        Impact of NR1I2, adenosine triphosphate-binding cassette transporters genetic polymorphisms on the pharmacokinetics of ginsenoside compound K in healthy Chinese volunteers

        Background: Ginsenoside compound K (CK) is a promising drug candidate for rheumatoid arthritis. This study examined the impact of polymorphisms in NR1I2, adenosine triphosphate-binding cassette (ABC) transporter genes on the pharmacokinetics of CK in healthy Chinese individuals. Methods: Forty-two targeted variants in seven genes were genotyped in 54 participants using Sequenom MassARRAY system to investigate their association with major pharmacokinetic parameters of CK and its metabolite 20(S)-protopanaxadiol (PPD). Subsequently, molecular docking was simulated using the AutoDock Vina program. Results: ABCC4 rs1751034 TT and rs1189437 TT were associated with increased exposure of CK and decreased exposure of 20(S)-PPD, whereas CFTR rs4148688 heterozygous carriers had the lowest maximum concentration ($C_{max}$) of CK. The area under the curve from zero to the time of the last quantifiable concentration ($AUC_{last}$) of CK was decreased in NR1I2 rs1464602 and rs2472682 homozygous carriers, while $C_{max}$ was significantly reduced only in rs2472682. ABCC4 rs1151471 and CFTR rs2283054 influenced the pharmacokinetics of 20(S)-PPD. In addition, several variations in ABCC2, ABCC4, CFTR, and NR1I2 had minor effects on the pharmacokinetics of CK. Quality of the best homology model of multidrug resistance protein 4 (MRP4) was assessed, and the ligand interaction plot showed the mode of interaction of CK with different MRP4 residues. Conlusion: ABCC4 rs1751034 and rs1189437 affected the pharmacokinetics of both CK and 20(S)-PPD. NR1I2 rs1464602 and rs2472682 were only associated with the pharmacokinetics of CK. Thus, these hereditary variances could partly explain the interindividual differences in the pharmacokinetics of CK.

        One-Pot Fast Synthesis of Leaf-Like CuO Nanostructures and CuO/Ag Microspheres with Photocatalytic Application

        A time- and energy- saving solution-based chemical precipitation method was developed to synthesize leaf-like CuO nanostructures. The morphology and size of the leaf-like CuO nanostructures could be simply manipulated by controlling the type and concentration of precursors, and the oriented attachment mechanism is responsible for the formation of leaf-like shape. With the concurrent reduction reaction and at appropriate concentration, CuO/Ag microspheres could be prepared and the growth mechanism is proposed. These two structures could serve as effective photocatalyst for the degradation of rhodamine B under visible light irradiation in the presence of hydrogen peroxide. Moreover, compared to pure CuO nanostructures, the photodecomposition activity of CuO/Ag microspheres increases by 42.9% due to plasmon-enhanced light absorption.

      • pH-Based immunoassay: explosive generation of hydrogen ions through an immuno-triggered nucleic acid exponential amplification reaction

        <P>In this work, we propose a novel concept and a proof-of-concept strategy for the fabrication of a pH-based immunoassay platform with a certain degree of universality and scalability to make it adaptable for different application scenarios. The immunoreactions for the target detection are converted to pH changes through an engineered and optimized isothermal nucleic acid amplification, named exponential amplification reaction (EXPAR). Thus, a variety of well-developed methods for pH analysis, <I>e.g.</I> pH indicators, pH-strips and pH meters, can be applied for immunoassay directly. Here, we show that this proof-of-concept strategy is applicable for both macromolecular and micromolecular antigens by adopting human platelet-derived growth factor-BB (PDGF-BB) and chloramphenicol (CAP) as the model targets, respectively. The detection can be achieved using a colorimetric pH indicator after a 15 min reaction of the immuno-triggered isothermal nucleic acid amplification. In addition, compared with the traditional enzyme-linked immunosorbent assay (ELISA), the performance of our strategy, especially the detection limits, is improved to varying degrees for different targets, making the strategy a promising alternative for diverse application scenarios of immunoassay.</P>

