        Sugar accumulation and characterization of metabolizing enzyme genes in leafy head of Chinese cabbage ( Brassica campestris L. ssp. pekinensis )

        Chinese cabbage ( Brassica campestris L. ssp. pekinensis ) is a widely cultivated vegetable crop in eastern Asia. The fl avor ofChinese cabbage was mostly aff ected by soluble sugar content. To make clear its accumulation and regulation mechanism,the content of diff erent sugars in the internal blade (IB), the internal midrib (IM), the external blade (EB) and the externalmidrib (EM) of the leafy head were determined during the leafy head formation. The results showed that fructose was themajor sugar accumulated in the internal tissues, followed by glucose. IM was the main tissue of sugar accumulation with thehighest contents of total soluble sugar and hexose at harvest. RNA sequencing data of the four tissues at 90 DAS showed thatthe number of diff erentially expressed genes (DEGs) in IB and IM was the least (2408), while the number of DEGs betweenthe external and internal tissues ranged from 6037 to 8674. The enzyme genes diff erentially expressed in ‘starch and sucrosemetabolism’ pathway was detected during the leafy head development. The expression of two sucrose synthase (SUS) genes,SUS1a (Bra002332) and SUS1b (Bra006587), were always higher in IM than in the other tissues, and positively correlatedwith the content of total soluble sugar and hexose, respectively. SUS1 was considered to play an important role in leafy headdevelopment and sugar accumulation in Chinese cabbage.

        SVC: Secure VANET-Assisted Remote Healthcare Monitoring System in Disaster Area

        With the feature of convenience and low cost, remote healthcare monitoring (RHM) has been extensively used in modern disease management to improve the quality of life. Due to the privacy of health data, it is of great importance to implement RHM based on a secure and dependable network. However, the network connectivity of existing RHM systems is unreliable in disaster area because of the unforeseeable damage to the communication infrastructure. To design a secure RHM system in disaster area, this paper presents a Secure VANET-Assisted Remote Healthcare Monitoring System (SVC) by utilizing the unique “store-carry-forward” transmission mode of vehicular ad hoc network (VANET). To improve the network performance, the VANET in SVC is designed to be a two-level network consisting of two kinds of vehicles. Specially, an innovative two-level key management model by mixing certificate-based cryptography and ID-based cryptography is customized to manage the trust of vehicles. In addition, the strong privacy of the health information including context privacy is taken into account in our scheme by combining searchable public-key encryption and broadcast techniques. Finally, comprehensive security and performance analysis demonstrate the scheme is secure and efficient.

        Ganoderic acid A attenuates high‑fat‑diet‑induced liver injury in rats by regulating the lipid oxidation and liver inflammation

        Ganoderic Acid A (GA) has many pharmacologicaleffects such as anti-tumor, antibacterial, anti-inflammatory,and immunosuppressive effects. However, the protectiveeffect of GA on liver injury has not been reported. Thisstudy aimed to investigate the action of GA on insufficientmethionine and choline combined with high-fat diet (HFD)-induced non-alcoholic fatty liver disease (NAFLD) in rats. NAFLD model was established by insufficient methionineand choline combined with high fat feeding to rats. The levelsof Acetyl-CoA carboxylase, fatty acid synthase, sterolregulatory element binding protein, liver X receptors, AMPactivatedprotein kinase, peroxisome proliferator-activatedreceptor α, PPARg coactivator 1α and NF-κB pathway inthe liver were detected by western blot. The results of thisstudy demonstrated that the expression of GA can not onlysignificantly decrease the live weight and liver weight perbody weight of HFD mice, but also restore the alanine aminotransferase,aspartate aminotransferase, total bilirubinlevels, triglyceride and cholesterol in serum. In addition,the expression of GA increased the levels of high-densitylipoprotein cholesterol in serum, ameliorated pathologicalchanges and decreased NAS score of mice’s liver. In conclusion,the treatment with GA could improve NAFLD in ratsby regulating the levels of signaling events involved in freefatty acid production, lipid oxidation and liver inflammation.

