        Meconopsis quintuplinervia Regel Improves Cutibacterium acnes -Induced Inflammatory Responses in a Mouse Ear Edema Model and Suppresses Pro-Inflammatory Chemokine Production via the MAPK and NF-κκB Pathways in RAW264.7 Cells

        Background: Acne vulgaris (AV) is a common adolescent skin condition which is mainly caused by Cutibacterium acnes overcolonization and subsequent inf lammation. Objective: Our previous studies demonstrated that ethanol extracts of Meconopsis quintu- plinervia Regel (EMQ) possess significant antimicrobial properties. However, their protec- tive effects and potential mechanisms against AV remain unclear. Methods: In the present study, the EMQ treatment potential for AV was evaluated in a C. acnes -induced mouse ear edema model, and the EMQ anti-inf lammatory mechanism was evaluated in lipopolysaccharide (LPS)-induced RAW264.7 macrophage cells. Results: The results showed that EMQ alleviated edema formation and inf lammatory cell infiltration in an acne mouse model by suppressing inf lammatory cytokines interleukin (IL)-6, IL-1β, and tumor necrosis factor α expression. Moreover, EMQ inhibited the phos- phorylation of MAP kinases (MAPKs) such as p38, JNK, and ERK, the phosphorylation and degradation of IκB-α and the nuclear translocation of nuclear factor kappa B (NF-κB) p65 in LPS-induced RAW264.7 cells. Conclusion: These findings suggest the potent anti-inf lammatory activity of EMQ is pos- sibly through the regulation of the MAPKs and NF-κB signaling pathways. Inhibition of C. acnes activity combined with a powerful anti-inf lammatory effect of EMQ indicated its potential as a novel therapeutic option for AV.

        Malus toringoides (Rehd.) Hughes Ameliorates Nonalcoholic Fatty Liver Disease with Diabetes via Downregulation of SREBP-1c and the NF-κB Pathway In Vivo and In Vitro

        Diabetic patients are more prone to developing nonalcoholic fatty liver disease (NAFLD) compared with healthy people. As a plant homologous to both medicine and food, Malus toringoides (Rehd.) Hughes has been used as an intervention for both NAFLD and diabetes. However, the effect and mechanism of M. toringoides on NAFLD on type 2 diabetes mellitus (T2DM) is unclear. The current investigation was designed to evaluate the ameliorative effects and mechanism of M. toringoides ethanol extract (CBTM-E375) on T2DM, and to identify the compounds in these extracts. The effects of CBTM-E375 on T2DM were verified using a high-fat diet-/streptozotocin-induced diabetic rat and free fatty acid (0.5 mM)-induced human hepatocellular carcinoma cell (HepG2) models. The components of CBTM-E375 were identified by high performance liquid chromatography-mass spectrometry/mass spectrometry. Our results demonstrate that CBTM-E375 ameliorated lipid accumulation (total cholesterol, triglyceride), oxidative stress (superoxide dismutase, catalase, malondialdehyde, glutathione peroxidase), and inflammation (tumor necrosis factor-α [TNF-α], interleukin [IL]-1β, IL-6, C-reactive protein [CRP]) in vivo and in vitro, these effects were associated with a CBTM-E375-mediated downregulation of SREBP-1c (sterol regulatory element binding protein 1c) and the NF-κB (nuclear factor κB) signaling pathway. A total of 20 chemical compounds were identified in CBTM-E375, including phlorizin, isoquercitrin, chlorogenic acid, quercetin, naringenin, and trigonelline, which have been reported to have positive effects on diabetes or on NAFLD.