        Dual-Coupled Inductor High Gain DC/DC Converter with Ripple Absorption Circuit

        High-gain DC/DC converters have become one of the key technologies for the grid-connected operation of new energy power generation, and its research provides a significant impetus for the rapid development of new energy power generation. Inspired by the transformer effect and the ripple-suppressed ability of a coupled inductor, a double-coupled inductor high gain DC/DC converter with a ripple absorption circuit is proposed in this paper. By integrating the diode-capacitor voltage multiplying unit into the quadratic Boost converter and assembling the independent inductor into the magnetic core of structure coupled inductors, the adjustable range of the voltage gain can be effectively extended and the limit on duty ratio can be avoided. In addition, the volume of the magnetic element can be reduced. Very small ripples of input current can be obtained by the ripple absorption circuit, which is composed of an auxiliary inductor and a capacitor. The leakage inductance loss can be recovered to the load in a switching period, and the switching-off voltage spikes caused by leakage inductance can be suppressed by absorption in the diode-capacitor voltage multiplying unit. On the basis of the theoretical analysis, the feasibility of the proposed converter is verified by test results obtained by simulations and an experimental prototype.

        The combined use of indium and polysorbate 20 (Tween 20) was considered as a new inhibition technique for zinc corrosion. Zn and Zn-In alloy coatings were prepared by electrodeposition and their morphology and composition were characterized by scanning electron microscopy (SEM) and inductively coupled plasma atomic emission spectrometry (ICP-AES). The corrosion inhibition effect of indium and Tween 20 on zinc was investigated by polarization curves and electrochemical impedance spectroscopy (EIS). The corrosion inhibition efficiencies obtained from Tafel and EIS analyses are well in agreement. Zinc corrosion can be inhibited to some extent by the individual use of indium and Tween 20 and higher corrosion inhibition efficiency can be obtained by the combined use of indium and Tween 20.

        Service Scheduling in Cloud Computing based on Queuing Game Model

        Cloud Computing allows application providers seamlessly scaling their services and enables users scaling their usage according to their needs. In this paper, using queuing game model, we present service scheduling schemes which are used in software as a service (SaaS). The object is maximizing the Cloud Computing platform`s (CCP`s) payoff via controlling the service requests whether to join or balk, and controlling the value of CCP`s admission fee. Firstly, we treat the CCP as one virtual machine (VM) and analyze the optimal queue length with a fixed admission fee distribution. If the position number of a new service request is bigger than the optimal queue length, it balks. Otherwise, it joins in. Under this scheme, the CCP`s payoff can be maximized. Secondly, we extend this achievement to the multiple VMs situation. A big difference between single VM and multiple VMs is that the latter one needs to decide which VM the service requests turn to for service. We use a corresponding algorithm solve it. Simulation results demonstrate the good performance of our schemes.

        Electrostatic spraying of membrane electrode for proton exchange membrane fuel cell

        In order to improve the performance of proton exchange membrane fuel cell (PEMFC), the optimization of electrostatic spraying of membrane electrode was conducted. The influence of the spraying voltage on morphology, elemental composition of catalyst layer, and performance of the PEMFC were investigated. The results show that increasing spraying voltage could reduce agglomeration of the carbon-supported platinum particles, leading to more uniform pore distribution. High voltage did not accelerate oxidation of platinum catalyst. A high electrochemical active surface area of 26.18m2/gpt was obtained when the platinum-carbon catalyst layer was deposited in cone jet mode. With further increasing spraying voltage, the total ohmic resistance and catalytic activity were changed slightly, whereas the charge transfer resistance was increased. Using the optimized electrostatic spraying parameters (injection rate=100 μL min−1, spraying voltage=8.5 kV, and working distance= 12 mm), a peak power density of 1.408Wcm−2 was obtained with an output voltage of 0.451 V.
