      • Regulatory Network of MicroRNAs, Target Genes, Transcription Factors and Host Genes in Endometrial Cancer

        Genes and microRNAs (miRNAs) have important roles in human oncology. However, most of the biological factors are reported in disperse form which makes it hard to discover the pathology. In this study, genes and miRNAs involved in human endometrial cancer(EC) were collected and formed into regulatory networks following their interactive relations, including miRNAs targeting genes, transcription factors (TFs) regulating miRNAs and miRNAs included in their host genes. Networks are constructed hierarchically at three levels: differentially expressed, related and global. Among the three, the differentially expressed network is the most important and fundamental network that contains the key genes and miRNAs in EC. The target genes, TFs and miRNAs are differentially expressed in EC so that any mutation in them may impact on EC development. Some key pathways in networks were highlighted to analyze how they interactively influence other factors and carcinogenesis. Upstream and downstream pathways of the differentially expressed genes and miRNAs were compared and analyzed. The purpose of this study was to partially reveal the deep regulatory mechanisms in EC using a new method that combines comprehensive genes and miRNAs together with their relationships. It may contribute to cancer prevention and gene therapy of EC.

        Multiple down-regulated cytochrome P450 monooxygenase genes contributed to synergistic interaction between chlorpyrifos and imidacloprid against Nilaparvata lugens

        Insecticide mixtures are an effective strategy in pest resistance management. The synergistic chlorpyrifos and imidacloprid mixture could significantly increase toxicity against rice pest, Nilaparvata lugens, despite their high levels of resistance. However, synergism mechanisms to explain this phenomenon remain unknown. Chlorpyrifos and imidacloprid at a 1:0.5 ratio showed significant synergism on N. lugens with a combination index value of 0.18 after topical exposure. We constructed a genetic database of the genes expressed in individual and synergistic chlorpyrifos and imidacloprid treatments of N. lugens using Illumina Hiseq™ X Ten, and 17 co-downregulated genes putatively involved in synergism were detected by comparative transcriptome analyses. Expression patterns of the 17 candidate synergistic genes matched with transcriptome sequencing data by quantitative real-time PCR analyses. Feeding of dsRNAs further reduced the expression levels of 10 of these candidate synergistic genes (from 1.68 to 4.13-fold). Nymphs fed with only dsRNAs of CYP4DE1, CYP6AY1v2, CYP353D1, and CYP439A1 experienced more high mortality rates (81.45–90.34%) to improve synergism between chlorpyrifos and imidacloprid. Multiple reductive expressed P450 genes were potentially associated with synergism of a mixture of chlorpyrifos and imidacloprid, as confirmed by comparative transcriptome analyses and RNAi assays. Our findings suggested that synergistic interactions between chlorpyrifos and imidacloprid might be controlled by P450s.

      • 2D Geometric Constraint Optimum Solving Based on Problem Decomposition

        Constraint solving is widely applied to many fields including computer aided design, 2 dimension (2D) model design and computer aided manufacturing. Geometric constraint solution is a difficult problem because there are a large number of entities and related parameters in 2D sketches. In this paper, a new method which decomposes geometric constraint relations based on entity-parameter graphs is proposed for reducing the size of constraint solution. A geometric constraint problem is decomposed into many independent sub-problems. Then, particle swarm optimization algorithm is used to solve constraint equations in each sub-problem. Solutions of all sub-problems are integrated to obtain the original problem’s solution. In experiments, the proposed method is applied to HUST-CAID system. Experimental results show that the method can effectively solve 2 dimension geometric constraints.