        Insulin resistance and overweight prolonged fertility-sparing treatment duration in endometrial atypical hyperplasia patients

        Objective: Our previous study showed that insulin resistance (IR) was related to endometrial hyperplasia as well as endometrial cancer. But the exact impact of IR on fertility-sparing treatment in endometrial hyperplasic disease is unclear. This study investigated how IR affects fertility-sparing treatment in endometrial atypical hyperplasia (EAH) patients. METHODS: The 151 EAH patients received fertility-sparing treatment were retrospectively investigated. All patients received high-dose progestin combined with hysteroscopy. Therapeutic effects were evaluated by hysteroscopy every 3 months during the treatment. RESULTS: The median age was 33.0 years old (range, 21-54 years old). Sixty-one patients (40.4%) were insulin resistant. Three patients were excluded from the analysis because they chose hysterectomy within 3 months after initiation of progestin treatment. The 141 out of 148 (95.3%) patients achieved complete response (CR). No difference was found in cumulative CR rate between those with or without IR (90.2% vs. 95.6%, p=0.320). IR significantly affected therapeutic duration to achieve CR (8.1±0.5 months with IR vs. 6.1±0.4 months without IR, p=0.004). Overweight (body mass index [BMI]≥25 kg/m²) was associated with higher risk of treatment failure (odds ratio=5.61; 95% confidence interval=1.11-28.35; p=0.040) and longer therapeutic duration to achieve CR (7.6±0.5 months vs. 6.3±0.4 months, p=0.019). EAH patients with both IR and overweight (IR+BMI+) had the longest therapeutic time compared with other patients (8.8±0.6 months vs. 5.6±0.7, 6.3±0.4, and 6.4±0.8 months for IR-BMI+, IR-BMI-, and IR+BMI-, respectively, p=0.006). CONCLUSION: IR and overweight were associated with longer therapeutic duration in EAH patients receiving progestin-based fertility-sparing treatment.

      • Research on the Pattern of "Dual Circulation" Development in Zhejiang Province Under Regional Integration Taking Digital Economy as an Example

        At present, the world economic situation is complex, and the epidemic has dealt a huge blow to global economic development. In this context, it is particularly important for China's economic construction to accelerate a pattern with the domestic circulation as the main body and the mutual promotion of domestic and double cycles between China and international cycles. From the perspective of the industrial chain, China is still in the middle and low end of the value chain, and the overall export competition and trade volume are low. Strengthening China's industrial chain with the help of regional economic integration is an important way to promote the "dual circulation", which can simultaneously stimulate the vitality of the two domestic and international markets, promote each other, and enhance the ability to resist risks. Among them, the Regional Comprehensive Economic Partnership Agreement (RCEP) has become a crucial platform for regional cooperation for China to build a current "dual circulation" development pattern. The development of digital economy relies on the rapid changes of digital technology. With this in-depth development of digital technology and the growing development trend of the digital economy, we are still deeply troubled by the data leakage, data abuse, digital security, network security and other technical issues, which also creates our trust in the digital economy. Studying the digital trust problem is also an indispensable step towards in achieving a healthy, efficient and sustainable digital ecological environment. Although scholars have conducted extensive studies on supply chain services in cross-border transactions and the development trend of the digital economy, the related research results have failed to address the following issues. First of all, due to cultural differences and customs, consumers in different countries often show differences in consumption behaviors. Therefore, it is necessary to conduct a comparative study on consumers in different countries to understand their behavioral preferences and differences. Secondly, most of the current literature on trust issues in cross-border transactions mainly focuses on consumer privacy and information security. It is necessary to sort out the framework of supply chain services and explore consumers' choice strategies when facing different types of services. Finally, scholars often focus on one or several types of influencing factors and ignore the interaction and influence of different types of factors. Therefore, this topic believes that in the current research, the trust relationship management model for the inclusive development of digital trade in the regional integration environment has not been constructed wholly and systematically, and many essential components have not been included in the research framework, which restricts the further development of the research in this field. So, this project will carry out analysis in five dimensions, from the technology and development of the digital economy, the reshaping of services and consumption, the acceptance of the customer, the labor market and ethical and moral safety, to deconstruct the connotation and the pattern of digital economy. This can not only describe the development picture and mechanism of digital economy in each dimension, but also try to clarify the relevance and influence among the dimensions. This project takes the trust of different stakeholders in the digital economy as the entry point. Also, it takes the users and enterprises under regional integration as the research objects to explore the specific factors affecting the trust behavior of the surrounding destiny community. The research process focuses on analyzing the trust management model in the overseas development of Zhejiang enterprises and exploring the changes that RCEP will bring to the overseas development. This project conducted an empirical study based on questionnaires, using a structural equation model through which the measurement and structural models were evaluated. The project verifies information reliability through empirical studies, examines the trust transfer function of trusted third parties in the digital environment through laboratory experiments, and analyzes the factors that moderate trust transfer by relying on partial least squares multigroup analysis. Ultimately, this topic realizes the evaluation of the effectiveness and overall effectiveness of the trust model. The study results verify that RCEP will strongly support Zhejiang enterprises' foreign development, and at the same time, identify some challenges faced by some Zhejiang enterprises in the process of foreign development. Also, the results will be beneficial to discover some new ideas to promote economic development further with RCEP.         The prognostic significance of estrogen and progesterone receptors in grade I and II endometrioid endometrial adenocarcinoma: hormone receptors in risk stratification