        Establishment of an efficient regeneration system using heading leaves of Chinese cabbage (Brassica rapa L.) and its application in genetic transformation

        To preserve and propagate valuable Chinese cabbage (Brassica rapa L.) using mature vegetative organs without seeds, we established a high efficiency regeneration system from heading leaf explants and further explored its application in genetic transformation. Our results showed that maximum shoot regeneration of cultivars ‘Beijing New No. 3’ (88.46 ± 1.65%) and ‘Chengyangqing’ (73.77 ± 1.73%) were obtained in the presence of 17.76 μM 6-benzylaminopurine (BA) and 5.37 μM α-naphthylacetic acid (NAA). The addition of silver nitrate (AgNO3) at 11.77 μM was essential for shoot regeneration from heading leaf explants. Genotypic differences in shoot regeneration frequencies (from 47.24 ± 3.65 to 88.46 ± 1.65%) were observed amongst eight Chinese cabbage cultivars, in addition to the number of shoots per explant (from 1.31 ± 0.02 to 2.02 ± 0.05) in Murashige and Skoog medium containing 17.76 μM BA, 5.37 μM NAA and 11.77 μM AgNO3. Low temperature (4 °C) had an effect on in vitro preservation of leafy heads with a delay in leaf wilting, and there were no significant differences in shoot induction frequency within 24 h for cultivars ‘Beijing New No. 3’ and ‘Chengyangqing’. In the genetic transformation experiments using selection with kanamycin (17.17 μM), a transformation efficiency of 0.6–1.2% was achieved, as assessed from PCR and Southern blot results. The above results suggested that the heading leaf explants can not only achieve efficient seed-independent propagation of Chinese cabbage, but also provide a feasible platform for genetic transformation.

        Phylogenetic relationships of ascomycetes and basidiomycetes based on comparative genomics analysis

        The relationship between ascomycetes and basidiomycetes, the two main phyla of non-flagellated fungi, has rarely been investigated. In this study, we performed a comparative genomics analysis of genome sequences of 55 ascomycetes and 26 basidiomycetes species and detected 81 universal markers, 875 homologous genes and a conserved contig in the glucose-regulated protein gene. In dendrograms based on simple sequence repeat markers and homologous genes, ascomycetes and basidiomycetes formed distinct clusters, with each set of taxa having a high coefficient of relatedness. Ascomycetes and basidiomycetes also constituted distinct groups in a phylogenetic tree based on a conserved contig in the glucoseregulated protein gene. These results provide evidence that basidiomycetes may be derived from ascomycetes but are definitely genetically differentiated at the genomic level. The phylogenetic relationships of ascomycetes and basidiomycetes uncovered in this study provide new insights for future research related to fungal classification and evolution.

        Research progress of single-atom coating and its application prospect in protective coatings

        Coating technology is the most economical and efficient approach to retard metal corrosion, and newcomposite coatings prepared by adding monoatomic fillers to a polymer matrix have excellent corrosionresistance and good mechanical properties. The single-atom coating formed by the immobilization ofmetal atoms alone on the matrix or filler represents the maximum utilization of atoms at the presentstage, which has unique coordination properties, active sites, and synergistic effects with the support. Compared with traditional fillers, single-atom fillers not only have the advantages of supporting fillersbut can also provide monatomic metals to form passivation films, promote the progress of catalytic passivationreactions, and combine organic compounds to generate new composite materials. This paperfocuses on the preparation methods and mechanisms of single-atom coatings in the field of batteriesand catalysts as well as the application of composite materials prepared by combining single-atom materialswith polymer matrices in the field of metal protection. The preparation methods, application prospects,and coating characteristics of various single-atom composites were summarized. In addition,we discuss the potential applications of single-atom coatings in the design of future composite coatings.

        Functional roles and mechanisms of ginsenosides from Panax ginseng in atherosclerosis

        Atherosclerosis (AS) is a leading cause of cardiovascular diseases (CVDs) and it results in a high rate of death worldwide, with an increased prevalence with age despite advances in lifestyle management and drug therapy. Atherosclerosis is a chronic progressive inflammatory process, and it mainly presents with lipid accumulation, foam cell proliferation, inflammatory response, atherosclerotic plaque formation and rupture, thrombosis, and vascular calcification. Therefore, there is a great need for reliable therapeutic drugs or remedies to cure or alleviate atherosclerosis and reduce the societal burden. Ginsenosides are natural steroid glycosides and triterpene saponins obtained mainly from the plant ginseng. Several recent studies have reported that ginsenosides have a variety of pharmacological activities against several diseases including inflammation, cancer and cardiovascular diseases. This review focuses on describing the different pharmacological functions and underlying mechanisms of various active ginsenosides (Rb1,-Rd, -F, -Rg1, -Rg2, and -Rg3, and compound K) for atherosclerosis, which could provide useful insights for developing novel and effective anti-cardiovascular drugs.

