mg/cap)] had the maximum DOB value, about 16 at 20 mim, while the formula composed of only 38 mg ^13C-urea had the maximum DOB value at 30 min. The results indicated that PEG improved the sensitivity of ^13C-urea in the human volunteers. The capsule [^13C-urea/PEG (38/1.9 mg/cap)] was stable for at least six months in 25 and 37℃. Thus, a PEG-containing capsule, [^13C-urea/PEG (38/1.9 mg/cap)] would be a more economical, sensitive and stable preparation for diagnosis of H. pylori.

        In the past few decades, the measuring instrument industries have made rapid progress with increase of the demand in industries such as precision instrument, chemistry, electronics, industrial measurement and automobile. Thus, development strategy and technical advancement are required for both securing the competitiveness of products in the world market and industrial development. Intensifying competitiveness of measuring instrument is needed in the world market. It can be achieved by development of measuring instrument holding comparative priority. A development strategy should be drawn up in the long-term point of view for coping with the increasing demand of measuring instrument and securing competitiveness in the world market.

      • 팥의 RAPD 분석 조건 최적화 연구

        The object of this study was to optimize PCR condition for RAPD analysis in adzuki bean. The best template DNA concentration was 20ng(0.25unit taq polymerase and 2.5mM MgCl2), 40ng(1unit taq polymerase and 2.5mM MgCl2, 1unit taq polymerase and 4.5mM MgCl2 and lunit taq polymerase and 7.0mM MgCl2), and 60ng(0.5unit taq polymerase and 2.5mM MgCl2, 0.5unit taq polymerase and 4.5mM MgCl2, 1unit taq polymerase and 2.5mM MgCl2, and 1unit taq polymerase and 4.5mM MgCl2) The best MgCl2 concentration was 2.5mM(40ng template DNA and 1unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq polymerase), 4.5mM(20ng template DNA and 0.25unit taq polymerase, 60ng template DNA and 0.5unit taq polymerase, and 60ng template DNA and 1unit taq polymerase), and 7.0mM(40ng template DNA and 1unit taq polymerase). Amount of taq polymerase was 0.25unit(20ng template DNA and 2.5mM MgCl2), 0.5unit(60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2) and 1unit(40ng template DNA and 2.5mM MgCl2, 40ng template DNA and 4.5mM MgCl2, 40ng template DNA and 7.0mM MgCl2, 60ng template DNA and 2.5mM MgCl2, 60ng template DNA and 4.5mM MgCl2, and 60ng template DNA and 7.0mM MgCl2). When we consider results from template DNA concentration, MgCl2 concentration, and amount of taq polymerase, the best condition for PCR optimization was 60ng template DNA, 4.5mM MgCl2, and 1unit taq polymerase. Reaction temperatures for the optimal PCR condition were 84℃, 32℃, 62℃; 90℃, 40℃, 72℃; and 92℃, 36℃, 72℃.

        프레탈정 (실로스타졸 100mg)에 대한 실로스탄정 (한국유나이티드 제약)의 생물학적 동등성

        Bioequivalence of one cilostazol tablets, the Pletaal^(R)(Korea Otsuka Pharmaceutical Co., Ltd.) and Cilostan^(R)(Korea united Pharmaceutical Co., Ltd.), was evaluated according to the guideline of KFDA. Sixteen normal male volunteers(age 20~30 years old)were divided into two groups and a randomized 2×2 cross-over study was employed. After one tablet containing 100㎎ of cilostazol were orally administered, blood was taken at predetermined time intervals and the concentration of cilostazol in plasma was determined with an HPLC method using UV detector. The pharmacokinetic parameters(C_(max), T_(max) and AUC_(t)) were calculated and ANOVA was utilized for the statistical analysis of parameter. The results showed that the differences in C_(max), T_(max) and AUC_(t) between one tablet were 16.08%, 18.88% and 17.57%, respectively. The powers (1-β) for C_(max), T_(max) and AUC_(t) were 85.03%, 83.92% and 80.12%, respectively. Detectable differences(Δ) and confidence intervals were all less than 20%, and confidence interval of all the parameters were also less than 20% at the significance level(α) of 0.05. All of these parameters met the criteria of KFDA for bioequivalence, indicating that Cilostan^(R) tablet is bioequivalent to Pletaal^(R) tablet.

        정밀부품 및 기기에 대한 환경시험기술

        정밀부품이나 기기는 개발과정이나 완료단계에서 부적절한 설계에 의하여 발생할지도 모르는 잠재적인 결함을 방지하기 위하여 시험평가를 실시하게 된다. 본 논문에서는 신뢰성 기술 분야의 기초가 되는 환경시험기술을 소개 하였으며 엔코더, 교통신호제어기, 계측기기 및 위성항법 수신기 등 제품단위의 대상품에 대한 시험조건 및 방법 등을 기술하고 시험사례를 보고하였다. 이러한 사례를 통하여 제품의 신뢰성 향상을 위한 복합환경시험의 중요성을 강조하였고 최근 환경시험기술에 대한 인식이 날로 증대되면서 시험기술자 양성, 지그설계기술 그리고 고장해석 및 대책기술의 확보가 매우 필요함을 제안하였다. Precision parts and instruments are tested to evaluate performance in development-process and product-step to prement a potential defect due to a failure design. In this paper, Environmental test technology, which is the basis of reliability analysis, is introduced with examples of test criterion, test method for products, encoder and traffic signal controller, and measuring instruments. Recently, as the importance of the environmental test technology is recognized, It is proposed that tranining of test technician and technology of jig design and failure analysis are very essential.