        Objectives: Although patients with grade I and II endometrioid endometrial adenocarcinoma (EEA) are considered with good prognosis, among them 15%–25% died in 5 years. It is still unknown whether integrating estrogen receptor (ER) and progesterone receptor (PR) into clinical risk stratification can help select high-risk patients with grade I–II EEA. This study was to investigate the prognostic value of ER and PR double negativity (ER/ PR loss) in grade I–II EEA, and the association between ER/PR loss and The Cancer Genome Atlas (TCGA) classification. Methods: ER and PR were assessed by immunohistochemistry on hysterectomy specimens of 903 patients with grade I–II EEA. ER and PR negativity were determined when <1% tumor nuclei were stained. Gene expression data were obtained from the TCGA research network. Results: Compared with ER or PR positive patients (n=868), patients with ER/PR loss (n=35) had deeper myometrial infiltration (p=0.012), severer FIGO stage (p=0.004), and higher rate of pelvic lymph node metastasis (p=0.020). In univariate analysis, ER/PR loss correlated with a shorter progression-free survival (PFS; hazard ratio [HR]=5.25; 95% confidence interval [CI]=2.21–12.52) and overall survival (OS; HR=7.59; 95% CI=2.55–22.60). In multivariate analysis, ER/PR loss independently predicted poor PFS (HR=3.77; 95% CI=1.60–10.14) and OS (HR=5.56; 95% CI=1.37–22.55) for all patients, and poor PFS for patients in stage IA (n=695; HR=5.54; 95% CI=1.28–23.89) and stage II–IV (n=129; HR=5.77; 95% CI=1.57–21.27). No association was found between ER/PR loss and TCGA classification. Conclusion: Integrating ER/PR evaluation into clinical risk stratification may improve prognosis for grade I–II EEA patients.

        Whole-genome bisulfite sequencing identified the key role of the Src family tyrosine kinases and related genes in systemic lupus erythematosus

        Background As a multisystemic autoimmune illness, the basic mechanisms behind the pathophysiology of systemic lupus erythematosus (SLE) remain poorly understood. Objective We aimed to investigate the possible significance of SLE’s DNA methylation and gain further insight into potential SLE-related biomarkers and therapeutic targets. Methods We used whole genome bisulfite sequencing (WGBS) method to analyze DNA methylation in 4 SLE patients and 4 healthy people. Results 702 differentially methylated regions (DMRs) were identified, and 480 DMR-associated genes were annotated. We found the majority of the DMR-associated elements were enriched in repeat and gene bodies. The top 10 hub genes identified were LCK, FYB, PTK2B, LYN, CTNNB1, MAPK1, GNAQ, PRKCA, ABL1, and CD247. Compared to the control group, LCK and PTK2B had considerably decreased levels of mRNA expression in the SLE group. Receiver operating characteristic (ROC) curve suggested that LCK and PTK2B may be potential candidate biomarkers to predict SLE. Conclusions Our study improved comprehension of the DNA methylation patterns of SLE and identified potential biomarkers and therapeutic targets for SLE.