        Risk factors affecting postoperative pulmonary function in congenital diaphragmatic hernia

        Purpose: It is well known that congenital diaphragmatic hernia (CDH) in infants impacts pulmonary function rehabilitation after surgery. However, the risk factors of postoperative pulmonary function are still unclear. In this research, we analyzed the potential risk factors of postoperative pulmonary function in CDH patients in order to improve the clinical management of CDH patients. Methods: Thirty-three cases CDH infants followed were enrolled from November 2016 to September 2018. Clinical data were reviewed. Tidal breathing pulmonary function testing was performed after surgery. Correlation between pulmonary function and clinical characteristics was evaluated using multivariate analysis of variance. Results: Pulmonary dysfunction was detected in 87.9% patients (29 of 33). The defect size was found to be significantly larger in patients with obstructed and mixed ventilatory disorders (P = 0.001). Diagnosis of gestational age (GA) was also significantly earlier compared to restrictive ventilatory disorders (P = 0.001). Larger defect size, and earlier prenatal diagnosis of GA were detected in severe obstructive ventilatory disorders (P = 0.007, P = 0.001, retrospectively). Conclusion: Most patients had various degrees of pulmonary dysfunction after surgery. Patients with larger defect size and earlier diagnosis time might be vulnerable to severe obstructive and mixed ventilatory disorders.

        Inhibition of chromosomal region maintenance 1 suppresses the migration and invasion of glioma cells via inactivation of the STAT3/MMP2 signaling pathway

        Chromosomal region maintenance 1 (CRM1) is associated with an adverse prognosis in glioma. We previously reported that CRM1 inhibition suppressed glioma cell proliferation both in vitro and in vivo. In this study, we investigated the role of CRM1 in the migration and invasion of glioma cells. S109, a novel reversible selective inhibitor of CRM1, was used to treat Human glioma U87 and U251 cells. Cell migration and invasion were evaluated by wound-healing and transwell invasion assays. The results showed that S109 significantly inhibited the migration and invasion of U87 and U251 cells. However, mutation of Cys528 in CRM1 abolished the inhibitory activity of S109 in glioma cells. Furthermore, we found that S109 treatment decreased the expression level and activity of MMP2 and reduced the level of phosphorylated STAT3 but not total STAT3. Therefore, the inhibition of migration and invasion induced by S109 may be associated with the downregulation of MMP2 activity and expression, and inactivation of the STAT3 signaling pathway. These results support our previous conclusion that inhibition of CRM1 is an attractive strategy for the treatment of glioma.

      • Fluorescent Microarrays of <i>in Situ</i> Crystallized Perovskite Nanocomposites Fabricated for Patterned Applications by Using Inkjet Printing

        <P>Perovskite materials have exhibited promising potential for universal applications including backlighting, color conversion, and anticounterfeiting labels fabricated using solution processes. However, owing to the tendency of those materials to have uncontrollable morphologies and to form large crystals, they cannot be utilized in discontinuous microminiaturization, which is crucial for practical optoelectronic applications. In this research, combining the effects of adding polyvinylpyrrolidone (PVP), precisely controlling the inkjet printing technique, and using a postprocessing procedure, we were able to fabricate <I>in situ</I> crystallized perovskite-PVP nanocomposite microarrays with perfect morphologies. The viscosity of the perovskite precursor increased with the addition of PVP, eliminating the outward capillary flow that induces the coffee-ring effect. In addition, because of the presence of metallic bonds with the C═O groups in PVP and the spatial confinement of such a polymer, we were able to fabricate regulated CsPbBr<SUB>3</SUB> nanocrystals capped with PVP and with a uniform size distribution. The as-printed patterns showed excellent homogeneity on a macroscale and high reproducibility on a microscale; furthermore, those patterns were invisible in the ambient environment, compatible with flexible substrates, and cost-efficient to produce, indicating that this technique holds promising potential for applications such as anticounterfeiting labels.</P> [FIG OMISSION]</BR>
