        Cloning of porcine chemerin, ChemR23 and GPR1 and their involvement in regulation of Lipogenesis

        Chemerin is a novel adipokine which is abundant in adipose tissue to promote adipocyte differentiation and with significant relativity to BMI and insulin sensitivity. We report here the molecular characterization of porcine chemerin and its receptors ChemR23 and GPR1, as well as their transcriptional regulation during lipogenesis. Chemerin was mainly expressed in liver, intestine, kidney and adipose tissue, consistent with the expression pattern of GPR1, but not ChemR23, which was predominantly present in spleen and temperately in adipose tissue. We further investigated the lipogenesis-related transcriptional activation of PPARγ and KLF15 on chemerin and its receptors. The data showed that KLF15, but not PPARγ, can up-regulate the mRNA level of chemerin, ChemR23 and GPR1, which was consistent with the results of luciferase assay that confirmed the effect of KLF15 on ChemR23 promoter. Taken together, our data provide basic molecular information for the further investigation on the function of chemerin in lipogenesis. [BMB reports 2010; 43(7): 491-498]

        Enhanced Sliding Mode Control and Online Estimation of Optimal Slip Ratio for Railway Vehicle Braking Systems

        This paper proposes an optimal anti-locking control scheme so as to improve the braking performance of railway vehicles. The controlling effect of sliding mode control is improved, and the optimal slip ratio is achieved by extreme seeking algorithm. Firstly, a substitute function for the conventional sign function is proposed. Secondly, a closed loop observer for braking systems is used to enhance the estimation value of adhesion force, which can also be used for calculating reference speed. Finally Sliding Mode Controlbased controller needs to be entered a reference slip ratio called optimal slip ratio, which is searched by extreme seeking algorithm from the functional relationship between slip ratio and friction coefficient. Thus, the maximum adhesion is achieved despite wheel/ rail surface changes. The simulation result demonstrates the effect of real-time adjustment for braking torque, which guarantees the braking performance.

      • Fusion Detection Algorithm for Infrared Dim Target based on Temporal-Spatial Domain Accumulation and Difference

        Aiming at the problem of the dim target real-time detection in Infrared search and track (IRST) systems, a fusion detection algorithm for infrared dim target based on temporal-spatial domain accumulation and difference is proposed. The proposed algorithm suppresses background and enhances targets by template filtering and difference processing in space domain, and the image sequences are processed by multi-frame energy accumulation and frame difference methods according to the target’s moving characteristics in time domain. After the fusion segmentation of the temporal and spatial processing results with definite rule, the target can be detected by the relation of the possible target’s position in adjoining frames according to target’s moving continuity and regularity. Experimental results show that the proposed algorithm enormously increases the target's SNR after the temporal-spatial fusion enhancement, and it has high detection probability and high detecting speed. In the meantime, the algorithm is easy to realize in hardware and can be applied effectively in the real-time target detection of the IRST systems.

        Statistical Analysis of Microhardness Variations in Plasma Sprayed $Cr_3C_2-NiCr$ Coatings

        The microstructure and properties of plasma-sprayed coatings depend on a great number of spraying parameters, random factors, which lead to vibration in these spraying parameters, may in some degree influence the microstructure and properties of the coatings. Therefore, the property values appear certain distributions, and the description and comparison of the properties of plasma-sprayed coatings should be performed employing statistical analysis. In this paper, $Cr_3C_2$-Nicr coatings of different thickness were sprayed onto stainless steel using atmosphere plasma system and adopting three kinds of gun translation speeds. Then the microhardness measurements were performed on polished surface of the coatings. Forty readings were taken and statistically analyzed by calculating the characteristic values, estimating and comparing the means, and assessing whether they belonged to the Normal or Weibull Distribution. This study has found that statistical analysis could discriminate influence of spraying parameters and coating design on microhardness of the $Cr_3C_2$-Nicr coatings from random vibration, which showed that the microharness of the $Cr_3C_2$-Nicr coatings were related to gun translation speed coating thickness.

        Updated SSDP Scheme for DDoS Attack Defense

        Abusing the Simple Server Discovery Protocol (SSDP) can induce an SSDP attack (including SSDP DoS, DDoS, DRDoS) posing a significant threat to UPnP devices. Rapid and extensive developments in computer technology, especially in regards to IoT, have made Upnp devices an indispensable part of our daily lives - but also render them susceptible to a variety of SSDP attacks without suitable countermeasures. This paper proposes the Two-dimensional table scheme, which provides high security at a reasonable computational cost. The feasibility and effectiveness of the proposed scheme are also validated by comparison against four other schemes (Stateless connections, Failing-together, Cookie, and Client puzzle).