        The Correlation between Thyrotropin and Dyslipidemia in a Population-based Study

        This study investigated the relationship between serum thyrotrophin levels and dyslipidemia in subclinical hypothyroid and euthyroid subjects. A total of 110 subjects with subclinical hypothyroidism and 1,240 euthyroid subjects enrolled in this study. Patients with subclinical hypothyroidism had significantly lower high density lipoprotein cholesterol (HDL-C) levels than those who were euthyroid. The lipid profiles were each categorized and mean thyrotrophin levels were higher in subjects in the dyslipidemia subclasses than subjects in the normal subclasses. Thyrotrophin was positively associated with serum triglyceride and negatively associated with serum HDL-C in women. Thyrotrophin was also positively associated with total cholesterol (TC) in the overweight population along with TC and LDL-C in overweight women. In the euthyroid population, thyrotrophin was positively associated with TC in the overweight population. In conclusion, serum thyrotrophin was correlated with dyslipidemia in subclinical hypothyroid and euthyroid subjects; the correlation was independent of insulin sensitivity.

        Cloning and Biochemical Characterization of a Hyaluronate Lyase from Bacillus sp. CQMU-D

        Hyaluronidase (HAase) can enhance drug diffusion and dissipate edema by degrading hyaluronic acid (HA) in the extracellular matrix into unsaturated HA oligosaccharides in mammalian tissues. Microorganisms are recognized as valuable sources of HAase. In this study, a new hyaluronate lyase (HAaseD) from Bacillus sp. CQMU-D was expressed in Escherichia coli BL21, purified, and characterized. The results showed that HAaseD belonged to the polysaccharide lyase (PL) 8 family and had a molecular weight of 123 kDa. HAaseD could degrade chondroitin sulfate (CS) -A, CS-B, CSC, and HA, with the highest activity toward HA. The optimum temperature and pH value of HAaseD were 40°C and 7.0, respectively. In addition, HAaseD retained stability in an alkaline environment and displayed higher activity with appropriate concentrations of metal ions. Moreover, HAaseD was an endolytic hyaluronate lyase that could degrade HA to produce unsaturated HA oligosaccharides. Together, our findings indicate that HAaseD from Bacillus sp. CQMU-D is a new hyaluronate lyase and with excellent potential for application in industrial production.

        The quorum sensing regulator OpaR is a repressor of polar flagellum genes in Vibrio parahaemolyticus

        Vibrio parahaemolyticus possesses two types of flagella: asingle polar flagellum (Pof) for swimming and the peritrichouslateral flagella (Laf) for swarming. Expression of Lafgenes has previously been reported to be regulated by the quorumsensing (QS) regulators AphA and OpaR. In the presentstudy, we showed that OpaR, the QS regulator at high cell density(HCD), acted as a negative regulator of swimming motilityand the transcription of Pof genes in V. parahaemolyticus. OpaR bound to the promoter-proximal DNA regionsof flgAMN, flgMN, and flgBCDEFGHIJ within the Pof geneloci to repress their transcription, whereas it negatively regulatesthe transcription of flgKL-flaC in an indirect manner. Thus, this work investigated how QS regulated the swimmingmotility via direct action of its master regulator OpaR onthe transcription of Pof genes in V. parahaemolyticus.

