        Target engagement of ginsenosides in mild cognitive impairment using mass spectrometry-based drug affinity responsive target stability

        Background: Mild cognitive impairment (MCI) is a transitional condition between normality and dementia. Ginseng is known to have effects on attenuating cognitive deficits in neurogenerative diseases. Ginsenosides are the main bioactive component of ginseng, and their protein targets have not been fully understood. Furthermore, no thorough analysis is reported in ginsenoside-related protein targets in MCI. Methods: The candidate protein targets of ginsenosides in brain tissues were identified by drug affinity responsive target stability (DARTS) coupled with label-free liquid chromatography-mass spectrometry (LC-MS) analysis. Network pharmacology approach was used to collect the therapeutic targets for MCI. Based on the above-mentioned overlapping targets, we built up a proteineprotein interaction (PPI) network in STRING database and conducted gene ontology (GO) enrichment analysis. Finally, we assessed the effects of ginseng total saponins (GTS) and different ginsenosides on mitochondrial function by measuring the activity of the mitochondrial respiratory chain complex and performing molecular docking. Results: We screened 2526 MCI-related protein targets by databases and 349 ginsenoside-related protein targets by DARTS. On the basis of these 81 overlapping genes, enrichment analysis showed the mitochondria played an important role in GTS-mediated MCI pharmacological process. Mitochondrial function analysis showed GTS, protopanaxatriol (PPT), and Rd increased the activities of complex I in a dose-dependent manner. Molecular docking also predicted the docking pockets between PPT or Rd and mitochondrial respiratory chain complex I. Conclusion: This study indicated that ginsenosides might alleviate MCI by targeting respiratory chain complex I and regulating mitochondrial function, supporting ginseng's therapeutic application in cognitive deficits.

        Expression of the Arabidopsis vacuolar $H^+$-pyrophosphatase gene AVP1 in peanut to improve drought and salt tolerance

        The Arabidopsis gene AVP1 encodes an $H^+$-pyrophosphatase that functions as a proton pump at the vacuolar membranes, generating a proton gradient across vacuolar membranes, which serves as the driving force for many secondary transporters on vacuolar membranes such as $Na^+/H^+$-antiporters. Overexpression of AVP1 could improve drought tolerance and salt tolerance in transgenic plants, suggesting a possible way in improving drought and salt tolerance in crops. The AVP1 was therefore introduced into peanut by Agrobacterium-mediated transformation. Analysis of AVP1-expressing peanut indicated that AVP1-overexpression in peanut could improve both drought and salt tolerance in greenhouse and growth chamber conditions, as AVP1-overexpressing peanuts produced more biomass and maintained higher photosynthetic rates under both drought and salt conditions. In the field, AVP1-overexpressing peanuts also outperformed wild-type plants by having higher photosynthetic rates and producing higher yields under low irrigation conditions.

        Construction of a Transposon-mediated Baculovirus Vector Hanpvid and a New Cell Line for Expressing Barnase

        In this study we developed the transposon-mediated shuttle vector `Hanpvid`, which composed of HaNPV (Heliothis armigera nuclear polyhedrosis virus) genomic DNA and a transposon cassette from Bacmid of Bac-to-Bac system. Hanpvid replicates in E. coli in the same way as Bacmid and retains infective function in cotton bollworm cells (Hz-AMU). Using Hanpvid we constructed a recombinant virus, which could infect Hz-AM1 cells and generate recombinant HaNPV (rHa-Bar) containing the barnase gene, a ribonuclease gene from Bacillus amyloliquefaciens. Since the expression vector carrying barnase gene cannot replicate in the absence of barstar, a specific inhibitor of barnase, we constructed a new cotton bollworm cell line (AMl-NB) using the marker rescue method. In AM1-NB barstar was integrated into the cellular chromosome to sustain the replication of rHa-Bar. To screen out recombinant HaNPV for potential use as biopesticide, Hz-AM! and AM1-NB cell lines were infected with rHa-Bar, respectively. The results obtained indicate that Viral progenies in AM1-NB were 23 and 160 times greater than those in Hz-AM1 48 h and 72 h after infection, respectively. With additional insertion of the polyhedron gene from AcNPV (Autographa californica nuclear polyhedrosis virus) into the Hanpvid genome, rHa-Bar regained the polyhedron phenotype and its pest-killing rate greatly improved. Toxic analysis showed that the lethal dosages (LD_(50)) and the lethal time(s) (LT_(50)) of rHa-Bar were reduced by 20% and 30%, respectively, compared to wt-HaNPV in the third instar larvae of cotton bollworm. This study shows that in AM1-NB barnase can be effectively produced and used as pest-killing agent for the biological control of cotton pests.