        Efficient Cross-Room Switch Mechanism for Indoor Room-Division-Multiplexing Based Visible Light Communication Network

        Visible light communication (VLC) is considered to be an attractive scheme to realize the broadbandwireless communication for an indoor environment. We present a room division multiplexing (RDM)mechanism for an indoor multi-room VLC network, which utilizes the spatial position of the LED lampin different rooms as a novel dimension of network resource for multiplexing, and thus the network capacityis increased. In such a network, the service interruption caused by user cross-room movement is animportant problem, and we propose a double-area-positioning based cross-room switch solution. Anexperimental platform demonstrates the RDM-deployed VLC network, and validates the performance ofthe presented switch mechanism

      • Two-stage damage identification for bridge bearings based on sailfish optimization and element relative modal strain energy

        Broad studies have addressed the issue of structural element damage identification, however, rubber bearing, as a key component of load transmission between the superstructure and substructure, is essential to the operational safety of a bridge, which should be paid more attention to its health condition. However, regarding the limitations of the traditional bearing damage detection methods as well as few studies have been conducted on this topic, in this paper, inspired by the model updating-based structural damage identification, a two-stage bearing damage identification method has been proposed. In the first stage, we deduce a novel bearing damage localization indicator, called element relative MSE, to accurately determine the bearing damage location. In the second one, the prior knowledge of bearing damage localization is combined with sailfish optimization (SFO) to perform the bearing damage estimation. In order to validate the feasibility, a numerical example of a 5-span continuous beam is introduced, also the noise robustness has been investigated. Meanwhile, the effectiveness and engineering applicability are further verified based on an experimental simply supported beam and actual engineering of the I-40 Bridge. The obtained results are good, which indicate that the proposed method is not only suitable for simple structures but also can accurately locate the bearing damage site and identify its severity for complex structure. To summarize, the proposed method provides a good guideline for the issue of bridge bearing detection, which could be used to reduce the difficulty of the traditional bearing failure detection approach, further saving labor costs and economic expenses.

        Investigation of Electrochemical Performance on SnSe2 and SnSe Nanocrystals as Anodes for Lithium Ions Batteries

        SnSe2 and SnSe nanocrystals were prepared using a simple solvothermal method by changing the molar ratio of SnCl2 · 2H2O and Se powder. When SnSe2 and SnSe are acted as lithium ion battery anodes, the SnSe hybrid structure shows more excellent electrochemical performance than that of SnSe2 interconnected nanosheet. It delivers a reversible capacity of 1023 mA h g -1 at a current density of 200 mA g -1, and maintaining a capacity of 498 mA h g -1 till 120 cycles. According to many present works, SnSe2 with interconnected thin nanosheet should possess more superior property than hybrid structured SnSe due to short charge transfer paths. However, in our research, the result is the opposite. Therefore, we consider that the superior electrochemical performance of SnSe is attributed to its highly reversible conversion reaction mechanism than SnSe2.

        Efficient Expression and Purification of Recombinant Alcohol Oxidase in Pichia pastoris

        In order to improve the production of alcohol oxidase (AOX), a recombinant Pichia pastoris (P. pastoris)system was constructed by transformation of the plasmid pPIC9K-AOX into P. pastoris GS115. The effects of different expression conditions on alcohol oxidase activity in the culture supernatant were investigated in the shake flask scale. The results showed that the highest extracellular activity (562 U/L) of alcohol oxidase was obtained after 56h induction with 4% methanol at OD600 1.0 in the medium containing 50 g/L maltose, which is about 4.2 folds higher than previously reported. High-purity functional recombinant AOX (>90%) was purified from the culture with the Ni-NTA affinity column and Sephadex G-100 chromatographical methods, with a total recovery rate of 68.9%. Further studies showed that the purified rAOX had similar enzymatic characteristics as the native enzyme, except that the thermal stability and resistance to H2O2 inhibition of rAOX were significantly greater compared to the previous report. The purified rAOX was well tolerant to various water-miscible organic solvents. This efficient expression and purification process will be promising for large-scale production of rAOX as an important diagnostic enzyme for alcohol detection in many areas.