        장예모 감독 영화의 여성 형상 연구: <붉은 수수밭>, <국두>, <홍등>, <황후화>를 중심으로

        최근에 중국 영화는 유수한 영화제를 통해 그 위상을 널리 떨치고 있다. 그 중에서도 장예모 감독의 영화는 특히 인상적이다. 장예모 감독의 영화는 그 예술성이나 상업성에 있어 전 세계 영화 관객들의 관심을 집중시키기에 충분한 매력을 가지고 있을 뿐만 아니라 장예모 감독의 여성 형상화 연구는 상당히 뛰어나다고 할 수 있다. 본 논문은 중국의 영화 감독인 장예모 감독의 대표적인 작품으로 알려진 <붉은 수수밭>, <국두>, <홍등> 등 세 편의 작품을 통하여, 장 감독이 "현대적 자주의식"이 코드화된 여성들, 다시 말하면 반 전통적인 "전통 여성"들의 비운을 그려 중국 역시 가부장적인 전통 관습의 비가시적인 여독의 해가 얼마나 큰지를 영상화해주는 면을 검토하여 보았다. 그러나 여성들을 그리면서 남성 감동이라는 한계 때문에 남성의 기준으로 여성을 길들여가는 전통 관습의 흔적도 보인다. 원작에서 성적으로 상당히 개방되어 있는 구야보다는 "일부종사"를 하는 <붉은 수수밭>에서의 구아, 남성 감독이 그려낸 <국두>의 "미친 여성"의 정형화 된 이미지, 그리고 항상 네 명의 부인중에서 누구를 제일 좋아하냐고 진좌천에게 묻는 <홍등>의 송련의 모습 등은 남성의 기준으로 여성을 길들여가는 전통 관습의 흔적이 조금씩 엿보이는 대목이 바로 그러한 것이다. 그럼에도 불구하고 장예모 감독은 그러한 한계를 극복하기 위한 관객과의 소통을 멈추지 않는다. 이러한 의미에서 장예모 감독의 영화에 구축된 다양하고 수많은 여성 형상은 여성주의 시각에서 접근할 수 있는 콘텐츠의 좋은 예들이다. 장예모 감독은 영화를 통해 지금도 중국의 문화를 상품화하고 코드화하는 작업을 멈추지 않는다. 그리고 그의 색깔은 초기의 단일화 된 색깔에서 점점 더 그 폭과 깊이를 더하고 있다. 변화하는 중국의 현실과 함께 더 폭 넒은 시대상을 제시하기도 한다. 끊임없이 도전하는 열정만큼이나 정형화된 자신의 틀을 깨는 장예모 감독의 긍정적인 변화 속에 그려질 여성 형상을 기대해본다. For the West, Zhang`s films are viewed as familiar "strangeness". The West poured out heaped praises on the "strange" exoticness which was reflected in Zhang`s films, because that is the oriental images in the imagination of the West. The "marginal remarks" of the marginal man in the marginal land is their familiar "strangeness", therefore, they exult over it. Zhang shaped the images of women, wearing Chinese traditional clothing but with a mind of anti tradition, which made a great impact on the fixed expectation of the Chinese and Korean audiences. It also swept over on the fixed expectation of the West. The Oriental Women which were known to the audience disappeared, and was taken place by the women with the western gender consciousness. This article for the film of <Red Sorghum>, <Judou> , <Raise the red lantern>, and <Curse of the Golden Flower> for example, analysis of female images. He share their interested in 3 themes. The first is interested in idealism, which shouws the ideal world through foucusing on freedom and liberaty of life. The second is returning to the real world from the ideal world, doing soul research to the traditional ethical code and pure humanity. The third is focusing on the intersection of traditional and modern. Zhang`s movies are the cultural text from which world audience can learn a lot, for it provides us a text of reflecting Chinese traditional culture and watching Chinese national thinking antitype.

        Influence of In-Plane Simple Shear Strain on the Grain Orientation Regulation and Stretch Formability of Pre-twinned AZ31 Magnesium Alloy Sheet

        Pre-twin is widely accepted as an efficient method for weakening the basal texture to improve the plasticity of magnesium alloys, particularly for formability. However, the enhancement is limited since the largest Schmid factor (SF) of basal slip cannot be achieved. In-plane simple shear strain is used to control the twin orientation in accordance with Schmid law to further decrease the basal texture and improve the formability of the Mg alloys sheet. Consequently, secondary regulation of initial twin orientation (SRITO) technology, a novel combination procedure, was developed. In this method, the AZ31 Mg alloys sheet is compressed to pre-twins with various volumes (1%, 3%, and 5%), then the twin orientation is regulated by in-plane simple shear strain for the second time. Compared with the as-received sample, the average fracture elongation of the annealed sample with the best properties improves from 15.3 to 28.8%, and the Erichsen value increases from 2.8 to 6.1 mm due to the regulation of twin orientation.