      • Role-based Morality, Ethical Pluralism, and Morally Capable Robots

        Dominant approaches to designing morally capable robots have been mainly based on rule-based ethical frameworks such as deontology and consequentialism. These approaches have encountered both philosophical and computational limitations. They often struggle to accommodate remarkably diverse, unstable, and complex contexts of human-robot interaction. Roboticists and philosophers have recently been exploring underrepresented ethical traditions such as virtuous, role-based, and relational ethical frameworks for designing morally capable robots. This paper employs the lens of ethical pluralism to examine the notion of role-based morality in the global context and discuss how such cross-cultural analysis of role ethics can inform the design of morally competent robots. In doing so, it first provides a concise introduction to ethical pluralism and how it has been employed as a method to interpret issues in computer and information ethics. Second, it reviews specific schools of thought in Western ethics that derive morality from role-based obligations. Third, it presents a more recent effort in Confucianism to reconceptualize Confucian ethics as a role-based ethic. This paper then compares the shared norms and irreducible differences between Western and Eastern approaches to role ethics. Finally, it discusses how such examination of pluralist views of role ethics across cultures can be conducive to the design of morally capable robots sensitive to diverse value systems in the global context.

        Limonoids from the roots of Trichilia sinensis and their cytotoxicities

        Six new compounds (1–4, 8, 10), along with six known limonoids (5–7, 9, 11, 12), were isolated from the roots of Trichilia sinensis. Their structures were elucidated on the basis of extensive spectroscopic methods including 1H NMR, 13C NMR, DEPT, HSQC, HMBC, 1H–1H COSY and ROESY experiments, as well as by comparison with the literature. All the compounds were evaluated for cytotoxicities against K562, SGC-7901 and BEL-7402 cell lines. Compounds 2, 7, 10, 11, and 12 showed weak inhibitory activity to the selected cell lines.

        Building an “Association of Trans-Korean Peninsula Nations” and China’s Role toward the RPG Approach

        Tensions ared for a time after North Korea’s fth and sixth nuclear tests as well as THAAD’s entry into South Korea. This paper extends the view that the root cause of these issues is the lack of regional public goods (RPGs) on the Korean Peninsula, and its insufficiency results from three major conflicts in the politico–economic situation of the Peninsula. In order to resolve the issues and maintain lasting peace, three specic aspects, i.e., in diplomacy, security and economy, are to be addressed in carrying out the long-term mission of supplying RPGs to the Peninsula. For effective operation and comprehensive management of the RPGs provision, an integration platform with powerful commitments is called for, i.e., “Association of Trans-Korean Peninsula Nations” (ATKPN) co-established by China, the United States, and the two Koreas. Apart from the dilemma commonly identified in the delivery of RPGs, the ATKPN also faces dilemmas specically from the Northeast Asian order. To overcome the ATKPN’s obstacles, concerted efforts are needed to promote the institutionalization of Sino–U.S. “new type of great power relations,” to transform the function of the alliance system, and to cultivate the awareness of “common external threats.”

        중국상업은행 유동성리스크의 영향요인에 관한 연구

        This paper is to analyze the Influential Factors of the Bank’s Liquidity Risk in Chinese Commercial Banks, and the empirical analysis is used panel data of 4 Chinese state-owned commercial banks and 11 share holding commercial banks over the period from 2005 to 2011. And also, the paper is to analyze if there are differences between Chinese state-owned commercial banks and share holding commercial banks. The results of the empirical analysis are as follows: Firstly, it shows there are significant differences between the Chinese state-owned commercial banks and shareholding commercial banks in 6 variables with Liquidity ratio, Loan-deposit ratio, Deposit structure, Ratio of loan, Capital ratio based on BIS and ROA, excepting Capital ratio. Secondly, we review Influential factors of the Bank’s Liquidity Risk. We run regression analysis with Liquidity ratio and Loan-deposit ratio as dependent variable and Deposit structure, Ratio of loan, Capital ratio, ROA, Ownership structure, Capital ratio based on BIS as independent variable. As the result, it shows that the Deposit structure, Ratio of loan, Capital ratio and Ownership structure affects significantly to the Liquidity ratio, excepting the Capital ratio based on BIS and ROA. On the other hand when the Loan-deposit ratio as dependent variable, Ratio of loan and Capital ratio based on BIS, Ownership structure and ROA affects significantly, excepting Deposit structure and Capital ratio. Among of them, Ratio of loan and Ownership structure affects significantly to the Liquidity ratio and Loan-deposit ratio.
